Abstract

PURPOSE: To obtain a reagent for measuring purine nucleoside phosphorylase (PNP) activity, adenosine deaminase(ADA) activity, and guanase activity in the blood, in the blood dissolving red corpsucle, and in a culture solution, by combining a substrate solution, coloring solution, enzyme solution, and reaction terminating agent.
CONSTITUTION: For PNP, a substrate solution consists of inosine, sodium azide, and 0.05W2mol buffer solution in pH 7W8, for ADA, it consists of adenosine, sodium, azide, and 0.05W0.2mol buffer solution in pH 7W8, and, for guanase, it consists of guanine, sodium azide, and 0.05W0.2mol buffer solution in pH 6W7. An enzyme solution is a solution of 1.5W4mol xanthine oxidase or xanthine oxidase and prine nucleoside phosphorylase in ammonium sulfate suspension or 40W60wt% glycerol solution. A reaction terminating agent consists of 0.1W0.3mol acitic acid solution, etc.
