알츠하이머병의 원인 물질인 아밀로이드 베타 단백질의 올리고머화(Oligomerization) 및 섬유화(fibrilization)을 방지하는 후보물질을 탐색하였다. 기존에는 일반적으로 아밀로이드 베타 단백질의 섬유화를 방지하는 물질을 알츠하이머병 치료제의 후보물질로 선정하였다. 그러나 아밀로이드 베타 단백질의 올리고머 형태가 세포 독성을 가지는 것으로 지적되는 점, 아밀로이드 베타의 섬유화 저해가 올리고머의 안정성을 높임으로써 발생할 수 있는 점을 고려할 때 섬유화 저해 효과만으로 유효물질을 적절하게 스크리닝 할 수 없을 가능성이 있다. 따라서 본 발명에서는 아밀로이드 베타 단백질의 올리고머화 및 섬유화 모두를 방지하는지를 검증하여 알츠하이머병 치료제 후보물질 CBRV1-04369를 선정하였다. MDS assay (Multimer Detection System assay)를 통해 CBRV1-04369의 올리고머화 저해효과를 검증하였다. Aβ를 CBRV1-04369와 반응시켜 샘플을 만들고 -80℃에 보관하였다. MDS-HTS를 통해 각 샘플의 Aβ 올리고머화 수준을 측정하였다. Negative 대조군에 비해 CBRV1-04369의 올리고머화 저해능이 약 70% 정도로 매우 높게 나타났다(도 3).The oligomerization inhibitory effect of CBRV1-04369 was verified through MDS assay (Multimer Detection System assay). Maleimide ELISA는 플레이트 표면에 maleimide 잔기가 있어 펩타이드의 시스테인(cysteine) 잔기와 결합한다. 펩타이드를 플레이트에 결합시키고 동일하게 Aβ42를 처리해 결합을 유도하고 항체로 결합 여부와 정도를 측정하였다. CBRV1-04369 펩타이드는 일반 ELISA 및 maleimide ELISA에서 높은 Aβ42 결합력을 보여주었다. The absorbance was measured by treating the plate with CBRV1-04369 and Aβ through ELISA. 96웰 플레이트의 각 웰에 신경모세포종 SH-SY5Y 세포를 5000개씩 시딩하고, Aβ와 펩타이드 CBRV1-04369를 포함하는 약물을 처리하여 세포의 생존율을 흡광도로 측정하였다. 미처리 대조군은 약물 및 Aβ를 처리하지 않았고, Aβ처리 대조군은 약물없이 Aβ만 처리하였다. 실험군은 5시간 동안 15uM의 펩타이드 CBRV1-04369를 포함하는 화합물로 전처리한 후 Aβ1-42(5M)를 처리해 배양하였다. Aβ 처리군은 세포 생존률이 64%에 불과하였으나, 펩타이드 CBRV1-04369를 함께 처리한 경우 생존률이 84%로 증가하여 해당 펩타이드의 세포 독성 저해 효과가 검증되었다 (도 5).5000 neuroblastoma SH-SY5Y cells were seeded in each well of a 96-well plate, treated with a drug containing Aβ and the peptide CBRV1-04369, and cell viability was measured by absorbance.
