RNAを含む凍結乾燥組成物
本発明は、リボ核酸(RNA)を含む凍結乾燥組成物及び該組成物の製造法に
関する。本発明はさらに、核酸の検出法における該組成物の使用及び該組成物を
含む核酸の検出用テストキットに関する。
RNAはかなり過敏であり且つ比較的不安定な物質であることが知られている
。溶液中のRNAは多かれ少なかれ急速に劣化しやすい。特にRNアーゼによる
分解はRNA調製物を取り扱う際の重要問題である。この遍在エンザイム(RN
アーゼは指先、ほこりなどの中に存在する)は熱安定性であり、その酵素活性を
得るのに補因子を必要としない。これは、RNA調製物中のRNアーゼの不活性
化を困難にする。RNアーゼを含まないRNAを得るいくつかの単離法(Sambro
ok J.,Fritch E.F.及びManiatis T.(1989)Molecular Cloning:A Laborator
y Manual,第2版,第1巻,Cold Spring Harbor Laboratory Press,Cold Spri
ng Harbor;Ausubel,F.M.ら編(1990)Current Protocols in Molecular Biol
ogy,第1巻,Green/Wiley-interscience.New York)があるが、精製又は単離
、即ちRNA
の可溶化という最終段階でRNアーゼの混入を回避するための予防処置を講じる
必要がある。一般的な方法は、ジエチルピロカーボネート(DEPC)処理水中
でRNAを可溶化することである(Chomczynski P.(1992),Solubilization
in formamide protects RNA from degradation,Nucleic Acid Research,第20
巻(14))。しかし、可溶化RNAは試料の取り扱い及び貯蔵の間に偶発的に汚
染されやすい。さらに、水に溶解したRNA試料の長期貯蔵には、分解を回避す
るために温度を-70℃まで低くする必要がある。代替案として、DEPC処理し
た水の代わりにホルムアミド中でRNAを可溶化することも可能である。ホルム
アミドは、RNAをRNアーゼによる分解から効果的に保護し、-20℃での長期
貯蔵を可能にする。しかし、RNA調製物を、輸送及び2〜8℃又はそれ以上の
温度での貯蔵を可能にする妥当な貯蔵寿命を有する実用製品にするためには、上
記方法では不十分である。
従って、容易に長期保存し得且つ取り扱い易い安定なRNA含有組成物が要望
されている。
本発明は凍結乾燥RNA含有粒子を提供する。
凍結乾燥法がRNAに有害な作用を及ぼすことなく且つ
全ての可能な不活性化プロセスを実質的に遅延させることが見いだされた。凍結
乾燥RNAにより、その構造を変化させることなく長期間保存し得る安定な製品
を得ることができる。従って、本発明の凍結乾燥RNA含有組成物は、例えば、
現在使用されているRNA含有試薬溶液の極めて有用な代替物である。本発明の
粒子は乾燥体であり、従って種々の分解プロセスに対して殆ど悲感受性である。
本発明は、多様なRNAに用いることが可能である。様々な長さのRNA配列
を凍結乾燥し得る。長さが例えば、数百〜数千の範囲で変化するヌクレオチドの
全てのRNA配列を本発明の凍結乾燥組成物に配合することが可能である。
本発明の凍結乾燥粒子は種々の粒状形態で存在し得る。驚くべきことには、該
粒子は、例えば、層状で凍結乾燥される物質より高い安定性を有し且つ極めて可
溶性である。これらの凍結乾燥粒子中のRNAは、2〜8℃から37℃までの温度
で少なくとも1年間の貯蔵の間にも安定である。RNAは球状粒子の形態で凍結
乾燥するのが好ましい。
球状凍結乾燥粒子〔アキュスフェア又はリオスフェア(accuspheres又はlyosp
heres)〕がPriceら(US3,655,83
8明細書)から公知である。これらの球状ビーズは免疫反応用の物質を含んでい
る。生物学的に活性な物質を含むリオスフェアは、例えば、USP3,932,943明細書
及び他の多くの特許明細書から公知となっている。リオスフェアの利点は、その
粒子の均一性、製品の取り扱い易さ、凍結乾燥プロセスが速いこと、凍結乾燥プ
ロセスの間の分解が少ないこと及び溶解特性が改良されていることである。
リオスフェアは、RNA水溶液の液滴を凍結乾燥することにより形成し得る。
所望サイズの液滴を得るために、該溶液を、氷浴中(例えば、DT2,140,747、EP8
1913又はUS3,928,566の各明細書に開示されている)、液体窒素中(例えばJ59 1
69,504明細書)に、又は冷却ドラム(例えばUSP3892876明細書)又は冷却プレー
ト(例えばUS4,501,719明細書)上に噴霧し得る。種々の他の方法も当業におい
て周知である。
本発明の凍結乾燥粒子は、RNAの他に、安定剤、例えばスクロース、マンニ
トール、デキストラン、ソルビトール及びグリセロールのようなポリヒドロキシ
化合物を含んでいてよい。
本発明の凍結乾燥粒子は、核酸検出法における診断試薬
として特に有用である。核酸を検出するための診断法は通常、臨床試料から単離
した核酸を増幅することを含む。本発明の凍結乾燥粒子の場合に用い得る増幅技
術のうちの一つは、NASBA、即ち、欧州特許出願EP0,329,822に開示されて
いる「核酸配列ベースの増幅(NASBA)」である。この技術を用いて、大量
のRNAを合成し得る。
標的配列の増幅を含む、核酸配列を検出するための診断法において、試料に内
部対照配列(internal control sequence)を添加す場合がある。そのような方
法が、同時係属で且つ本出願人所有の出願であるPCT/EP93/02284明細書に記載さ
れている。該明細書に記載の方法に用いられている内部対照配列は、標的配列か
ら区別し得る核酸配列を含み、該核酸配列は標的配列と同一の増幅試薬を用いて
増幅し得る。
これらの「内部対照配列」は本発明の凍結乾燥粒子に配合し得、PCT/EP93/022
84明細書に記載の方法に用いるのに好適である。
試料中の核酸の定量法にも凍結乾燥粒子を用いることができる。
標的核酸に対応する既知数の核酸配列分子を未知数の標
的核酸配列を含む試料に添加することを用いる核酸の定量法が、EP525882の下に
公開された同時係属で且つ本出願人所有の欧州特許出願明細書に記載されている
。該明細書に記載の方法は、既知量の明確に規定された突然変異体配列を添加し
た未知濃度の野生型標的核酸を含む試料から核酸を増幅するという原理に基づい
ている。増幅は、標的及び突然変異体配列にハイブリダイズし得る1つのプライ
マーセットを用いて行われる。EP525882に記載の競合増幅は、固定量の試料と段
階希釈した突然変異体配列、又は段階希釈した試料と固定量の突然変異体配列を
用いて行うことができる。この場合も、上記定量法の実施に必要とされるプライ
マー及びプローブを含み得る凍結乾燥粒子に突然変異体配列を配合物してもよい
。それぞれが被検体である核酸から区別可能であり且つ被検体である核酸と共増
幅し得る、それぞれ異なる量の種々の核酸構築物からなる試料に添加することを
含む、試料中の被検体である核酸の定量法が、同時係属で且つ本出願人所有のPC
T/EP94/02295明細書に記載されている。用いられた全てのRNA配列を、それぞ
れの量で本発明の凍結乾燥粒子に配合し得ることは明らかである。
本発明の凍結乾燥粒子は球体の形態で形成するのが好ましい。本発明の球状粒
子は、1種以上の安定剤を含むRNA溶液を調製する段階、該溶液から好ましく
は該溶液を液体窒素中に滴下することにより凍結乾燥粒子を形成する段階、液滴
をそれらが完全に凍結するまで液体窒素中に保持する段階、及び該液滴を凍結乾
燥する段階からなる方法により形成するのが好ましい。
本発明の凍結乾燥粒子は、適当なエンザイム、プライマー及びプローブのよう
な使用する検出法に用いられる他の試薬と共に、核酸の検出又は定量用テストキ
ットに組み込むことが可能である。
実施例実施例1:RNA含有凍結乾燥粒子の形成
A.用いられた全ての化学物質は試薬等級1の品質を有するものであった。水中
の15w/w%のサッカロース、5w/w%のマンニトール及び5w/w%のデキストラン
T40を用いて安定剤混合物を調製した(該サッカロースはSigma社から、マン
ニトールはBaker社から、デキストランT40はFarmacia社から入手した)。別
に、濃縮RNAストックをDEPC処理水で100倍希釈してRNA溶液を調製し
た。5
0mlの安定剤溶液を50mlのRNA溶液と混合した。リオスフェアは以下の方法で
RNA/安定剤溶液から形成した:
50μlのRNA/安定剤混合物を沸騰窒素中に滴下した。球体をそれらが完全
に凍結するまで液体窒素中に保持した。凍結球体を凍結乾燥して、残留含水量が
2w/w%以下であり且つ凍結球体と実質的に同一の粒径を有する球体を得た。こ
のようにして形成されたアキュスフェアは1.22×107個のRNA分子を含んでい
た。
B.あるいは、RNA含有凍結乾燥粒子を以下の方法で形成することもできる:
アキュスフェア1個につき各種のRNAからなる1012個の分子を含む600個の
アキュスフェアを形成するためにRNA溶液混合物を調製した。該溶液は、それ
ぞれが9mlの3種の異なるRNA(合計27ml)からなるものであった。該RNA
溶液は1.5Mの塩化ナトリウム(該塩化ナトリウムはJanssen Chimicaから入手し
た)を含んでいた。A.に用いたものと同じ安定剤を乾燥成分としてRNA混合
物に加えた。RNA混合物を含む管に、2.25gのサッカロース、0.75gのマンニト
ール及び0.75gのデキストランT40を加えた。RNA/安定剤混合物に、最終
容量が30mlになるまで
水を加えた。
A.に記載のようにして凍結乾燥した。
C.同様にRNAを含む凍結乾燥粒子も以下の方法で形成し得る:
アキュスフェア1個につき7.6×104個のRNA分子を含む600個のアキュスフ
ェアを形成するためにRNA溶液を調製した。3.0gのスクロース、1.0gのマンニ
トール及び1.0gのデキストランT40を40.0mlの水に溶解した。30mlのスクロー
ス/マンニトール/デキストランT40含有溶液に、1μl当たり106個のRNA
分子を含むRNAストック溶液45μlを加えた。
A.に記載のようにして凍結乾燥した。実施例2:RNA含有凍結乾燥粒子の安定性
凍結乾燥球体としてのRNAの安定性を、本発明による凍結乾燥球体及び独立
の基準物質に由来するRNAを用いるNASBA反応の増幅効率を比較すること
によって試験した。凍結乾燥球体を、実施例1の“A”項に述べたようにして製
造した。基準物質の野生型(WT)は、電子顕微
粒子/mlと定量された、in vitro生成させたH
IV−1細胞系である(Iayne等,1992)。この基準物質を溶解緩衝液
で稀釈し、7,300RNA分子/μlを含有する単一使用100μlアリコー
トとして−70℃で貯蔵した。
この安定性試験において試験した凍結乾燥球体は、2〜8℃、20〜25℃及
び35〜38℃で貯蔵した四つの異なるバッチのRNA球体であった。
安定性の測定には次のプロトコルを用いた。
稀釈した基準物質の10μlアリコートを900μlの溶解緩衝液に添加し、
混合した。乾燥したRNA球体を550μlの溶離緩衝液(pH8.5の1mm
ol/l トリス/HCl)で稀釈し、3種のRNA即ちQa、Qb及びQcの
濃度をそれぞれ溶離緩衝液50μl当たり1,000,000、100,000
及び10,000RNA分子とした。Qa、Qb及びQcは、約1,000〜1
,500ヌクレオチドの長さを有するin vitro合成RNA配列である。
溶解RNA球体の50μlアリコートを溶解緩衝液管に添加し、混合した。Bo
om等が述べている単離操作(R.Boom,Sol CJA,J.van d

495−503,1990)を用いてRNA試験対象(calibrators
)及びWT基準物質を単離した。Kievits等が述べている方法(T.Ki

増幅物(WT、Qa、Qb及びQc)を分離するべく増幅試料のアリコートを
、各々前記増幅物のうちの一つに特異的である4種のハイブリダイゼーション液
に添加した。この例では各増幅物をビーズーオリゴ(即ち固相として機能するス
トレプトアビジン被覆磁気ビーズに結合させたビ
オチン−オリゴ)、及びルテニウムで標識したプローブとハイブリダイズさせた
。
ハイブリダイズした増幅物/プローブ複合体を保持する磁気ビーズを磁石によ
って電極の表面に捕捉した。前記電極に印加される電圧が電気化学ルミネセンス
(ECL)反応を惹起する。ハイブリダイズしたルテニウム標識プローブによっ
て発せられる光は増幅物の量に比例する。4種の増幅物の相対量に基づく計算に
よって、WT基準物質の量が明らかとなる。
WT基準RNAの量を上述のプロトコルに従って決定した。結果を表1に示す
。
RNA球体を14週間以下の期間にわたりいずれの試験温度で貯蔵しても、定
量的NASBAアッセイにおける通常の変動以外の甚だしいRNA減少は観察さ
れない(表1)。
このような結果は、バッチ毎の、及び毎日の変動に関わりなく、様々な経過時
間及び温度において計算したWT基準物質量に球体中のQa、Qb及びQc R
NAの量の減少を示唆する甚だしい増大は起こらないことを示している。
実施例3:凍結乾燥粒子の安定性(2)
実施例1のプロトコルCに従って製造した、約1,500ヌクレオチドのin vitro
合成RNA配列を含有するアキュスフィア(accusphere
s)を200μlの溶離緩衝液(pH8.5の10mMトリス/HCl)に溶解
させた。得られた溶液20μlを9mlの溶解緩衝
液(5Mチオシアン酸グアニジン、1%(v/v) Triton X−100
、20mM EDTA、H6.4の50mMトリス/HCl)に添加した。その
後、70μlの活性シリカ(0.1N HCl中の1mg/mlサイズ選択懸濁
液)を溶解混合物に添加して核酸と結合させた。シリカを洗浄及び乾燥後、核酸
を100μlの溶離緩衝液中に溶出させた。
核酸増幅はKievits等が述べているようにして実施するが、その際次の
ような改良を行なう。
反応混合物の最終量は25μlとし、その際pH8.5の40mMトリス/H
Cl、12mM MgCl2、40mM KCl、5mM DTT、1mMの各
dNTP、2mMの各NTP、15%(v/v)DMSO、0.1μg/μl
BSA、0.2μlの各オリゴヌクレオチドプライマー、8.0u AMV−R
T、0.1u RNアーゼH、及び40.0u T7 RNAポリメラーゼを含
有させる。一つのプライマーはT7 RNAポリメラーゼ認識部位を有する。各
増幅反応のために8μlの単離RNAを用いた(約200分子のRNAを含む)
。増幅反応は遠心用マイクロ試験管内で、41℃で90分間行なう。増幅
生成物を酵素結合ゲルアッセイ(ELGA)によって分析する。増幅したRNA
を、配列特異的HRP 5′−標識オリゴヌクレオチドプローブを用いる溶液中
非放射性ハイブリダイゼーションによって検出する。ハイブリダイゼーション後
、ハイブリダイズしなかったプローブを相同ハイブリダイゼーション生成物から
ゲル電気泳動によって分離した。未結合HRPプローブ及びハイブリダイゼーシ
ョン生成物はポリアクリルアミドゲル中で、ゲルをHRP基質溶液中でインキュ
ベートすることにより直接可視化した。
上述の操作を、4℃、20〜25℃及び35〜38℃で貯蔵した三つの異なる
凍結乾燥RNA調製物に適用した。
上記ELGA検出を用いたところ、8ヵ月(バッチ3)及び1年(バッチ1及
び2)の貯蔵後にシグナル間の相違は観察されなかった。この結果を、−70℃
で貯蔵した(実施例2に述べた)基準RNAを用いて確認した。基準RNAと凍
結乾燥RNAとの同時単離、同時増幅及び検出後、基準RNA及び凍結乾燥RN
AのECL比間に甚だしい相違は見出されず、このことは本発明によるRNA含
有アキュスフィアが4℃、20〜25℃及び35〜38℃で貯蔵された場合に安
定であることを示している。実施例4:ゲルマーカーRNA含有凍結乾燥粒子の安定性
ゲルマーカーRNAを含有する、実施例1のプロトコルBに従って製造したア
キュスフィアを25μlのRNアーゼ、DNアーゼ及びプロテアーゼを含有しな
い水に溶解させた。得られた溶液に、25μlの西洋ワサビペルオキシダーゼ(
HRP)5′−標識オリゴヌクレオチドプローブ溶液を添加する。配列特異的な
前記オリゴヌクレオチドとゲルマーカーRNAとのハイブリダイゼーションを4
1℃で15分間生起させる。ハイブリダイゼーション後、ハイブリダイズしなか
ったプローブを相同ハイブリダイゼーション生成物からゲル電気泳動によって分
離した。未結合HRPプローブ及びハイブリダイゼーション生成物はポリアクリ
ルアミドゲル中で、ゲルをHRP基質溶液(テトラメチルベンジジン;TMB)
中でインキュベートすることにより直接可視化した。
三つの異なるゲルマーカーRNAアキュスフィアの各ロットを、長期安定性試
験において先に述べたようにして分析した。最初の二つのバッチは、ゲルマーカ
ーRNAアキュスフィアを4℃で52週間貯蔵してもRNA分解の兆候を
示さない。第三のバッチを36週間後に試験したが、やはり予測どおりの、分解
を伴わないゲルパターンを示した。RNAの分解が起こっていれば、ゲル中に見
出された個別バンドに替わってRNA汚点(smears)が現われることによ
り示唆されたはずである。TECHNICAL FIELD The present invention relates to a lyophilized composition containing ribonucleic acid (RNA) and a method for producing the composition. The invention further relates to the use of the composition in a method for detecting nucleic acid and a test kit for detecting nucleic acid containing the composition. RNA is known to be a fairly hypersensitive and relatively unstable substance. RNA in solution is more or less prone to rapid degradation. In particular, degradation by RNase is an important issue when handling RNA preparations. This ubiquitous enzyme (RNase is present in fingertips, dust, etc.) is thermostable and does not require cofactors to obtain its enzymatic activity. (1990) Current Protocols in Molecular Biology, Volume 1, Green / Wiley-interscience. New York). Preventive measures must be taken to avoid RNase contamination at the final step of RNA solubilization. A common method is to solubilize RNA in water treated with diethylpyrocarbonate (DEPC) (Chomczynski P. (1992), Solubilization in formamide protects RNA from degradation, Nucleic Acid Research, Volume 20 (14)). . However, solubilized RNA is subject to accidental contamination during sample handling and storage. Furthermore, long-term storage of RNA samples dissolved in water requires lowering the temperature to -70 ° C to avoid degradation. Alternatively, RNA can be solubilized in formamide instead of DEPC treated water. Formamide effectively protects RNA from degradation by RNase and allows long-term storage at -20 ° C. However, the above method is not sufficient for making RNA preparations into practical products with reasonable shelf life allowing transport and storage at temperatures of 2-8 ° C. or higher. Therefore, there is a need for a stable RNA-containing composition that can be easily stored for a long period of time and is easy to handle. The present invention provides freeze-dried RNA-containing particles. It has been found that the lyophilization method does not have a deleterious effect on RNA and substantially delays all possible inactivation processes. Lyophilized RNA makes it possible to obtain a stable product that can be stored for a long period of time without changing its structure. Therefore, the freeze-dried RNA-containing composition of the present invention is, for example, a very useful alternative to the RNA-containing reagent solution currently used. The particles of the present invention are dry and therefore almost immune to various degradation processes. The present invention can be used for various RNAs. RNA sequences of various lengths can be lyophilized. It is possible to incorporate into the lyophilized composition of the present invention all RNA sequences of nucleotides which vary in length, for example from hundreds to thousands. The lyophilized particles of the present invention can exist in various granular forms. Surprisingly, the particles are, for example, more stable than layers of lyophilized material and are very soluble. The RNA in these lyophilized particles is stable during storage for at least one year at temperatures from 2-8 ° C to 37 ° C. RNA is preferably lyophilized in the form of spherical particles. Spherical lyophilized particles (accuspheres or lyosp heres) are known from Price et al. (US 3,655,838). These spherical beads contain substances for immune reaction. The freeze-dried particles of the present invention are particularly useful as a diagnostic reagent in nucleic acid detection methods. Diagnostic methods for detecting nucleic acids typically involve amplifying nucleic acids isolated from clinical samples. One of the amplification techniques that can be used in the case of the lyophilized particles of the present invention is NASBA, ie “Nucleic Acid Sequence Based Amplification (NASBA)” disclosed in European Patent Application EP 0,329,822. In diagnostic methods for detecting nucleic acid sequences, including amplification of target sequences, an internal control sequence may be added to the sample. The internal control sequences used in the methods described herein include nucleic acid sequences that are distinguishable from the target sequence, which nucleic acid sequence can be amplified using the same amplification reagents as the target sequence. These "internal control sequences" may be incorporated into the lyophilized particles of the present invention and are suitable for use in the method described in PCT / EP93 / 02284. Lyophilized particles can also be used in the method for quantifying nucleic acids in a sample. A method for quantifying nucleic acids using the addition of a known number of nucleic acid sequence molecules corresponding to a target nucleic acid to a sample containing an unknown number of target nucleic acid sequences is disclosed in EP525882, a co-pending and applicant-owned European patent. The method described therein is based on the principle of amplifying nucleic acid from a sample containing an unknown concentration of wild-type target nucleic acid spiked with a known amount of a well-defined mutant sequence. Amplification is performed with one primer set that is capable of hybridizing to the target and mutant sequences. The competitive amplification described in EP525882 can be performed with a fixed amount of sample and serially diluted mutant sequence, or with serially diluted sample and fixed amount of mutant sequence. Again, the mutant sequence may be incorporated into lyophilized particles which may contain the primers and probes required to carry out the above quantitation method. Nucleic acid that is the analyte in the sample, including addition to samples that are different from each other and that can be co-amplified with the nucleic acid that is the analyte, and that each consist of different amounts of different nucleic acid constructs Is described in co-pending and applicant's owned PC T / EP 94/02295 specification. It is clear that all the RNA sequences used can be incorporated into the lyophilized particles according to the invention in respective amounts. The freeze-dried particles of the present invention are preferably formed in the form of spheres.
