Field of invention
The present invention relates to methods and systems for detecting magnetic nanoparticles and observing cell or tissue samples using confocal microscopy. More specifically, the present invention provides an optical change (scattering, absorption) caused by displacement of magnetic nanoparticles activated by a magnetic field after injecting magnetic nanoparticles into a cell or tissue sample and applying a magnetic field using a magnetic field generator. And a fluorescent or the like), and a method and system for selectively observing a cell or tissue sample by measuring with a confocal microscope.
BACKGROUND OF THE INVENTION
A representative bioimaging device for precisely diagnosing cells or tissues is magnetic resonance imaging (MRI). MRI works on the following principle: When a strong magnetic field is applied to the living body outside the living body, the molecules constituting the living body and the electrons present in the atoms are aligned in a certain direction by the magnetic field. When the magnetic field applied to the electrons disappears, the electrons return to their original intrinsic position, and the time taken to return to the intrinsic position is called the 'relaxation time'. This regression rate is an intrinsic property of an atom or molecule.
MRI measures the regression velocity of each part of the living body and uses it to produce 3D images. That is, since the regression speed of the specific diseased area is different from the regression speed of the normal area, it is a device to estimate the area and extent of the disease through the quantified image.
As can be seen from this principle of operation, the identification of disease by MRI is possible only when the change of tissue caused by a specific disease causes a difference in regression rate. Therefore, a disease with a regression rate similar to that of the surrounding normal region has a problem that cannot be distinguished by MRI.
In order to solve the above problems, magnetic nanoparticles are used as contrast agents to maximize the contrast of MR images to identify diseases. The magnetic nanoparticles mainly use iron oxide-based materials having a strong magnetic iron (Iron oxide) in the center and a coating agent wrapped around them, and have a size of several nanometers to several hundred nanometers.
MRI imaging after magnetic nanoparticles were introduced into the body showed that the magnetic properties (ie regression rate) in the region where the cells trapped magnetic nanoparticles were present showed a significant difference compared to the surroundings due to magnetic nanoparticles. Thus, it is possible to distinguish the areas where cells that have captured magnetic nanoparticles exist. This technology has made it possible to diagnose certain diseases that were not possible with conventional MRI.
However, the MRI method using the contrast agent has a problem that it is difficult to observe a cell or tissue having a spatial resolution of several hundred nanometers because the spatial resolution is several hundred micrometers.
Confocal Microscope is a microscope that uses a laser as a light source, unlike a general optical microscope that uses sunlight or a halogen lamp as a light source. While scanning a laser beam adjusted to a size of about 1 μm in three dimensions, light or fluorescent light reflected at each point is measured using an optical sensor having excellent sensitivity. The measured analog information is made into 3D image using computer software.
In addition, recently, various kinds of fluorescent materials have been developed, and new technologies for studying the interaction of various materials and structures at the same time have been developed one after another, and the scope of application of confocal microscopes has been expanded. Today, major microscope manufacturers such as Zeiss, Olympus, and Nikon manufacture and sell commercially available confocal microscopes.
However, in the case of a cell or tissue region where light scattering, absorption, or change in fluorescence is small, there is a problem that it is difficult to observe with the confocal microscope because the contrast is not large.
The present inventors studied a system for solving the problems of the observation method by the MRI and the method using a confocal microscope. This led to the development of a system for observing cells or tissues that could not be easily observed in the past, with spatial resolution of hundreds of nanometers to hundreds of micrometers while increasing the contrast of light.
The present invention relates to a method and system for identifying a cell or tissue sample to which magnetic nanoparticles are selectively captured or bound using a confocal microscope from a cell or tissue sample that is not.
By "selectively captured or bound" is meant herein that "a relatively high amount of magnetic nanoparticles are ingested, captured or upragulated" compared to other cell or tissue samples.
The present invention also relates to a method and system for easily observing the structure of the vascular or lymphatic system, which was difficult to observe with conventional confocal microscopy.
An object of the present invention is to apply an external magnetic field around a cell or tissue sample in which the magnetic nanoparticles are selectively captured or bound, and then measure the optical change caused by the magnetic field with a confocal microscope to capture or bind the magnetic nanoparticles or To provide a method and system for selectively observing a tissue sample.
Another object of the present invention is to provide a method and system for observing a cell or tissue sample which has a spatial resolution of up to several hundred nanometers and at the same time increases contrast, which has been difficult to observe with conventional technology.

DETAILED DESCRIPTION Specific contents of the present invention will be described below in detail with reference to the accompanying drawings.
Detailed Description of the Invention
As shown in Fig. 1A, the confocal microscope 40 enters a strong laser (incident light) of a single wavelength into the object to be observed. As a result, fluorescence and scattered light generated in the observation target are separated from incident light by using a dichroic mirror or a beam splitter, and then measured by a photo-detector. The measured analog signal is made into an image through computer image processing.
At this time, the incident light is scanned inside and outside the observation material in three dimensions by using a scanner composed of a mirror and a motor. During scanning, optical information (fluorescence and scattering, etc.) generated at each position of the observation material is measured to produce a three-dimensional image. Stored images can be combined in the X, Y, and Z directions or played back in units of time using computer software, eliminating microscope observation time and spatial constraints.
Conventional confocal microscopy systems have been difficult to observe due to the low contrast in cells or tissue regions with small changes in light scattering, absorption and fluorescence.
However, the observation system of the present invention changes the scattering and absorption of light due to the movement of magnetic nanoparticles activated by the magnetic field by capturing or binding magnetic nanoparticles to a cell or tissue sample to be observed and by applying a magnetic field from the outside. Fluorescent light can be varied to increase contrast.
Through this, the
A system for confocal microscopy of cells or tissues in which the magnetic nanoparticles are selectively captured or bound is possible under all conditions in vivo, ex vivio or in vitro.
The difference between the present invention and MRI using magnetic nanoparticles is that the physical quantities measured to produce an image are different. MRI using magnetic nanoparticles is to measure the regression time of electrons in the molecule (phenomena occurring inside the molecule), and the present invention is characterized by measuring the actual displacement (displacement) of magnetic nanoparticles caused by the magnetic field.
Specifically, as shown in Figure 1a, the
(Conjugation with antibody or small molecule coating)되지 않고 세포 또는 조직에 포획 또는 결합된 다음, 자기장에 의하여 활성화된 자기나노입자들의 변위로 야기되는 산란광(back-scatterd light), 자기형광(autofluorescence) 또는 형광의 변화를 계측하여 세포나 조직 샘플을 관찰할 수 있다.The magnetic nanoparticles are scattered light caused by the displacement of magnetic nanoparticles activated by a magnetic field after being captured or bound to a cell or tissue without the antibody or molecule coating (Conjugation with antibody or small molecule coating) on the surface Cell or tissue samples can be observed by measuring back-scatterd light, autofluorescence or changes in fluorescence.
(Conjugation with antibody or small molecule coating)되어서 특정 세포 및 조직에 선택적으로 포획 또는 결합된 다음 이용될 수 있다. In contrast, the magnetic nanoparticles may be used after being selectively captured or bound to specific cells and tissues by chemically coating an antibody or molecule on the surface thereof. The chemical coating is a technique well known to those skilled in the art. Representative coating materials include, but are not limited to, dextran, chitosan, and the like. The size of the coated nanoparticles is preferably 20 to 500 nm.
The magnetic nanoparticles may be used without a fluorescent material attached to the surface thereof. When the magnetic nanoparticles are activated by a magnetic field, a confocal microscope is used to measure changes in scattered light (reflected light or back-scattered light), light absorption, or autofluorescence caused by displacement of the magnetic nanoparticles. The cells or tissues can be observed.
The magnetic nanoparticles may be used by attaching a fluorescent material on its surface. When the magnetic nanoparticles are activated by a magnetic field, cells or tissues may be observed by measuring the change in fluorescence caused by displacement of the magnetic nanoparticles using a confocal microscope.
Conventional MRI or conventional confocal microscopy systems using magnetic nanoparticles have low spatial resolution and low contrast, which limits the observation of cells or tissues. However, since the observation method and the system of the present invention have a spatial resolution of several hundred nanometers to several hundred micrometers and have high contrast, it is possible to clearly observe and study cells, etc. which have been difficult to observe.
Cell or tissue samples that can be more effectively observed through the present invention are i) bone marrow, liver, lymphatic vessels, interest or Kupffer cells in the circulatory system, ii) bone marrow, liver, lymphatic, interest or macrophage in the circulatory system. iii) cells that selectively trap the magnetic nanoparticles in cells circulating in the vascular or lymphatic system, iv) atherosclerotic plaques in the arteries, v) inflammation sites, vi) the magnetic nanoparticles Cancer cells that selectively capture, vii) Reticuloendothelial system (RES) that selectively captures the magnetic nanoparticles, viii) endothelial cells of blood vessels or lymphatic vessels, or iX) chemical coating of the magnetic nanoparticles Due to this, there are cells or tissues bound thereto.
The cell or tissue sample may be used in an in vivo, ex vivio or in vitro form which is conventional in the art. 2 schematically illustrates this.
When observing the cells or tissues according to the observation method of the present invention, it can also be observed using a confocal microscope combined with endoscopy.
The cells or tissues are all capable of selectively capturing or binding magnetic nanoparticles. When the magnetic nanoparticles composed of iron are trapped in the cells or tissues, for example, Cooper cells or macrophages, large iron particles are formed. When a magnetic field is applied outside the Cooper cells or macrophages including the magnetic nanoparticles, the light reflection, absorption change or fluorescent light change occurs due to the movement of the magnetic nanoparticles activated by the magnetic field, resulting in contrast. Higher, which can be clearly observed with a confocal microscope.
Since the spatial resolution of MRI images is up to several hundred microns, the spatial resolution of confocal microscopy is up to several hundred nanometers, so cells with a size that is 1,000 times smaller than those observed by MRI can be observed.
Confocal microscopy is widely used for biopsy, which is widely used for cancer cells or abnormal cells (malignant cells). However, due to poor contrast, there are many cells and tissues that were indistinguishable from conventional confocal microscopy.
The observation method and system of the present invention can be used to diagnose cancer because the distribution of macrophages can be accurately known. The magnetic nanoparticles of the present invention are ingested by Cooper cells of the liver, and most malignant tumors occurring in the liver do not include Cooper cells, so liver cancer can be easily detected. In addition to macrophages and Cooper cells, the present invention can be applied to the histology of the aforementioned cells.
In addition, using the observation method and system of the present invention, it is possible to easily observe the structure of the vascular or lymphatic system, which was difficult to observe with a conventional confocal microscope. When magnetic nanoparticles are injected into the vascular or lymphatic system, the magnetic nanoparticles circulate for a certain period of time before being selectively captured by a specific cell population. At this time, when a magnetic field is applied to the vascular or lymphatic system from the outside, the displacement of the magnetic nanoparticles in circulation is activated and changed by the magnetic field. By measuring the scattered light or fluorescence in this manner with a confocal microscope, it becomes possible to observe a vascular or lymphatic system including capillaries that was impossible to observe with a general confocal microscope. Thus, the present invention is very useful in the field of angiogenesis.
