Abstract
PURPOSE: To measure a very small amount of kallikrein contained in human urine in high sensitivity by adding prolyl-phenyl-alanyl-arginine-4-methylcoumarin-amide into human urine whose pH is adjusted in a specified range, and measuring the product by a fluorescent method.
CONSTITUTION: An enzyme reaction is made by contacting prolyl-phenylalanyl- arginine-4-methylcoumarin-amide with human urine whose pH range is adjusted from 8 to 12 and 7-amino-4-methyl-coumarin is produced, then, it is measured by a fluorescent method. For example, 0.5ml of 0.1-M tris-HCl buffer solution (pH 8.0) containing 0.15M of NaCl is mixed with 5W100μl of human urine at 37°. After 5 minutes, 5μl of 10-mM L-prolyl-L-phenylalanyl-L-arginine-4-methylcoumarin-amide is added, thus 7-amino-4-methylcoumarin is produced, and its fluorescent intensity at 460nm is measured.
