BACKGROUND ART Edible plants such as vegetables, mushrooms and seaweed are
It is well known that living organisms are important as a source of functional components such as minerals and dietary fiber, as well as vitamins, and are indispensable ingredients for growth and growth.
In addition, in recent years, these edible plants have attracted attention for their ability to scavenge active oxygen, which is closely related to maintaining and improving health, preventing aging, preventing adult diseases, and suppressing carcinogenesis. Was. Aging of the living body and various diseases associated with this aging, for example, cancer, arteriosclerosis, liver cirrhosis, myocardial infarction, stroke, cataract, Parkinson's disease, rheumatism, Alzheimer's dementia, etc. There are many points. However, in recent years, as an inducing mechanism of these intractable diseases, active oxygen such as superoxide, which is rich in reactivity with oxygen having unpaired electrons, and a resulting hydroxy radical and hydrogen peroxide, is a target molecule in cells,
For example, it has been considered that membrane lipids, proteins, DNA, and the like are oxidized to cause oxygen damage, resulting in aging of living bodies and various diseases associated with aging.

1.6) and antioxidants such as L-ascorbic acid and α-tropherol, and the concentration of active oxygen is usually kept extremely low. However, when exposed to a large amount of ultraviolet rays, radiation, electromagnetic waves, etc., exercises intensely, receive mentally intense stress, and in addition to these, as aging progresses, it increases the amount of active oxygen generated It is thought that the oxygen scavenging ability cannot keep up, and as a result, compounds oxidized by excessive active oxygen accumulate, causing the above-mentioned oxygen damage. In order to improve this, for example, Japanese Patent Application Laid-Open
No. 68435, as disclosed in JP-A-8-143466, etc., utilizing the high active oxygen scavenging ability contained in edible plants, supplementing the active oxygen scavenging ability that is apt to be lacking in living organisms, There have been proposals to maintain and improve health. However, even when using these edible plants having relatively high active oxygen scavenging ability, they may be subjected to processing such as squeezing, extraction, heating, drying, etc. It has been found that there is a disadvantage that the original active oxygen scavenging ability possessed is significantly reduced or, in severe cases, completely eliminated. In order to solve this drawback, it is strongly desired to establish a completely new method for suppressing the reduction of the active oxygen scavenging ability. And for the living body,
It is desirable to establish a method that does not raise concerns about toxicity.

The trehalose used in the present invention is α, α
-Trehalose is preferred, regardless of its origin and properties as long as it can suppress the reduction of the active oxygen scavenging ability. The origin of trehalose is described in, for example, JP-A-7-246.
No. 097, Trehalose from yeast, JP-A-58-21669, JP-A-7-17097
Trehalose from maltose described in Japanese Patent Application Laid-Open No. 7-143876 and Japanese Patent Application Laid-Open No.
Various trehalose-containing saccharides such as trehalose from starch described in JP-A-213283 and the like can be appropriately employed.
In addition, as its properties, syrup, honey-containing crystal powder, hydrated crystal, anhydrous crystal, amorphous solid and the like can be appropriately adopted. Of course, it is also advantageous to use commercially available high-purity trehalose hydrous crystals, high-purity trehalose anhydrous crystals, or the like.
The content of trehalose in the carbohydrate used is not limited as long as it can be contained in a vegetable edible food or the like so as to exert the effect of suppressing the reduction of the active oxygen scavenging ability of the vegetable edible food or the like. It is preferably at least 10 w / w% (hereinafter, w / w% is simply indicated by% unless otherwise specified), preferably at least about 20%, more preferably at least about 50%. In addition, the edible food and / or the plant antioxidant-containing composition of the present invention, as trehalose, is usually about 1% or more, preferably about 5%, in terms of anhydride, as trehalose.
%, More preferably about 20% or more. In general, if trehalose is less than about 1% in terms of anhydride, the effect of suppressing the reduction of the active oxygen scavenging ability is insufficient, and if it is about 30% or more, the resulting composition tends to be sweet.

% Can be advantageously implemented. At this time, trehalose present in the plant tissue may be contained in an amorphous state or in a hydrated crystalline state, and it can be advantageously carried out in any state. Further, it is possible to advantageously carry out a sterilization and / or drying step together with the step of containing trehalose in an aqueous medium. The solid composition thus obtained is not only excellent in suppressing the reduction of the active oxygen scavenging ability, but also has low hygroscopicity, is stable in an indoor environment, and is easy to handle. In addition, the solid composition is suitable as a solid food in which the plant tissue maintains its shape well and is firm by trehalose, and can enjoy a chewy feel.

In the present invention, the term "contain" means to coexist. When the trehalose of the present invention is contained in a substance having a plant-derived active oxygen-eliminating ability, the plant-derived active oxygen-eliminating ability is thereby reduced. It is sufficient that the effect of suppressing the reduction can be exerted, and there is no particular limitation on the method of incorporation and the relative ratio between the two. Desirably, trehalose is contained in the vegetable edible food and / or the plant antioxidant by bringing the trehalose into contact with the aqueous medium as uniformly as possible. More specifically, for example, when the vegetable edible food or the like is in a multi-juice state such as a liquid or a suspension, a solid trehalose such as a powder or a crystal is mixed and dissolved as uniformly as possible. The syrup-shaped trehalose may be mixed and contained as uniformly as possible. Further, when the vegetable edible food or the like is in a solid state, it is converted into a liquid or a suspension like water in a multiple juice state with water and then treated as described above for inclusion, or syrup. Trehalose in the form of a solid, and disperse, dissolve or suspend a solid plant edible substance or the like into the trehalose, and make the edible substance and / or the plant antioxidant contain trehalose by contacting as uniformly as possible. I just need. Also, when the plant edible food is such as a raw plant tissue cut, shredded or sliced, sprinkle with solid trehalose and mix to dissolve or contain the trehalose solution. Trehalose may be contained as uniformly as possible in the tissue of the plant by immersion or the like. Conditions such as temperature and time for bringing trehalose into contact with a plant edible food or the like can be appropriately selected as necessary.

More specifically, the method for producing a composition of the present invention in which a reduction in the ability to scavenge active oxygen is suppressed is as follows. For example, an edible portion of an edible plant is heat-sterilized, if necessary, and then mixed. , Crushed or cut with a juicer, shredder, cutting machine, slicer, etc. to make a paste or suspension or cut, into which trehalose is mixed and dissolved and contained, or edible plant edible The part may be extracted with a solvent such as hot water or alcohol, filtered, and, if necessary, concentrated to a liquid or paste-like substance, and trehalose may be mixed and dissolved therein to contain it. The composition thus obtained is usually a syrup-like or paste-like composition or a cut-containing multi-juice composition, having a high water content,
The disadvantage is that cold storage is required to maintain stability.
These syrup-to-paste compositions or cut material-containing multi-juice compositions are dried or dried, powdered as they are, or after heat sterilization, dried or dried, powdered, etc. A solid composition in the form of a strip or a strip is obtained. Known methods can be appropriately used for the drying and powdering methods. For example, after vacuum drying, ventilation drying, drum drying,
After crushing, sieving, spray drying, fluid drying, sieving, or further mixing anhydrous saccharides such as anhydrous maltose and anhydrous trehalose, dehydrating and drying, and then sieving. It can be advantageously used in the form of a powder product, which may be used in an appropriate form such as granules, tablets, capsules, etc., if necessary. By increasing the degree of processing to such a solid composition, not only the effect of suppressing the reduction of the active oxygen scavenging ability is further enhanced, but also the storage stability is improved and the handling becomes easy. In addition, the solid plant edible food-containing composition thus obtained contains trehalose in an amorphous state or in a hydrous crystalline state, and the edible plant as a raw material originally has, for example, It is characterized by suppressing deterioration of pigments such as chlorophyll, carotene, anthocyanins, flavonoids, etc., and keeping the original color tone well. In addition, there is a feature of masking unpleasant tastes such as bitterness and astringency.

[Experiment 1] <Effects of various carbohydrates on suppression of reduction of active oxygen scavenging ability> Raw ginseng was crushed with a mixer, and various saccharides were converted to anhydrous carbohydrate weight based on this crushed product.
%, And dried under reduced pressure at 40 ° C. for 2 days, followed by vacuum drying at the same temperature for 24 hours, and pulverized with a mixer to prepare a powdered ginseng-containing composition. The carbohydrates used were glucose, mannitol, sorbitol, maltose, sucrose,
Commercial reagents were used for both trehalose and pullulan. About 100 g of the prepared powdery composition was added to 500 ml
The storage capacity at 40 ° C. for 7 days was sealed in a polystyrene container, and the ability to scavenge active oxygen was compared with that at the start of storage. According to the nitro blue tetrazolium (NBT) method [Toshio Imanari et al., History of Medicine, 101, 496-497 (1977)], the amount of superoxide generated in a xanthine-xanthine oxidase system was measured by measuring the reactive oxygen scavenging ability. The amount of formazan produced by reduction of NBT was measured by colorimetry. Purified water was used as a test liquid as a control. The activity that inhibits the amount of formazan produced by 50% as compared with the control was defined as one unit of the activity of scavenging active oxygen. Table 1 summarizes the composition of the prepared powdery composition and the ability to scavenge active oxygen per gram of the composition before and after the start of storage. As is clear from Table 1, among the carbohydrates tested, those using trehalose have the highest residual rate of active oxygen scavenging ability, and it is found that trehalose is remarkably effective in suppressing reduction of active oxygen scavenging ability. did. The effect of trehalose on inhibiting the ability to scavenge active oxygen was significantly stronger than glucose, a constituent sugar of trehalose, or maltose, a disaccharide composed of two glucose units similar to trehalose. Therefore, trehalose is suitable as an active ingredient of the active oxygen scavenging ability reduction inhibitor. In addition, pullulan, after trehalose, also had a high effect of suppressing the reduction of active oxygen scavenging ability.

[Experiment 4] <Effect of trehalose on reducing the ability to scavenge active oxygen for superoxide dismutase and various antioxidants> Using a 45% trehalose aqueous solution, per 1 ml of this was added horseradish-derived superoxide dismutase (Sigma). 5 μg, α-glucosyl hesperidin (manufactured by Hayashibara Biochemical Laboratory) 10 mg, α-glucosylrutin (manufactured by Hayashibara Biochemical Laboratory) 10 mg, gallic acid (manufactured by Wako Pure Chemical Industries) 250 μg and 50 μg of catechin (manufactured by Wako Pure Chemical Industries)
Were respectively dissolved to obtain liquid compositions, each of which was placed in a glass test tube, sealed with a rubber stopper, and stored under light shielding. As a control, water was used instead of the trehalose aqueous solution. Table 4 shows their storage conditions and the ability to scavenge active oxygen before and after storage. As shown in Table 4, in addition to superoxide dismutase, α-glucosyl hesperidin, α
-Regarding the active oxygen scavenging ability of antioxidants derived from plants such as glucosyl rutin, gallic acid and catechin, it was also recognized that trehalose exerts a remarkable reduction suppressing effect. Separately, the trehalose hydrated crystal powder and each antioxidant substance powder were separately prepared as mixed powders, and they were similarly subjected to a storage test. However, no change was observed in any of the active oxygen scavenging ability. From this, it has been found that trehalose is hard to exhibit its effect only by mixing it in a powder form, and it is found that the effect is exhibited by containing trehalose in contact with the trehalose in a dissolved state, in other words, by incorporating it in an aqueous medium.

[Experiment 5] <Effect of reducing active oxygen scavenging ability on herbal extract> 10 l of water was added to 500 g of dried sardine (car foreground grass, Pharmacopoeia, manufactured by Daido Pharmaceutical Co., Ltd.), and the mixture was boiled and boiled for 90 minutes. The decoction was collected with a basket-type centrifuge. The decoction was further boiled down to obtain 2 kg of a liquid having a concentration of about 5%. This boiled liquid 1
Trehalose and pullulan were converted to anhydrous carbohydrate weight, and 240 g and 10 g, respectively, were mixed and dissolved, and then spray-dried with a desktop spray dryer (SD-1, manufactured by Tokyo Rika) to obtain a powder. A composition containing the leafy tobacco extract was prepared. As a control, a similar powdery composition was prepared using dextrin (DE about 11) instead of trehalose and pullulan. These powdery compositions were sealed in a polystyrene container and stored at 40 ° C. for 7 days in the same manner as in Experiment 1, and the effect of suppressing the reduction of the active oxygen scavenging ability was examined. At the start of storage, both were about 5000 units per gram, and after storage for 7 days, the composition using trehalose and pullulan was about 4300 units per gram, whereas the composition using dextrin was Thing is 1g
About 3100 units per unit. Therefore, the use of trehalose and pullulan is clearly superior to the effect of dextrin in suppressing the reduction of active oxygen scavenging ability.
