먼저, 인플루엔자바이러스 헤마글루티닌(Hemagglutinin, HA) 항원의 감별을 위한 타입별 항체 개발을 위하여, 국내외 돼지 및 인체 감염 인플루엔자바이러스의 염기 및 아미노산서열을 해외 GenBank(미국), 유전체 데이터베이스에서 확보하고 각 혈청형별 multialign을 실행하여 각 헤마글루티닌 단백질 부위의 도메인별로 약 10개 이상의 보존적인 아미노산서열을 상동성 지역으로 선정하였다. 이와 동시에 동일 인플루엔자바이러스 동일 또는 타입간의 분리연도 및 지역별 바이러스 변이(mutation) 양상을 분석하여 고변이부위(hyper variable region)와 고보존 부위(highly conserved region)으로 구분하여 분석하였다.
