Description
技術分野:
本発明はアクリル酸エステル生産菌を用いたア
クリル酸エステルの製造方法、特に水酸基を有す
るアクリル酸エステルの製造方法に関する。
従来技術:
エステル合成をエステラーゼの逆反応で行う研
究は、古くから行われている。その一つに、リパ
ーゼを用いる反応において、酸成分とアルコール
成分を種々変えたときの研究報告がある(辻阪、
岩井、奥村ら、Biochemica et Biophysica
Acta、575(1979)p.156−165)。そこでは、酸
成分として酢酸、プロピオン酸、酪酸をはじめス
テアリン酸、オレイン酸、リンゴ酸、コハク酸な
どが検討され、アルコール成分としては1級アル
コール、1級ジオール、フエノール類、2級アル
コール、2級ジオール、3級アルコール、糖アル
コールなどのあらゆるアルコール類が検討されて
いる。そこに使用されているリパーゼ類は4種で
あり、それぞれの起源はAspergillus niger、
Rhizopus delemer、Geotrichum candidum、
Penicillium cyclopiumである。そのうちの前二
者が特に低分子量の酸に対してもエステル合成能
を有することが示されている。しかし、いずれに
しろ、この研究には酸成分として、アクリル酸が
用いられていない。本発明者は上記報告に開示さ
れたリパーゼ(天野製薬(株)製よび生化学工業(株)
製)を用いて検討したが、アクリル酸はいかなる
アルコール類ともエステル合成し得ないことを確
認した。その他の従来のエステル合成に関する報
告においても酸成分にアクリル酸を用いた反応は
報告されていない。
アクリル酸とアルコールとのエステル合成によ
り得られるアクリル酸エステルは、ビニル基を有
するため、種々の工業材料となり得る重合体を合
成するための単量体として極めて有用である。例
えば、このアクリル酸エステルを他の単量体と共
重合して得られる高分子は、粘着剤、接着剤、お
よびフイルムなどとして用いられる。さらに、ま
た、このアクリル酸エステルが水酸基を有するの
で親水性を持つ官能基としても作用し、それより
得られる重合体は易架橋性ポリマーとなり、接着
剤、塗料、その他の多くの用途を有しかつ新規な
用途開発も期待される。このようなアクリル酸エ
ステルの合成は、化学合成により可能ではある。
しかし、化学合成によると、反応条件が過酷であ
るため、そのための付帯設備とスペースと費用が
かさむ。しかも、作業環境が汚染されやすく環境
公害の原因にもなる。このエステル合成を微生物
を用いた生物学的反応により行うと、低温・低圧
という温和な条件下で反応できしかも基質特異性
を有するため、環境汚染のおそれがないうえに設
備費なども安くつく。
発明の目的:
本発明の目的は、アクリル酸と二価アルコール
とにより水酸基を有するアクリル酸エステルを生
産する新規な微生物を用いて、生物学的反応によ
りアクリル酸エステルを製造する方法を提供する
ことにある。本発明の他の目的は、生物学的反応
により安全かつ安価にアクリル酸エステルを製造
する方法を提供することにある。本発明のさらに
他の目的は、アクリル酸エステル生産菌を用い
て、工業材料となり得る有用な重合体を合成する
ための単量体としてのアクリル酸エステルを製造
する方法を提供することにある。
発明の要旨:
本発明のアクリル酸エステルの製造方法は、ア
ルカリゲネス属に属する菌により、アクリル酸と
一般式HOROH(ただし、Rは炭素数2〜6の直
鎖のアルキレン基)で表されるアルコールとから
水酸基を有するアクリル酸エステルを生産するも
ので、そのことにより上記目的が達成される。ア
ルカリゲネス属菌はアルカリゲネス・フエーカリ
スであり、そのうちでも特に、アルカリゲネス・
フエーカリスTH836株およびアルカリゲネス・
フエーカリスTK4935株のうちの少なくとも一方
である。二価アルコールとしては両末端に水酸基
を有する炭素数2〜6の直鎖状アルコールであ
り、それはエチレングリコール、1・3ジヒドロ
キシプロパン、1・4ジヒドロキシブタン、1・

本発明のアクリル酸エステルの製造に用いられ
る微生物は、化学工業原料を扱つている工場など
の土壌(茨木市、豊橋市)から分離・採集した新
菌株である。この新菌株は、後述する菌学的性状
をもとに「Bergey’s Manual of
Determinative Bacterology、8th editor、1974」
の細菌分類書を参考にして同定され、アルカリゲ
ネス属に属する細菌であるところから、それぞ
れ、アルカリゲネス・フエーカリス
(Alcaligenes faecalies)TH836株(受託番号:
微工研菌寄第7083号(FERM P−7083))およ
びアルカリゲネス・フエーカリス(Alcaligenes

菌学的性質
本発明のアクリル酸エステルの製造に用いられ
るAlcaligenes faecalis TH836株とAlcaligenes
faecalis TK4935株の菌学的性質を第1表に示
す。

However, it was confirmed that acrylic acid cannot be ester-synthesized with any alcohol. Even in other reports on conventional ester synthesis, there are no reports of reactions using acrylic acid as the acid component. Acrylic acid esters obtained by ester synthesis of acrylic acid and alcohol have vinyl groups and are therefore extremely useful as monomers for synthesizing polymers that can be used as various industrial materials. For example, polymers obtained by copolymerizing this acrylic ester with other monomers are used as adhesives, adhesives, films, and the like. Furthermore, since this acrylic acid ester has a hydroxyl group, it also acts as a hydrophilic functional group, and the polymer obtained from it becomes an easily crosslinkable polymer, which has many uses such as adhesives, paints, and others. 菌株の同定
次に本発明に用いられる菌株をAlcaligenes属
に属する菌株であると同定した根拠を以下に示
す。
本菌株TH836株およびTK4935株はいずれも上
記試験結果より「グラム陰性であり、***により
増殖する好気性細菌であり光合成によつては増殖
しない」という特徴を有する。このような細菌
は、Bergey’s Manualによれば第7部に分類
されている。ここに属する科および属は次のよう
になる。

本菌株は、いずれもAlcaligenes faecalisと
は、プロピオン酸と酪酸の資化性能を欠く点が異
なるにすぎず、他の特徴はすべてこれと一致す
る。よつて、本菌株をAlcaligenes faecalisにき
めて近縁の菌株でありAlcaligenes faecalisと同
定した。
培養条件
培地としては格別である必要はなく、肉エキ
ス、酵母エキス、麦芽エキス、ペプトンなどの有
機栄養源、およびリン酸塩、マグネシウム、ナト
リウム、カリウム、マンガン、鉄などの無機栄養
源等を適宜含有する通常の培地でよい。
炭素源としては、炭素源資化性試験において基
質となつた各種アミノ酸、TCAサイクルメンバ
ーの有機酸などが用いられる。窒素源としては、
硝酸塩、亜硝酸塩およびアンモニウム塩などの無
機窒素やアミノ基の有機窒素などが用いられる。
培養温度は20℃〜35℃好ましくは25℃〜30℃であ
る。培養PHは5〜8好ましくは6.5〜7.5であり、
1日〜3日間好気的に撹拌又は振とうしながら培
養を行なう。
反応組成
本発明のエステル反応は、このような培養条件
のもとで本菌株を培養し、その休止菌体、菌体抽
出液(粗酵素液)、精製酵素、固定化菌体および
固定化酵素状態のうちの少なくとも一つを適当に
選んで用いる。本発明の水酸基を有するアクリル
酸エステルの合成に必要な反応系の基本組成は第
5表に示される。なお、この合成に当つて媒体を

得られ、280nmの吸光度(A280)が0.1以上、通
常は0.5〜20.0の範囲で用いられる。
反応PHは5〜9の範囲にあればよい。好ましく
は6.5〜7.5である。
アクリル酸の使用可能な濃度は、0.1%(v/v)〜
5%(v/v)であり、好ましくは0.5%(v/v)〜2%
(v/v)の範囲に選ばれる。アクリル酸の使用によ
り反応系組成のPHが低下しアクリル酸の消費と共
にPHが上昇する。このようなPH変動を極小にし反
応系のPHが常時7前後になるようにするために本
基本組成ではバツフアーとして0.2Mのリン酸バ
ツフアーが用いられる。リン酸バツフアーに代え
て重炭酸−炭酸Naバツフアー、トリス塩酸バツ
フアーなどを用いることもできる。バツフアー濃
度も0.2Mに限定されることはなく、使用される
アクリル酸量や反応速度などに応じて適宜選択さ
れる。
前記二価アルコールは、酵素の安定化に悪影響
を与えることがないため量的な制限は特にない。
しかし、アクリル酸1モル当り、少なくとも0.5
モルの二価アルコールが用いられる。上記アルコ
ールの使用量の上限は前記のようにいくら多くて
もよいが、通常0.5%(v/v)〜10%(v/v)で用いられ
る。モル比ではアクリル酸1モル当り通常0.5〜
50モル、好ましくは1〜20モルの二価アルコール
が用いられる。ジヒドロキシアルコールの添加量
に応じて生成する水酸基を有するアクリル酸エス
テルの量も多くなる。反応温度は20℃〜40℃の範
囲内であれば良い。好ましくは25℃〜35℃であ
る。反応時間は基質添加量により幾分異なるが約
10分以上であれば検出可能である。
生成物の検出
反応終了後の系から菌体を遠心分離により除
く。その上澄液を直接ガスクロマトグラフあるい
は液体クロマトグラフにかけ生成物質の検出定量
を行なうか、もしくはその上澄液を必要に応じて
さらに熱処理(例えば100℃にて1分間)し夾雑
する蛋白を除去してからガスクロマトグラフもし
くは液体クロマトグラフにかけ生成物質の検出定
量を行なう。検出は次のような条件で行なわれ
る。

例えば、アクリル酸と1・4ブタンジオールと
の反応で生成するヒドロキシブチルアクリレート
についてはガスクロマトグラフでは約5分後、液
体クロマトグラフでは約4分後にピークが現われ
る。二価アルコールがエチレングリコールであれ
ばヒドロキシエチルアクリレートが生成され、液
体クロマトグラフでは3.2分にピークが見られ
る。
生成物質の確認
生成物質の確認は次のようにして行なつた。
1 アクリル酸、二価アルコールおよび酵素の3
者が同時に存在したときだけ、生成物のピーク
が見られ(液体クロマトグラフにて)、そのピ
ークが時間とともに増加する。
2 ガスクロマトグラフおよび/もしくは液体ク
ロマトグラフにより既知物質と同じリテンシヨ
ンタイムを示した。(既知物質は化学合成にて
入手した)
3 GC−Mass(日立製)によるCI法により生成
物質が既知物質と同じ分子量であることを確認
した。
生成されたアクリル酸エステルを採取するには
常法の蒸留もしくは溶剤抽出が用いられる。
酵素の安定性
Alcaligenes faecalis TH836株のエステル化能
を有する酵素の安定性について、超音波破砕時
(海上電気(株)のTA4280振動子4280S;2〜4Aにて
使用)の酵素の安定性を第1図に示す。図はソニ
ツク処理時間とヒドロキシブチルアクリレートの
生成反応活性との関係を示している。活性は、1
分間にヒドロキシブチルアクリレート1μmol生
成したとき1ユニツトとした。第1図から本菌株
TH836株のエステル化酵素は比較的安定である
ことがわかる。
エステル化反応におけるPH
反応系組成とそこに用いる各種バツフアーを下
記のように調整し、本菌株TH836株を用いたヒ
ドロキシブチルアクリレートの生成に及ぼすPHの
影響を調べた。その結果を第2図に示す。図から
酵素活性はPH5〜10付近までであることがわか

実施例:
以下に本発明を実施例により具体的に説明す
る。
実施例 1
(菌体懸濁液の調製)
肉エキス10g、ポリペプトン10g、NaCl 5
g、蒸留水1、そしてPH7.0の組成の肉汁液体
培地100mlを調製した。これを500ml容坂口フラス
コに入れ殺菌して後、これにAlcaligenes
faecalis TH836株を植菌した。これを30℃にて
24時間振とう培養した。この培養液を遠心分離
(10000rpm、10min.)し、沈澱した菌体をPH7.0
のリン酸バツフアー0.05Mにて1回洗浄した。こ
の菌体を同じバツフアーにてOD660=10.0になる
ように希釈し、これを菌体懸濁液とした。
(粗酵素液の調製)
得られた菌体懸濁液を海上電気製超音波破砕機
にて氷冷水で冷却しつつ20分間破砕した。これを
遠心分離(15000rpm、30分間)し、その上澄液
を粗酵素液とした。
(反応組成)
次の反応組成にて30℃で反応し、得られた生成

(結 果)
1・4ブタンジオール濃度のエステル生成にお
よぼす影響を調べた。その結果を第3図に示す。
エステル生成量は対アクリル酸収率(モル%)で
表示される。1・4ブタンジオール量が多くなる
につれ生成するヒドロキシブチルアクリレートの
量も多くなることがわかる。また、アクリル酸10
%(最終濃度)と1・4ブタンジオール10%(最
終濃度)との(1:1の)反応でもジアクリレー
トの生成は生成アクリル酸エステルの1%以下で
あつた。しかし、アクリル酸と二価アルコールと
の比が1:2以上になるとジアクリレートはまつ
たく生成しなかつた。化学合成においてはアクリ
ル酸と二価アルコールとを1:1で反応させる
と、生成したアクリル酸エステルに対し5〜15%
のジアクリレートが生成する。ジアクリレートは
重合時の反応系をゲル化させるため、それが生成
しないようにすることが必要である。化学合成で
はそれは極めてむつかしい。それゆえ、重合時の
仕込み濃度を低くおさえねばならないという欠点
があつた。本発明のエステル反応においてはジア
クリレートの生成は微量もしくは皆無であるた
め、このような問題はない。
実施例 2
実施例1で示した培地にて同様にAlcaligenes
faecalis TH836株とAlcaligenes faecalis
TK4935株をそれぞれ培養した。得た各菌体を
OD660=10.0になるように0.05Mのリン酸バツフ
アー(PH7.0)にて、菌体懸濁液を調製した。こ
の菌体懸濁液を実施例1と同様に処理して粗酵素
液を調製した。この菌体懸濁液もしくは市販の酵
素液を用いて次の反応組成にて30℃で2時間反応
を行つた。市販のリパーゼとしては、
Aspergillusに属する糸状菌から生産されたリパ
ーゼ(リパーゼAP:天野製薬(株))およびMucor
に属する糸状菌から生産されたリパーゼ(リパー

発明の効果:
本発明のアクリル酸エステルの製造方法では、
菌株Alcaligenes faecalis TH836およびTK4935
を用いて、アクリル酸と二価アルコールとを含有
する反応系から水酸基を有するアクリル酸エステ
ルを製造蓄積させることができる。エステル化に
必要な酵素は、培地の種類に無関係に、菌体増殖
に比例して生産される。この生成物アクリル酸エ
ステルはビニル基を有するため、工業材料となり
うる重合体を合成するための単量体として極めて
有用である。この生成物を他の既知単量体と共重
合して得られる高分子は粘着剤、接着剤、フイル
ムなどとして用いられうる。しかも、この生成物
は水酸基を有するため、親水性をもちしかも官能
基としても作用するので、他の多くの用途に利用
されうる。
さらに、本発明のエステル反応において、ジア
クリレートがほとんど生成されない。ジアクリレ
ートが系内に生成されなければ、重合反応中にゲ
ル化が起こらず、したがつてエステル反応出発物
質の高濃度仕込みが可能となる。その結果、新規
な特徴をもつ高分子を得ることも可能である。微
量ながらジアクリレートが系内に生成されても、
本菌株はいずれもこれに対応するアクリル酸エス
テルとアクリル酸とに加水分解する能力を有する
