Claims (7)
- 밀배아 추출액, 대장균 추출액 또는 집토끼의 망상 적혈구 추출물 중 어느 하나를 함유하는 합성반응용액(반응상)과 기질 및 에너지원 공급용액(공급상)을 직접 접촉시켜 접촉계면을 매개로 하여 공급상의 기질 및 에너지원을 자유확산에 의해 반응상으로 연속적으로 공급하고,상기 반응상에서 생성된 부산물은 공급상으로 이동하여 희석되어 제거됨을 특징으로 하는 확산연속 회분식에 의한 무세포 단백질 합성방법.

- 제 1항에 있어서, 반응상과 공급상의 사이에 형성된 직접적인 접촉계면이 수직면상에 있는 것을 특징으로 하는 무세포 단백질 합성방법.
- 제1항에 있어서, 상기 무세포 단백질 합성반응용액은 밀배아 추출액을 포함하는 경우 상기 반응용액을 26℃에서 15분 ~ 30분간 프리인큐베이션한 후에 기질 및 에너지원 공급용액을 첨가하여 반응용액에 포함된 밀배아 추출액을 7~12%로 희석시켜 합성반응을 실시하는 단계를 추가로 포함함을 특징으로 하는 무세포 단백질 합성방법.
- i) 회분식 무세포 단백질 합성방법을 이용하여 단백질 합성반응을 개시하고,ii) 합성반응 정지 후의 반응용액을 겔여과칼럼 또는 반투막에 통과시켜 합성반응에서 생성된 부산물을 제거하고,iii) 단백질 합성에 필요한 아미노산, 아데노신삼인산(adenosine triphosphate; ATP), 구아닌삼인산(guanine triphosphate; GTP) 및 크레아틴인산(creatine phosphate)을 포함하는 원료 및 에너지원 용액을 상기 반응용액에 재공급하고,iv) 회분식 무세포 단백질 합성방법을 이용하여 iii) 단계의 반응용액의 단백질 합성을 다시 개시하고, 및v) 상기 ii) 내지 iv) 단계를 1회 이상 반복하는 단계를 포함함을 특징으로 하는 회분식 무세포 단백질 합성방법.
