Abstract

PURPOSE: To obtain a lactam useful as a remedy for peptic ulcer, etc. and an intermediate for other medicines in high yield industrially and efficiently, by reacting a 1-benzoyl-pyrrolidin-5-one-2-carboxylic acid halide with a secondary amine.
CONSTITUTION: N-Benzoylglutamic acid is reacted with a phosphorus halide under mild reaction conditions to give a 1-benzoyl-pyrrolidin-5-one-2-carboxylic acid halide of formula I (X is halogen) in high yield. The resultant compound of formula I is then preferably without isolation from the reaction mixture reacted with an amine of formula II (R1 and R2 are 1W6C alkyl or together with the adjacent N link to form a heterocyclic ring) under mild condition in the absence of a solvent in a short time of about 1W2hr to give a lactam of formula III almost quantitatively. According to the method, the lactam of formula III can be prepared in high yield by using the conventional reactor with saved energy.
