실시예Example 2: 2: 펩타이드Peptide 고정화 및 Immobilization and 펩타이드Peptide -변형 금 표면 분석-Deformation gold surface analysis
금 기판은 DC 마그네트론 스퍼터링(magnetron sputtering) 방식 및 실리콘 기판을 이용하여 제조하였다. 금 증착하기 이전에 Cr을 매개체로 증착하였다. 금과 크롬의 두께는 각각 43 nm와 2 nm이다. 펩타이드 막을 만들기 전에 결합되어 있는 이물질을 제거하기 위하여, 금 표면을 피라나 용액(piranha solution, 70% vol H2SO4, 30% H2O2)을 이용하여 세척하였다. 금 표면 상의 펩타이드 박막은 기판을 펩타이드 용액(0.1 mg/ml)으로 12 시간 처리한 후, PBS(Phosphate buffered saline)를 사용하여 세척하고, N2 가스를 이용하여 건조시켜 제작하였다. 제작된 펩타이드 박막의 표면 분석을 위하여, 금 기판과 펩타이드 1, 펩타이드 2 또는 펩타이드 3으로 개질된 금 표면을 AFM(atomic force microscopy, NTEGRA spectra, NT-MDT, Russia)를 이용하여 조사하였다.
