Abstract

PURPOSE: To reduce the dose of piromidic acid, and to suppress the side effects of a piromidic acid antibacterial composition such as renal disorder as far as possible, by compounding piromidic acid with trimethoprim.
CONSTITUTION: A composition composed of (A) 1pt.wt. of piromidic acid of formula I and (B) 0.25W0.02 part of trimethoprim of formula II. Piromidic acid which is a known chemotherapeutic agent having strong antibacterial activity against gram-negative bacteria, has a disadvantage of causing side effects such as renal disorder. On the other hand, trimethoprim inhibits the metabolism of di-hydrogen folic acid to tetra-hydrogen folic acid. An excellent synergistic effect can be attained by the combined use of both compounds, which results in the reduction of the dose of piromidic acid. It can be administered as capsules or tablets.
