本発明のカルボキシペプチダーゼ活性検出蛍光プローブは、外科手術において微小がん部位を高感度に可視化・検出するシステムなどの構築を図るのに有用である。
また、本発明のカルボキシペプチダーゼ活性検出蛍光プローブは、乳がんのイメージングにも有効に用いることが可能である。
アミノ酸残基として、好ましくは、グリシン、アラニンである。
式(1)のNHがPと結合する。
Sのアミノ酸残基としては、L体アミノ酸、D体アミノ酸の残基のいずれであってもよい。
このような蛍光団としては、以下の式(3)〜(5)で表される吸収・蛍光波長変化型の蛍光団、式(6)で表される蛍光強度変化型の蛍光団が挙げられる。
また、式(3)において、R2は、水素、カルボキシル基又はエステル基(−COOR:Rは、炭素数1〜6のアルキル基を表す)を示す。
式(3)において、*は、−O−P(=O)(−R1)−T−Sと結合する部位を表す
mは、0〜4の整数である。mが2以上の場合は、各々のR2は同じであっても異なっていてもよい。
R3又はR4がアリール基を示す場合には、アリール基は単環の芳香族基又は縮合芳香族基のいずれであってもよく、アリール環は1個又は2個以上の環構成ヘテロ原子(例えば窒素原子、酸素原子、又は硫黄原子など)を含んでいてもよい。アリール基としてはフェニル基が好ましい。アリール環上には1個又は2個以上の置換基が存在していてもよい。置換基としては、例えばハロゲン原子、カルボキシ基、スルホニル基、水酸基、アミノ基、アルコキシ基などが1個又は2個以上存在していてもよい。
本発明の1つの好ましい側面において、mは0である。
R2及びR3がアルキル基を示す場合には、該アルキル基にはハロゲン原子、カルボキシ基、スルホニル基、水酸基、アミノ基、アルコキシ基などが1個又は2個以上存在していてもよく、例えばR2又はR3が示すアルキル基はハロゲン化アルキル基、ヒドロキシアルキル基、カルボキシアルキル基などであってもよい。R2及びR3がそれぞれ独立に水素原子又はハロゲン原子であることが好ましい。R2及びR3がハロゲン原子である場合は、R2及びR3がともにフッ素原子であるか、塩素原子である場合がより好ましい。
本発明の1つの好ましい側面においては、R2及びR3は水素原子である。
R7又はR8がアリール基を示す場合には、アリール基は単環の芳香族基又は縮合芳香族基のいずれであってもよく、アリール環は1個又は2個以上の環構成ヘテロ原子(例えば窒素原子、酸素原子、又は硫黄原子など)を含んでいてもよい。アリール基としてはフェニル基が好ましい。アリール環上には1個又は2個以上の置換基が存在していてもよい。置換基としては、例えばハロゲン原子、カルボキシ基、スルホニル基、水酸基、アミノ基、アルコキシ基などが1個又は2個以上存在していてもよい。
また、後述するYがリン原子の場合は、R7、R8のいずれか一方は、=Oである。
本発明の1つの好ましい側面においては、Yは酸素原子である。

本発明の1つの好ましい側面において、mは0である。
即ち、本発明のもう1つの態様は、一般式(I)で表される化合物又はその塩を含む蛍光プローブである。
また、本発明のもう1つの態様は、細胞内のカルボキシペプチダーゼ活性を検出する方法であって、(a)一般式(I)で表される化合物又はその塩を細胞内に導入する工程、及び(b)当該化合物又はその塩が細胞内でカルボキシペプチダーゼと反応することにより発せられる蛍光を測定する工程を含む方法、である。
一般式(I)で表される本発明の化合物又はその塩は、カルボキシペプチダーゼのない環境において実質的に無蛍光であるか、弱い蛍光のみを有するが、カルボキシペプチダーゼのある環境において強い蛍光を発する特徴を有する。
本発明の検出方法の1つの好ましい側面においては、上記工程(a)における測定対象である細胞の試料は、がん細胞であり、好ましくは乳がん細胞である。
合成に使用した全ての化学物質は、東京化成工業(株)、和光純薬工業(株)、シグマアルドリッチ(株)から購入した。 さらに精製することなく使用した。
NMRスペクトルは、重水素化溶媒中を用い、JEOL JNM-LA400[1H 400 MHz, 13C 100 MHz]もしくはBruker NMR AVANCE III 400分光計[31P(162MHz)]で得た。
高分解能ESI質量スペクトルは、JEOL JMS-T100LC AccuToF (ESI)で得た。
HPLC精製は、Inertstil−ODS−3カラム(Φ10×250mm(セミ分取)およびΦ20×250mm(分取))を備えたJASCO PU−2080 Plusポンプ(GL Science Co.、Ltd.)およびMD-2015検出器(JASCO)で行った。
HPLCに使用した溶媒は、和光(株)より入手した。シリカゲルカラムクロマトグラフィーは、シリカゲル60N(球状、中性、63〜210μm;関東化学株式会社製)を用いて行った。
TLCは、シリカゲルプレートF254(0.25mm(分析);Merck、AKG)で行った。
UV−visスペクトルは、Shimadzu UV−1800分光光度計で得た。
蛍光スペクトルは、F-7100 (日立)で取得した。
蛍光団として青色蛍光を有する4−MU、一般式(I)におけるTとしてアラニン残基、R1としてエトキシ基、Sとしてフェニルアラニン残基およびアルギニン残基を夫々有する化合物A、Bの合成を行った。尚、化合物AとBをはじめとして本分子設計においてはジアステレオマーが存在するが、それらの単離は行わず混合物を用いて評価を行った。
合成は、以下のスキーム1により、エチルジクロロホスフェート(1)に対して4−MU、別途調整したジペプチド(2、3)を順次縮合させ、その後の酸性条件での保護基の除去を行うことで目的化合物を得た。

Fmoc−Ala−OH(622mg、2.00mmol)、H−Phe−OtBu塩酸塩(515mg、2.00mmol)、N,N−ジイソプロピルエチルアミン(1032μL、5.92mmol)をN,N−ジメチルホルムアミド5mLに溶解し、HATU(837mg、2.20mmol)を加えて室温にて一晩攪拌した。反応溶液に酢酸エチルおよび飽和塩化アンモニウム水溶液を加え、分液操作によって酢酸エチル層を抽出した。抽出した有機層を硫酸ナトリウムで乾燥させたのち、減圧除去によって溶媒を除いた。残渣をN,N−ジメチルホルムアミド8mLに溶解し、ピペリジン2mLを加えて室温にて一晩攪拌した。反応溶液に酢酸エチルおよび飽和塩化アンモニウム水溶液を加え、分液操作によって酢酸エチル層を抽出した。抽出した有機層を硫酸ナトリウムで乾燥させたのち、減圧除去によって溶媒を除いた。残渣をアミノシリカゲルカラムクロマトグラフィーにより精製し(酢酸エチル/ヘキサン=1/2→2/1)、化合物2(498mg、85%)を得た。

Fmoc−Ala−OH(595mg、1.91mmol)、H−Arg(Pbf)−OtBu塩酸塩(992mg、1.91mmol)、N,N−ジイソプロピルエチルアミン(986μL、5.73mmol)をN,N−ジメチルホルムアミド15mLに溶解し、HATU(799mg、2.10mmol)を加えて室温にて1時間攪拌した。反応溶液に酢酸エチルおよび飽和塩化アンモニウム水溶液を加え、分液操作によって酢酸エチル層を抽出した。抽出した有機層を硫酸ナトリウムで乾燥させたのち、減圧除去によって溶媒を除いた。残渣をN,N−ジメチルホルムアミド8mLに溶解し、ピペリジン 2 mLを加えて室温にて90分攪拌した。反応溶液に酢酸エチルおよび飽和塩化アンモニウム水溶液を加え、分液操作によって酢酸エチル層を抽出した。抽出した有機層を硫酸ナトリウムで乾燥させたのち、減圧除去によって溶媒を除いた。残渣をアミノシリカゲルカラムクロマトグラフィーにより精製し(ジクロロメタン→ジクロロメタン/メタノール=95/5)、化合物3(745mg、71%)を得た。

エチルホスホロジクロリダート(21μL、0.171mmol)をテトラヒドロフラン1.5mLに溶解し、アルゴン雰囲気下、−78°Cにて10分攪拌した。テトラヒドロフラン1mLに溶解した4−メチルウンベリフェロン(31mg、0.171mmol)、トリエチルアミン(47μL、0.342mmol)を順次ゆっくり滴下した後、室温まで昇温し、アルゴン雰囲気下で2時間攪拌した。反応溶液を再び−78°Cにて10分間攪拌した後、テトラヒドロフラン1mLに溶解した化合物2(100mg、0.342mmol) 、トリエチルアミン(94μL、0.684mmol)を順次ゆっくり滴下した後、室温まで昇温し、アルゴン雰囲気下で一晩攪拌した。水を加えて反応を停止した後、ジクロロメタン、酢酸エチルを用いて水層を抽出し、有機層を合わせたものを、硫酸ナトリウムを用いて乾燥させ、溶媒を減圧除去した。残渣をシリカゲルカラムクロマトグラフィー(酢酸エチル)にて一部精製した。目的化合物を含む画分を集めた後に減圧除去によって溶媒を除き、残渣をアセトニトリル1mLに溶解させた後、トリフルオロ酢酸3mLを加えて2時間室温で攪拌した。溶媒を減圧除去して得た残渣をHPLC(eluent A (H2O 0.1%TFA)and eluent B(CH3CN 80%、H2O 20%、0.1% TFA)(A/B=80/20 to 0/100 in 30min))で精製して目的化合物(7.2mg、8.4%)を得た。

The UPLC (eluent; A/B = 95/5 to 5/95 in 4 min) chromatogram after purification is shown. Absorbance at 310 nm was detected. Eluent A (H2O containing 0.1 % formic acid) and eluent B (80 % acetonitrile and 20 % H2O containing 0.1 % formic acid) were used for UPLC analysis.
エチルホスホロジクロリダート(21μL、0.171mmol)をテトラヒドロフラン1.5mLに溶解し、アルゴン雰囲気下、−78°Cにて10分攪拌した。テトラヒドロフラン1mLに溶解した4−メチルウンベリフェロン(31mg、0.171mmol)、トリエチルアミン(47μL、0.342mmol)を順次ゆっくり滴下した後、室温まで昇温し、アルゴン雰囲気下で2時間攪拌した。反応溶液を再び−78°Cにて10分間攪拌した後、テトラヒドロフラン1mLに溶解した化合物3(189mg、0.341mmol)、トリエチルアミン(94μL、0.684mmol)を順次ゆっくり滴下した後、室温まで昇温し、アルゴン雰囲気下で一晩攪拌した。飽和塩化アンモニウム水溶液を加えて反応を停止した後、ジクロロメタン、酢酸エチルを用いて水層を抽出し、有機層を合わせたものを、硫酸ナトリウムを用いて乾燥させ、溶媒を減圧除去した。残渣をシリカゲルカラムクロマトグラフィー(酢酸エチル→酢酸エチル/メタノール=97/3)にて一部精製した。目的化合物を含む画分を集めた後に減圧除去によって溶媒を除き、残渣にトリフルオロ酢酸3mLを加えて1時間室温で攪拌した。溶媒を減圧除去して得た残渣をHPLC(eluent A (H2O 0.1% TFA) and eluent B(CH3CN 80%、H2O 20%、0.1% TFA)(A/B=80/20 to 0/100 in 30min))で一部精製した。目的化合物を含む画分を集めた後に減圧除去によって溶媒を除き、残渣をHPLC(eluent C (H2O containing 100mM triethylammonium acetate) and eluent D(80%acetonitrile and 20%H2O containg 100 mM triethylammonium acetate)(C/D=80/20 to 0/100 in 30min))で一部精製した。目的化合物を含む画分を集めた後に減圧除去によって溶媒を除き、残渣を再度HPLC(eluent A (H2O 0.1% TFA) and eluent B(CH3CN 80%、H2O 20%、0.1% TFA)(A/B=80/20 to 0/100 in 30min))で生成し、目的化合物(18mg、17%)を得た。

合成した化合物A、Bを用いた酵素アッセイを行った。結果として、化合物A及びBはそれぞれの標的酵素であるカルボキシペプチダーゼA(CPA)、カルボキシペプチダーゼB(CPB)とインキュベーションすることで蛍光が上昇し、その蛍光上昇はそれぞれの阻害剤である(S)−benzylsuccinic acidおよびMGTAの添加によって抑制された。また、化合物Aの酵素アッセイにおいてはproCPAの活性化のためにトリプシンが共存しているためトリプシンのみを添加した条件も検討したが、顕著な蛍光上昇は見られなかった。一方で、わずかではあるか酵素非存在下においても徐々に蛍光が上昇してきており、本プローブは非酵素的な分解反応を受けることが明らかとなった(図2、図3)。

次に、リン原子上の置換基として新たにtBu基を導入し、一般式(I)のSのアミノ酸としてアルギニンを有する化合物Cの合成を以下のスキームにより行った。
(a) 1) 4-MU, TEA, THF, 2) H-Ala-Arg(Pbf)-OtBu, TEA, THF, 3) TFA, MeCN, 14 % in 3 steps (diastereomer mixture).
tert−ブチルホスホン酸ジクロリド(30mg、0.171mmol)をテトラヒドロフラン1mLに溶解し、アルゴン雰囲気下、−78°Cにて10分攪拌した。テトラヒドロフラン1mLに溶解した4−メチルウンベリフェロン(31mg、0.171mmol)、トリエチルアミン(47μL、0.342mmol)を順次ゆっくり滴下した後、室温まで昇温し、アルゴン雰囲気下で3時間攪拌した。反応溶液を再び−78°Cにて10分間攪拌した後、テトラヒドロフラン 1 mLに溶解した化合物3(189mg、0.341mmol)、トリエチルアミン(94μL、0.684mmol)を順次ゆっくり滴下した後、室温まで昇温し、アルゴン雰囲気下で一晩攪拌した。飽和塩化アンモニウム水溶液を加えて反応を停止した後、ジクロロメタン、酢酸エチルを用いて水層を抽出し、有機層を合わせたものを、硫酸ナトリウムを用いて乾燥させ、溶媒を減圧除去した。残渣をシリカゲルカラムクロマトグラフィー(酢酸エチル→酢酸エチル/メタノール=97/3)にて一部精製した。目的化合物を含む画分を集めた後に減圧除去によって溶媒を除き、残渣をアセトニトリル2 mLに溶解し、トリフルオロ酢酸8mLを加えて1時間室温で攪拌した。溶媒を減圧除去して得た残渣をHPLC (eluent A (H2O 0.1% TFA) and eluent B (CH3CN 80%、H2O 20%、0.1% TFA)(A/B=80/20 to 0/100 in 30min))で精製し、化合物C(15mg、14%)を得た。

合成した化合物CとCPBを用いて酵素アッセイを行った。結果として化合物CはCPBの基質となり蛍光上昇を示した一方、化合物Bと比較すると酵素非存在下では蛍光の上昇が見られず、生理的条件であるpH7.4において安定であることが明らかとなった(図4)。

一方、生細胞や臨床検体におけるカルボキシペプチダーゼのライブイメージングを行う上では、細胞毒性・自家蛍光の観点から蛍光波長を長波長化させることが望ましい。そこで次に、蛍光団として緑色蛍光を有し、フェノール性水酸基の保護・脱保護反応を、スピロ環化平衡の変化を介して蛍光のOFF/ONに繋げられることが報告されているHMRefを有する新たなプローブHMRef−tSoul−ARの合成を以下のスキームにより行った。尚、HMRefは文献(D. Asanuma, M. Sakabe, M. Kamiya, K. Yamamoto, J. Hiratake, M. Ogawa, N. Kosaka, P.L. Choyke, T. Nagano, H. Kobayashi, Y. Urano, Nat. Commun., 2015, 6, 6463.)に従って合成した。

HMRef−tSoul−ARの合成
tert−ブチルホスホン酸ジクロリド(30mg、0.171mmol)をテトラヒドロフラン1mLに溶解し、アルゴン雰囲気下、−78°Cにて10分攪拌した。テトラヒドロフラン1mLに溶解したHMRef(68mg、0.170mmol)およびトリエチルアミン(47μL、0.342mmol)をゆっくり滴下した後、35°Cまで昇温し、アルゴン雰囲気下で4時間攪拌した。反応溶液を再び−78°Cにて10分間攪拌した後、テトラヒドロフラン1mLに溶解した化合物3(188mg、0.334mmol)、トリエチルアミン(94μL、0.684mmol)を順次ゆっくり滴下した後、60°Cまで昇温し、アルゴン雰囲気下で一晩攪拌した。飽和塩化アンモニウム水溶液を加えて反応を停止した後、酢酸エチルを用いて水層を抽出した。有機層を硫酸ナトリウムにより乾燥させ、溶媒を減圧除去した後、残渣をアミノシリカゲルカラムクロマトグラフィー(ジクロロメタン→ジクロロメタン/メタノール=93/7)にて一部精製した。目的化合物を含む画分を集めた後に減圧除去によって溶媒を除き、残渣にトリフルオロ酢酸2mLを加えて2時間室温で攪拌した。減圧除去によって溶媒を除き、HPLC(eluent A (H2O 0.1% TFA) and eluent B(CH3CN 80%、H2O 20%、0.1% TFA)(A/B=80/20 to 0/100 in 30min))で一部精製し、目的化合物を含む画分を集めた後に減圧除去によって溶媒を除いた。残渣をHPLC(eluent C (H2O containing 100mM triethylammonium acetate) and eluent D (80% acetonitrile and 20% H2O containg 100mM triethylammonium acetate)(C/D=80/20 to 0/100 in 30min))で一部精製し、目的化合物を含む画分を集めた後に減圧除去によって溶媒を除いた。残渣をHPLC(eluent E(H2O) and eluent F(80% acetonitrile and 20% H2O)(E/F=80/20 to 0/100 in 30min))により精製し、目的化合物HMRef−tSoul−AR(2.2mg、1.7%)を得た。

次に、CPBとの反応性およびpH7.4の緩衝液中での安定性の検討を行った。結果として、HMRef−tSoul−ARはCPBとの反応によって3320倍もの蛍光増大を引き起こす一方、酵素非存在下では少なくとも1時間程度は安定であることが明らかとなった。その結果を図5に示す。
図5の(a)は、HMRef−tSoul−ARとCPBの反応スキームを示す。図5の(b)は、共溶媒として0.1%DMSOを含む30μgCPBとインキュベートしたPBS(−)中の1μM HMRef−tSoul−ARの蛍光増加を示す。25℃でインキュベートした。励起波長は498nmであった。図5の(c)は、PBS(−)中の1μM HMRef−tSoul−ARの25℃で50分間の吸収変化を示す。
生細胞におけるカルボキシペプチダーゼ活性のライブイメージング
上記で合成したHMRef−tSoul−ARを用いて、生細胞におけるカルボキシペプチダーゼ活性のライブイメージングを行った。具体的には、HMRef−tSoul−ARを基質とすることが明らかとなったCPMを発現するMDCK細胞にプローブを添加することで蛍光の上昇が見られるか否かを検討した。尚、CPMはCPBとは異なり活性型として細胞膜上に発現することが報告されているため、トリプシンなどの添加は不要である。
その結果、MDCK細胞からの経時的な蛍光上昇が観測された一方、その蛍光上昇は阻害剤であるMGTAの添加によって完全に抑制されることが明らかとなった(図6)。
ここで、図6の(a)、(b)は、10μM MGTAの存在下(a)又は非存在(b)での、HBSSに10μM HMRef−tSoul−ARを含むMDCK細胞の蛍光画像を示す。Ex/Em=488/500−550nm レーザー出力5%、PMT 600V。10μM Hoechst 33342を焦点面の補正のために共インキュベートした。Hoechst信号のイメージングでは、Ex/Em=405/430−460nm。レーザー出力3%、PMT 1000V、SP8。
また、20μMのHMRef−tSoul−ARを20時間インキュベーションした後の外液を回収しUPLCで分析したところ、確かにHMRefが生成していることが確かめられた(図7)。
ここで、図7は、10μM MGTAの存在下または非存在下で20μM HMRef−tSoul−ARとインキュベートしたMDCK細胞の培地のUPLC分析の結果を示す。MDCK細胞を、10μM MGTAの存在下または非存在下、37°Cで20時間、HBSS中の20μM HMRef−tSoul−ARとインキュベートした。溶出は、C/D=95/5〜5/95の線形勾配で4分で行い、254nmで検出した。
これらの結果は、生細胞におけるCPM活性のライブイメージングに成功した世界で初めての例である。
PSMA活性検出蛍光プローブの開発
これまでに開発してきたtSoulを用いて、前立腺がんのバイオマーカーとして確立しているProstate specific membrane antigen(PSMA)活性検出蛍光プローブの開発を行った。PSMAはC末端のグルタミン酸を切断する活性を有することが報告されている。そこで、蛍光団として青色蛍光を有する4−MU、一般式(I)におけるTとしてアラニン残基、R1としてtBu基、Sとしてグルタミン酸残基を有する4MU−tSoul−AEの合成を行った。

Fmoc−Ala−OH(994mg、3.00mmol)、H−Glu(OtBu)−OtBu塩酸塩(886mg、3.00mmol)、N,N−ジイソプロピルエチルアミン(1548μL、9.29mmol)をN,N−ジメチルホルムアミド5mLに溶解し、HATU(1254mg、3.30mmol)を加えて室温にて3時間攪拌した。反応溶液に酢酸エチルおよび飽和塩化アンモニウム水溶液を加え、分液操作によって酢酸エチル層を抽出した。抽出した有機層を硫酸ナトリウムで乾燥させたのち、減圧除去によって溶媒を除いた。残渣をシリカゲルカラムクロマトグラフィーにより一部精製し(酢酸エチル/ヘキサン=1/1)、目的化合物を含む画分を集めた後に減圧除去によって溶媒を除いた。残渣をN,N−ジメチルホルムアミド8mLに溶解し、ピペリジン2mLを加えて室温にて90分攪拌した。反応溶液に酢酸エチルおよび飽和塩化アンモニウム水溶液を加え、分液操作によって酢酸エチル層を抽出した。抽出した有機層を硫酸ナトリウムで乾燥させたのち、減圧除去によって溶媒を除いた。残渣をアミノシリカゲルカラムクロマトグラフィーにより精製し(ジクロロメタン→ジクロロメタン/メタノール=97/3)、化合物5(841mg、85%)を得た。

ここで、TBSバッファー中の10μM 4MU−tSoul−AEを、共溶媒として0.1%DMSOを含む10μM阻害剤(2−PMPA)の存在下または非存在下で22ng PSMAと37℃でインキュベートした。Ex/Em=355/460nm。n=3。
この結果を基に、プローブの改良を行った。PSMAの酵素認識にはC末端のグルタミン酸だけでなく、隣接するアミノ酸残基も重要であることが示唆されている。そこで一般式(I)におけるTを異なるアミノ酸に変えた誘導体の合成を行うこととした。PSMAの酵素ポケットが比較的小さいことを考慮して、分子サイズの小さな蛍光団として青色蛍光を有する4−MU、一般式(I)におけるTとしてグリシン残基、R1としてtBu基、Sとしてグルタミン酸残基を有する4MU−tSoul−GEの合成を行った。

tert−ブチルホスホン酸ジクロリド(30mg、0.171mmol)をテトラヒドロフラン2mLに溶解し、アルゴン雰囲気下、−78°Cにて10分攪拌した。テトラヒドロフラン1mLに溶解した4−メチルウンベリフェロン(30mg、0.170mmol)、トリエチルアミン(48μL、0.349mmol)を順次ゆっくり滴下した後、35°Cまで昇温し、アルゴン雰囲気下で2時間攪拌した。反応溶液を再び−78°Cにて10分間攪拌した後、テトラヒドロフラン1mLに溶解した化合物6(108mg、0.341mmol)、トリエチルアミン(96μL、0.698mmol)を順次ゆっくり滴下した後、60°Cまで昇温しアルゴン雰囲気下で一晩攪拌した。飽和食塩水を加えて反応を停止した後、酢酸エチルを用いて水層を抽出した。有機層を合わせたものを飽和塩化アンモニウム水溶液で洗浄した後、硫酸ナトリウムを用いて乾燥させ、溶媒を減圧除去した。残渣をアミノシリカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン=1/1→酢酸エチル)にて一部精製した。目的化合物を含む画分を集めた後に減圧除去によって溶媒を除き、残渣をアセトニトリル2mLに溶解し、トリフルオロ酢酸6mLを加えて2時間室温で攪拌した。溶媒を減圧除去して得た残渣をHPLC(eluent A(H2O 0.1% TFA) and eluent B(CH3CN 80%、H2O 20%、0.1% TFA)(A/B=80/20 to 0/100 in 30min))で精製し、化合物C(18mg、22%)を得た。

13C NMR (100 MHz, CD3OD): δ 17.3, 23.4, 26.7, 26.9, 29.5, 29.6, 32.8 (d, J = 130.3 Hz), 43.9 (d, J = 4.8 Hz), 51.2, 51.5, 108.8 (d, J = 4.8 Hz), 109.9 (d, J = 4.8 Hz), 113.1, 113.2, 117.0, 117.2, 117.4 (d, J = 4.8 Hz), 117.5 (d, J = 4.8 Hz), 126.3, 126.4, 153.3 (d, J = 10.5 Hz), 153.5 (d, J = 10.5 Hz), 154.3 (containing 2 peaks), 161.4 (containing 2 peaks), 171.3 (containing 2 peaks), 172.9 (containing 2 peaks), 174.7 (containing 2 peaks)

The UPLC (eluent; A/B = 95/5 to 5/95 in 4 min) chromatogram after purification is shown. Absorbance at 310 nm was detected. Eluent A (H2O containing 0.1 % formic acid) and eluent B (80 % acetonitrile and 20 % H2O containing 0.1 % formic acid) were used for UPLC analysis.
ここで、図9の(a)は、4MU−tSoul−GEとPSMAの反応スキームを示す。図9の(b)では、TBSバッファー中の10μM 4MU−tSoul−GEを、共溶媒として0.1%DMSOを含む10μM阻害剤(2−PMPA)の存在下または非存在下で22 ng PSMAと37℃でインキュベートした。Ex/Em=355/460nm。n=3。
HMRef−tSoul−ARを用いた乳がんイメージング
上記の実施例3〜4の検討でHMRef−tSoul−ARはin vitroにおいてCPBおよびCPMと反応すること、またMDCK細胞のCPM活性をライブイメージング可能であることが明らかとなった。これらの結果を基に、HMRef−tSoul−ARをがん患者由来の臨床検体に適応することでがんイメージングが可能であるか否かを検討することとした。
生体内には表2にまとめるように、塩基性アミノ酸を基質として好むカルボキシペプチダーゼが複数種存在し、いくつかはがんとの関わりが報告されている。HMRef−tSoul−ARがこれらの基質となるのかは現状わかっていないが、CPB、CPM以外のサブタイプとも反応することは十分に期待できる。そこで、これらサブタイプのうちCPD、CPE(変異体)、CPNの関連が報告されている乳がんに対するイメージングを試みることとした。これらのカルボキシペプチダーゼはCPMと同様活性体として存在しているため、トリプシンなどの添加は不要である。
HMRef−tSoul−ARを終濃度50μMになるようにPBSに溶解し(1%DMSOを含む)、100μM MGTA存在下・非存在下で乳がん患者からの臨床検体に適用した。蛍光像は、in vivoイメージング蛍光装置Maestro (PerkinElmer,Inc.,MA,USA)で取得し、励起光フィルターとして455nm(435−480nm)バンドパスフィルター、蛍光フィルターとして490 nm ロングパスフィルターを使用した。
尚、上昇が見られた場合にそれが塩基性カルボキシペプチダーゼ活性由来であるか否かを検討するため、同時に阻害剤MGTAを添加した群も検討した。MGTAは塩基性アミノ酸を好むカルボキシペプチダーゼを幅広く阻害することが知られている。
以上の結果より、開発したプローブHMRef−tSoul−ARを用いることにより乳がんのイメージングが可能であり、その蛍光上昇は乳がん組織におけるカルボキシペプチダーゼ活性の亢進によるものであることが示された。
Claims (5)
- 請求項1に記載の化合物又はその塩を含むカルボキシペプチダーゼ活性検出蛍光プローブ。
- 請求項2に記載の蛍光プローブを含む、カルボキシペプチダーゼ検出用キット。
- カルボキシペプチダーゼの活性を検出する方法であって、
(a)請求項1に記載の化合物又はその塩を細胞内に導入する工程、及び
(b)当該化合物又はその塩が細胞内でカルボキシペプチダーゼと反応することにより発せられる蛍光を測定する工程
を含む方法。 - 蛍光イメージング手段を用いて前記蛍光応答を可視化することを特徴とする、請求項4に記載の検出方法。
