Abstract

PURPOSE: To carry out the concentration and purification of β-amylase in high efficiency, by adding a salt of a specific metal and a halogen-free anion to a β-amylase extract liquid, and concentrating the mixture through a membrane.
CONSTITUTION: A raw material such as soybean, malt, sweet potato, etc. is extracted with water or a buffer solution to obtain the extract containing β-amylase. Proteins are removed from the extract by acid precipitation, salting out at a relatively low degree of saturation, addition of an alkaline earth metal, etc. The extract is mixed with ≥0.01% of a salt of a metal selected from Ca, Mg and Al and a halogen-free anion, e.g. CaSO4, magnesium acetate, aluminum borate, etc., adjusted to pH4W6, and concentrated with a membrane, e.g. by ultrafiltration, reverse osmosis, etc. at 50W55°C. By this process, the putrefaction of β-amylase is prevented while keeping the high enzymatic activity.
