Abstract

PURPOSE: To prepare the titled compound useful as an intermediate of a 4-substituted indole compound having remarkable pharmaceutical activity, e.g. ergot, by cyclizing a 1-alkoxycarbonylpyrrole derivative in the presence of an acid catalyst.
CONSTITUTION: The objective compound of formula III (R1 is alkyl which may have an oxygen-substituent group) is prepared, in high yield, by hydrolyzing a compound of formula II (R2 is lower alkyl) with caustic alkali in methanol. The compound of formula II can be obtained by reacting 1 mole of a 1-alkoxycarbonylpyrrole derivative of formula I with 3W5 moles of tin tetrachloride in the presence of an acid catalyst (a mineral acid or a Lewis acid) pref. in a chlorinated alkane solvent, with ice cooling under agitation; and the compound of formula I is prepared, in high yield, by reacting a compound of formula IV with an alkyl magnesium halide, and then oxidizing the reaction product with chromic anhydride.pyridine adduct.
