Abstract

PURPOSE:To stably perform quantitative analysis with high accuracy by a method wherein a solution containing saccharides is separated through a separating column of liquid chromatograph and hydrolized under heating while continously adding an acid or alkali solution thereto, and then colorimetry is effected by continuously adding reducing sugar coloring reagent to the solution hydrolized. CONSTITUTION:A given quantity of a solution 10 containing saccharides such as non-reducing saccharides, oligosaccharides or polysaccharides is sampled by a sampler 3 and eluting liquid 1 is sent to the sampler by a quantitative pump 2. The saccharide solution within the sampler 3 is fed to a separating column 4 of liquid chromatograph and separated with the eluting solution 1 fed thereto subsequently, so that the eluted solution enters a mixing portion 5. Meanwhile, a saccharide hydrolizing reagent (acid or alkali solution) is sent to the mixing portion 5 from a bottle 22 by a quantitative pump 20 in such a manner that its pulsatory current is reduced through a resistance coil 40. Then, the mixed solution is fed to a heating reaction tube 6 and hydrolized at temperature of 70-150 deg.C. Thereafter, the solution is fed to a mixing portion 7 to be mixed with reducing sugar coloring reagent and is reacted in a coloring reaction tube 8 at temperatur of 80-120 deg.C. The reacted solution is measured in a detecting section 9 by a spectrophotometer or colorimeter and the result is recorded on a recorder 38.
