Abstract

PURPOSE:To obtain a reagent exhibiting specific agglutination reaction of high sensitivity by sensitizing an antibody (antigen) with the latex obtd. by polymerizing styrene by the use of a water soluble radical initiator in the presence of a silicic acid anhydride of ultrafine particles then treating the same with an antibody (antigen)-contg. serum. CONSTITUTION:A silicic acid anhydride of ultrafine particles of about 5-50mmu grain sizes (''Aerosil '': Product of Nihon Aerosil Co., Ltd., or the like) is dispersed in water, and strene is polymerized by using a water soluble radical initiator such as K2S2O7 or the like in water, whereby latex of small variance in grain sizes of about 0.384mu average grain sizes is prepd. This latex is sensitized with an antibody (or antigen) at 7-8.6pH. A serum contg. an antibody (antigen) is further added to the sensitized latex and these are stirred for a short time, after which the serum treated latex is again suspended, whereby a latex reagent is prepd. When the resultant reagent is caused to react with a specimen contg. an antigen (an antibody when the reagent is sensitized with the antigen), it makes specific agglutination reaction, and the antigen is detected without the disturbance by the other protein or the like in the specimen and with high sensitivity. In this way, this reagent is adapted for detection of hepatitis B virus and the like.
