Abstract

PURPOSE:To obtain a stable measured value by oxidation and condensation with a color former, such as 4-amino antipyrin by using a composition which having as a coloring-matter forming substance an N-sulfoalkyl aniline derivative or its salt. CONSTITUTION:An N-sulfoalkyl aniline derivative shown by the formula (R<1> is a substituent selected among a hydrogen atom, lower alkyl group and sulfoalkyl group of 1-4 carbon number; R2 is a sulfoalkyl group of 1-4 carbon number; R<3> is a substituent selected among a hydrogen atom, alkoxy group, hydroxyl group, and lower alkyl group; and R<4> is a substituent selected among a hydrogen atom, alkoxy group and lower alkyl group) or its salt is dissolved in an acetate buffer solution together with glucose oxidase peroxidase and 4-amino antipyrin to obtain a coloring reagent. Then, the coloring regent is added to blood serum in a test tube for reaction and then the light bsorbance in contrast, to the blank teston regent is measured and contrasted with a calibration line to calculate the concentration of glucose in the serum.
