Abstract
PURPOSE: To prepare slow-releasing complex, by denaturating a system consisting of an aqueous solution of a bioactive substance and a protein as a whole, thereby dispersing a sufficiently large amount of the bioactive substance in the coagulated protein.
CONSTITUTION: One or more bioactive substances (e.g. 5-fluorouracil, insulin, etc.) are brought into contact with one or more proteins (e.g. serum albumin, γ-globulin, etc.) in an aqueous solution, and treated physically or chemically (e.g. heating in a water-bath at 80W90°C for a short time) to denaturate the proteins. By this process, a slow-release complex composed of a bioactive substance dispersed and included in a denaturated protein matrix can be obtained. The process affords the inclusion of a sufficiently large amount of the bioactive substance, and enables the release of the substance slowly for a long period when administered to a living body.
