Abstract

PURPOSE: In contacting and mixing of alkaline eluent with strong acidic cation exchange resin containing adsorbed low soluble amino acid, rapid flowing and agitation accelerates elution of amino acid, allowing the use of only a small amount of high concentrated eluent solution.
CONSTITUTION: Commercial strong acidic cation exchange resin layer which has adsorbed low soluble amino acid such as cystine, glutamic acid, tryptophan and the like is treated. Through the resin layer, alkaline eluent such as aqueous solution of ammonium chloride 0.1 N or more concentration is passed upwardly at a rate of 5 m/hr or more. The alkaline eluent is left stationary for a while in the resin column or the resin layer is agitated. Alternatively, air is jetted from the bottom of the resin layer and the alkaline eluent is passed from either the bottom or the top. After elution treatment, water or alkali solution of about 0.1W1 N is passed from the top to remove the eluent.
