Description
Claims (1)
- 【特許請求の範囲】 1.哺乳動物に移植するための最終滅菌骨原性デバイスであって; (a)不溶性の合成ポリマーキャリア材料と、該キャリア材料中に配置されて 哺乳動物中に移植された場合、軟骨性骨形成または関節軟骨形成の誘導能を有す る単離された生物活性骨原性タンパク質とを組み合わせて含む骨原性デバイスを 提供する工程;および (b)工程(a)の骨原性デバイスを、デバイスを滅菌するに十分な量のイオン 化放射線に曝露し、それにより哺乳動物における軟骨性骨形成または関節軟骨形 成の誘導能を有する最終滅菌骨原性デバイスを製造する工程 により製造される、デバイス。 2.前記不溶性キャリアがポリ乳酸、ポリ酪酸、ポリグリコール酸またはその混 合物を含む、請求項1に記載のデバイス。 3.哺乳動物に移植するための最終滅菌骨原性デバイスであって; (a)不溶性のポリマーキャリア材料と、該キャリア材料中に配置されて哺乳 動物中に移植された場合、軟骨性骨形成または関節軟骨形成の誘導能を有する単 離された生物活性骨原性タンパク質とを組み合わせて含む骨原性デバイスを提供 する工程;および (b)工程(a)のデバイスを、デバイスを滅菌するに十分な量のイオン化放射 線に曝露し、それにより哺乳動物における軟骨性骨形成または関節軟骨形成の誘 導能を有する最終滅菌骨原性デバイスを製造し、ここで該最終滅菌骨原性デバイ スは、滅菌前の骨原性デバイスの少なくとも約20%の生物活性を有する、工程 により製造されるデバイス。 4.前記キャリア材料と前記骨原性タンパク質とが重量比で約1:1〜約250,000 :1の範囲で組み合わされる、請求項1または3に記載のデバイス。 5.キャリアと骨原性タンパク質との前記重量比が、約40:1〜約50,000:1の範 囲である、請求項4に記載のデバイス。 6.前記イオン化放射線がガンマ線である、請求項1または3に記載のデバイス。 7.前記イオン化放射線が約0.5〜約4.0メガラドの範囲内の線量で与えられる、 請求項6に記載のデバイス。 8.前記イオン化放射線が約2.0〜約3.5メガラドの範囲内の線量で与えられる、 請求項7に記載のデバイス。 9.前記イオン化放射線が電子線である、請求項1または3に記載のデバイス。 10.前記イオン化放射線が約0.5〜約4.0メガラドの範囲内の線量で与えられる、 請求項9に記載のデバイス。 11.前記イオン化放射線が約2.0〜約3.5メガラドの範囲内の線量で与えられる、 請求項10の記載のデバイス。 12.前記不溶性キャリア材料が多孔性粒子を含む、請求項1または3に記載のデバ イス。 13.前記不溶性キャリア材料が充填粒子を含む、請求項1または3に記載のデバイ ス。 14.前記不溶性キャリア材料が約70〜約850μmの範囲内の粒子サイズを有する粒 子を含む、請求項1または3に記載のデバイス。 15.前記不溶性キャリア材料が約125〜約450μmの範囲内の粒子サイズを有する 粒子を含む、請求項14に記載のデバイス。 16.前記不溶性キャリア材料がインビボ生分解性ポリマーを含む、請求項1また は3に記載のデバイス。 17.前記不溶性キャリア材料が合成ポリマーを含む、請求項3に記載のデバイス 。 18.前記不溶性キャリア材料がポリ乳酸、ポリ酪酸、ポリグリコール酸、または その混合物を含む、請求項17に記載のデバイス。 19.前記不溶性キャリア材料が天然ポリマー材料を含む、請求項3に記載のデバ イス。 20.前記天然ポリマー材料がヒドロキシアパタイト、リン酸三カルシウム、コラ ーゲンまたはその混合物を含む、請求項19に記載のデバイス。 21.前記天然ポリマー材料が同種異系または異種骨である、請求項19に記載のデ バイス。 22.前記骨原性タンパク質がOP-1、OP-2、BMP-2、BMP-3、BMP-4、BMP-5、BMP-6 、BMP-8、BMP-9、DPP,Vg1、Vgr-1またはそれらの機能的等価物である、請求項1 または3に記載のデバイス。 23.前記骨原性タンパク質がホモダイマーである、請求項22に記載のデバイス。 24.前記骨原性タンパク質がOP-1またはその機能的等価物である、請求項1また は3に記載のデバイス。 25.前記骨原性タンパク質がホモダイマーである、請求項24に記載のデバイス。 26.前記最終滅菌骨原性デバイスが哺乳動物の無血管位での関節軟骨形成の誘導 能を有する、請求項1または3に記載のデバイス。 27.前記骨原性タンパク質がOP-1、OP-2、BMP-2、BMP-3、BMP-4、BMP-5、BMP-6 、BMP-8、BMP-9、DPP、Vg1、Vgr-1またはそれらの機能等価物である、請求項26 に記載のデバイス。 28.前記骨原性タンパク質がOP-1またはその機能的等価物である、請求項26に記 載のデバイス。 29.哺乳動物のあらかじめ選定した位置での軟骨性骨または関節軟骨の形成の誘 導に使用するための、請求項1または3に記載の最終滅菌骨原性デバイス。 30.前記関節軟骨形成が哺乳動物の無血管位で行われる、請求項29に記載の使用 。 31.前記骨原性タンパク質がOP-1またはその機能的等価物である、請求項29に記 載の使用。 32.哺乳動物のあらかじめ選定した位置に軟骨性骨または関節軟骨の形成を誘導 するための医薬の製造に使用するための、請求項1または3に記載の最終滅菌骨原 性デバイス。 33.前記関節性軟骨形成が哺乳動物の無血管位で行われる、請求項32に記載の使 用。 34.前記骨原性タンパク質がOP-1またはその機能的等価物である、請求項32に記 載の使用。 35.哺乳動物に移植するための最終滅菌骨原性デバイスの製造法であって; (a)不溶性の合成ポリマーキャリア材料と、該キャリア材料中に配置されて 哺乳動物中に移植された場合、軟骨性骨形成または関節軟骨形成の誘導能を有す る単離された生物活性骨原性タンパク質とを組み合わせて含む骨原性デバイスを 提供する工程;および (b)工程(a)の骨原性デバイスを、デバイスを滅菌するに十分な量のイオン 化放射線に曝露し、それにより哺乳動物における軟骨性骨形成または関節軟骨形 成の誘導能を有する最終滅菌骨原性デバイスを製造する工程 を含む、方法。 36.哺乳動物に移植するための最終滅菌骨原性デバイスの製造法であって; (a)不溶性のポリマーキャリア材料と、該キャリア材料中に配置されて哺乳 動物中に移植された場合、軟骨性骨形成または関節軟骨形成の誘導能を有する単 離された生物活性骨原性タンパク質とを組み合わせて含む骨原性デバイスを提供 する工程;および (b)工程(a)のデバイスを、デバイスを滅菌するに十分な量のイオン化放射 線に曝露し、それにより哺乳動物における軟骨性骨形成または関節軟骨形成の誘 導能を有する最終滅菌骨原性デバイスを製造し、ここで該最終滅菌骨原性デバイ スは、滅菌前の骨原性デバイスの少なくとも約20%の生物活性を有する、工程 を含む、方法。 37.前記骨原性タンパク質が、OP-1、OP-2、BMP-2、BMP-3、BMP-4、BMP-5、BMP- 6、BMP-8、BMP-9、DPP、Vg1、Vgr-1またはそれらの機能的等価物である、請求項 35または36に記載の方法。 38.前記骨原性タンパク質がOP-1またはその機能的等価物である、請求項35また は36に記載の方法。
