Abstract

PURPOSE: To determine spermidine with good reproducibility quickly and easily, by reacting a spermidine-containing solution with spermidine dehydrogenase in the pressence of an electron acceptor.
CONSTITUTION: A spermidine-containing solution, e.g., the human urine, an extracted solution of the human organization, is reacted usually with 0.1W5.0 enzyme units of spermidine dehydrogenase in the presence of an electron acceptor, e.g., potassium ferricyamide. A SH group-containing compound (e.g., β-mercaptoethanol) is then added to the reaction solution to reduce the excess electron acceptor so that the reaction solution becomes colorless. Prepared 1-pyrroline shown by the formla 1 is submitted to colorimetry to determine spermidine. By using this determination of sperimidine, a solution containing both spermidine and putrescine can be analyzed quantitatively to determine putrescine with good reproducibility quickly and easily.
