Abstract

PURPOSE: To prepare high purity L-malic acid dehydrogenase (MDH) having high stability and low Km value to oxalacetic acid, and useful for clinical diagnosis, by culturing MDH-producing bacteria belonging to Alcaligenes or Klebsiella genus.
CONSTITUTION: MDH-producing bacteria belonging to Alcaligenes or Klebsiella genus, such as Alcaligenes faecalis ATCC 8750, Klebsiella pneumoniae L ATCC 8307, are aerobically cultured in a nutrient medium at pH7-8 for 1-2 days under stirring. After culturing, bacterial cells are separated by filtering or centrifuging, and crushed. High purity MDH can be separated from the curshed cells. The production rate of the MDH can be improved by adding an enzyme-inducing material, such as fumaric acid, malic acid, etc., to the culturing medium
