Abstract
PURPOSE:To control the dissolving speed of a powder mixture to water, by roughly dividing the crystal particles in each lot of a drug crystal powder hardly soluble in water into two or more particle groups according to a specific surface area and mixing other one or more lot different in the specific surface area or content of a particle group. CONSTITUTION:A powder of a hardly water-soluble drug is suspended in proper immersion liquid for measuring refractive index using a polarization microscope to be pressed on slide glass by cover glass and upper and lower nicols (polarizing filters) are inserted to perform observation. Magnification is respectively set to about 40, 100, 200 according to a purpose. A crystal habit parameter is used to make it possible to calculate a volume V, a surface area S and a specific surface area SSA according to formulae V=abc, S=2(ab+bc+ca), SSA=2(ab+bc+ca)/abcrho (rho: density). For example, a parameter (b) is actually measured with respect to all of crystal particles appearing in a visual field of 100 magnifications and the particles are successively divided into 5 groups according to the sizes of (b) and the number of particles are substituted with respect to the b-value of each group to calculate an average value. This particle size distribution graph is formed at every lot and a safe and effective preparation is formed in relation to the data of biological utilization capacity.
