Abstract
Description
The present invention relates to a liquid patterning apparatus and a liquid patterning method, and more particularly, to a non-channel type liquid patterning apparatus capable of patterning a liquid at a desired position by a surface tension between a microstructure composed of a plurality of microstructures and a liquid, To a liquid patterning method.
In general, microfluidic-based studies as an example of liquids have several advantages over conventional lab-level analytical processes because they provide faster and more sensitive detection results using smaller volume formulations.
Studies on these microfluidic fluids are increasingly being used more and more in the practice of cell-based studies and other applied studies.
Microfluidic technology can successfully mimic real organs using microfluidics, fluid separation, cell positioning, three-dimensional complex cell culture, harmonized flow, and constructive changes. For high-efficiency screening and long-term-on-chip utilization, the fixed region of interest (ROI) was an important item for data analysis and cell positioning. In the past, it has been known that chemical patterning to coat a surface with chemicals, ECM patterning, optical tweezers, droplet formation, patterning with ultraviolet curable materials, We used various methods to fix the ROI, such as the method.
However, the methods described above can efficiently fix the region of interest (ROI), but require additional processing (e.g., continuous fluid flow, the use of photocurable materials) or equipment (e.g., a light irradiation device).
In order to solve this problem, a channel type liquid patterning method has been proposed in which a liquid is patterned at a desired position on the surface of a microfluidic platform using only fine structure and liquid surface tension without additional process or additional equipment.
This channel type liquid patterning method has been developed using surface tension and surface dynamics, a new technique for rapid liquid patterning on microfluidic platforms. A microfluidic device containing a post arrangement with specific geometry was devised and made. After the microfluidic device is manufactured, the microchannel is filled with a liquid (mixture), and during liquid patterning, the liquid (mixture) inside the microchannel is sucked using a suction pump of 60 kPa pressure, Was trapped inside the post array, and using this method, the patterned liquid could be obtained in less than 5 seconds without additional surface treatment or equipment.
However, since the conventional channel-type liquid patterning method fills the microchannel with liquid, i.e., the upper portion of the microchannel is not open, the surface of the microfluidic device must be hydrophilic in order to use a liquid based on water , The patterning becomes uneven due to the affinity between the microfluid and the post material, and the patterning area is affected by the size of the microchannel, so that the patterning area is very limited.
Further, in the conventional channel type liquid patterning method, since the liquid is filled in the microchannel and then the liquid is sucked at a constant pressure by using the suction pump, the patterning is complicated and the patterning time is long, It is not easy to form a liquid pattern of a shape and a size, and recovery of a patterned liquid is not easy.
Further, in the conventional channel-type liquid patterning method, in order to pattern two or more liquids, each microchannel must be filled with a liquid and then patterned, so that it takes a lot of time for patterning as well as a problem of difficulty in patterning.
An object of the present invention is to provide a non-channel type liquid patterning apparatus capable of patterning a small amount of liquid in a desired shape and size only at a desired position, without being limited by the shape and size of the pattern, as compared with the conventional channel type patterning method.
Another object of the present invention is to provide a liquid patterning apparatus capable of uniformly patterning liquids, in particular irregardless of affinity between microfluidic and microstructure materials.
Another object of the present invention is to provide a liquid patterning apparatus capable of patterning with relatively large area as compared with the conventional channel type.
Further, the present invention is intended to provide a liquid patterning apparatus capable of performing two or more liquid patterning quickly and simply using a plurality of nozzles.

In addition, the type of the liquid 300 to be patterned is not limited. However, a liquid having an excessively high surface tension or an excessively high viscosity may reduce patterning efficiency and uniformity. Therefore, the kind of liquid to be patterned and the patterning Depending on the speed, the size of the surface tension and the viscosity of the viscosity can be changed (adjusted).

Another embodiment of the present invention is a substrate having a substrate having at least a flat bottom surface and a surface and a plurality of unit fine columns formed in a shape protruding perpendicularly to the surface of the substrate, A method of patterning a liquid inside of a microstructure of an apparatus comprising at least one microstructure, the method comprising: moving a liquid in one direction of the substrate in a different direction so that a surface between the microstructure and liquid generated on the upper and side surfaces of the microstructure Provided is a liquid patterning method for patterning a liquid in a microstructure using a tension.
This may be a liquid patterning method using a device of a structure including the substrate except for the liquid moving portion and the microstructure on the substrate in the liquid patterning device described above.
Such a liquid patterning method can be applied not only to the side surface of the microstructure, but also to the meniscus generated depending on the surface tension between the liquid and the microstructure generated at the upper side. Thus, the liquid can be patterned in the microstructure by combining the manifold generated at the side and the meniscus generated at the upper side, and the liquid can be patterned regardless of the affinity between the liquid and the microstructure.
The substrate may be of an open top shape. By opening the upper part, the use of the maniskers on the upper part of the microstructure can be facilitated, and recovery after liquid patterning can be facilitated.
The material of the unit micropillar may be a hydrophilic material or a hydrophobic material. This is made possible by utilizing the meniscus generated from the upper part, and it is possible to use hydrophilic or hydrophobic materials as the material of the unit fine pillar irrespective of affinity with the liquid.
Hereinafter, the operation of the liquid patterning apparatus according to one embodiment of the present invention will be described with reference to FIGS. 1 to 6. FIG.
A liquid patterning apparatus according to an embodiment of the present invention includes a
The liquid patterning can be divided into two steps, liquid imbibition and liquid isolation, as shown in Figs. 3 to 6.
When the liquid 300 is wiped, a meniscus is formed between the unit micropipes 210 and the liquid 300, and the liquid 300 is pushed into the
In addition, when the
If the liquid 300 is continuously wiped after the liquid 300 enters the
Further, since the liquid patterning apparatus of the present invention is a method using liquid surface tension, it is possible only when the effect of surface tension is larger than the influence of inertia upon liquid movement. Therefore, using the Weber number, which shows the relationship between the surface tension and the inertial force, it is possible to calculate and predict the kind of fluid that can be patterned, the distance between the unit fine pillar and the wiping speed.
We=(2)/We = ( 2 ) /
ρ=density of liquid(액체의 밀도)ρ = density of liquid
σ=surface tension of liquid(액체의 표면장력)σ = surface tension of liquid
L=length between two microstructures(2개의 단위 미세기둥 사이의 거리)L = length between two microstructures (distance between two unit microstructures)
u=velocisy of moving liquid (wiping speed)[액체의 이동 속도(와이핑 속도)]u = velocity of moving liquid (wiping speed) [liquid moving speed (wiping speed)]
We=(2)/≪1At this time, the greater the influence of the surface tension means that the Weber number is smaller than 1, so We = ( 2 ) / 1
The patterning can be performed only under the conditions that satisfy the following conditions.
For example, when water is patterned on microstructures arranged at intervals of 100 μm
We=(?^2)/=(1000x100x(10)6x2)/0.07≪1 We = (? ^ 2) / = (1000x100x (10) 6 x 2) /0.07«1
Calculating this equation, it can be seen that the wiping speed should be slower than about 0.84 m / s in order to pattern a liquid, for example, water, on the microstructures arranged at intervals of 100 um (200).
In this way, the liquid patterning device and the automatic patterning system can be designed in consideration of the patterning of liquid other than water, the result of patterning in the
Referring to FIG. 7, it can be seen that as the distance between the unit micro-pillars increases, wiping must be performed slowly as described below.
Umax@100um = 0.8367 m/sU max @ 100um = 0.8367 m / s
Umax@200um = 0.5916 m/sU max @ 200um = 0.5916m / s
Umax@300um = 0.4830 m/sU max @ 300um = 0.4830 m / s
Umax@400um = 0.4183 m/sU max @ 400um = 0.4183 m / s
Umax@500um = 0.3742 m/sU max @ 500 um = 0.3742 m / s
Umax@600um = 0.3416 m/sU max @ 600um = 0.3416 m / s
Umax@700um = 0.3162 m/sU max @ 700um = 0.3162 m / s
Umax@800um = 0.2958 m/sU max @ 800um = 0.2958m / s
Umax@900um = 0.2789 m/sU max @ 900 um = 0.2789 m / s
8 to 10, the non-channel type liquid patterning apparatus of the present invention is faster and simpler than the conventional channel type liquid patterning apparatus, and has no limitation on the pattern shape and size.

12 is a photograph showing a Hydrogel + microalgae mixture patterned between unit fine columns.
12, it can be seen that a minute amount of cells or liquid can be patterned only at a desired position by using the liquid patterning apparatus of the present invention (ROI fix, only a very small amount of cells and liquid are needed ).
13 and 14, since the liquid patterning apparatus according to the present invention is formed on the surface of the substrate with the top opened, it is possible to recover the material that was impossible with the conventional patterning method in the microchannel.
