pGTKOLacZneoTK 벡터의 제작을 위해서 우선, pBluescript Ⅱ (+) (Stratagene)의 XhoⅠ과 HindⅢ 제한효소 위치로 HSV 프로모터가 연결되어 있는 음성 선별 마커인 TK (thymidine kinase) 유전자인 HSV-TK유전자를 XhoⅠ과 HindⅢ 제한효소로 잘라서 삽입하여서 pBstTK 벡터를 제작하였다 (도 2). TK 유전자는 배지에 간시클로버(gancyclovir)라는 물질이 존재하면, 이를 분해하여 세포에 독성이 있는 효소 산물을 만들어 낸다. 상동재조합이 일어나게 되어 타겟팅 벡터의 TK 유전자가 탈락된 (도 6) 세포주는 간시클로버가 첨가된 배지에서 자랄 수 있으나, 무작위적인 삽입에 의해 타겟팅 벡터가 염색체의 임의의 위치에 끼워 들어가면 TK 유전자가 함께 들어가 발현되기 때문에 간시클로버가 함유된 배지에서 성장하지 못하고 죽게 된다. 완성된 pGTKOLacZneoTK벡터는 NotⅠ제한효소에 의해 선형화된 후, 체세포내로 도입될 수 있다 (도 5). pGTKOLacZneoTK 타겟팅 벡터와 내생적 (endogeneous) 알파 1,3-갈락토실트랜스퍼라아제 유전자 간의 동일 서열인 레프트 암과 라이트 암 부분의 이중교차에 의한 결과로서 내생적 알파 1,3-갈락토실트랜스퍼라아제 유전자의 염기서열이 타겟팅 벡터의 염기서열로 교체됨으로서 알파 1,3-갈락토실트랜스퍼라아제 유전자의 발현은 제거되고, LacZ 유전자는 알파 1,3-갈락토실트랜스퍼라아제 유전자의 프로모터 하에서 알파 1,3-갈락토실트랜스퍼라아제 유전자와 동일한 mRNA로 발현되나 IRES 염기서열의 특징으로 인하여 독립적인 단백질로 발현하게 된 다. 또한, neo유전자는 별도의 PGK 프로모터와 polyA가 연결되어 있으므로 독립적인 전사체로 발현되게 된다. 음성 선별 마커인 HSV-TK 유전자는 상동재조합의 결과 탈락하게 된다 (도 6).
