Abstract
NEW MATERIAL:An amikacin derivative of formula I (R is OH or H) and its acid addition salt.
USE: Remedy for bacterial infectious diseases, causing little damages to the faculty of hearing.
PROCESS: The objective compound of formula I wherein R is H is prepared e.g. by (1) protecting the four NH2 groups of amikacin of formula II with amino-protecting groups, (2) protecting the two OH groups with one bivalent OH-protecting group, (3) protecting the remaining five OH groups at 2', 3', 4', 2", and 2''' positions or six OH groups at 5, 2', 3', 4', 2", and 2''' positions, with one OH-protecting group, (4) eliminating the OH-protecting groups at the 4" and 6" positions to obtain a compound of fomula III (A is H; B is monovalent amino-protecting group, etc.; D is acyl-type OH-protecting group; E is H or acyl-type OH-protecting group), (5) converting the 6" OH group or 4" and 6" OH groups to sulfonic acid ester, (6) iodizing the product, and (7) eliminating the iodine by catalytic reduction.
