Abstract
본 발명은 옥수수 유래 다당을 포함하는 면역증강용 조성물에 대한 것이다. 상기 면역증강용 조성물은 식품 조성물, 사료 조성물, 또는 약학적 조성물일 수 있다.
Description
본 발명은 옥수수 및 옥수수 가공 부산물 유래 아라비노자일란을 포함하는 면역 증강용 조성물에 대한 것이다.
옥수수는 그 열매로부터 전분 등을 제조하기도 하고, 최근 옥수수 수염으로부터 유효 성분을 추출하기도 하는 등(한국등록특허 10-1535835호), 널리 연구되고 이용되는 식물이다. 그런데 옥수수를 이용하여 특정 제품을 제조 시 그 부산물들이 발생되는데 그 처리가 곤란한 경우가 많다.
에컨대, 옥수수 전분 제조 시에는 옥수수 침지 폐액(corn steep liquor, CSL)과 추출시 제거되었던 배아(corn germ)부분 및 과피(pericarp, bran)가 부산물로서 발생한다. 원료의 침지가 끝난 액에는 옥수수 중 가용성 물질의 대부분이 용출되며, 용출된 당류에 의해 발효된 다량의 젖산이 함유되어 있으며 침지가 끝난 액을 농축시키면 CSL이 되고, 이 액은 다량의 탄수화물, amino acid, peptides, vitamin, essential minerals 및 천연식물의 항산화제로 알려진 phytic acid를 함유하고 있음이 보고되어 있어 발효공업에서 영양원이 우수한 유기질소원으로 사용되고 있으며, 그의 일부가 사료용으로 이용되고 있다. 하지만 CSL은 점도가 매우 높은 액상으로 존재하기 때문에 취급상 불편하고, 그 일부가 발효용 배지 및 사료로 이용되고 있지만 대부분의 옥수수 전분 공장에서는 다량 발생하는 양의 CSL을 폐수로 방류할시 높은 COD부하를 야기시키기 때문에 경비를 들여 해양에 투기하고 있는 실정이다. 하지만 이 또한 2016년부터 법률적 제재에 의해 전면 금지될 예정으로 이의 처리는 심각한 문제에 있다.
한편, 대식세포(macrophage)는 세균이나 이물질을 탐식, 제거하는 과정에서 여러 가지를 분비하여 면역현상을 조절하며, 항원에 대한 면역작용의 중추적인 역할을 하는 세포로써, 항원제시와 림프구의 비특이적인 면역작용에 관계하며,종양세포에 대해서는 직접적인 상해활성을 나타낸다. 또한, TLR(toll like receptor)에 반응하는 물질(LPS 혹은 천연물)은 macrophage를 활성화하여 T세포와 B세포의 증식, 식균 작용을 위한 대식세포의 활성화, 미생물 감염에 대한 방어 등의 2차 면역반응을 조절할 수 있는 IL-1,IL-6, lL-12 및 TNF-α와 같은 사이토카인을 생산한다고 알려져 있다. IL 6 및 TNF-α는 대식세포에 의해 유도되는 대표적인 사이토카인으로서 세균감염에 따른 염증반응에 있어서 중추적인 역할을 하며 염증병소에서 그 양이 증가되는 것으로 알려져 있다. IL 6는 IL 1과 협동적으로 작용하여 T 세포와 B 세포의 분화에 관여하고 항암효과가 있다고 보고되었으며, TNF-α는 특정 암세포에 대한 세포독성과 항 바이러스성 작용이 있고, 급성 및 만성 염증질환에서 일어나는 여러 가지 생체반응에도 중요한 역할을 한다고 알려져 있다. 한편, IL 12는 NK cell의 활성화 및 Th1 type의 면역 반응을 유도하는 사이토카인으로써 암세포와 같은 세포성 외래물질에 대한 반응성을 높이는 작용을 한다고 알려져 있다.
본 발명자들은 옥수수 전분 제조 시 발생하는 부산물들의 이용에 대하여 연구하던 중 이로부터 분리한 특정 다당이 면역기능 증강 활성이 있는 것을 확인하고 본 발명을 완성하였다.
본 발명의 목적은 면역 증강용 조성물을 제공하는 것이다.
상기 목적을 달성하기 위하여 본 발명은 옥수수 유래 다당을 포함하는 면역증강용 조성물을 제공한다.
본 발명의 면역증강용 조성물은 사이토카인 분비 증강능, 자연살해세포의 활성 증강능, 종양의 전이 억제능 등을 갖는다.
도 1은 옥수수로부터 다당들을 분리 정제하는 공정을 나타낸다.
도 2는 Sephadex G-100의 칼럼 상 CBP1S 획분의 겔 여과를 나타낸다. CBP1S를 Sephadex G-100 칼럼 (2.5×90 cm)에 처리하고 1 mL/분 속으로 50 mM ammonium formate 버퍼 (pH 5.5)로 용출시킨 것이다.

도 7은 엑스 비보(ex vivo)에서 NK 세포들의 세포살해 활성에 대하여 CBP-0이 미치는 영향을 나타낸다. NK 세포들은 37℃ CO2 인큐베이터에서 24시간 또는 6 시간 동안 CBP-0의 존재 또는 부존재 하 YAC-Ⅰ타겟과 함께 배양되었다. 다른 윗첨자들(a-d)을 갖는 평균들은 Duncan's multiple range test에 의할 때, p<0.05의 중요한 차이를 나타낸다.
도 8은 Colon26-M3.1 cacinoma 세포들의 정맥 내(i.v.) 접종에 의하여 만들어진 폐 전이에 CBP1S의 경구 투여가 미치는 영향을 보여준다. 그룹 당 4 마리의 BALB/c 마우스들이 CBP1S의 지시된 투여량으로 경구투여되었으며, 그 다음으로 3×104 Colon26-M3.1 cacinoma 세포들로 정맥내 접종되었다. 마우스들은 종양 평가를 위하여 종양접종 14일 후 희생되었다. 다른 윗첨자들(a-b)을 갖는 평균들은 Duncan's multiple range test에 의할 때, p<0.05의 중요한 차이를 나타낸다.

이하, 본 발명을 자세히 설명한다.
옥수수 유래 다당
본 발명은 옥수수 유래 다당을 포함하는 면역증강용 조성물에 대한 것이다.
상기 옥수수는 옥수수 열매 또는 그 부산물일 수 있다. 상기 부산물은 옥수수의 가공 과정에서 발생하는 옥수수 유래 부산물을 가리킨다. 상기 부산물은 옥수수로부터 전분 제조 공정에서 발생하는 옥수수 유래 부산물일 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 다당은 옥수수 유래 다당이다. 상기 다당은 아라비노자일란인 것이 바람직하며, 더욱 바람직하게는 아라비노스 및 자일로스의 함량이 8 중량% 이상이고, 더더욱 바람직하게는 아라비노스 및 자일로스의 함량이 20 중량% 이상이며, 가장 바람직하게는 아라비노스 및 자일로스의 함량이 50 중량% 이상이다.
본 발명의 다당은 하기 실시에들에 기재된 CBP-0, CBP1S 또는 CBP1S-I의 제조 방법에 따라 분리, 정제하여 제조할 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 다당은 아라비노자일란으로, 이는 주쇄에 자일란을 포함하고, 측쇄에 아라비노스를 포함하는 다당류이다. 바람직하게는 본 발명의 다당은 자일로스가 1 -> 4 결합으로 연결된 자일란을 주쇄로 포함하며, 또한 상기 자일로스의 C3위치에서 연결된 하나의 측쇄 또는 C2 및 C3 위치에서 연결된 두 개의 측쇄를 포함하고, 상기 측쇄는 아라비노스를 포함한다. 이 때, 상기 아라비노스는 말단에 4-메틸-글루콘산, 아세틱 페룰릭산 또는 육탄당이 연결된 것이 바람직하다.
본 발명의 다당은 인터루킨-6, 인터루킨-12 또는 TNF-알파(TNF-α)의 분비를 증가시키며, 또한 자연살해 세포의 활성을 증진시키고, 종양의 전이를 억제하는 특성을 갖는다.
면역증강용 조성물
본 발명은 옥수수 유래 다당을 포함하는 면역증강용 조성물에 대한 것이다. 상기 면역증강용 조성물은 식품 조성물, 약학적 조성물 또는 사료 조성물일 수 있다.
식품 조성물
본 발명의 상기 식품이란 건강보조식품, 건강기능식품, 기능성 식품, 운동 보조제 등이나 이에 제한되는 것은 아니며, 천연식품, 가공식품, 일반적인 식자재 등에 본 발명의 옥수수 유래 다당을 첨가한 것도 포함된다.
본 발명의 옥수수 유래 다당을 포함하는 식품 조성물은, 상기 조성물을 그대로 첨가하거나 다른 식품 또는 식품 조성물과 함께 사용될 수 있으며, 통상적인 방법에 따라 적절하게 사용될 수 있다. 유효 성분의 혼합양은 그의 사용 목적(예방 또는 건강 유지 목적)에 따라 적합하게 결정될 수 있다. 일반적으로, 본 발명의 옥수수 유래 다당을 식품 또는 음료의 제조시에 원료에 대하여 0.01 내지 70.00 중량%, 바람직하게는 0.01 내지 30.00 중량%의 양으로 첨가할 수 있으며, 더욱 바람직하게는 0.01 내지 10.00 중량%의 양으로 첨가할 수 있다.
상기 식품의 종류에는 특별한 제한은 없다. 상기 옥수수 유래 다당을 유효성분으로 포함하는 식품 조성물은 정제, 경질 또는 연질 캅셀제, 액제, 현탁제 등과 같은 경구투여용 제제의 형태로 이용될 수 있으며, 이들 제제는 허용 가능한 통상의 담체, 예를 들어 경구투여용 제제의 경우에는 부형제, 결합제, 붕해제, 활택제, 가용화제, 현탁화제, 보존제 또는 증량제 등을 사용하여 조제할 수 있다.
상기 옥수수 유래 다당을 첨가할 수 있는 식품의 예로는 육류, 소세지, 빵, 쵸코렛, 캔디류, 스넥류, 과자류, 피자, 라면, 기타 면류, 껌류, 아이스크림류를 포함한 낙농제품, 각종 스프, 음료수, 차, 드링크제, 알콜 음료, 파우더 제제 및 비타민 복합제 등을 들 수 있으나 이들 종류의 식품으로 제한되는 것은 아니다.
사료 조성물
본 발명의 옥수수 유래 다당을 유효성분으로 포함하는 사료 조성물은 통상적인 사료와 함께 배식할 수 있으며, 본 발명의 사료 조성물을 통상적인 사료 조성물에 첨가하여 기능성 사료 조성물을 제조하여 이용할 수도 있다. 또한 본 발명의 사료 조성물은 본 발명의 옥수수 유래 다당 외 기능성 성분을 추가로 포함할 수도 있다. 상기 통상적인 사료 조성물과 본 발명의 옥수수 유래 다당이 혼합된 기능성 사료 조성물의 제조 시, 본 발명의 옥수수 유래 다당은 총 사료 조성물에 대하여 0.01 내지 30.00 중량%, 바람직하게는 0.01 내지 20.00 중량%의 양으로 첨가될 수 있다. 사료 조성물의 상기 옥수수 유래 다당의 유효용량은 상기 식품 조성물의 유효 용량에 준해서 사용할 수 있으나, 지속적인 면역 증강을 목적으로 하거나 또는 건강 조절을 목적으로 하는 장기간의 섭취의 경우에는 상기 범위 이하일 수 있으며, 유효성분은 안전성 면에서 아무런 문제가 없기 때문에 상기 범위 이상의 양으로도 사용될 수 있다.
본 발명의 사료 조성물은 가축 또는 가금을 대상으로 한다. 상기 가축 또는 가금은 소, 돼지, 닭, 말, 양, 당나귀, 노새, 멧돼지, 토끼, 메추라기, 집오리, 장닭, 투계용 닭, 비둘기, 칠면조, 개, 고양이, 원숭이, 햄스터, 생쥐, 래트, 구관조, 앵무새, 잉꼬, 카나리아 등이나 이들에 제한되는 것은 아니며 가정 내에서 사육 가능한 인간 이외의 포유 동물 또는 조류이면 본 발명의 사료 조성물의 대상이라 할 것이다.
약학적 조성물
본 발명의 옥수수 유래 다당을 포함하는 약학적 조성물은, 투여자의 면역 활성을 증강시킨다. 본 발명의 옥수수 유래 다당을 포함하는 약학적 조성물은 면역결핍, 면역저하 또는 면역계 손상으로 인한 질환의 예방 및 치료용 약학적 조성물일 수 있다. 또한 본 발명의 옥수수 유래 다당을 포함하는 약학적 조성물은 화학요법 및 방사선요법과 같은 항암요법에 의한 면역기능의 저하 또는 골수이식 후 면역저하로 인한 질환, 면역계 손상으로 인한 에이즈 및 면역기능의 저하로 인한 암질환의 예방 및 치료용 약학적 조성물일 수 있다.
본 발명의 약학적 조성물은 상기 본 발명의 옥수수 유래 다당을 0.01 내지 80 중량% 포함할 수 있으며, 바람직하게는 0.02 내지 65 중량% 포함할 수 있다. 그러나 이는 투약자의 필요에 따라 증감할 수 있으며, 식생활, 영양 상태, 병의 진행 정도, 비만의 정도 등 상황에 따라 적절히 증감할 수 있다.
본 발명의 약학적 조성물은 경구 또는 비경구로 투여가 가능하며 일반적인 의약품 제제의 형태로 사용될 수 있다. 바람직한 약제학적 제제는 정제, 경질 또는 연질 캅셀제, 액제, 현탁제 등과 같은 경구투여용 제제가 있으며 이들 약제학적 제제는 약제학적으로 허용 가능한 통상의 담체, 예를 들어 경구투여용 제제의 경우에는 부형제, 결합제, 붕해제, 활택제, 가용화제, 현탁화제, 보존제 또는 증량제 등을 사용하여 조제할 수 있다.
본 발명의 옥수수 유래 다당을 포함하는 약학적 조성물의 투여 용량은, 환자의 상태, 연령, 성별 및 합병증 등의 다양한 요인에 따라 전문가에 의해 결정될 수 있지만 일반적으로는 성인 1kg 당 0.1㎎ 내지 10g, 바람직하게는 10 mg 내지 5g의 용량으로 투여될 수 있다. 또, 단위 제형당 상기 약학적 조성물의 1일 용량 또는 이의 1/2, 1/3 또는 1/4의 용량이 함유되도록 하며, 하루 1 내지 6 회 투여될 수 있다. 그러나 건강 및 위생을 목적으로 하거나 또는 건강 조절을 목적으로 하는 장기간의 섭취의 경우에는 상기 양은 상기 범위 이하일 수 있으며, 유효성분은 안전성 면에서 아무런 문제가 없기 때문에 상기 범위 이상의 양으로도 사용될 수 있다.
본 발명의 이점 및 특징, 그리고 그것들을 달성하는 방법은 상세하게 후술되어 있는 실시예들을 참조하면 명확해질 것이다. 그러나, 본 발명은 이하에서 개시되는 실시예들에 한정되는 것이 아니라 서로 다른 다양한 형태로 구현될 것이며, 단지 본 실시예들은 본 발명의 개시가 완전하도록 하며, 본 발명이 속하는 기술분야에서 통상의 지식을 가진 자에게 발명의 범주를 완전하게 알려주기 위해 제공되는 것이며, 본 발명은 청구항의 범주에 의해 정의될 뿐이다.
<재료 및 방법>
옥수수 전분 제조 시 발생하는 부산물인 옥수수 침지 폐액 (Corn steep liquor, CSL)을 ㈜산돌식품 및 재단법인 홍천메디칼허브연구소로부터 구입하여 이용하였다. 밀 유래 아라비노자일란은 Megazyme (Wicklow , Ireland)를 구입하여 사용하였다.
<실시예 1> 옥수수 유래 면역 다당의 분리
옥수수 전분은 옥수수를 아황산 수용액에 침지하여 얻어지며 이때 부산물로 옥수수 침지 폐액(Corn steep liquor, CSL)과 추출 시 제거되었던 배아부분 및 과피가 발생 된다. CSL을 최종농도 20 Brix가 되도록 약 3배 희석하고 최종 부피의 4배 에탄올 (EtOH)을 첨가하여 하룻밤 방치한 후, 원심분리기(6000 rpm, 30 min, 4 ℃)를 이용하여 침전물을 회수하였다. 상기 침전물을 CBP-0라 명명하였다. 그리고 상기 침전물, 즉 CBP-0을 다시 증류수(DIW):에탄올을 1:1 부피비로 혼합한 용액을 이용하여 침전시킨 후, 원심분리기(6000 rpm, 30 min, 4 ℃)를 이용하여 침전물 및 상등액 회수를 3번 반복하였다. 이후 회수된 상등액을 회전감압 농축장치를 이용하여 재차 농축한 후, 동결건조하여 상기 동결건조물을 회수하였다. 회수된 동결건조물을 이하 CBP1S로 명명하여 시험에 이용하였다(도 1).
<실시예 2> 다당의 정제
상기 실시예 1에서 수득한 옥수수 유래 다당 침전물인 CBP1S의 활성 본체를 규명하기 위하여, 하기 공정에 따라 다당의 정제를 수행하였다. 먼저, 증류수(DIW):에탄올을 1:1 부피비로 혼합한 용액을 이용하여 분리한 상등액의 동결건조물인 CBP1S 획분을 소량 물에 용해하고, 이를 50 mM ammonium formate buffer (pH 5.5)로 평형화된 Sephadex G-100 column (4×120 cm)을 이용하여, gel permeation chromatography (GPC)를 수행하여 정제하였다. GPC 결과 얻어진 용출액은 5 mL씩 120개의 획분으로 분획하였으며, 각 획분은 중성당 및 단백질 함량 분석실험(도 2)을 거쳐 1kDa 내지 100kDa까지 회수하여, 분자량이 80 kDa인 다당 분획물(CBP1S-I)을 수득하였다(도 3).
<실시예 3> CBP0 및 CBP1S 의 구성당 분석
구성당 분석은 Albersheim 등(Jones TM, Albersheim P. A gas chromatographic method for the determination of aldose and uronic acid constituents of plant cell wall polysaccharides. Plant Physiol. 49: 926-936 (1972))의 방법을 일부 변형하여 수행하였다. 각 다당 시료, 즉, 분획물들을 가수분해한 후 각 구성당들을 alditol acetate로 유도체화 하여 GC (Gas Chromatography)를 이용하여 분석하였다. 구체적으로는 먼저, CBP-0 및 CBP1S 다당 분획물인 다당 시료를 2 M TFA (trifluoroacetic acid) 중에서 121℃, 1.5시간 동안 반응시켜 가수분해한 후, 상기 가수분해물을 1 mL의 1 M NH4OH (Ammonia solution)에 용해하여 10 mg의 NaBH4로 4시간 환원시켰다. 그리고 Acetic acid를 적당량 가하여 잔존 NaBH4를 제거한 후, methanol을 가하며 반복 건조함으로써 과량으로 가해진 acetic acid를 제거하여 각 구성당에 상당하는 alditol로 전환하였다. 이 후 각각의 alditol 에 1 mL의 acetic anhydride를 가하여 121 ℃에서 30분 동안 반응시켜 alditol acetate로 전환시켰으며 이를 chloroform/H2O 2상 용매계로 분리하여 추출하고, 추출물은 건조 후 소량의 acetone에 용해하여 GC 분석용 시료로 사용하였다. Alditol acetate 유도체의 GC분석 조건은 표 1과 같으며, 각 구성당의 mole%는 각 유도체의 피크(peak) 면적, 분자량 및 FID (Flame ionization detector)에 대한 molecular response factor를 각각 산출하여 계산하였다.
구성당 분석을 위한 가스-액체 크로마토그래피의 분석 조건들
그 결과는 하기와 같다. 먼저, 전분 부산물인 CSL의 조다당인 CBP-0의 화학적 조성을 분석한 결과 중성당 70.7±0.4%, 산성당 0.9±0.3% 및 단백질 28.4±0.9%이 검출되는 특성을 보였다. 또한, CBP-0를 가수분해하여 alditol acetate 유도체로 전환하여 구성당을 분석한 결과, CBP-0는 글루코스(glucose) (46.8%), 아라비노스(arabinose) (11.7%), 자일로스(xylose) (7.0%)가 높은 비율로 함유하고 있었으며, 그 외 만노스(mannose)와 갈락토스(galactose)가 미량 검출되었다(표 2).
한편, 50% 에탄올 처리에서 얻어진 상등액 획분인 CBP1S의 화학적 특성을 살펴본 결과, CBP1S의 경우, 중성당 81.0%, 산성당 9.5% 및 단백질 9.5%로 구성되어 있었으며, 식물에서 발견되는 KDO 물질은 함유하고 있지 않았다. 또한 CBP1S의 구성당을 분석한 결과 글루코스 (33.5%), 아라비노스 (22.3%), 자일로스 (14.3%)로 구성되어 있었으며, 미량의 갈락토스와 만노스가 검출되었다.
옥수수 및 옥수수 침지 폐액으로부터 분리한 CBP-0 및 CBP1S의 화학적 특성
<실시예 4> CBP-0 및 CBP1S의 분자량 분포 측정
전분 부산물에서 정제한 CBP-0 및 CBP1S의 분자량 분포 측정을 위해 superdex 200 GL 칼럼을 이용하여 HPLC (High performance liquid chromatography)를 수행하였다.
그 결과, 전분 부산물로부터 분리한 조다당 CBP-0의 경우 고분자 물질과 저분자 물질이 혼합된 형태로 구성되어 있다는 것을 확인할 수 있었다. 또한 CBP1S의 분자량 분포를 확인한 결과, CBP-0에 비해 CBP1S는 대부분 저분자 획분이 높은 비율로 함유되어 있었다(도 4). 하지만 저분자일수록 HPLC의 RI detector의 반응성이 크므로 CBP1S는 전체적으로는 고분자와 저분자 물질이 혼합되어 있다고 판단되었다.
<실시예 5> 대식세포 (Macrophage) 활성능
옥수수의 전분 부산물 유래 다당 시료의 직접적인 자극에 의한 macrophage의 cytokine 생산을 in vitro에서 측정하였다. 구체적인 측정 방법은 하기와 같다. 먼저 PBS를 이용하여 2000 μg/mL 농도로 조제한 다당 시료 용액에 RPMI 1640를 가하여 1000 μg/mL에서 1 μg/mL의 농도가 되도록 10배수로 연속 희석하고 flat bottomed 96-well microplate에 100 μL씩 분주하였다. 여기에 BALB/c (♀, 6 weeks) 마우스의 복강에 5% thioglycollate medium 1 mL를 주입하여 72시간 동안 유도된 대식세포(macrophage)를 복강으로부터 회수하여 세척 후, 세포 수 (RPMI 1640의 2.5×106 cells/mL)를 조정하여 대식세포 현탁액을 조제하고 이를 100 μL를 가한 후 37 ℃, 5% CO2 배양기에서 24시간 배양하였다. 배양 종료 후 1,500 rpm, 4 ℃에서 5분간 원심분리하여 세포배양액 150 μL를 회수하고 상등액 중에 유도된 cytokine의 함량을 측정하였다.
대식세포에 의해 생산된 사이토카인(cytokine)의 함량은 sandwich ELISA (enzyme-linked immunosorbent assay)에 의해 분석하였다. 각 사이토카인의 항체는 coating buffer에 희석하여 flat-bottomed 96-well microplate에 코팅한 후 4℃에서 12시간 방치하였다. 코팅이 완료된 microplate는 워싱 버퍼(PBS with 0.05% tween 20, PBST)를 이용하여 3차례 세척하고, assay diluent (PBS with 10% FBS or 2% skim milk) 200 μL를 가하여 1 시간 동안 방치하여 항체가 붙지 않은 well 표면을 blocking하였다. Blocking 완료 후 각 well은 washing buffer를 이용하여 재차 3회 세척하고, 각 well에 연속 희석한 표준물질(recombinant mouse cytokine)과 면역세포배양액을 각각 50 μL씩 분주하였다. 이를 실온에서 2시간 동안 방치한 다음 washing buffer로 세척하고 detection antibody (in assay diluent) 100 μL를 처리하여 실온에서 1시간 방치한 후 재차 세척하였다. 그리고 세척 후 Enzyme reagent (avidinhorseradish peroxidase conjugate) 100 μL를 처리하여 실온에서 30분간 결합시킨 후 substrate solution [tetramethylbenzidine (TMB) and hydrogen peroxide] 100 μL를 가하여 암소에서 30분간 반응시킨 후 50 μL의 stop solution [(1 M H3PO4) or (2 N H2SO4)]을 처리하여 450 nm에서 흡광도를 측정하였다.
그 결과, 활성다당 CBP-0 및 CBP1S는 IL-6, IL-12 및 TNF-α의 생산을 촉진하는 것이 확인되었다(도 5). 특히 CBP-0에서 분리된 CBP1S는 1000 μg/mL의 고농도에서 10 μg/mL의 저농도까지 IL-6, IL-12 및 TNF-α의 생산능이 높게 유지되는 것이 확인되었다(도 5). 이러한 결과들로 미루어 보아 CBP-0의 활성 다당은 CBP1S에 주로 존재하는 것으로 확인되었는바, 이하, CBP1S을 대상으로 추가 분석 실험을 행하였다.
<실시예 6> CBP-0 및 CBP1S의 항보체 활성 평가
건강한 성인의 혈액을 채취하여 실온에서 약 15분간 방치하여 응고시킨 후, 응고된 혈액을 절단하고 약 5분간 상온에서 방치시켰다. 이 혈액을 다시 4℃에서 약 20분간 방치한 다음 원심분리(2,200 rpm, 15 min, 4℃)하여 혈청을 분리한 뒤 미량 원심분리용 튜브에 1 mL씩 분주하여 -70℃에서 냉동 보관하면서 실험에 사용하였다.
항보체 활성은 Meyer법을 이용하여 시료에 의한 보체 소비(complement consumption) 후 잔존하는 보체에 의한적혈구 용혈 정도에 근거를 둔 complement fixation test 방법으로 측정하였다. 여러 농도로 증류수에 용해시킨 시료를 GVB++ (gelatin veronal buffer, pH 7.4, 0.1% gelatin, 0.15 mM Ca++, 0.5 mM Mg++함유) 및 정상인의 혈청과 각각 50 μL씩 혼합하여 37℃에서 30분간 1차 반응시켰다. 동반응액에 GVB++ 350 μL를 가하고, 이를 10 내지 160배까지 연속 희석시킨 후, 750 μL의 GVB++와 양의 감작적혈구(IgM-sensitizated sheep erythrocyte, EA cell, 1×108cells/mL)를 250 μL를 가하여 37℃에서 60분간 2차 반응시키고, PBS (phosphate bufferedsaline, pH 7.4) 2.5 mL를 가하여 반응을 정지시켰다. 반응액은 2,000rpm에서 10분 간 원심분리 하였으며, 얻어진 상등액을 412 nm에서 흡광도를 측정하여 잔존 용혈활성을 측정하였다. 항보체 활성은 정상인의 혈청과 GVB++, 증류수만을 반응시킨 음성대조군의 총 보체용혈(50% total complement hemolysis, TCH50, %)에 대한 저지율(inhibition of 50% total complement hemolysis, ITCH50, %)로써 나타내었다. 양성대조군으로는 운지버섯 유래 면역증강제인 PSK (polysaccharide-K)를 사용하여 비교하였다.
음성대조군에서의 활성화 정도를 ITCH50 0%로 하여 CBP-0 및 CBP1S 시료의 활성화능을 확인한 결과, 활성다당 CBP-0는 1,000 μg/mL의 농도에서 동일 농도를 가한 양성대조군의 50%에 해당하는 활성을 보였으며 CBP1S는 1,000 μg/mL의 농도에서 동일 농도를 가한 양성대조군에 준하는 우수한 항보체 활성을 보였다(도 6). 양성대조군인 PSK는 현재 항암제로 시판되는 면역활성 다당이며, 일반적으로 1,000 μg/mL의 농도에서 50% 이상의 항보체 활성을 나타내는 다당체는 그 약리성이 통상적으로 인정된다고 알려져 있기 때문에, CBP1S의 보체계 활성화능이 우수한 것이 확인되었다.
<실시예 7> CBP-0의 자연 살해 세포에 의한 종양 세포 살해능 평가
전분 가공 부산물로부터 분리한 CBP-0 다당 시료를 BALB/c (6 weeks, ♀) mouse에 1,000 μg을 정맥 주사하고 3일 후에 경추탈구법으로 치사시켜 무균적으로 비장(spleen)을 적출하였다. Stainless steel mesh를 이용, PBS 상에서 마쇄(100 mesh) 및 여과(200 mesh)하여 비장세포를 획득하였다. 0.2% NaCl 5 mL을 15~30초 동안 가하여 혼입된적혈구를 파괴하고 무혈청 배지로 2~3회 세척 후 세포수가 1×106 cells/mL이 되도록 조정하고 이를 effector cell로 사용하였다. 자연 살해 세포에 대한 감수성이 높은 종양세포 YAC-1을 target cell로 하여 roundbottomed 96-well microplate에 effector cell과 target cell의 비율(E/T ratio)이 100, 50, 25이 되도록 조정 하여 가하였다. 37℃, 5% CO2 배양기에서 6시간 배양한 후 effect cell의 살해능에 의해 target cell로부터 유리되는 lactate dehydrogenase (LDH)의 발생양을 EZ-LDH (Dogen, Seoul, Korea)를 사용하여 측정하였다. 이 때, 자연 살해 세포의 종양세포 살해능은 하기 <식 1>에 의해 계산하였다.

그 결과, CBP-0 1,000 μg을 정맥주사하여 얻어진 비장 유래 세포(splenocytes, NK-cells)는 target cell (YAC-1)에 대한 effect cell (splenocytes)의 비율(E/T ratio)이 증가할수록 종양세포에 대해 높은 살해능을 나타내는 것이 확인되었다. 특히 자연살해 세포의 살해능은 E/T=100에서 가장 활성이 우수한 것이 관찰되었다. 또한, NK cell에서 분비되는 지표물질인 IFN-γ와 granzyme도 마찬가지로 E/T=100에서 가장 우수한 활성이 관찰되었다(도 7). 이로써 옥수수 전분 부산물 유래 다당 CBP-0는 종양세포에 대한 살해능을 가지는 NK cell의 활성화에 기여함을 확인할 수 있었으며, 따라서 옥수수 전분 부산물 유래 다당인 CBP-0는 NK cell의 활성화에 의한 항암활성을 가질 것으로 판단되었다.
<실시예 8> CBP1S의 종양 세포에 대한 항전이 활성능
시료의 항전이 활성은 폐(lung)에 대한 고전이성 종양세포주인 carcinoma Colon26-M3.1을 이용한 실험동물 종양전이 모델을 이용하여 평가하였다. 시료에 의한 종양전이 효과를 관찰하기 위하여 시료는 각 농도 별로 15일간 1일 2회 12시간 간격으로 실험동물에 경구투여 하였고 이후, Colon26-M3.1 lung carcinoma (3×104cells/mouse)를 BALB/c mouse에 정맥 주사하였다. 종양접종 14일 뒤 마우스를 치사시키고, 종양세포의 표적기관인 폐를 적출하여 Bouin's solution (Sigma)에서 전이된 종양을 고정시킨 후, 전이된 종양의 colony를 계수하였다. 시료에 의한 항종양전이 효과는 종양만을 접종한 대조군과 비교하여 산출하였다. 즉, 시료를 투여하지 않은 대조군을 100% 전이된 것으로 하여 시료의 항전이 활성을 평가한 것이다.
그 결과, 1000 μg의 CBP1S를 투여한 군에서 전이가 약 70%만이 진행됨으로써 30%의 우수한 항전이 효과가 확인되었다. 그러나, 10 μg 투여군 및 100 μg 투여군에서는 별다른 항전이 활성을 나타나지 않았다. 상기 결과로부터 1000 μg의 시료 투여시 종양의 전이를 억제하는데 유효한 것이 확인되었다(도 8).
<실시예 9> CBP1S-I, CBP1S-II 및 CBP1S-III의 대식세포 활성능 평가
CBP1S-I, CBP1S-II 및 CBP1S-III의 대식세포의 활성능을 평가하였다. 이는 macrophage cytokine 생산 유도능을 통하여 평가하였으며, 구체적인 평가는 상기 실시예 4와 동일한 방법으로 수행하였다.
그 결과, CBP1S-I은 1000 μg/mL의 고농도에서 10 μg/mL의 저농도까지 IL-6, IL-12 및 TNF-α의 생산능이 높게 유지되는 반면, CBP1S-II 및 CBP1S-III은 그러하지 않은 것이 확인되었다(도 9). 그러므로 대식세포 활성능을 갖는 CBP1S의 활성 다당은 CBP1S-I에 주로 존재하는 것이 확인되었다. 따라서, CBP1S-I를 대상으로 이후의 구조분석 실험을 수행하였다.
<실시예 10> CBP1S-I, CBP1S-II 및 CBP1S-III의 구성당 분석
CBP1S-I에 대하여 일반 화학특성을 알기 위하여 그 구성당을 분석하였다. 이 때 구체적인 분석은 상기 실시예 2와 동일한 방법으로 수행하였다.
그 결과, CBP1S-I은 중성당 88.9%, 산성당 9%로 구성되어 있었으며 식물추출물에서 검출되는 단백질 및 KDO물질은 소량 함유하고 있었다. 한편 CBP1S-I, CBP1S-II 및 CBP1S-III을 가수분해하여 alditol acetate 유도체로 전환하고 구성당을 분석한 결과, CBP1S-I은 arabinose와 xylose를 각각 37.9%, 29.8%의 높은 비율로 함유하고 있었으며, 그 외 glucose와 uronic acid 및 기타 구성당을 소량 함유하고 있었다(표 3). 이러한 결과는 CBP1S-I이 옥수수에 존재하는 세포벽 다당 중 hemicellulose의 한 종류인 arabinoxylan을 높은 비율로 함유하고 있을 가능성을 강력히 시사하였다.
CBP1S로부터 분리한 CBP1S-I, CBP1S-II 및 CBP1S-III의 화학적 특성
<실시예 11> CBP1S-I의 구조 분석
<11-1> Methylsulfinyl carbanion의 조제
Methylsulfinyl carbanion을 조제하기 위하여 무수 NaH 1.26 g에 무수 DMSO (dimethylsulfoxide) 20 mL을 첨가한 후 질소로 충진하며 90℃ 오일배쓰(oil-bath) 하에서 약 10 내지 15분간 반응시켰다. 반응액이 엷은 녹색을 띠는 시점을 종말점으로 하여 반응을 종결하고 상온까지 냉각시킨 후 3,000 rpm, 30℃에서 원심분리하였다. Methylsulfinyl carbanion이 함유된 상등액은 공기의 접촉이 없도록 질소로 치환하여 소량씩 분주한 후 냉동보관하며 사용하였다.
<11-2> 메틸화
다당 시료의 결합위치를 결정하기 위한 메틸화는 Hakomori 방법을 이용하여 실시하였다(Hakomori, S. A. (1964) A rapid permethylation of glycolipid and polysaccharide catalyzed by methylsuphinyl carbanion in dimethyl sulfoxide. J. Biochem. 55: 205-208). 데시케이터에서 1 내지 2일간 충분히 건조한 각 다당 시료(0.5 mg)에 1 mL의 무수 DMSO를 가하고 교반하여 완전히 용해시킨 후, 500 μL methylsulfinyl carbanion (MSCA)를 가하여 4시간 동안 반응시켰다. 이때 다당이 완전히 polyalkoxide로 전환될 수 있도록 필요한 경우 MSCA를 추가로 첨가하였으며 미반응 MSCA의 잔존 여부는 triphenylmethane으로 확인하였다. Polyalkoxide로 전환된 시료는 과량의 CH3I를 가하여 메틸화 하였으며, 잔존 CH3I는 N2 gas flushing을 통해 제거 후 Sep-pak C18 cartridge를 이용하여 회수하였다.
<11-3> 메틸화 다당의 가수분해 및 아세틸화
메틸화된 시료는 2 M TFA 1 mL을 가하여 121℃, 1.5시간 반응시켜 가수분해를 행한 후 건조하였다. 가수분해한 시료는 25% NH4OH가 수 방울 첨가된 에탄올에 용해하고 10 mg의 NaBH4를 가하여 4시간 동안 개환 및 환원하였으며, 아세트산을 적당량 가하여 잔존 NaBH4를 제거하고, 메탄올을 가하며 반복 건조함으로써 과량으로 가해진 아세트산을 제거하였다. 이후 1 mL의 아세트산무수물(acetic anhydride)을 가하고, 121℃에서 3시간 동안 반응시켜 partially methylated alditol acetate로 전환하였으며, 이는 2상 용매계(chloroform, H2O)로 분리, 추출하여 아세톤에 용해시켜 GC 및 GC-MS로 분석하였다.
<11-4> 부분적으로 메틸화된 alditol acetate의 GC 및 GC-MS 분석
GC 분석은 SP-2380 capillary column (0.25 mm×30 m, 0.2 mm film thickness, Supelco)이 장착된 Young-Lin ACME-6100 GC를 사용하여 최적온도 조건 [60℃ (1 min), 60℃ → 180℃ (30℃/min), 180℃ → 250℃ (1.5℃/min), 250℃ (5 min)]에서 splitless injection mode (1/20)로 분석하였으며, 이때 carrier gas (N2)의 flow rate는 1.5 mL/분으로 조정하였다. 한편 GC-MS는 SP-2380 capillary column (0.2 mm film, 0.25 mm i.d.×30 m, Supelco, Bellefonte, PA, USA)을 장착한 Agilent 6890N GC system과 5973N mass spectrophotometer (Agilent Technologies, Palo Alto, CA, USA)를 이용하여 GC 분석과 동일한 최적온도 조건에서 splitless injection mode (He flow rate : 1.5 mL/분)로 분석하였다. 메틸화된 시료의 유도체는 GC-MS에 의한 fragment ion 분석과 GC의 relative retention time을 조합하여 동정하였으며 각 피크의 molar %는 peak area 및 molecular response factor로부터 환산하였다
그 결과, 옥수수 전분 가공부산물 다당인 CBP1S-I는 총 8 종의 당쇄가 결합에 참여하고 있었으며 아라비노스 결합 및 자일로스 결합이 높은 비율로 확인되었다. 특히, 아라비노스 잔기의 경우 terminal-Araf가 높은 비율(12.2%)로 검출되었는데 이러한 사실은 비환원성 말단에 존재하는 아라비노스(arabinose)가 많으며, 따라서 측쇄(side chain)에 아라비노스(arabinose)가 고도로 분지된 형태임을 추정할 수 있었다. 2-linked-Araf및 5-linked-Araf가 높은 비율로 존재하는 사실로부터 아라비노스로 이루어진 측쇄는 1→2결합, 1→5결합으로 연결된 형태로 비환원 말단을 구성하는 것으로 추정되었다(표 4).
한편 자일로스 잔기의 경우 비환원 말단의 자일로스(xylose)는 거의 검출되지 않은 반면 4-linked Xylp와 3,4-branched Xylp, 2,3,4-branched Xylp가 높은 비율로 검출되었다. 이러한 결과로부터 CBP1S-I의 주쇄(main chain)가 (1→4) 결합의 자일란(xylan) 형태로 존재하며, 자일로스(xylose)의 C3위치에서 다시 한가닥의 측쇄가 뻗어져 나가거나 C2 및 C3위치에서 동시에 두가닥의 측쇄로 뻗어져 나갈 것으로 생각되었다. 특히 옥수수 전분 부산물에서 유래한 CBP1S-I 다당에는 2,3,4-linked Xylp 잔기가 특히 높은 비율로 검출되는 특이한 양상을 보였는데 이러한 사실은 (1→4) 결합으로 연결된 자일로스 주쇄에 두가닥으로 아라비노스(arabinose)가 측쇄로 연결된 형태로 확인되었다.
이러한 결과들로부터 시판되는 밀(wheat) 아라비노자일란(arabinoxylan)보다 옥수수 전분 가공부산물에서 추출한 아라비노자일란(arabinoxylan)의 경우 arabinose로 구성된 측쇄가 훨씬 많이 존재하는 것이 시사되었다(도 10). 또한, xylan 주쇄에서 뻗어나간 arabinan 측쇄도 시판 wheat arabinoxylan에 비해 상대적으로 긴 형태임을 확인할 수 있었다. 따라서 옥수수 전분 부산물 유래 면역활성 다당 CBP1S-I은 고도로 분지된 arabinoxylan에 기인하는 것으로 최종 판단되었다. 한편, 본 발명의 옥수수 전분 가공부산물에서 추출한 아라비노자일란(arabinoxylan)에 대하여 methylation을 분석한 결과, 자일로스 측쇄 수에 비해 arabinose의 말단 수가 현저히 적게 검출되는 것이 확인되었는데, 이로써 arabinan 측쇄 말단에 다른 당류가 결합되어 존재할 것으로 생각되었다.
CBP1S으로부터 정제된 CBP1S-I의 메틸화 분석
<실시예 12> CBP1S-I의 세부구조 분석
상기 실시예 9에서 사이토카인 분비를 증가시키는 것으로 확인된 CBP1S-I에 대하여 세부구조를 분석하였다.
<12-1> Endo-1,4-β-D-Xylanase에 의한 단편조제
CBP1S-I 20 mg에 0.1 M sodium phosphate buffer (pH 6.5)를 가하여 용해시킨 후, 10 units 의 endo-1,4-β-D-xylanase (from Cellvibrio mixtus, Megazyme International Ireland Ltd., Wicklow, Ireland)를 가하여 43 ℃ 항온수조에서 72시간 가수분해를 행하고, 100 ℃에서 30분간 중탕하여 잔존효소를 실활시켰다. 이 반응물을 원심분리하여 침전된 불활성 효소 단백을 제거하고 그 상등액은 50 mM ammonium formate buffer (pH 5.5)로 평형화된 Superdex 200 GL column을 이용, HPLC상에서 분획하고 중성당 함량 분석을 실시하였다. 이때 CBP1S-I-A, CBP1S-I-B, CBP1S-I-C 세 개의 획분을 얻었고, 이를 동결건조하여 endo-1,4-β-D-xylanase 처리 획분을 조제하였다(도 11).
<12-2> MALDI-TOF 분석
Matrix-assisted laser desorption ionization-time of flight (MALDI-TOF) 에 의한 분자량 측정을 위해 matrix는 10 mg/mL의 2,3,4-trihydroxyacetophenone (THAP)를 0.1% TFA가 함유된 증류수 및 아세토니트릴(acetonitrile, ACN)을 1:1의 부피비(v/v)로 용해하여 제조하였다. Matrix 용액과 시료를 1:1의 부피비로 혼합한 후, MALDI target plate에 1 μL씩 점적하고 건조하였으며, UltraflexⅢ TOF/TOF (Bruker Daltonics, Bremen, Germany)를 이용하여 가속전압 25 kV에서 시료의 분자량 결과를 얻었다.
CBP1S-I-C를 구성하는 oligosaccharide의 정확한 분자량 확인 및 구조 예측을 위하여 MALDI-TOF 분석을 수행하였다. 2,3,4-THAP를 matrix로 하여 MALDI-TOF를 행한 결과, 여러 종류의 분자량 peak가 관찰되었다. 첫 번째로 가장 단순한 결합으로 추정되는 m/z 833 [P6+Na]+의 분자량은 6개의 pentose에 [P = (132*6)+18]에 Na+ 이온 (MW: 23)이 부가된 것으로 판단되었다. 또한 m/z 833 분자량에 pentose 잔기가 1개씩 차례로 첨가된(+132) m/z 965, 1097, 1229 및 1361 peak가 관찰되었으며 이는 [P7+Na]+, [P8+Na]+, [P9+Na]+ 및 [P10+Na]+에 해당된다(도 12). 두 번째로 m/z 759 [P4+M+Na]+의 분자량은 4개의 pentose [P = (132*4)+18]에 1개의 4-methyl-glucuronic acid (MW: 190)가 결합되고 Na+ 이온 (MW: 23)이 부가된 것으로 판단되었으며, 이후 pentose가 1개씩 부가된 분자량의 peak가 역시 관찰되었다(도 13). 세 번째로 m/z 731 [P4+H+Na]+의 분자량은 4개의 pentose [P = (132*4)+18]에 1개의 hexose (MW: 162)가 결합되고 Na+ 이온 (MW: 23)이 부가된 것으로 판단되었다(도 14). 네 번째로 m/z 741 [P+Acf+Na]+의 분자량은 2개의 pentose [P = (132*2)+18]에 2개의 acetyl ferulic acid (Acf: 176+42)가 결합되고 Na+ 이온 (MW: 23)이 부가된 것으로 판단되었다(도 15). 마지막으로 m/z 713 [P+M+Acf+Na]+의 분자량은 2개의 pentose [P = (132*4)+18]에 1개의 4-methyl-glucuronic acid (MW: 190)와 1개의 acetic ferulic acid (Acf: 176+42)가 결합되고 Na+ 이온 (MW: 23)이 부가된 것으로 판단되었다(도 16). 이상의 결과로부터 옥수수 전분 부산물에서 분리된 arabinoxylan은 일반적으로 식물 다당에서 자주 검출되지 않는 4-methyl-glucuronic acid와 acetly ferulic acid가 측쇄 말단에 결합되어 있음을 확인할 수 있었다.
<실시예 13> 활성 다당 CBP1S-I의 전체 구조 분석
상기 실시예 9에서 사이토카인 분비를 증가시키는 것으로 확인된 CBP1S-I에 대하여 전체 구조를 분석하였다. 전분 부산물의 면역활성 다당 CBP1S-I의 메틸화 분석에 의한 당쇄 결합양식의 분석과 효소처리 과정을 거쳐 얻어진 정보를 바탕으로 CBP1S-I의 전체 구조를 도식화하였다(도 17). CBP1S-I은 분자량이 약 80 kDa인 다당체로, 전체 약 600 여개의 당으로 이루어져 있다.
옥수수 전분 부산물의 면역활성 다당인 CBP1S-I의 최종 구조 특성은 하기와 같다. ① xylose가 (1→4) 결합으로 연결된 xylan이 주쇄(main chain)를 구성하고 있으며, 주쇄의 대부분은 xylose의 C3위치에서 한가닥 또는 C2 및 C3위치에서 동시에 두가닥의 측쇄(side chain)가 측쇄 구조로 연결되어 존재한다. ② 측쇄 구조는 arabinose가 단당 상태로 연결되어 있거나, 몇몇 잔기는 C2, C3 또는 C5위치에서 아라비노스 잔기가 연결된 구조로 존재한다. ③ 아라비노스 말단에는 4-methyl-glucuronic acid, acetic ferulic acid와 hexose가 결합되어 있다. 그러므로 옥수수 전분 부산물이 갖는 면역 활성은 arabinose와 특이적인 당이 고도로 분지된 arabinoxylans에 의한 것임을 확인하였다.
<비교예 1> 밀 유래 아라비노자일란의 대식세포 활성능 평가
밀 유래 아라비노자일란의 대식세포의 활성능을 평가하였다. 이는 macrophage cytokine 생산 유도능을 통하여 평가하였으며, 구체적인 평가는 상기 실시예 4와 동일한 방법으로 수행하였다.
그 결과, 농도에 상관없이, 밀 유래 아라비노자일란은 IL-6, IL-12 및 TNF-α의 생산능을 증진시키지 않는 것이 확인되었다(도 18). 그러므로 모든 아라비노자일란이 면역 증강능을 갖는 것이 아니라는 것이 확인되었다.
<비교예 2> 밀 유래 아라비노자일란의 구조 분석
실시예 11과 동일한 방법으로 밀 유래 아라비노자일란의 구조를 분석하였다. 그 결과는 하기 표 5 및 도 19와 같다.

- 옥수수를 아황산 수용액으로 처리하여 얻어지는 옥수수 폐액 유래의 다당을 포함하는 면역증강용 식품 조성물로,
상기 다당은 말단에 4-메틸-글루콘산이 연결된 아라비노스를 포함하는, 면역증강용 식품 조성물.
- 제 1항에 있어서,
상기 옥수수는 옥수수 열매 또는 그 부산물인 것을 특징으로 하는 면역증강용 식품 조성물.
- 제 1항에 있어서,
상기 다당은 아라비노자일란인 것을 특징으로 하는 면역증강용 식품 조성물.
- 제 1항에 있어서,
상기 다당은 아라비노스 및 자일로스의 함량이 8 중량% 이상인 것을 특징으로 하는 면역증강용 식품 조성물.
- 제 1항에 있어서,
상기 다당은 인터루킨-6, 인터루킨-12 또는 TNF-알파의 분비를 증가시키는 것을 특징으로 하는 면역증강용 식품 조성물.
- 제 1항에 있어서,
상기 다당은 자일로스가 1 -> 4 결합으로 연결된 자일란을 주쇄로 포함하며, 또한 상기 자일로스의 C3위치에서 연결된 하나의 측쇄 또는 C2 및 C3 위치에서 연결된 두 개의 측쇄를 포함하고, 상기 측쇄는 아라비노스를 포함하는 것을 특징으로 하는 면역증강용 식품 조성물.
- 제 1항에 있어서,
상기 다당은 말단에 아세틱 페룰릭산 또는 육탄당이 연결된 아라비노스를 더 포함하는 것을 특징으로 하는 면역증강용 식품 조성물.
- 제 1항에 있어서,
상기 다당은 자연살해 세포의 활성을 증진시키는 것을 특징으로 하는 면역증강용 식품 조성물.
- 제 1항에 있어서,
상기 다당은 종양의 전이를 억제하는 것을 특징으로 하는 면역증강용 식품 조성물.
- 옥수수를 아황산 수용액으로 처리하여 얻어지는 옥수수 폐액 유래의 다당을 포함하며, 상기 다당은 말단에 4-메틸-글루콘산이 연결된 아라비노스를 포함하는, 암의 전이를 억제하는 약학적 조성물.
