Abstract
본 연구에서 구현한 그래핀 기반의 바이오 소자는 적은 노이즈로 피질전도를 측정하고 효율적으로 전기 자극을 가하여 발작 신호를 성공적으로 완화하였다. 이러한 성능을 구현하기 위해 매우 얇은 초박막 형태의 소자를 구현하였으며, 기존 금 전극보다 성능이 우수한 D4L-Gr을 이용하였다. 이 연구에서 시행한 유연하고 얇은 그래핀 전극 어레이를 이용한 간질을 위한 전기요법 결과는 간질뿐 아니라 파킨슨 병, 조현병, 이명과 우울증 등 다양한 뇌 관련 질환 치료의 가능성을 열어주었다.
Description
본 발명은 미세 가공된(microfabricated) 그래핀 미소전극(microelectrode)을 전기 신호를 기록하고 대뇌 피질 영역으로 전달하기 위한 전기 요법용 그래핀 바이오 소자에 관한 것이다.
흔히 간질 발작은 반복적인 경련이 특징인 신경 질환의 한 유형으로, 유병률은 전 세계적으로 약 0.7 % 이다. 이는 뉴런에 과다 흥분성 활동을 포함하여 인지 기능이 제대로 발휘하지 않는다. 결과적으로, 에피소드는 갑자기 급증하는 경련으로 인한 2 차 신체 상해를 유발할 수 있으며, 이는 생명을 위협할 수 있다. 시상피질 시스템은 일반 발작의 주기적인 뇌 활동에 대한 해부학적 네트워크를 제공한다고 제안되었다. 동물 모델을 이용한 발작의 시험관 및 생체 내 기록 기법(recording technique)은 발작 활동의 발생과 동기화에 있어서 시상(thalamus)과 신피질(neocortex) 사이의 상호 작용을 보여준다. 이러한 연구는 시상에서 시상 그물핵(thalamic reticular nucleus, TRN)과 중계 뉴런(relay neuron) 사이의 상호 연결(reciprocal connection)이 일반화된 발작을 개시한다는 것을 밝혔다. 이러한 시상내 진동(intrathalamic oscillation)은 시상피질 네트워크의 밀접한 상호성을 통해 주로 피질(cortex)로 전파하고 동기화 한다.
간질의 1차 치료법은 항경련제(anticonvulsant) 투약이다. 간질 완화를 위하여 GABA 억제 전달을 증가시키는 화학물질(예를 들어, 바르비투레이트 및 벤조디아제핀)이 사용되었다. 반면에 흥분성 발작을 감소시키는 치료제(예를 들어 페니토인 및 카르바마제핀)가 대체재(alternative)이다. 사실, 발작은 일부 약물로 부분적으로 조절할 수 있다. 그러나 약물에는 전뇌에 걸친 전역 효과(global effect)로 인해 많은 부작용을 가진다. 발작이 시작되는 뇌 영역의 절제술은 일부 환자에 있어서 또 다른 실행 가능한 선택이다. 예측한 바와 같이, 신경 조직의 절제는 돌이킬 수 없으며, 따라서 종종 심각한 합병증과 연관된다. 환자의 발작이 약물 및 절제술에도 반응하지 않는 경우, 일부 환자에 대해서는 미주 신경 자극(vagus nerve stimulation, VNS), 뇌 심부 자극(deep brain stimulation, DBS) 및 반응 자극 (responsive stimulation, RNS)과 같은 신경 자극(neurostimulation)이 적용되어 효과적으로 시행된다. 전기 자극은 수술 후보자로 간주되는 환자에서 가역성(reversibility)과 조정 가능성(adjustability)의 장점을 가지고 있다. 목에 있는 미주 신경(vagus nerve)을 따라 설치된 전기 펄스 발생기 VNS는 임상 실험에서 항 간질 치료제로서 효과적이 었다. 소뇌(cerebellum), 청반(locus coeruleus), 중심내측 시상핵(centromedian thalamic nucleus), 미상핵(caudate nucleus), 시상 하부(subthalamus), 해마(hippocampus) 및 신피질 발작 지점(neocortical seizure foci)과 같은 깊은 두뇌 구조에 설치된 DBS는 몇몇 치료 효과를 나타낸다. 뇌에 이식된 신경 자극기인 RNS는 경막하 피질 스트립 리드(strip lead)를 통해 간질 활동을 감지하고 발작이 일어나기 전에 전기 자극기를 통해 치료한다. 이러한 장치는 난치성 발작 환자에게 보완적인 치료 옵션을 제공하였으나, 아직 출혈이나 감염과 같은 확인되지 않은 부작용과 합병증을 가지고 있다.
그래핀(Graphene)은 우수한 전기적, 기계적, 열적 특성으로 인하여 최근에 가장 많이 연구된 2차원 재료 중 하나로 부각되고 있다. 이와 같이 그래핀 기반 전자 기기는 전자 노이즈(electronic noise)가 낮아 신경 신호 및 분자의 획기적인 검출 감도를 갖는 센서 기술로 이어진다. 또한 장기적인 생체 적합성과 취약한 뇌 조직에 대한 기계적 유연성은 그래핀의 잘 알려진 특성이다. 이와 같은 그래핀의 유망한 특성은 생의학 응용 분야에서 그 사용을 증가시켰다.
본 발명자는 간질 치료를 위해 그래핀 전극을 사용할 것을 제안하고, 미세가공된(microfabricated) 그래핀 미소전극(microelectrode)을 전기 신호를 기록하고 대뇌 피질 영역으로 전달하기 위하여 사용하였다. 그래핀 전극으로 검출된 간질형 방전(Epileptiform discharge)은 그래핀 다중 채널 어레이의 서브세트에 내장된 전기 자극을 적용함으로써 제거되었다. 본 그래핀 기술은 많은 뇌질환에 대하여 치료적 개입을 위한 임상 적용을 가능하게 한다.

상기 과제를 해결하기 위하여, 본 발명은 플렉서블 기판; 상기 플렉서블 기판 상에 그래핀으로 형성된 전극; 상기 그래핀 전극 상이 절연층을 포함하며,상기 그래핀 전극은 그라운드(ground) 전극, 기준(reference) 전극, 기록(recording) 전극 및 자극(stimulation) 전극을 포함하는 것을 특징으로 하는 전기 요법용 그래핀 바이오 소자를 제공한다.
또한 본 발명은 상기 그래핀 바이오 소자; 사인파 고주파 자극 신호를 발생하는 신경 자극기; 상기 그래핀 바이오 소자로부터 뇌 신호를 입력 받아 증폭하는 증폭기; 상기 증폭된 뇌 신호를 사용자 단말기에 전송하는 인터페이스를 포함하는 신경 자극 장치를 제공한다.
또한 본 발명은 인간을 제외한 포유류의 신경세포 또는 신경세포를 포함하는 생체조직내에 상기 그래핀 바이오 소자를 이용하여 상기 신경세포 또는 신경세포를 포함하는 생체조직으로부터의 전기신호를 수신하거나, 또는 상기 신경세포 또는 신경세포를 포함하는 생체조직에 전기 자극을 제공하는 방법을 제공한다.

플렉서블 기판(substrate)은 그래핀 전극을 지지하는 지지기판으로 사용된다. 플렉서블 기판은 PET(Polyethylene terephthalate), PC(polycarbonate), PES(polyethersulfone), PI(polyimide), PMMA(PolymethlymethAcrylate), COP (Cyclo-Olefin Polymers) 등의 필름으로, 플렉서블한 특성을 갖는 물질로 이루어질 수 있다. 이 같이 플렉서블 기판은 물질 특성에 따라 플렉서블한 특성을 갖는다.
그래핀은 투명의 전극 재료로 높은 전하 이동도(~ 200,000cm2/V·s)와 열전도도 (~ 5000W/mK) 및 뛰어난 내화학성을 갖는다. 또한, 그래핀은 밴드갭이 없어 전파장 영역대의 빛을 고르게 흡수하지만, 두께가 탄소원자 한층에 불과하여 투명도가 97.7%에 달하고 좋은 전기 전도도와 뛰어난 기계적 강도 (Young's modulus ~1.0TPa) 및 높은 유연성 (ε~ 25%)을 갖기 때문에 투명 전극 소재로 사용될 수 있다.
그래핀은 얇은 필름 형태로 제조할 수 있어 플렉서블한 특성 및 신축성을 가지며, 변형되더라도 전기적 특성에 영향을 받지 않기 때문에 플렉서블한 장치에 적용 가능한 감지 전극으로 이용될 수 있다.
절연층은 절연시키는 물질로 이루어질 수 있으며, 바람직하게는 OCR(optical clean resin), OCA(optical clean adhesive) 또는 SU-8로 이루어질 수 있다.
도 4a는 본 발명의 실시 예에 따른 신경 자극 장치의 모식도로서, 뇌의 피질에 신경 자극을 가하는 모습이다. SCS나 DBS 장치와 같은 신경 자극 기반의 생체 이식형 의료 장치는 크게 나누어 신경의 전기적 자극을 위한 펄스 신호를 발생하는 신경 자극기(neurostimulator), 치료할 신경 부위에 삽입되며, 신경 자극 신호를 해당 신경 부위에 가하는 적어도 하나의 전극을 포함한다. 도 4a에서는 뇌의 두개(頭蓋)를 관통하는 DBS의 예를 들었지만, 이러한 신경 자극용 전극은 척수, 말초 신경 또는 뇌의 어느 특정 신경 부위에 삽입 또는 이식할 수도 있다. SCS 장치의 경우 환자의 큰가슴근 부위, 복부, 엉덩이를 관통하여 생체 이식형 전극을 삽입할 수 있다. 그리고 신경 자극 기반의 생체 이식형 장치에서 이러한 하나 이상의 전극은 도 4a에 나타낸 바와 같이 긴 전극도선(lead)에 연결되는 것이 보통이며, 이 전극도선은 전극과 신경 자극기 사이의 전기적 연결을 담당한다. 종종 이러한 장치의 전극은 환자의 피하 조직층 속에 삽입되는데, 실제 전극이 닿는 신경의 위치는 몸 안의 더 깊숙한 지점인 경우가 많다. 따라서 신경자극기와 전극도선 사이를 이어주는 연결부가 있고, 신경자극기와 연결부는 체외에 위치하는 형태가 많다.
신경 자극 장치는 증폭기를 더 포함할 수 있다. 증폭기는 상기 그래핀 바이오 소자의 기록 전극에서 검출한 뇌의 전기 신호를 증폭할 수 있다. 또한 본 발명의 신경 자극 장치는 상기 증폭기에 의하여 증폭된 뇌 신호를 사용자 단말기에 전송하는 인터페이스를 더 포함할 수 있다.
상기 단말기는 검출된 뇌 전기 신호를 시각적으로 제공할 수 있으며, 이를 위한 디스플레이장치, 스피커 등을 포함할 수 있다. 상기 단말기로는, 휴대폰, 스마트폰(smart phone), 노트북 컴퓨터(laptop computer), 데스크탑 컴퓨터(desktop computer), PDA(personal digital assistants), PMP(portable multimedia player), 내비게이션, 디지털 TV 등이 활용될 수 있다.
이하, 본 발명을 실시예와 함께 상세히 설명한다.
실험방법
1. 그래핀 전극 제조 및 특성화
25㎛ 두께의 구리 호일에 화학증착법(CVD)을 이용하여 단층 그래핀을 성장시켰다. Cu의 롤 형태의 호일(두께 : 25 μm 및 크기 : 210 x 297 mm2, Alfa Aesar Co.)이 석영 튜브 내에 로딩되었고, 이후 상압 하에서 1,000℃로 가열되었다. 탄소 소스를 포함하는 가스 혼합물(CH4 : H2 : He = 50 : 15 : 1000 sccm)을 공급하여 그래핀을 상기 Cu 호일 상에 성장시킨 후, 퍼니스(furnace)를 이동시켜주면서 Ar 또는 He를 흘려 주어 단시간에 ~10℃/s의 속도로 실온으로 냉각하여, 상기 Cu 호일 상에 성장된 그래핀 층을 수득하였다.
다층 그래핀 전극을 만들기 위하여, PMMA가 그래핀 지지층으로 사용되었다. Cu 필름 상에서 그래핀 합성 후에 지지층인 PMMA를 스핀 코팅한 후, (B) 상기 구리 촉매를 용해시키기 위하여 약 0.1 M의 (NH4)2S2O8 용액 상에 상기 필름을 띄웠다(Floating). 상기 구리를 제거한 후에, 다른 그래핀 성장된 Cu 호일을 사용하여 상기 PMMA/G 필름을 들어올렸다. 상기 에칭 및 전사 방법을 반복하여 다층막을 형성 하였다. 암모늄 설페이트 수용액을 이용하여 Cu 호일을 에칭한 후 얻은 PMMA 코팅된 그래핀은 Cu 호일 위에 있는 다른 그래핀 위에 옮겨졌다. 그 다음, 상기 그래핀을 SU-8 에폭시 기판으로 옮겼다. 전달된 그래핀은 포토리소그래피(photolithography) 및 산소 플라즈마 에칭(oxygen plasma etching)을 사용하여 패턴화 되었다. 질산(Nitric acid)은 그래핀의 화학 도핑에 사용되다.
2. 시상피질 절편 제조
동물 실험에 관한 모든 사항은 인천대학교 실험동물위원회 (Institutional Animal Care and Use Committee at Incheon National University)의 승인을 얻은 후 시행하였다. 뇌절편을 위하여 C57BL/6 마우스(생후 3~6주)는 2% 이소플루란으로 깊이 마취시켰다. 뇌를 신속히 제거하고 냉각(4℃), 산소화(5% CO2 및 95% O2)된 절편 미디움(in mM, 212 sucrose, 5 KCl, 1.23 NaH2PO4, 26 NaHCO3, 11 glucose, 1.5 MgCl2, 및 2.5 CaCl2) 안에 넣었다. 감각피질(somatosensory cortex)과 시상(thalamus)을 포함하는 시상피질 절편(400㎛)을 프로토콜(Agmon, A. et al. Neuroscience 41, 365-379 (1991))에 따라 절단하였다. 이어서 뇌절편을 124 NaCl, 4 KCl, 1.23 NaH2PO4, 26 NaHCO3, 10 글루코오스, 1.5 MgCl2 및 2 CaCl2(in mM)로 구성된 산소 처리 생리 식염수(physiological saline)를 함유하는 홀딩 챔버로 옮겼다. 약 1시간의 리커버리(recovery) 후, 각각의 절편을 레코팅(recording) 챔버로 옮겼다. 산소 처리 생리 식염수를 1.5 ml/min의 속도로 연속적으로 과주입(superfuse) 하였다. 시상피질 절편에 대한 비쿠쿨린(bicuculline, 1mM)의 국소 주사는 2가지 주요 유형의 자발적 활동을 유도했다; 3-5Hz의 발작과 유사한 단조로운 파열(monotonic burst) 이는 뇌파 측정(EEG recording)의 빈도와 기간에 대한 인간에서 일반화된 발작과 유사함, 및 발작(interictal)과 유사한 활동.
3. In vivo animal surgery 생체내(in vivo) 동물 실험
정위(stereotaxic) 장치에 머리를 고정시킨 할로탄-마취 마우스(1200 mg/kg, i.p.)를 이용하여 생체내(in vivo) 실험을 수행 하였다. 5주된 수컷 마우스를 실험에 사용했다. 개두술(craniotomy)로 피질의 5 x 5mm2 영역을 노출시켰다. 대뇌척수액 (cerebral spinal fluid , CSF)은 뇌부종의 가능성을 줄이기 위해 배액(drain)하였다. 이 과정에서 척수 꼭대기의 축 위쪽에 대수조(cisterna magna)가 드러나도록 후두골에서 근육을 비절개 박리(blunt dissected) 하였다. 경질(dura)은 날카로운 나이프로 찢었고, 면 심지(cotton wick)가 CSF를 배액하는데 사용되었다. 아이 클램프(eye clamp) 및 정위 장치에 고정된 머리를 사용하여, 깨끗한 수술 조건하에서 두피 절개가 이루어 졌으며, calvaria로부터 bone window를 절개하는 개두술(craniotomy)을 수행하였다. 이것은 뇌의 표적 영역에서 신경 생리학적 기록을 가능하게 하였다. 경막(dura mater)은 동물 기록(animal recording)동안에 절제되었으며, 이것은 동물 기록 연구에서 경막경유(transdural)하여 도입된 그래핀 전극에 의하여 손상되지 않았다.
4. 생체외 및 생체내 신경 레코딩
신경 레코팅 설정은 도 4a에 도시된 것과 같이 구축되었다. 그래핀 기반 전자장치에 의하여 검출된 뇌신호는 다채널 신경 증폭기 및 수집(acquisition) 시스템(Intan Technologies, LCC, Los Angeles, CA, USA)을 사용하여 처리되었으며, 자극은 자극기(NI-9263, National Instruments, Austin, TX, USA)에 의하여 제공되었다. 신경 기록 데이터는 Matlab을 이용하여 분석하였다.
결과
전기 요법(electrotherapy)을 위한 그래핀 전극의 제조 및 실현 가능성은 도 1에 나타내었다. 전극 어레이는 레코딩 및 전기 요법를 위해 각각 10 및 20 개의 채널을 포함한다. 그래핀은 기존 화학기상증착법(CVD)을 사용하여 얇은 구리 포일 위에 성장시킨 다음 폴리메틸메타크릴레이트(PMMA) 지지층을 사용하여 SU-8 에폭시 필름(~ 5 μm 두께)으로 옮겼다. 옮겨진 그래핀은 포토리소그래피와 산소 플라즈마 에칭을 사용하여 패턴화되었으며, 전기 전도성을 향상시키기 위해 질산 증기에 노출되어 화학적으로 도핑되었다. 모든 비전도 영역과 금선(gold line)은 얇은 SU-8 에폭시 층(~ 0.5μm)으로 캡슐화되었다(도 1a). 제조가 완료된 후 상기 어레이는 마우스 뇌의 피질에 직접 장착되었다. 여기에서 사용된 그래핀의 주요 특징은 피질의 심한 변형을 견디기 위하여 뇌피질에 대한 바람직한 표면 지형(topography)을 가진다는 것이다. 이 특성은 낮은 휨 강성(flexural rigidity)으로 인한 것이다(도 1b 및 c). 이 초박막 그래핀 전극 어레이가 피질 표면 전체에 걸친 간질 방전을 감지하면, 다중 채널 어레이의 서브세트에 내장된 전기펄스(electrical pulse)는 파열 방전(bursting discharge)을 막기 위하여 간질 부위로 전달될 것이다.
본 발명자는 4가지 유형(단층, 2층 및 4층 그래핀 및 금)의 전극에 대해 전기화학적 성질을 테스트 하였다. 적층된 층의 수가 증가함에 따라, 그래핀 전극의 임피던스는 금 전극과 같이 감소하였다(도 2a 및 도 5). 도핑된 4층 그래핀 전극의 임피던스는 낮은 지역장 전위범위(1-100Hz) 내에서 더 낮은 값을 나타내었으며, 대다수의 간질 방전이 나타났다. 저주파수 범위에서 낮은 임피던스는 더 효율적인 뉴런 스파이킹(neuronal spiking)을 포함하여, 전기 신호의 흐름을 도울 수 있으며, 따라서 전자 노이즈를 감소시킨다. 다음으로, 제작된 그래핀 전극이 전하 이동에서 우수한 성능을 보이는지 여부를 조사하기 위해 각각의 4개의 전극에 순환전압전류법(cyclic voltammetry, CV)을 측정하였다. 측정 결과 4층 그래핀 전극이 326.7 F/g의 비전정용량(specific capacitance)을 가지며 금 전극 (77.5 F/g)보다 훨씬 높다는 것을 발견하였다(그림 2b 및 표 1 & 2).

여기서 I는 전류 밀도(A/cm2), ΔV는 전압 스캔 범위, m은 전극 질량(g), s는 스캔 속도(mV/s)이다.
이것은 4 층 그래핀 전극이 전기 자극이 가해질 때 전하 이동의 양을 증가시킬 가능성이 있음을 나타낸다. 이러한 전하 전달 능력의 향상은 전극의 치수를 감소시켜 전력 소비 감소에 영향을 미치므로 전기 자극에 의한 뇌에 대한 물리적 손상의 가능성이 낮다. 이러한 전하 전달 능력은 전극의 크기(dimension)를 감소시켜 전력 소비 감소에 영향을 미치며, 따라서 전기 자극에 의한 뇌의 물리적 손상 가능성을 낮춘다. 인공 뇌척수액 (artificial cerebrospinal fluid, ACSF)에서 도핑된 4층 그래핀 전극의 장기(long-term) 안정성을 조사하기 위해 0일, 1일 및 5일에 전기화학 임피던스(electrochemical impedance spectroscopy, EIS)를 측정하였다(도 2c). 발작이 감지되었을 때 전기 신호 전력의 가장 분산된 주파수 범위인 특정 주파수 범위, 즉 15에서 25 Hz에서 임피던스의 변화를 관찰하였다(도 6). 도핑된 4층 그래핀 전극은 무시할 수 있을 정도의 분해율을 나타내었다(1일 및 5일에 5.5%). 또한, 상대적 임피던스는 9 x 104 회 이상의 주기적인 전기 자극 테스트에서 약 5 %의 약간의 변화를 나타냈다(그림 2d). 이러한 결과는 제작 된 그래핀 전극이 장기 안정성을 통해 뇌에서 작동할 수 있음을 보여준다.
4층 그래핀 다중 채널 전극에 의한 간질 활동 검출 가능성을 테스트하기 위해 60 x 60 μm2 그래핀 전극을 시상막 조각에 장착했다(그림 3a).
GABAA 수용체 길항제인 1 mM 비쿠쿨린(bicuculline)을 국소 도포한 후 5분 이내에 2가지 유형의 자발적 활동, 예를 들어 발작기(ictal)과 같은 파열(bursting) 및 발작간기(interictal)와 같은 활동이 관찰되었다. 대부분의 기록 단위에서 5 ~ 20초 동안 발작기(ictal)과 같은 파열(bursting) 활동(도 3b)과 발작간기(interictal)와 같은 활동(도 3c)이 있었다. 이러한 결과는 이 장치가 대뇌 피질 앙상블(ensemble)의 빠른 스파이킹 반응을 동시에 포착할 수 있음을 보여주었다.
다음으로, 마취된 동물에서 그래핀 전극으로 생체 내 뇌 반응을 측정하였다. 그래핀 장치는 체성 감각(somatosensory) 및/또는 운동 피질 영역(그림 4a 및 b)에 배치되었다. 표피 그래핀의 유연성은 주름진 피질 표면에서 뉴런 신호의 샘플링을 가능하게 하였다. 15 mM 비쿠쿨린(bicuculline)의 국소 적용은 실험 대상의 10 마리 동물에서 시험관내 뇌 절편 실험과 유사한 2 가지 자발적 활동을 유도했다; 발작기(ictal)과 같은 파열(bursting) 및 발작간기(interictal)와 같은 활동을 한 경우, 발작과 유사한 활동이 대뇌 피질 영역의 대부분의 기록 지점(spot)에서 발견되었다(도 4c).
진폭은 아마도 피질에서 그래핀의 접촉 정도, 비쿠쿨린의 영향을 받는 부위 또는 두 가지 모두에 따라 다양할 수 있다. 특히, 발작 에피소드에서 뇌 신호의 패턴은 모든 지점에서 유사했다.
다음으로 본 발명자는 사인파 고주파 자극(sinusoidal high frequency stimulation, sHFS)에 의해 간질 활동이 제거될 수 있는지 확인하였다. 진폭(1V), 주파수(100Hz) 및 지속 시간(30초)의 자극 매개 변수는 뇌심부 자극술(Deep brain stimulation, DBS) 시스템을 사용하는 이전 참조로부터 적용되었다. 이는 간질 발작을 치료하기 위한 장치를 개발하기 위해 고안된 것이다. sHFS 자극은 자극 후 2 분까지 파열(bursting) 활동을 차단하였다(도 4d). 이 후 억압된 반응은 비쿠쿨린에 의해 유발된 상태로 되돌아 갔고, 고주파 자극 그 자체가 조직이나 반응 손상을 일으키지 않았다. 실험의 하위 집합에서, sHFS가 정상 신호 전송에 미치는 영향을 알아보기 위하여, 동일한 자극 프로토콜을 발작간기(interictal)와 같은 활동에 적용해 보았다. sHFS 자극은 발작간기(interictal)와 같은 활동을 차단하지 못했다(도 4e). 오히려 그것은 간질성(epileptiform) 활동에서 sHSF의 선택성을 보여주었다. 발작기(ictal) 및 발작간기(interictal)와 같은 활동에 대한 sHFS의 효과는 세 가지 매개 변수, 즉 주파수, 스파이크 카운트 및 진폭으로 분석되었다. 상기 빈도는 발작기(ictal)와 같은 파열(P <0.05, paired t-test)에서는 1.68 ± 0.5 %에서 0.94 ± 0.27 %로 감소하였으나, 발작간기(interictal)와 같은 활동에서는 8.80 ± 3.08 %에서 11.54 ± 3.91 %로 변하였다(P <0.05, 도 4f, 상단). 발작기와 같은 파열에서의 상기 스파이크 카운트(spike count)는 66.64 ± 7.92에서 44.91 ± 6.67로 감소하였으며(P <0.001, 그림 4f, 가운데), 반면에 발작간기(interictal)와 같은 활동에는 영향을 받지 않았다(P>0.1). 발작기와 같은 파열의 평균 진폭은 0.28 ± 0.03 (upward deflection) 및 -0.35 ± 0.05 mV (downward deflection)에서 0.26 ± 0.02 및 -0.33 ± 0.05로 감소하였으며(P = 0.003, 도 4f, 하단), 반면에 발작간기(interictal)와 같은 활동의 진폭은 변동이 없었다(P>0.1, 도 4f, 하단).
고찰
본 명세서에서 개시된 기술은 초박형 및 유연한 그래핀을 뇌파 측정법(electrocorticography)에 접목하였으며, 이는 종합적인 피질 앙상블 활동(진단 목적)과 뇌 질환 치료(치료 목적)를 시도할 수 있도록 한다. 본 발명의 다중 채널 그래핀 전극은 높은 효율 시공간 해상도로 뇌의 신호를 성공적으로 검출하였으며, 약물 유발성 발작과 같은 활동을 중단시켰다. 이렇게 동시에 기록과 자극을 통합하는 것은 진단 및 치료용 의학 장치에 대한 접근 방식의 근본적인 변화로 이어질 수 있다.
종래에는 운동 장애, 발작 및 정신병적 증상과 같은 비정상적인 뉴련 활동을 제어하기 위하여 sHFS가 사용되었다. 전형적으로 AC 자극에 의해 모방된 sHFS는 임상 DBS 시스템에서 1-10 V의 세기와 100-165 Hz의 펄스를 가지며, 이는 표피 전자장치를 위한 본 발명자의 프로토콜과 유사하다(동물 모델의 경우 1 V, 100 Hz). 간질 활동에 대한 억제 역치값은 펄스 트레인 HFS(pHFS)에서 보다 sHFS에서 더 낮기 때문에, 본 발명자는 캐리어 주파수의 엔벨로프 (envelope)로서 정현파 (sinusoidal wave)를 구현했다. 실제로 사인파 자극은 체세포 신경 활동과 파열 뉴런의 축삭 전도를 효과적으로 억제하였다. 이러한 결과는 신경돌기(axonal) 전도 및 시냅스 효능에 억제 효과가있는 pHFS를 사용한 연구와는 다르다. 예를 들어, 펄스 트레인 자극에서와 같이 가파른 상승 시간은 흥분 반응(excitatory response)을 높일 수 있다. sHFS가 간질 뇌에서 세포 외 칼륨의 증가와 연관되어 있다는 것은 매우 흥미로운 점이다. 상승된 세포 외 칼륨 농도는 활동 전위 진폭을 감소시키고, 시냅스 이전(presynaptic) 전위를 낮추고, 신경돌기(axonal) 신호 전달에 영향을 미치고, 이어서 탈분극 차단(depolarization blockade)을 일으키는 것으로 알려져 있다. 반면에 sHFS는 본 발명에 영향을 받지 않는 정상 전기 신호를 남겼다. 간질파(epileptic discharge)에 선택적 효과를 나타내는 sHFS는 치료 자극을 위한 유용한 매개 변수가 될 수 있다 (그림 4).
DBS와 RNS는 뇌에 침투하여 신체적 손상과 유해한 면역 반응을 일으키는 경우가 많다. 본 명세서에서는 비침습적으로 간질을 조절하는 표피 전자장치를 소개한다. 금속 Ag는 종래의 전극에 사용되며 종종 부식성이 있으며 다양한 생물학적 환경에서 황화(sulfidation) 및 산화를 통해 쉽게 분해된다. 독성과 불안정성 때문에 노출된 Ag는 신경 임플란트로 사용하기에 제한적이다. 한편, Ag를 그래핀으로 코팅하면 Ag 표면의 전기 화학 반응을 제거할 수 있다. 순환전압전류법(CV) 및 potentiodynamic 분극 시험에 의하면 그래핀 코팅은 인산염 완충 생리 식염수에서 Ag 부식을 크게 감소시키는 것으로 나타났다. 이전 연구와 일관되게, 본 발명의 데이터는 그래핀 전극이 오래 견딜 수 있는 안정성과 생체 적합성을 가지고 있음을 보여주었다. 중요한 것은 축색 돌기(axons)와 수상 돌기(dendrites)와 같은 뉴런의 풍부한 디지털 부분이 존재하는 피질 표면에서의 표피 그래핀은 손상될 수 있는 세포체가 존재하는 뇌의 깊숙한 곳으로 전극이 침투하는 것보다 해롭지 않다는 것이다. 마지막으로 표피 전자장치의 기계적 유연성과 순응성은 신경 신호의 신뢰성있는 검출을 보장합니다.
현재의 표피 전자장치는 인간의 간질에 더 잘 맞는 구조와 기능에 따라 더 발전되어야 한다. 일단 그래핀 기반 접이식 전자 장치가 피질 표면의 간질파(epileptic discharge)를 감지하면 다 채널 배열의 하위 집합에 포함된 전기 펄스가 간질 발작을 막기 위하여 제공된다. 주머니에 들어갈 수 있는 휴대형 무선 두뇌 자극기는 편리하게 사용할 수 있도록 전기펄스의 전력은 전환할 것이다. 본 발명은 간질에 유용한 장치이며 파킨슨 병, 조현병, 이명과 우울증 등 다양한 뇌 관련 질환 치료의 도구로 사용될 수 있다.

플렉서블 기판;
상기 플렉서블 기판 상에 그래핀으로 형성된 전극;및
상기 그래핀 전극 상에 절연층을 포함하며,
상기 그래핀 전극은 그라운드(ground) 전극, 기준(reference) 전극, 기록(recording) 전극 및 자극(stimulation) 전극을 포함하고,
상기 그래핀은 1 ~ 4층(layer)으로 이루어지고,
상기 그래핀 바이오 소자는 체성감각(somatosensory) 또는 운동 피질 영역(motor cortical area)에 배치되는 것을 특징으로 하는 전기 요법용 그래핀 바이오 소자.

- 제1항 내지 제3항, 제5항, 제7항 및 제8항 중 어느 한 항에 따른 그래핀 바이오 소자;
사인파 고주파 자극 신호를 발생하는 신경 자극기;
상기 그래핀 바이오 소자로부터 뇌 신호를 입력 받아 증폭하는 증폭기;및
상기 증폭된 뇌 신호를 사용자 단말기에 전송하는 인터페이스를 포함하는 신경 자극 장치.
- 인간을 제외한 포유류의 신경세포 또는 신경세포를 포함하는 생체조직내에 제1항 내지 제3항, 제5항, 제7항 및 제8항 중 어느 한 항에 기재된 그래핀 바이오 소자를 이용하여 상기 신경세포 또는 신경세포를 포함하는 생체조직으로부터의 전기신호를 수신하거나, 또는 상기 신경세포 또는 신경세포를 포함하는 생체조직에 전기 자극을 제공하는 방법.
- 제10항에 있어서,
상기 신경세포를 포함하는 생체조직은 포유류의 뇌 또는 척수인 것을 특징으로 하는 생체조직으로부터의 전기 신호를 수신하거나, 또는 상기 생체조직에 전기 자극을 제공하는 방법.
- 제10항에 있어서,
상기 전기 자극은 사인파 고주파 자극(sinusoidal high frequency stimulation, sHFS)인 것을 특징으로 하는 생체조직으로부터의 전기 신호를 수신하거나, 또는 상기 생체조직에 전기 자극을 제공하는 방법.
