9개의 하이브리도마 세포주에서 생산한 항체를 정제하기 위해 Protein G 크로마토그래피 방법(Pierce?? Chromatography Cartridges Protein G, Thermo)을 사용하였다. Protein G 컬럼 카트리지과 모든 버퍼를 상온에서 평형(equilibrate)을 유지시킨 후 버퍼를 채워 항체 정제를 준비하였다. Protein G 컬럼은 바인딩 버퍼(Protein G IgG Binding Buffer, Thermo)를 1 ml/분의 유속으로 흘려주었다. 항체를 포함한 배지와 바인딩 버퍼를 1:1 비율로 희석하여 준비가 끝난 Protein G 컬럼에 넣어서 1~2 ml/분의 유속으로 흘려주었다. 그 다음 컬럼의 5~10배 양의 바인딩 버퍼를 넣어서 세척을 진행하였다. 세척이 끝난 Protein G 컬럼에 그 양의 2배의 Elution 버퍼(Pierce IgG Elution Buffer, Thermo)를 넣어서 항체를 수집하였다. 수집이 끝난 항체 1 ml당 100 ul의 중화 버퍼(High ionic strength alkaline buffer/1M Tris at pH 7.5)를 넣어서 수집된 항체를 pH를 바로 조정하였다. 수집된 항체 샘플을 SDS-PAGE로 분리한 뒤 Coomassie blue 염색(InstantBlue Coomassie protein stain, Abcam)을 이용하여 상온에서 15분 동안 반응시킨 후 정제가 잘된 것을 확인하고, 버퍼를 PMSF(phenylmethylsulfonyl fluoride, protease inhibitor)가 포함된 PBS로 교체하기 위해 투석(Slide-A-Lyzer?? Dialysis Cassettes)을 진행하였다. 정제 후 1개의 마우스 RSV F 특이 항체(8E9)가 RSV F 항원에 대한 결합력이 우수한 것을 확인하였다(도 2 및 표 2).Protein G chromatography method (Pierce Chromatography Cartridges Protein G, Thermo) was used to purify the antibodies produced by the 9 hybridoma cell lines. After separating the collected antibody samples by SDS-PAGE, they were reacted at room temperature for 15 minutes using Coomassie blue staining (InstantBlue Coomassie protein stain, Abcam), confirming that the purification was successful, and using the buffer as PMSF (phenylmethylsulfonyl fluoride, protease inhibitor). ) Dialysis (Slide-A-Lyzer?? ELISA 코팅 용액(BupH carbonate-bicarbonate buffer)과 RSV F 단백질(sino bio, #11049-V08B)을 섞어준 후 ELISA 플레이트(넝크 임뮤노 플레이트(Nunc-Immuno Plate)에 500 ng/well로 분주하였다. 4℃에서 오버나잇(overnight)으로 반응시켜 RSV F 단백질을 플레이트에 코팅하였다. 실험 전, 항원단백질을 코팅한 플레이트를 PBST로 3회 세척 후, 5% skim milk/PBS로 5시간 동안 블로킹을 진행하였다. 항체의 결합력을 확인하기 위해 본 발명의 RSV F 특이 항체(8E9)와 팔리비주맙을 RSV F 단백질이 코팅된 플레이트에 분주하여 최종 농도가 0, 0.01, 0.1, 1 ㎍/ml 되도록 하였다. 4℃에서 오버나잇(overnight)으로 반응시킨 후 PBST로 3회 세척하였다. 2차 항체(anti-mouse IgG-HRP)를 5% skim milk/PBS에 1:1000비율로 희석한 후 플레이트(100 ul/well)에 넣어준 후 상온에서 1시간 반응시킨 후 PBST로 7회 세척하였다. TBS/Tween에서 3회 세척 후 멤브레인을 화학발광 검출 시약(ECL plus western blotting detection reagent, Thermo)으로 5분간 인큐베이션 후 본 발명의 항체(8E9)와 팔리비주맙의 RSV F 단백질에 대한 결합력을 분석하였다.
