Abstract

PURPOSE: To prepare the titled compound in high productivity, by treating a specific compound with immobilized enzyme obtained by adsorbing acid amidase to a porous ion exchange resin.
CONSTITUTION: An acid amidase is adsorbed to e.g. a strongly basic anion exchange resin (e.g. Amberlite IRA904) having a specific surface area of about 40W60m2/g, prove volume of about 0.85W0.9ml/g, mode pore diameter of about 650W750Å, and ion exchange group of -N-(CH3)3-X, and the adsorption is fortified with a polyfunctional protein crosslinking agent such as glutaraldehyde. A compound of formula I (R is -COOH, etc.; X is nucleophilic residue such as H) is passed through a column packed with the above immobilized enzyme to obtain the 7-amino-cephem compound of formula II by enzymatic reaction. The reaction is carried out in an aqueous medium in the presence of pH-buffering agent.
