More specifically, the present invention is characterized in that, in the step of removing liquid from a slurry containing liquid and solid by solid-liquid separation, the liquid removal state of the liquid is sensed and the timing for ending solid-liquid separation is determined. And a solid-liquid separation device including a means for removing liquid from a slurry containing liquid and solid by solid-liquid separation, and a means for sensing a liquid removal state.
Description
The present invention relates to a solid-liquid separation method and a solid-liquid separation apparatus for obtaining a high-purity solid from a slurry containing a liquid and a solid.
As a method for isolating and purifying a target component from a solution containing components such as natural products and synthetic products such as amino acids and peptides, crystallization by, for example, concentrated crystallization is performed. In such a crystallization process, a slurry containing a mother liquor and crystals is obtained, and crystals are recovered from the slurry by solid-liquid separation.
Centrifugal filtration is known as one of solid-liquid separation methods. For centrifugal filtration, basket-type centrifugal filtration devices are widely used. When the slurry is subjected to centrifugal filtration, if the mother liquor is not sufficiently drained and remains attached to the crystals, the purity of the product is lowered. Therefore, crystal cleaning is performed for the purpose of washing away the adhered mother liquor.
A conventional crystal separation and crystal washing process using a general basket type centrifugal filtration device will be described with reference to FIG.
A general basket-type centrifugal filtration device is programmed so that all processes are transferred by a timer, and after supplying the slurry, the mother liquor is removed for a certain period of time to separate the solid crystals and the liquid mother liquor. . After that, when moving to the crystal washing step, the attached mother liquor is washed away by spraying water or an organic solvent such as methanol onto the crystal surface as a washing liquid from the washing nozzle attached to the centrifugal filtration device. It has become. Cleaning is also performed by a timer, and cleaning is terminated after a certain period of time. Thereafter, the washing solution is drained for a certain time. In some cases, washing and draining are repeated several times, and finally the liquid is drained for a predetermined time to complete the crystal washing.
In centrifugal filtration, the size of the crystal form greatly affects the drainage of the mother liquor or cleaning solution. However, in the conventional crystal separation and crystal cleaning methods, as described above, since all processes are transferred by a timer, it is not possible to cope with a change in crystal form, and all processes are transferred at the same set time regardless of the crystal size. It will be.
For this reason, if the crystals are small and the mother liquor is very poor in liquid removal, the crystal separation process ends without the mother liquor being sufficiently drained, adversely affecting the next process such as drying and washing, and the purity decreases. Will get the crystal. If the crystals are large and the mother liquor is very good, the centrifugal operation is continued even though the mother liquor has been drained, and time is wasted.
Further, when considering the cleaning efficiency when performing crystal cleaning, it is generally considered that the most efficient cleaning method is that the cleaning liquid pushes out the mother liquor by the piston flow. Therefore, it is necessary to shift to a cleaning process just before the mother liquor on the crystal surface is drained. However, if the mother liquor is very good as described above, the mother liquor is drained at an early stage, and the remaining mother liquor is dried due to the effect of high-speed rotation, and the salts contained in the mother liquor are removed. There is a risk that it will be precipitated and the purity of the crystal will be lowered.
On the other hand, if the mother liquor has not yet been drained and the process proceeds to the washing step, a large amount of the mother liquor and the washing liquid are mixed, and the washing efficiency may be lowered and the crystal purity may be lowered.
Therefore, there has been a demand for a method of sensing the desorption state of the mother liquor and ending the crystal separation step in an ideal state, or a method of shifting to a cleaning step.
Japanese Patent Application Laid-Open No. 2004-231642 discloses a method for producing high-purity terephthalic acid by increasing the operation pressure at the time of centrifugation higher than the final crystallization pressure in the step of crystallization by flash cooling. Japanese Laid-Open Patent Publication No. 2004-261710 discloses a crystal separation and cleaning method in which the crystal grain size is adjusted to improve the stability and efficiency of cleaning, and both methods detect the desorption state of the mother liquor. However, no consideration is given to ending the crystal separation step in an ideal state or shifting to a cleaning step.
An object of the present invention is to detect a liquid draining state in a step of draining a liquid in order to recover the solid from a slurry containing the liquid and the solid by solid-liquid separation, and finish the solid separation step at an appropriate time. In other words, the solid washing efficiency is improved by shifting to the solid washing step.
According to the present invention, (1) in the step of removing the liquid from the slurry containing the liquid and the solid by solid-liquid separation, the time when the solid-liquid separation is terminated by detecting the liquid removal state or (2) (a ) Detecting the liquid drainage state during the transition from the step of draining the liquid from the slurry containing liquid and solid by solid-liquid separation to the step of (b) washing the solid separated using the cleaning liquid, Determine the timing of the transition from the dewatering process to the cleaning process.
The solid washing step may be performed before or after the step of removing the liquid from the slurry containing liquid and solid by solid-liquid separation in the above (1), and may not be performed if not necessary.
Although the slurry containing a liquid and solid is not specifically limited, For example, the slurry containing mother liquid and a crystal can be mentioned.
According to the present invention, the liquid removal state of the mother liquor is detected by the solid-liquid separation method used, and the crystal separation process is completed at an optimal time, or the process moves to the crystal washing process. The mother liquor does not remain in the crystal, and salts contained in the mother liquor do not precipitate on the crystal surface due to an excessive liquid removal step.
As a result of diligent research on a method for detecting the mother liquor drainage state, the present inventors have found that when the mother liquor is drained by solid-liquid separation, the mother liquor generates acoustic emission (hereinafter referred to as “AE”). It was found that the mother liquor drainage state can be accurately sensed by measuring.
In general, AE is defined as a signal in the frequency band of the ultrasonic region of several kHz to several MHz that is generated when the material stores the strain energy stored inside as an elastic wave. For example, AE measurement is performed. The AE measurement includes various measurement items such as the number of occurrences of AE, frequency, or energy.
As a specific example of the present invention, when general centrifugal filtration is used for the solid-liquid separation of the slurry, the AE measurement value of the signal generated when the mother liquor to be drained collides with the inner wall of the centrifugal filter and the removal of the mother liquor. It was found that there was a correlation between the liquid state and the AE sensor was used to measure the AE, and it was confirmed that the mother liquor can be immediately detected. For example, by using an AE sensor, it is possible to detect with high sensitivity the signal generated when the drained mother liquor collides with the inner wall without being affected by the noise and vibration generated from the centrifugal filtration device itself. Can be grasped.
Further, when the AE value becomes the minimum, it can be accurately determined that the mother liquor is completely drained. Therefore, by measuring AE, it becomes possible to end the crystal separation process at an appropriate time when the AE value is minimum for any crystal form. Further, in the crystal separation method including the crystal washing step, it is possible to shift to the washing step at an appropriate time before the AE value is minimized.
That is, according to the present invention, in solid-liquid separation, in particular, centrifugal filtration, the timing of ending the crystal separation depending on the size of the crystals can be appropriately determined by detecting the desorption state of the mother liquor from the measurement value by the AE sensor. It is characterized by determining. In the crystal separation method including the crystal washing process, the desorption state of the mother liquor is sensed from the measurement value by the AE sensor, and the process proceeds to the washing process immediately before the mother liquor on the crystal surface is desorbed. It is characterized by high crystal washing.

The solid-liquid separation device of the present invention is not particularly limited as long as it is a device capable of solid-liquid separation, and preferably a crystal separation device.
Similarly to the conventional method, the method of the present invention shifts from the crystal separation step to the cleaning step by the step shown in FIG. In the conventional method, each step proceeds with a timer after slurry supply, but in the method of the present invention, the timing for shifting from the mother liquor drainage to the cleaning is determined by the measured value of the AE sensor. By doing so, it became possible to finish the crystal separation at an optimal time and to perform ideal cleaning.
Furthermore, while the conventional method performed dehydration for a fixed time, in the method of the present invention, the time is arbitrarily changed according to the dehydrated state, so that it is optimal for any crystal form. Crystal separation can be completed at a certain time, and even when the crystal is washed, it is possible to always move to the washing step in an ideal state. Moreover, useless time can be saved, leading to shortening of the centrifugal filtration process time.
With reference to FIG. 2, FIG. 3, and FIG. 4, the process of separating the crystal from the slurry by the method of the present invention and the process of shifting from this process to the process of washing the crystal will be specifically described.

The drained filtrate 31 (here, mother liquor) collides with the inner wall of the

FIG. 5 shows an example of AE measurement during the centrifugal filtration operation according to the method of the present invention. In this measurement, since the slurry is supplied while rotating the
When the slurry supply is completed and the process proceeds to the liquid removal step (B region), the mother liquid is gradually discharged, and the amount of the mother liquid that collides with the inner wall of the centrifugal filtration device decreases, so the AE measurement value also decreases.
Next, when the cleaning process is started, the sprayed cleaning liquid is drained, so that the AE measurement value rises again. When the spraying of the cleaning liquid is interrupted and the liquid removal process is started, the AE measurement value decreases. This is repeated several times (C region). After washing is completed, the liquid is finally drained (D region) to recover the crystals.
At the stage where the AE value is decreasing in the region B of FIG. 5 (AE value: about 0.09 V → about 0.02 V), it can be determined that the mother liquor remains on the crystal surface. In this AE value reduction stage, it is possible to shift from the dehydration process of the mother liquor to the cleaning process, but it is possible to move at a time when the decrease rate of the AE value is greatly relaxed, where the mother liquor has not been completely dehydrated. preferable.
On the other hand, in the liquid removal process in the C region and the final stage in the D region, the minimum AE value (about 0.015 V) is shown. At this time, the liquid is completely drained. Therefore, if the minimum AE value is known in advance, a large amount of the mother liquor does not remain in the crystal at the time of washing, and salts contained in the mother liquor are precipitated on the crystal surface by an excessive liquid removal step. It is possible to more accurately determine the most suitable state.
As described above, when the AE sensor is used, it is possible to sense the liquid removal state of the centrifugal filtration. The appropriate timing for completing the above-mentioned crystal separation process and the appropriate timing for shifting from the liquid removal process to the washing process are the state and amount of slurry to be liquid discharged, the substances contained, the target crystal purity, and the solid-liquid separation used. Depends on the device.

Example 1
L-valine crystals were precipitated by concentrated crystallization from the filtrate obtained by separating the cells from the L-valine fermentation broth. Thereafter, the crystals and the mother liquor were separated by a basket type centrifugal filtration device (manufactured by Tanabe Wiltech) equipped with an AE sensor (Win-PCI2 system manufactured by Nippon Physical Acoustics Co., Ltd.). In the mother liquor draining step, the test was conducted in two patterns, the conventional method and the method of the present invention. In the conventional method, the AE sensor is not used, and all processes are terminated according to the timer setting. On the other hand, in the method of the present invention, an AE sensor was used in the mother liquor draining step, and the process was terminated when the AE measurement value was decreasing (0.06 V). Thereafter, the crystal washing step and the liquid removal step were terminated according to the timer setting. In addition, a test without a crystal washing step according to a conventional method was also conducted for a comparative test. As shown in Table 1, in the method of the present invention, the mother liquor drainage time of the conventional method could be shortened to ½.
It is known that crystals are colored when heat is applied to the mother liquor adhering to the crystals. Therefore, if the degree of coloration of the heated crystal is measured, the amount of the mother liquor adhering to the crystal can be quantified, whereby the crystal washing efficiency can be quantified.
Specifically, the L-valine crystals finally obtained by each method in Table 1 were heated for 1 hour or 24 hours with a 105 ° C. dry heat oven. Absorbance (OD 430 ) at 430 nm of a 25 g / L solution obtained by dissolving heated L-valine crystals in deionized water was measured, and the value was defined as the degree of coloring. The results are shown in Table 2.
As shown in Table 2, since the method of the present invention showed a lower value than the conventional method, it was found that the amount of the mother liquor attached was small and the cleaning efficiency was high. Therefore, by terminating the mother liquor draining process at an optimal time in AE measurement, not only the effect of shortening the draining process time but also high purity crystals could be obtained.

The L-leucine crystals finally obtained by each method were dried at 60 ° C. under vacuum for about 12 hours. The absorbance (OD 430 ) at 430 nm of a 50 g / L solution obtained by dissolving heated L-leucine crystals in 1N hydrochloric acid was measured, and the value was defined as the degree of coloring. The results are shown in Table 4.
As shown in Table 4, since the method of the present invention showed a lower value than the conventional method, it was found that the amount of the mother liquor attached was small and the cleaning efficiency was high. Therefore, by terminating the mother liquor draining step and the draining step after crystal washing at the optimal time in AE measurement, not only the effect of shortening each draining step time but also high purity crystals could be obtained.
