Abstract
The present invention relates to a portable whole blood preprocessing device and a preprocessing method using the same for application to a field test. Since it is possible to rapidly separate/remove blood cells and plasma proteins in whole blood with just a simple operation of adding , high-purity serum samples can be effectively obtained. Since it is possible to sufficiently remove viscous substances that hinder the flow of blood and fluid, high selectivity and high sensitivity detection of target biomarkers in blood can be performed.
Description
The present invention relates to a portable whole blood preprocessing device for application to a field test and a preprocessing method using the same.
Biological samples such as blood or saliva contain not only blood sugar but also various biomarkers indicating myocardial infarction, liver function, tumor presence, and various infections, so they are the core of health examinations that provide health-related values. In point of care testing (POCT), a test method that uses a portable diagnostic tool such as a biosensor to detect biomarkers in real time in a biological sample is attracting attention. Because it is a fluid, if it does not undergo pretreatment, reproducibility and stability and accuracy of device operation may be reduced. Therefore, there is an urgent need for a method for quickly and simply preprocessing a biological sample in real time in the field.
Blood is classified into blood cells such as red blood cells, white blood cells, and platelets, and plasma components. There are more than six types of plasma proteins such as albumin, fibrinogen, and immunoglobulin in plasma, but only a very small amount of less than 1% of the major proteins having clinical significance exist. Even genetic materials such as proteins, DNA, RNA, etc. of infectious bacteria that must be found to test for infectious diseases are generally present at ppm or ppb or less, so it is possible to specifically and accurately detect a very small amount of a specific substance to be analyzed. It is very important.
Therefore, when applied to a diagnostic biosensor for biomarker detection without isolating blood cells or plasma proteins from whole blood, the signal disrupting substances in the whole blood form an electrode fouling on the signal sensing part of the sensor, It is difficult to detect with high sensitivity because it can inhibit the sensitivity of the detection signal or non-specifically bind to chip electrodes, microparticle surfaces, antibodies, etc.
For conventional blood pretreatment, a method of obtaining plasma in a supernatant after centrifugation of blood cells is generally used. However, this can only be performed by experts because it requires expensive and huge analysis equipment and skilled techniques, and since it often uses power supplied by voltage, it is difficult to carry and consumes a large amount of samples.
In addition, in many cases, samples at the plasma level are used after pretreatment with blood cells, which means that plasma proteins other than the markers to be detected not only interfere with accurate analysis, but also increase the viscosity of the sample, making it difficult to move the sample in the microchannel. Because of this problem, serum samples from which these plasma proteins have been removed must be used. However, in order to obtain serum, there is a difficulty in that a complex and difficult process of removing plasma proteins such as albumin and fibrinogen through additional treatment is required.
In accordance with this demand, development studies using various driving forces such as gravity, centrifugal force, electromagnetic force, optical power, and sonic wave power have been conducted in order to obtain plasma from whole blood on a biochip. However, since it requires complex physical operation methods such as rotational speed and centrifugal force, it is difficult to drive and has a disadvantage in that it is difficult to carry and expensive. In addition, there are functional limitations in generally satisfying the use of a small amount of sample blood, high blood cell removal efficiency, simple operation, non-dilution, rapidity, reproducibility, low-cost disposable use, and versatility.
In the case of on-site inspection, the pretreatment process must be carried out easily and quickly even with a small amount of sample, and marketability needs to be performed at low cost must also be met. In the field of biosensors, various high-efficiency whole blood separation technologies have been studied (Korean Patent Publication No. 10-2015-0022701), but it is necessary to construct a pretreatment system that can be operated more easily and simply and at a lower cost.

3 and 5, the

Example 1. Fabrication of whole blood pretreatment device
In order to design a microstructure including a
For the fabrication of the PDMS
For the fabrication of the
The PDMS
Two
Experimental Example 1. Plasma extraction amount evaluation according to plasma separation conditions
In order to establish more effective plasma separation conditions, the effect on the amount of plasma extraction when the hardness of the
In order to confirm the plasma extraction effect according to the thickness of the
7 is data comparing the amount of plasma extraction according to the type of membrane.
As a result, as shown in FIG. 7, when Fusion 5 (GE Healthcare, thickness 0.37 mm) and VF2 (GE Healthcare, thickness 0.785 mm) were used, a lot of blood cells passed through the membrane or the membrane was too thick and the blood A phenomenon that does not come out occurred, and it was confirmed that a similar problem existed even when other conditions were changed using the membrane. However, when using the FR1 (mdi, thickness 0.6 mm) membrane, it was confirmed that a sufficient amount of plasma was extracted and the reproducibility was high.
In addition, in order to confirm the plasma extraction effect according to the number of
8 is data comparing the amount of plasma extraction according to the number of stacked membranes.
As a result, as shown in FIG. 8 , it was confirmed that blood cells escaped through the membrane when one sheet of membrane was stacked, whereas plasma was not extracted properly due to the thick membrane thickness when three sheets were stacked. When two membranes were stacked, the amount of obtainable plasma was slightly less than that of one sheet, but blood cells did not pass through the membrane, so the number of membrane stacks was established as two.
In addition, in order to confirm the effect of plasma extraction according to the hardness of PDMS, the curing agent compared to the silicone-based elastomer, which is a material used for fabricating PDMS of the
9 is data comparing the amount of plasma extraction according to the curing degree of PDMS of the first substrate and the second substrate.
As a result, as shown in FIG. 9, as a result of setting both the PDMS production ratio of the
In addition, in order to confirm the plasma extraction effect according to the waiting time from loading the whole blood to the whole blood preprocessing device until pressure is applied, the plasma extraction amount was compared and analyzed by varying the waiting time.
10 is data comparing the amount of plasma extraction according to the waiting time until pressure is applied after loading the whole blood into the whole blood pretreatment device.
As a result, as shown in FIG. 10, when pressure was applied immediately after loading whole blood into the first hole of the
Accordingly, it was confirmed that the PDMS-based whole blood preprocessing device set to the above conditions could obtain about 20 μl of plasma out of 60 μl of whole blood, thus obtaining a sufficient amount of plasma for application to a biosensor.
Experimental Example 2. Evaluation of Plasma Protein Removal Efficiency by Antibody Coating
Based on the whole blood pretreatment device prepared in Example 1 and the plasma separation conditions set in Experimental Example 1, plasma protein removal efficiency and serum extraction amount according to antibody coating were evaluated.
Specifically, a membrane to which only albumin-specific antibody was adsorbed, a membrane to which only fibrinogen-specific antibody was adsorbed, a membrane to which both albumin-specific antibody and fibrinogen-specific antibody were adsorbed, and no antibody as a control. After loading 60 μl of whole blood into a whole blood pretreatment device equipped with a non-adsorbed membrane, SDS-PAGE (Sodium Dodecyl Sulfate - Polyacrylamide Gel Electrophoresis) was performed on the pretreated samples to determine the amount of residual albumin and fibrinogen. did
Figure 11 is SDS-PAGE data comparing the amount of albumin and fibrinogen in the extracted serum when using a membrane to which the albumin-specific antibody and the fibrinogen-specific antibody are adsorbed, and comparison of the amount of the extracted serum This is the measured data.
As a result, as shown in FIG. 11, compared to the control group to which no antibody was adsorbed, the amount of albumin decreased when only the albumin-specific antibody was adsorbed, and the amount of fibrinogen when only the fibrinogen-specific antibody was adsorbed. It was confirmed that this decrease. On the other hand, when the two antibodies were adsorbed together, it was confirmed that the amount of albumin and fibrinogen both significantly decreased and the amount of extracted serum also increased.
Experimental Example 3. Evaluation of Plasma Protein Removal Efficiency According to Antibody/Silica Magnetic Particle Complex Coating
In Experimental Example 2, it was confirmed that albumin and fibrinogen could be removed using the antibody-coated membrane, but it was not completely removed, so albumin and fibrinogen removal efficiency was improved. To this end, by using silica binding protein - protein G (SBP-ProG) that directly binds to silica magnetic beads (SiMBs) with a high surface area per volume to the silica surface without physical and chemical treatment, SiMBs The antibody was stably immobilized on the surface.
Specifically, in the preparation of the silica magnetic microparticle complex, first, a 1.5ml tube for replacing the buffer of the silica magnetic particles (AccuBeadTM Silica Coated Magnetic Beads, 0.5g/25ml, D.W, BIONEER, CAT no. TS-1010-1) 50 μl of SiMBs was added, mixed with 125 μl of PBS (Phosphate-Buffered Saline, pH 7.4), vortexed, and the supernatant (supernatant ) was removed. Remove the tube from the magnetic rack, add 125 μl of PBS again, vortex to mix, and then sonicate in a sonicator for 30 seconds to release the agglomerated SiMBs individually. At the same time, 1 mg/ml of silica-bonded fusion protein (SBP-ProG) was prepared, and 25 μl was added to SiMBs immediately after sonication, inserted into a rotator (F6, 60 rpm), and reacted at room temperature for 30 minutes to obtain silica magnetic microparticles. SBP-ProG was attached to the surface. Then, the SiMBs/SBP-ProG mixture was placed on a magnetic rack, the supernatant was removed, mixed with 125 μl of fresh PBS, vortexed, and then ultrasonicated in a sonicator for 30 seconds to release the clumped mixture individually. Immediately after sonication, 25 μl of 100 μg/ml albumin-specific antibody or fibrinogen-specific antibody was added to the mixture and reacted for 30 minutes at room temperature by inserting it into a rotator (F6, 60 rpm), so that SBP-ProG and each Protein-specific antibodies were bound. Thereafter, the SiMBs/SBP-ProG/Antibody mixture was placed on a magnetic rack, the supernatant was removed, mixed with 125 μl of fresh PBS, vortexed, and then ultrasonicated in a sonicator for 30 seconds to release the clumped mixture individually. Immediately after sonication, add 25 μl of 1% BSA (Bovine Serum Albumin) at 1 mg/ml to the mixture and put it on a rotator (F6, 60 rpm) to react at room temperature for 10 minutes to cover the empty silica surface to reduce non-specific binding. made it Finally, after removing the supernatant by putting the SiMBs/SBP-ProG/Antibody/BSA mixture on a magnetic rack, it was mixed with 125 μl of fresh PBS.
60 μl of the completed silica magnetic beads complex (SiMBs complex) was dropped on the membrane, and then placed in a refrigerator at 4 ° C. overnight for 12 hours to adsorb the complex to the membrane and at the same time dry the membrane. Thereafter, multiple membranes were stacked on the PDMS
12 is SDS-PAGE data comparing the amounts of albumin and fibrinogen in serum extracted when using a membrane adsorbed with a silica magnetic particle complex to which an albumin-specific antibody and a fibrinogen-specific antibody are bound, and Data obtained by comparing and measuring the amount of extracted serum.
As a result, as shown in FIG. 12, compared to the control group in which only SiMBs were adsorbed without antibody, when only SiMBs complex bound to albumin-specific antibody was adsorbed, the amount of albumin decreased, and only SiMBs complex bound to fibrinogen-specific antibody When adsorbed, it was confirmed that the amount of fibrinogen decreased. Finally, when both SiMBs complexes were adsorbed, the amounts of both albumin and fibrinogen were significantly reduced, and the amount of extracted serum was also effectively increased. As a result, compared to FIG. 11 in which only antibodies were adsorbed without SiMBs complexes It was confirmed that albumin and fibrinogen could be removed much more efficiently.
Summarizing the above results, the whole blood preprocessor manufactured in Example 1 and the PDMS-based whole blood preprocessor including a membrane coated with an antibody/silica magnetic particle complex based on the plasma separation conditions set in Experimental Example 1 produced 60 μl This means that it is possible to obtain serum from which albumin and fibrinogen are effectively removed from whole blood.
