Abstract

PURPOSE: To provide a method of determination of deoxyribonucleic acid-binding protein useful for a clinical diagnosis, by separating it from serum by a specified column chromatography and affinity chromatography, and providing the gel cataphoresis and dyeing before performing scanning by means of a densitometer.
CONSTITUTION: DNA-binding protein is separated from the serum inspected by the column chromatography using diethylaminoethyl ether (DEAE)-Sephadex and the affinity chromatography using the deoxyribonucleic acid (DNA)-binding carrier. After a more precise separation is done by the gel cataphoresis, scanning is performed by means of a densitometer in order to determine DNA-binding protein of molecular weight approximately 64,000. The microcolumn, filler, various buffer solutions, eluate, precipitant, gel filler, dyeing plate, and reagent for measuring protein which are used in the above method, are made to be a set to provide a quantitative kit.
