Abstract
PURPOSE: When lactic dehydrogenase is measured using lactic acid and NAD as a substrate, pyruvic acid is determined by use of pyruvic oxidase, thus permitting the accurate measurement of the activity of lactic dehydrogenase.
CONSTITUTION: The reaction of lactic acid with nicotinic amide-adenine dinucleotide is effected in the presence of unknown concentration of lactic dehydrogenase to form pyruvic acid, which is oxidized with a pyruvic oxidase, e.g., one produced by Lactobacillus delbruckii, to form hydrogen peroxide. The hydrogen peroxide is made to act on a pigment precursor in the presence of peroxidase and the color developed is determined by colorimetry.
