外来性ポリペプチドの発現のためのベクターとしてのメンゴウイルス
関連出願との前後参照
本出願は、1993年6月3日に出願された米国出願番号第08/090,5
31号の部分継続出願である。この出願の開示全体が、依拠され、参照によりこ
こに明白に包含される。
発明の背景
本発明は、免疫学的及び非免疫学的目的のための1つ以上の外来性ポリペプチ
ドをコードする核酸を含有するよう改変されたメンゴウイルス(mengoviruses)
に関する。本発明の改変メンゴウイルスは、組換えウイルス及び/又はキメラウ
イルスであることができる。
メンゴウイルスは、カルディオウイルス(cardiovirus)属に属するピコルナ
ウイルスの一種である。メンゴウイルスの自然宿主はマウスであり、感染により
急性マウス髄膜脳炎をもたらすが、メンゴウイルスは広い宿主範囲を有する。マ
ウスに加えて、メンゴウイルスは、ブタ、ゾウを含む種々の動物種及びヒトを含
む霊長類に感染することもできる。
ピコルナウイルスは、病原性ウイルスの科である。ピコルナウイルスの例とし
ては、一般的な風邪の原因となるライノウイルス、手足口病(foot and mouse d
isease)ウイルス(FMDV)、コクサッキーウイルス、A型肝炎ウイルス、及
びメンゴウイルス及び脳心筋炎ウイルスを含むマウスのカルディオウイルスが挙
げられ
る。
ピコルナウイルスは、小さい正鎖(+)センスRNAゲノムを含むエンベロー
プを欠いたキャプシドを有する。全てのピコルナウイルスのキャプシドは、5:
3:2の正20面体対称を有する60サブユニットのタンパク質の殻から構成さ
れ、各サブユニットは4つの同一でないポリペプチド鎖(VP1、VP2、VP
3及びVP4)を含んでいる。この殻は、単一コピーの正鎖センスRNAゲノム
を包んでいる。メンゴウイルスの三次元構造は、X線結晶解析により原子の解像
度で決定されている。Luo et al.,Science 235:182-191(1987)。
メンゴウイルスのウイルスゲノムは、長さ約7,800ヌクレオチドの正鎖R
NA分子である。ゲノムは、その3′末端でポリアデニル化されており、その5
′末端で共有結合により小さいウイルスポリペプチドVPgに連結している。メ
ンゴウイルスゲノムは、相補的DNA(cDNA)分子の形態でクローニングさ
れている。ゲノムは、ウイルスのポリタンパク質をコードする単一のオープンリ
ーディングフレームを含む。
ウイルスタンパク質は、ポリタンパク質のN末端からC末端へ、L−P1−P
2−P3の順でポリタンパク質内に位置している。ポリタンパク質は、一連の切
断事象によりプロセシングされ、全ての構造及び非構造タンパク質を与える。こ
のプロセシングの詳細は、Ann C.Palmenbergにより、「ピコルナウイルスポリ
タンパク質のタンパク分解的プロセシング(Proteolytic Processing ofPicorna viral Polyprotein
)」、44 Ann.Rev.Microbiol.603
(1990)に論評されており、この文献は参照によりここに包含されるものとする
。Lは、カルディオウイルス及びアフトウイルスに存在するリーダーポリペプチ
ドを表す。P1は、構造タンパク質VP1、VP2、VP3及びVP4(これら
はそれぞれ1D、1B、1C及び1Aとも特定される)の前駆体である。P2及
びP3は、ウイルスRNAの複製及びポリタンパク質のプロセシングに必要な非
構造ウイルスタンパク質の前駆体である。
このウイルスRNAは、感染性である。これは、許容細胞へ導入されると、そ
れが、感染性ウイルスを再生する完全なウイルス増殖サイクルを開始することが
できる、ということである。完全長のウイルスcDNAからRNAポリメラーゼ
によりインビトロ(invitro)で合成されたRNA転写物も、感染性であること

メンゴウイルス及び脳心筋炎ウイルスのようなマウスのカルディオウイルス、
ならびにアフトウイルスは、その5′非コーディング配列内に長いホモポリマー
状ポリ(C)トラクトが存在することにより、他の正鎖RNAウイルスから区別
することができる。その長さ(一般に60〜350塩基である)、及びしばしば
このホモポリマー状配列を中断する配列の不連続性(例えばウリジン残基)は、
天然のウイルス分離株を特徴づけるために役立ってきたが、このポリ(C)領域
の正確な生物学的機能は明らかではない。メンゴウイルスポリ(C)トラクトの
、cDNA媒介性の短縮は、マウスにおけるこのウイルスの病原性を弱毒化する

メンゴウイルスの弱毒株であるvM16は、Duke et al.,「5′非コーディ
ングポリ(C)トラクトの遺伝子工学によるメンゴウイルスの弱毒化(Attenuat
ion of Mengovirus through Genetic Engineering of the 5'Non-coding Poly(
C)Tract)」、Nature343:474(1990)(これは参照によりここに包含されるも
のとする)、及びDuke及びPalmenberg,「短い別個のポリ(C)トラクトを含む
感染性カルディオウイルスRNAのクローニング及び合成(Cloning and Synthe
sis of Infectious Cardiovirus RNAs Containing Short,Discrete Poly(C)T
racts)」、J.Virol.,63:1822(1989)(これも参照によりここに包含される
ものとする)により記載されている。この弱毒株は、そのゲノムの5′非コーデ
ィング領域のポリ(C)トラクト中に欠失を有している。この弱毒株は、毒性(
virulent)メンゴウイルス及び脳心筋炎ウイルス(EMCV)を用いる抗原投与
からマウスを保護する。
組換えDNA技術の進歩は、生の組換えワクチンの開発を可能にしている。
ヒト又は動物の病原体のポリペプチド及び/又はエピトープもしくは抗原を包
含させることができ、ワクチンに用いることができる改変生ウイルスを生じうる
好適なベクターが、当業界で必要とされている。
発明の概要
本発明は、特に、メンゴウイルスの構造又は非構造タンパクが非相同アミノ酸
配列を含む、生存可能な改変弱毒メンゴウイルス、上記生存可能な改変メンゴウ
イルスの融合タンパク、上記生存可能な
改変メンゴウイルスに感染した許容細胞、上記改変メンゴウイルスの全長配列を
含む組換え核酸(RNA又はDNA)、ワクチン、及び免疫応答を誘発する方法
を提供することにより、当業界の上記の要望を満足することを助けるものである
。
本発明は、中でも、弱毒株であり、非相同ヌクレオチド配列を含む、生存可能
な改変メンゴウイルスに関する。本発明のある態様は、弱毒株であり、非相同ペ
プチド又はタンパク質をコードする非相同ヌクレオチド配列を含む、生存可能な
改変メンゴウイルスに関する。さらなる態様においては、生存可能な改変メンゴ
ウイルスは、メンゴウイルスのゲノムの5′非コーディング領域のポリ(C)ト
ラクトに突然変異又は欠失を有する弱毒株である。
特に、本発明のある態様は、メンゴウイルスのゲノムの5′非コーディグ領域
のポリ(C)トラクトに欠失を有する弱毒株であり、かつ、この組換えメンゴウ
イルスのリーダーポリペプチドが完全長のものであって非相同アミノ酸を含む、
生存可能な組換えメンゴウイルスに関する。本発明のある特定の態様においては
、組換えメンゴウイルスは、リーダーポリペプチドのアミノ酸6の後に挿入され
た、HIV−IのMN分離株のgp120のアミノ酸299〜466を含む。
本発明の別の態様は、非相同アミノ酸配列が挿入されている、弱毒メンゴウイ
ルス株の全長リーダーポリペプチドを含む融合タンパク質に関する。
さらなる態様においては、本発明は、本発明の組換えメンゴウイルスに感染し
た許容細胞に関する。特定の態様においては、許容
細胞は、HeLa、VERO、BHK21、及びP815細胞である。
本発明のさらなる態様は、メンゴウイルス核酸配列及び非相同核酸配列を含む
組換え核酸分子(RNA又はDNA)に関する。好ましくは、非相同配列は、全
長リーダーポリペプチドをコードするメンゴウイルス配列内に挿入されている。
より好ましくは、組換え核酸分子は、全長の弱毒メンゴウイルス配列、及び全長
リーダーポリペプチド配列内に挿入された非相同配列を含む。
さらに別の態様においては、本発明は、本発明の組換えメンゴウイルスのウイ
ルスゲノムに関する。
本発明の別の態様は、本発明の組換えメンゴウイルスを含むワクチンに関する
。特定の態様においては、ワクチンは、医薬的に許容可能な担体と混合された組
換えメンゴウイルスを含む。
本発明のさらなる態様は、免疫応答を誘発しようとするヒト又は動物のような
生物に、非経口又は経口ルートで本発明の組換えメンゴウイルスを投与すること
を含む、免疫応答の誘発方法に関する。本発明は、免疫原性組成物にも関する。
このような組成物は、医薬的に許容可能な担体と混合された組換えメンゴウイル
スを含む。
本発明のさらなる態様は、非相同アミノ酸配列とリーダーポリペプチドとの間
にプロテアーゼ切断部位をさらに含む、本発明の生存可能な組換えメンゴウイル
スに関する。本発明の特定の態様においては、プロテアーゼ切断部位は、プロテ
アーゼ3C切断部位である。
さらに別の態様においては、本発明は、本発明の生存可能な組換
えメンゴウイルスに感染した許容細胞に関し、この許容細胞は、ネイティブな形
態で非相同アミノ酸配列を発現する。

発明の詳細な説明
本書で記載され、特許請求される発明がより充分に理解されうるように、本発
明の態様の詳細な説明を以下に与える。
本説明においては、当業界の種々の用語が用いられる。これらの用語は、その
通常の、よく認知された意味において一般に用いられている。本説明を通じて用
いられる種々の用語を、以下で定義する。
本書で用いる場合、「組換えウイルス」という用語は、遺伝的に改変されたウ
イルスをいう。組換えウイルスは、少なくとも1つの他の生物からのタンパク質
又は核酸を含みうる。したがって、組換えウイルスは、非構造非相同ポリペプチ
ドを発現するウイルス、ならびに構造要素として非相同ポリペプチドを含むウイ
ルスを指すことができる。さらに、組換えウイルスは、キメラウイルスであるこ
とができる。
本書で用いる場合、「弱毒株」という用語は、低下した疾患生成能及び/又は
病原性を有する株をいう。
本書で用いる場合、「ゲノム」という用語は、ある1つの種の全ての遺伝子を
含む核酸をいう。ゲノムを作り上げている核酸は、その種の性質によって、RN
A又はDNAであってよい。例えば、ピコルナウイルス又は他のRNAウイルス
のゲノムは、RNAで構成されているが、一方、ヒトのゲノムはDNAで構成さ
れている。
本書で用いる場合、「非相同(性)」という用語は、ある与えられた種に天然
には見出されない物質をいう。例えば、特定のウイルスを指して用いる場合の「
非相同アミノ酸配列」という用語は、そのウイルス、例えばそのウイルスのタン
パク質に見出されないアミノ酸配列を指す。
本書で用いる場合、「ヌクレオチド又は核酸配列」という用語は、隣り合うヌ
クレオチドの3′及び5′炭素間の共有結合により連結されたヌクレオチドの線
状の連なりをいう。ヌクレオチド又は核酸配列は、RNA配列又はDNA配列で
あってよい。
本書で用いる場合、「アミノ酸配列」という用語は、共有結合により連結され
たアミノ酸の線状の連なりをいう。
本書で用いる場合、「融合タンパク質」という用語は、少なくとも2つのアミ
ノ酸配列を含むタンパク質であって、1つのアミノ酸配列が通常は天然にそれ以
外のアミノ酸配列と一緒には見出されないものをいう。例えば、メンゴウイルス
融合タンパク質は、非相同アミノ酸配列に共有結合により連結されたメンゴウイ
ルスアミノ酸配列を含むことができる。
本書で用いる場合、「エピトープ」という用語は、タンパク質におけるアミノ
酸のコンフィギュレーション(形状)であって、そのアミノ酸のコンフィギュレ
ーションが免疫応答と関連しているものをいう。例えば、エピトープは、抗体に
より認識され、免疫応答を誘発しうる抗原性モチーフにより定義されることがで
きる。エピトープは、アミノ酸配列の線状配列であってよいが、それに限定され
ない。
本書で用いる場合、「許容細胞」という用語は、ウイルスによって生産的に感
染されうる細胞をいう。したがって、メンゴウイルスの許容細胞は、メンゴウイ
ルスにより感染されうる細胞である。
本書で用いる場合、「組換え核酸分子」という用語は、インビトロの操作によ
って一緒に配置された少なくとも2つのヌクレオチド配列を含むハイブリッドヌ
クレオチド配列(RNA又はDNA)又はそのクローンをいう。
本書で用いる場合、「cDNA」という用語は、相補的DNAをいう。ゲノム
がDNAで構成されている生物の場合、cDNAは、mRNA又はそのフラグメ
ントに対して相補的である。ゲノムがRNAで構成されている生物の場合には、
cDNAは、その生物のゲノム又はそのフラグメントに対して相補的である。
本書で用いる場合、「ポリペプチド」という用語は、ペプチド結合によって互
いに連結されたアミノ酸の線状の連なりをいう。「ポリペプチド」という用語は
タンパク質を含むが、それに限定されない。
本書で用いる場合、「発現」という用語は、構造遺伝子からポリペプチドが産
生される課程をいう。
本書で用いる場合、「ポリタンパク質」という用語は、2つ以上のタンパク質
を含む、共有結合により連結されたアミノ酸の線状の連なりをいう。ある場合に
は、ポリタンパク質を構成しているタンパク質は、特異的プロテアーゼによる内
的タンパク分解的切断により遊離されうる。
メンゴウイルスのゲノム編成を図1に示す。この図は、ゲノムへ
のウイルスポリペプチドのマッピング、ならびにウイルスの成熟成分へのポリタ
ンパク質のプロセシングにおける種々の中間体を描いている。
ポリ(C)領域も、図1において確認される。Duke et al.(前出)、及びDuk
e及びPalmenberg(前出)により記載されているように、この領域における欠失
は、弱毒表現型に関連している。結果として、ポリ(C)領域に突然変異(例え
ば置換及び欠失)を有するメンゴウイルスのゲノムのcDNAを含むプラスミド
は、弱毒表現型を呈する組換えメンゴウイルスに関する本発明の種々の態様の構
築のための、メンゴウイルスDNAの供給源として用いることができる。
本発明の好ましい態様においては、メンゴウイルス核酸の好適な供給源は、プ
ラスミドpM16である。pM16は、フランスのパリにあるthe Collection N
ationale de Cultures de Micro-organismes(CNCM)に、受託番号I−13
13として1993年6月2日付で寄託されている。pM16の部分的プラスミ
ド地図を図2に示し、pM16の配列を図16に示す。このプラスミドは、突然
変異したポリ(C)トラクト、C13UC10をコードするが、それ以外は、二本鎖
複製型ベクターpBluescribeM13(+)のEcoRI及びBamHI制限部位
の間に挿入されたメンゴウイルスの全長ゲノムに相当するDNA配列を含む。結
果として、このプラスミドは、T7プロモーターの下流にメンゴウイルスcDN
Aを含んでいる。
本発明の他の態様においては、メンゴウイルス核酸の好適な供給
源はpM16−1である。pM16−1もまた、1993年6月2日に受託番号
I−1312としてCNCMに寄託されている。pM16−1の配列を図17に
示す。
本発明の他の態様においては、他のプラスミドをメンゴウイルス核酸の供給源
として用いてもよい。例えば、C8ポリ(C)トラクトをコードするpM18、
又はC12ポリ(C)トラクトをコードするpM19、又はポリ(C)トラクトの
完全な欠失を有するプラスミドを用いることができる。弱毒表現型が必要とされ
ない場合には、野生型のポリ(C)トラクトであるC50UC10をコードするpMwt
を用いることができる。これらのプラスミドの各々がポリ(C)トラクトの外
にメンゴウイルスゲノムに相補的なDNAを含むので、種々のインビトロ操作に
よって、あるメンゴウイルスプラスミド(又は野生型メンゴウイルスDNA)か
ら別のプラスミドを構築することができる。1つの可能性は、あるプラスミドか
らのポリ(C)トラクトを含むEcoRV−AvrII制限フラグメントを、構築
すべきプラスミドの適切なEcoRV−AvrIIフラグメントで置換することで
ある。
DNA形態の弱毒メンゴウイルスゲノムをコードするベクターがいったん得ら
れると、組換えメンゴウイルスにより発現されるべきアミノ酸配列をコードする
非相同ヌクレオチド配列を、そのメンゴウイルスゲノムのコーディング領域内に
挿入することができる。本発明の特定の態様においては、その核酸配列は非相同
抗原又はエピトープをコードする。
非相同ヌクレオチド配列を挿入する部位は、制限部位であること
ができる。本発明の特定の態様においては、この部位は、メンゴウイルスゲノム
のヌクレオチド729にわたるNcoI制限部位である。
非相同ヌクレオチド配列を制限部位に挿入する場合、メンゴウイルスDNAベ
クターを適切な酵素で制限消化し、DNAベクターを切断する。次いで、非相同
DNA配列を制限消化したメンゴウイルスベクターに連結し、メンゴウイルスゲ
ノムを含む組換えDNA分子(今や非相同ヌクレオチド配列を含んでいる)を生
成する。
非相同ヌクレオチド配列を制限部位に挿入しない場合、当業者は、その挿入部
位を含むDNAベクターの制限フラグメントを選択することができる。次に、選
択した制限フラグメントに対応するが、望ましい部位に挿入された非相同ヌクレ
オチド配列をさらに含む、合成DNAフラグメントを合成することができる。こ
の合成DNAフラグメントは、選択した制限フラグメントのかわりにDNAベク
ターに挿入し、組換えメンゴウイルスゲノムcDNAを含む組換えDNA分子を
生成することができる。
非相同ヌクレオチド配列は、様々な方法で調製することができる。例えば、配
列は、組換えメンゴウイルスにより発現されるべき非相同ポリペプチドをコード
するcDNAを特異的に切断することによって得てもよい。例えば、これは適切
な制限酵素を用いて達成してもよい。あるいは、非相同ヌクレオチド配列は、当
業界において周知の方法を用いて化学的に合成することができる。
望ましい非相同ポリペプチド(例えば抗原又はエピトープ)を含む組換えメン
ゴウイルス及びそのタンパク質もしくは発現産物は、
組換えメンゴウイルスゲノム中に挿入された非相同ヌクレオチド配列を含む組換
えDNA分子からRNA転写物を生成することによって得ることができる。例え
ば、pM16及びpM16−1の場合には、RNA転写物は、T7RNAポリメ
ラーゼを用いてインビトロで産生させることができる。代替的なプロモーター及
び対応するポリメラーゼで置き換えることもできる。転写混合物のアリコートを
、許容細胞をトランスフェクトするために使用することができる。例えば、DE
AEデキストラン、リン酸カルシウム、ポリ−オルニチン、電気穿孔法、及び合
成トランスフェクション剤を用いて、HeLa、VERO、BHK21、及びP
815のような哺乳類細胞をトランスフェクトすることができる。子孫ウイルス
の産生は顕微鏡により監視することができ、ウイルスは、細胞破砕の周知の方法
、例えば凍結解凍により放出させることができる。
本発明の特定の態様においては、非相同ポリペプチドはリーダーポリペプチド
(L)中に挿入される。メンゴウイルスポリタンパク質のN末端から6アミノ酸
のところでの外来性エピトープの挿入は、Lを機能的でなくすることがなく、し
たがって、インビトロでもインビボでもウイルスの増殖に干渉しないことがわか
った。
他の挿入部位は、挿入物(インサート)がウイルスの生活環に重要な機能と干
渉しない位置での挿入のために、ウイルスゲノム内で選んでもよい。好適な挿入
部位に制限部位がない場合には、例えば、部位特異的突然変異誘発によりそれら
を導入することができる。したがって、原理的には、よく定義された機能的領域
、例えば、3Cの触媒性3つ組(トリアッド)を除き、ゲノム内の全ての
制限部位及び他の位置を、挿入のために考慮することができる。好ましい部位と
しては、ゲノムの非構造領域、例えば、P2、P3及び/又はウイルスタンパク
質のNもしくはC末端に相当する領域が挙げられる。
抗原又はエピトープのような外来性ポリペプチドを含む融合タンパク質を製造
するために、外来性ヌクレオチド配列を、リーディングフレームを保存するよう
な方法で、メンゴウイルスcDNA中にLポリペプチドをコードする配列内に挿
入する。そうすると、組換えウイルスは、ウイルスポリタンパク質の一部として
外来性配列を発現することができる。ポリタンパク質は、特に、挿入された非相
同ポリペプチドを含むLポリペプチドを有する融合タンパク質の形態にプロセシ
ングされる。融合タンパク質は、次いで、感染細胞の細胞質から得ることができ
る。
本発明に関しては、メンゴウイルスベクターに挿入される非相同DNA配列は
、いかなるアミノ酸配列をコードすることもできる。本発明のある種の態様にお
いては、アミノ酸配列は、非相同エピトープを含む。これらの態様においては、
非相同アミノ酸配列は、いくつかの外来性エピトープ又は単一の外来性エピトー
プを含むことができ、あるいは他のアミノ酸、例えば、メンゴウイルスアミノ酸
と一緒にエピトープを定義することもできる。他の態様においては、アミノ酸配
列は、実質的に非相同エピトープからなることができる。一例として、本発明の
種々の態様のアミノ酸配列を表1に列挙する。
本発明の態様においては、挿入されるべき非相同DNA配列は、一般に約70
0〜1,100塩基の範囲にある。表1に与えられている配列は、本発明の組換
えメンゴウイルスの構築のための非相同配列の例である。この表に与えられてい
ない種の非相同配列を含む
組換えメンゴウイルスは、本書に示すようにして構築することができる。非相同
配列が、大きすぎてメンゴウイルスにおいて発現させることができない場合には
、各々が与えられたタンパク質の一部を発現する組換えメンゴウイルスのセット
を構築することができる。
本発明の非相同DNA配列は、2つ以上のポリペプチドをコードすることがで
きる。ポリペプチドをコードするDNA配列は、互いに直接連結することもでき
、あるいは連結配列によって分離することもできる。特定の態様においては、こ
れらの連結配列は切断部位をコードすることができる。
本発明の態様においては、組換えウイルスのLポリペプチドは、HIV−Iの
gp120のセグメントを含む。特定の態様においては、Lポリペプチドは、H
IV−IのMN株のgp120のアミノ酸299〜446を含む。より特定の態
様においては、HIV−IのMN株のgp120のアミノ酸299〜446は、
Lポリペプチドのアミノ酸6の後に挿入されている。このHIV配列は、主要中
和決定基(PND)を構成するV3ループをコードする配列及びgp120分子
のCD4レセプターに対する結合に関与する下流配列を含む。結果として得られ
る組換えメンゴウイルスは、HIV−I特異的抗体により認識される、抗HIV
−I抗体を動物に誘起する、HIV−I gp120−メンゴウイルスL融合タ
ンパク質を発現する。
HIV−I gp120−メンゴウイルスL融合タンパク質は、動物において
gp120特異的細胞毒性免疫応答をも誘発する。
抗原性については、外来性エピトープを含むL融合タンパク質は、ネイティブ
なタンパク質配列に対する抗体を誘起し、それに結合する能力を保持することが
できる。それゆえ、この戦略は目的の抗原性特性を呈するいかなる外来性タンパ
ク質にも適用しうる。結果として、単一のよく定義された短いエピトープの場合
には、これらのエピトープをより大きいタンパク質中に含む組換えメンゴウイル
スの構築が、選択すべき戦略である。
本発明の組換えウイルスは、マウス及びカニクイザルにおいて抗体を誘起し、
これが、その外来性配列が由来するタンパク質と結合する、及び/又は病原体を
中和する、ということが示されている。したがって、メンゴウイルスに感染しう
る他の動物種、例えばヒトにおける免疫応答の誘発は、得られたインビトロ及び
インビボの結果に基づいて予測される。したがって、防御免疫応答は、本発明の
組換えウイルスにより誘起することができる。例えば、狂犬病のGタンパク質の
配列を発現する組換えウイルスは、マウスを含む動物におけるワクチンとしての
使用のために、本発明にしたがって工学的に作製することができる。同様に、H
TLV−Iの糖タンパク質からの配列を発現する組換えウイルスを、マカク、他
の霊長類、又はヒトでの使用のために得ることができる。
組換えメンゴウイルスにより発現される外来性エピトープを含むタンパク質の
免疫原性及び抗原性は、種々の方法により改良することができる。外来性エピト
ープをコードする非相同ヌクレオチド配列のサイズは、最大約350アミノ酸の
大きさまでの外来性抗原のより大きいセグメント(又は抗原全体)を発現させる
ように増大さ
せることができ、外来性抗原は、融合タンパク質としてよりもネイティブな形態
で発現させることもでき、そして、融合タンパク質の抗原性及び/又は免疫原性
に重要であり得るグリコシル化のような翻訳後修飾を可能にするために、適切な
細胞区画(コンパートメント)に対する外来性抗原の選択的ターゲティングを達
成することができる。
本発明の態様においては、組換えメンゴウイルスは、1つのタンパク質の複数
の配列を発現することができる。その上、組換えメンゴウイルスは、異なるタン
パク質からの複数の配列を含むことができる。したがって、本発明は、複数の病
原体に対して同時に免疫する、又は免疫応答を誘導する、ということを可能にす
る。
本発明のある種の態様においては、非相同ポリペプチドは、その非相同ポリペ
プチドのアミノ酸配列とメンゴウイルス配列との間にプロテアーゼ切断部位を入
れることによって、ネイティブな形態で発現させることができる。
本発明の好ましい態様においては、メンゴウイルスプロテアーゼ3C切断部位
を用いる。内在性メンゴウイルスプロテアーゼ3Cは、メンゴウイルスポリタン
パク質のほとんどの切断に関与する。例えば、プロテアーゼ3Cは、前駆体タン
パク質L−P1−2AをQ−Gアミノ酸連結で特異的に切断することにより、L
ペプチドとVP4(1A)との間の切断を媒介し、遊離のLペプチド及びP1−
2Aを生じる。
図13は、示されているこのプロテアーゼ3C切断部位を有するメンゴウイル
スゲノムの図である。図13のアミノ酸配列は、プロ
テアーゼ3Cによる切断の充分な基質である。Parks et al.,J.Viro1. 63:1054
(1989)を参照されたい。この文献の開示全体が、参照によりここに包含される
ものとする。本発明の態様においては、プロテアーゼ3C切断部位をコードする
DNA配列は、挿入されるべき非相同DNA配列の各々の末端に入れることがで
きる。その上、ウイルスタンパク質2A及び2Bの間の切断部位であるAsn-Pro-
Gly-Pro配列を用いることもできる。2Aと2Bとの間の切断は、自己触媒的メ
カニズムによりグリシン−プロリン結合を切ることによって起こる、という証拠
がある。このような非相同DNA配列を、組換えメンゴウイルスを構築するため
に用いる場合、結果として得られる非相同ポリペプチドは、感染の間に自己触媒
的切断によってメンゴウイルスポリペプチドから遊離させることができる。
非相同ポリペプチドは、特定の細胞区画に対してターゲティングすることがで
きる。例えば、SV40のT抗原のもののような核局在性配列を包含させること
により、タンパク質が核へターゲティングされることになる。これに対して、β
−2−ミクログロブリンのもののようなシグナル配列は、ポリペプチドを小包体
へと向かわせ、グリコシル化のような翻訳後修飾及び培地中への分泌を可能にす
ることになる。さらに、HIV−Iのgp41の膜貫通配列のような配列は、ア
ンカー配列として用いて、ポリペプチドを細胞膜の中に挿入させることを可能に
することができる。
さらなる態様においては、2以上の異なるタンパク質を、2以上の組換えウイ
ルスによる標的細胞の同時トランスフェンクションに
より、同じ細胞において同時発現させてもよい。これは、同時トランスフェクシ
ョンを受けた標的細胞における種々の分子間相互作用を可能にすることができる
。
本発明の組換えメンゴウイルスは、ワクチンとして、又は免疫原性組成物の一
部として使用することができる。ワクチン接種されるべき対象としては、人体、
霊長類、ヒト以外の霊長類、哺乳類、マウス、又はメンゴウイルスウイルスに感
染しうる他のあらゆる動物を挙げることができる。本発明の態様においては、組
換えメンゴウイルスは、医薬的に許容可能な担体と混合された状態でワクチン又
は免疫原性組成物中に存在する。好適な担体の例としては、ウイルスの安定性を
支持し、動物又はヒトにおいての使用に許容されうるあらゆる緩衝剤が挙げられ
る。生ワクチンには、アジュバントは不要である。本発明の他の態様においては
、ワクチン又は免疫原性組成物は、本発明のメンゴウイルス融合タンパク質を、
医薬的に許容可能な担体との混合物の状態で含み、その融合タンパク質は抗原を
含む非相同アミノ酸配列を含む。
メンゴウイルスは、以下の特性により、広い潜在能力を有する生ワクチン又は
免疫原として特に魅力的である。
1.本書に記載したvM16株及び他の弱毒株は、その弱毒化が欠失に起因す
るため、容易には毒性に復帰しにくい。本発明の文脈において、ポリ(C)トラ
クト全体が欠失している遺伝子工学的に作製されたメンゴウイルスを用いること
が可能である。
2.メンゴウイルスの広い宿主範囲は、医学的又は獣医学的に重要な多様な病
原体に対する多くの異なる動物種におけるワクチン又
は免疫原としてのその利用を可能にし、例えば、HAV、HIV、HTLV、狂
犬病、FMDV、ウシのようなコロナウイルス、ヘルペスウイルス、麻疹、ムン
プス(おたふくかぜ)及びレスピラトリーシンシチアルウイルス(RSV)が挙
げられる。
3.非経口又は経口投与後に生体内で複製することができる生ワクチンの形態
でのワクチンにおけるメンゴウイルスの利用は、体液性免疫応答及び細胞性免疫
応答の両方、特に細胞傷害性T細胞応答の誘発を可能にすることになる。
4.経口免疫後に腸において増殖するメンゴウイルスの能力は、全身性の一般
的免疫応答及び局所的な粘膜応答の両方の誘発を可能にする。これは、HIV、
HPV、HTLV、TGEV又はロタウイルスのような病原体の場合に、特に適
切である。
本発明の組換えウイルスは、種々の非免疫学的方法において用いることができ
る。例えば、組換えウイルスは、大量の目的核酸又はタンパク質を産生させるた
めに、例えば組織培養において、クローニング又は発現ベクターとして用いるこ
とができる。組換えメンゴウイルスに感染した細胞において発現される非相同タ
ンパク質は、感染細胞から大量に精製することができる。
その上、本発明の組換えウイルスは、種々の病原体、又は疾患に関与する細胞
機能の特異的阻害剤を、種々の細胞位置又は細胞以下の位置に送り込むために用
いることができる。
以下の例は、本発明のよりよい理解を容易にするための説明の方法として与え
るものであり、本発明を制限することを意図するものではない。さらに、詳細な
説明は、本発明の好ましい態様を示しつ
つも、説明の方法としてのみ与えられていることも、理解されるべきである。こ
れは、本発明の精神及び範囲内における種々の変更及び改変は、詳細な説明から
当業者には明白になるであろうためである。
実施例
材料と方法
vMLN450の構築
全てのDNA操作は標準的手順(7)にしたがって行った。T7プロモーター
(5)の下流にvM16メンゴウイルスcDNAを含むプラスミドpM16から
の5.8kbのSphI−SphIフラグメント(プラスミド塩基1928〜77
75)の欠失によって、サブクローンpMRA1を生成した。HIV−IMN
gp120特異的DNAを、オリゴヌクレオチド5′VCN(ATATGTTGACCATGGA
ACAAATTAATTGTACAAGACCC)及び3′VCN(TAATCCATGGCGGTCAACGTGGGTGCTACTCC
TAATGG)を用いて、M.P.Kieny氏から好意により提供されたプラスミドpTG5
156(Transgene SA,ストラスブール)からPCRにより増幅させた。まず、
PCRフラグメントを、NcoI部位(ウイルス塩基729)でpMRA1にク
ローニングし、結果としてプラスミドpMRA2を得た。pMRA2のSphI
部位にpM16の5.8kbフラグメントをトランスファーすることによって、全
長cDNAを再度確立し、プラスミドpMRA3を得た。PCRにより、クロー
ニングされた配列の正しい方向性を決定し、E.coli DH5α(Promega)にお
いてプラスミドを増幅させた。
pMRA3から由来する感染性のRNAの転写物を、T7RNAポリメラーゼ
を用いて調製し、記述されているとおりに(5)HeLa細胞にこれをトランス
フェクションし、その結果、組換えメンゴウイルスvMLN450を得た。He
La細胞での継代によりvM16及びvMLN450のストックを作成し、記述
されているとおりに(8)滴定し、これを、メンゴウイルス特異的オリゴヌクレ
オチドである5′194−(TAGGCCGCGGAATAAGGCCGGTGTGC)及び3′1094−
(GGAGCATGTTCGAGAAAGCATTGAC)を用いて、挿入されたHIV−I配列の存在に
ついてRT−PCRにより分析した。
免疫及びELISA試験
10週齢のBALB/cマウスを、0日目及び56日目の2回、PBS中の1
06pfu(プラーク形成単位)のvMLN450又は106pfuのvM16を用いて
、腹腔内で免疫した。対照動物には、PBSを単独で与えた。成体カニクイザル
は、PBS中の106pfuのvMLN450又はvM16を用いて、筋内で免疫し
た。ELISAプレート(Nunc Maxisorb)を1ウェルにつき50ngの精製した
組換えgp160LAI又はgp160MN−LAI(Transgene)でコーティ
ングし、血清の段階希釈液を用いてインキュベートし、続いてホースラディッシ
ュペルオキシダーゼ結合ヒツジ抗マウスIgG(H+L)抗体(Diagnostics Pa
steur)又はウサギ抗サルIgG(H+L)抗体(Nordic)と共にインキュベー
トした。ELAVIA試験(Diagnostics Pasteur)は、メーカーのプロトコー
ルにしたがって実施した。HIV−I MN中和検定
は、3.5×105のMT4細胞を用いて、記述されたとおり(9)に行った。
6日目と10日目の間、融合細胞(シンシチウム)形成を監視した。
例1
pMRA−3の構築
メンゴウイルスcDNAのNcoI部位729へのクローニングを容易にする
ため、2つのNcoI部位を含む5.8kbのSphI(1928)−SphI(
7775)フラグメントを削除することによって、pM16の欠失クローンを構
築した。結果として得られたpMRA−1と呼称されるクローン(図4)は、位
置729に唯一のNcoI部位を含んでいる。
以下のオリゴヌクレオチドを用い、プラスミドp05156Sからポリメラー
ゼ連鎖反応(PCR)により、478bpのHIV−I−MN特異的DNAフラグ
メントを生成した:
プラスミドp05156Sは図3に描かれており、Trangene S.A.(フランス
、ストラスブール)から入手した。p05156Sについては、特許出願WO9
2/19742の中で記述されている。ゲノムのMN配列は、Gurgo et al.,19
88,Virology 164:531-536(1988)によって公開されており、この文献は参照
により本書に包含されるものとする。
増幅された配列Δgp120−VCNは、HIV−I−MNの糖タンパク質(
gp)120のアミノ酸299〜446に対応する。
Δgp120−VCNを、5ngの鋳型上で約100mMの濃度のオリゴヌクレオチ
ド5′VCN及び3′VCNを用いて生成し、p05156Sを、Stratageneか
ら入手したPFUポリメラーゼを用い、同じくstratageneから得たPFU増幅緩
衝液を用いて、標準的温度サイクル条件で生成した。
その後、Δgp120−VCNフラグメントをpMRA−1のNcoI部位7
29にクローニングして、pMRA−2を生成した。pMRA−2は図5に描か
れている。pMRA−2内のΔgp120−VCNインサートの正しい配向及び
長さは、オリゴヌクレオチドM−1094、5′VCN及び3′VCNを用いて
、PCRにより確認した。M−1094のオリゴヌクレオチド配列は次のとおり
である:
正しいクローンは、M−1094及び5′VCNで800bpのPCRフラグメ
ントを生じ、M−1094及び3′VCNでは増幅されたDNAを全く生じなか
ったが、一方、正しくない方向性でΔgp120−VCNを含むクローンは、反
対の結果を示した。
pM16からの5.8kbのSphI(1928)−SphI(7775)フラ
グメントをpMRA−2へクローニングして戻すことにより、全長cDNAを再
構築し、その結果、プラスミドpMRA−3を得た。pMRA−3は、図6に描
かれている。このプラスミドは、メンゴウイルスcDNAのL−ペプチドコーデ
ィング領域内にフレーム内で挿入された付加的な459bpを含んでおり、25.
8kDの融合タンパク質LN450をコードする(図
7)。
pMRA−3内のpM16の5.8kbのSphI(1928)−SphI(7
775)フラグメントの正しい方向性は、BamHI(Boehringer Mannheim)
及びHindIII(Boehringer Mannheim)での二重制限消化によって確認した。
正しい方向性にあるプラスミドのこの二重制限消化は、結果として30塩基及び
11356塩基の2つのDNAフラグメントをもたらす。一方、正しくないクロ
ーンは、長さ5821塩基及び5565塩基の2つのDNAフラグメントを示す
。
例2
vMLN450の生育特性
vMLN450及びvM16 cDNAの感染性RNA転写物を、BamHI
で消化されたプラスミドpM16及びpMRA−3とT7RNAポリメラーゼ(
Pharmacia)を適当な緩衝液中でインキュベートすることによって、インビトロ
で産生させた。この転写混合物のアリコートを、1mg/ml DEAEデキストラン
に調整し、その後、Sylvie van der Werf et al.の「精製されたT7RNAポリ
メラーゼによる感染性ポリオウイルスRNAの合成」,Proc.Natl.Acad.Sci.
(USA)83:2330(1986)により記述されているとおりに、単層細胞上でこの溶
液を30分間インキュベートすることによってHeLa細胞にこのアリコートを
トランスフェクションした。上記文献の開示全体が、参照により本書中に包含さ
れるものとする。
HeLa細胞内へのプラスミドpMRA−3から由来するRNA
のトランスフェクションは、結果としてウイルスvMLN450の産生をもたら
した。後代ウイルスの産生は、顕微鏡で監視した。ストック(保存)ウイルスは
、感染多重度(MOI)1でHeLa細胞の集密(コンフルエント)単層を感染
させることによって生育させた。完全なcpe(細胞変性効果)の後、ウイルス
を収獲した。細胞内ウイルスは、細胞を3回凍結及び解凍させ、遠心分離により
細胞砕片及び核をペレット化することによって放出させた。完全なcpeは、p
M16RNAで細胞をトランスフェクションした場合の48時間に比べて、72
時間後に観察された。vMLN450は高い力価まで生育するが、vM16に比
べると約3分の1〜4分の1である。
組換えウイルスのプラーク表現型を試験した。ウシ胎児血清(FCS)無しで
ダルベッコの改変イーグル培地(DMEM)中でウイルス原液を希釈し、HeL
a細胞の集密性単層を感染させるためにこれを用いた。30分のインキュベーシ
ョンの後、0.9%の軟寒天を含むDMEMで細胞をカバーした。次に細胞を3
7℃で72時間インキュベートし、無傷の(インタクトな)細胞を0.2%のク
リスタルバイオレット溶液で染色した。
培養上清から収獲した組換えウイルスのプラーク表現型は、野生型ウイルスプ
ラークのものの約65%の平均直径をもつ中サイズのプラークを示す(図8)。
例3
vMLN450の遺伝的安定性
vMLN450を含むトランスフェクション上清を、HeLa細
胞上で3回継代し、ウイルスRNAを抽出した。組換えウイルスvMLN450
の継代3からのウイルスRNAを、オリゴヌクレオチド5′VCN、3′VCN
、M−1094及びM−VDW−1を用いてRT−PCR(逆転写PCR)によ
り分析した。オリゴヌクレオチドM−VDW−1の配列は以下のとおりである:

us 14:31-33(1992)によって記述されている、サンガーのジデオキシ配列決定
方法から誘導されたPCR配列決定方法により、RT−PCR産物について配列
分析を行った。低融点アガロースゲルからPCRフラグメントを抽出し、各々異
なる濃度のdNTP及びddNTP、及び4pmolの32P−末端標識プライマーを
含む4つの反応混合物の各々に、約200ngのDNAを添加した。反応は、TA
Qポリメラーゼ(Amersham)を添加することによって開始し、標準的温度サイク
ル条件で20回の増幅の間インキュベートした。配列決定反応は、標準的条件下
でのポリアクリルアミドゲル電気泳動(PAGE)により分析した。
オリゴヌクレオチド対M−VDW−1/M−1094及び5′VCN/M−1
094での分析は、それぞれ1,382bp及び932bpの特異的DNAフラグメ
ントがvMLN450のRNAから増幅されうることを明らかにした。これらの
DNAフラグメントとプラスミドpMRA−3から生成されたDNAフラグメン
トとの間には、明らかなサイズの差はなかった。これらの実験の結果を図9に示
す。
vM16様のウイルスへの復帰突然変異を示す野生型サイズのフラグメントは
全く検出できなかった。1,382bpのフラグメントを部分的に配列決定して、
非特異性をさらに排除した。vMLN450について得られた部分配列は、図1
0に下線の付された配列として示されており、これは当初のインサートのものと
の差異を全く示していない。

HeLa細胞におけるLN450の発現
融合タンパク質LN450の発現を分析するために、単層のHeLa細胞をM
OI10のvMLN450に感染させ、基本的に参照により本書に包含されるLe
e及びWimmerのVirology 166:405(1988)に記述されているとおりに、感染から
51/2〜71/2時間後に35Sメチオニンで標識した。感染後7時間で、細胞をNP
40で溶解させ、細胞質抽出物を、参照により本書に包含されるHarber et al.
,Journal of Virology 65:326(1991)に記述されているとおりに調製した。
その後、35Sメチオニンで標識されたタンパク質をSDSポリアクリルアミドゲ
ル電気泳動(SDS−PAGE)により分析した。ゲル電気泳動及びオートラジ
オグラフィは、標準的条件にしたがって実施した。
HeLa細胞をvMLN450及びvM16それぞれで感染させ、ウイルスタ
ンパク質を35S−メチオニンで標識した。感染後71/2時間に細胞質抽出物を調
製し、その後、アリコートをSDS−PAGEにより分析した。vMLN450
に感染した細胞は、vM16感染又は偽感染細胞(図11、a、c)には存在し
ない
26kDaの見かけの分子量をもつ単一の付加的なタンパク質の存在を示す(図1
1、e)。このタンパク質の分子量は、LN450について予想されるサイズに
相当する。
例5
抗原性
LN450の抗原特性を、モノクローナル抗体50.1(Repligenから入手)
を用いて、免疫沈降法により検定した。35Sメチオニンで標識したvMLN45
0感染細胞のライセートのアリコートを、1μgのMAb50.1を用いて、4
℃で1時間インキュベートし、その後、同量のプロテインA−セファロース懸濁
液を加え、混合物を1時間インキュベートした。プロテインA−セファロース複
合体をペレット化し、3回洗浄し、SDS−PAGEとそれに続いてのオートラ
ジオグラフィにより分析した。
vMLN450、vM16感染及び偽感染HeLa細胞の細胞抽出物を、モノ
クローナル抗体50.1(MAb50.1)で免疫沈降させた。この抗体は、H
IV−I−MNの主要な中和決定基であるHIV−IのMN株のV3ループを認
識する。図11、b)、d)及びf)に示すデータは、vMLN450感染細胞
中で産生される26kDのタンパク質のみがMAb50.1で免疫沈降しうること
を明らかにし、LN450としての26kdタンパク質の同一性(正体)を確認し
ている。

免疫原性
1つの実験において、10週齢のBalb/c又はCBAマウス
を、PBS中の106pfuのvMLN450(Balb/cについては4匹、CB
Aについては5匹)又は106pfuのM16(Balb/cについては4匹、CB
Aについては2匹)を用いて、腹腔内で免疫した。3匹(Balb/c)又は2
匹(CBA)の動物には、ウイルス無しの対照としてPBSを与えた。動物に、
等用量のウイルスを用いて、初回免疫から8週間後に追加免疫を施した。免疫後
14日に血液試料を採取し、血清を調製した。
PBS中のvMLN450(動物3匹)又はvM16(動物1匹)約4×105
pfuを用いて、カニクイザル(Macaca fascicularis)を筋内で免疫した。免疫
から28日後に血液試料を採取し、血清を調製した。
ELISAプレート(Nunc Maxisorb)を、1ウェルにつき100μlの組換
えgp160MN−LAI又はgp160 LAI(PBS中、500ng/ml)
でコーティングした。gp160MN−LAIは、MNウイルス株からのgp1
20部分とIIIB(LAI)株からのgp41とを含む融合タンパク質である。
プレートを洗浄し、マウス又はサルの血清の段階希釈液を用いてインキュベート
した。37℃で2時間インキュベートした後、プレートを洗浄し、ホースラディ
ッシュペルオキシダーゼ結合モノクローナル抗体[抗マウスIgG(H+L)(
Diagnostics Pasteur)又は抗サルIgG(H+L)(Nordic Immunology)]を
添加し、37℃で1時間インキュベートした。最終的洗浄の後、OPD(670
μg/ml)、H2O2(0.042%)溶液を添加することによって、試験を発色さ
せた。4NH2SO4を添加することにより反
応を停止し、Dynatech ELISAプレートリーダーで490nmでODを読み取
った。
マウス血清の1/100希釈液から出発した2中1の段階希釈液を、融合細胞
阻害検定で試験した。融合細胞形成を誘発する最低の濃度でHIV−I−MNを
含むウイルス溶液に、希釈した血清を添加した。混合物を1時間インキュベート
し、3.5×105のMT4細胞の懸濁液を添加した。細胞を48ウェルのプレ
ート中で生育させ、3日目に4中1の割合で希釈し、6日目と10日目の間で融
合細胞形成を監視した。
Balb/cマウス、CBAマウス又はカニクイザルを、マウスの場合には1
06pfuのvMLN450もしくはサルの場合には2×105pfuのvMLN450
、106pfuのvM16又はウイルス無しの対照のいずれかを用いて免疫した。免
疫から2週間後及び免疫から10週間後(追加免疫から2週間後)にマウスを採
血した。血清を、抗原として組換えgp160 MN−LAI又はgp160
LAIを用いて、ELISAにより分析した。サルは、免疫から4週間後に採血
し、同様に分析した。図12及び表3に示すデータは、特異的抗HIV抗体が、
vMLN450に感染させたマウスからの血清中にのみ検出されたことを明らか
にしている。
抗HIV−I力価は、動物をvMLN450で追加免疫した後、約35倍に上
昇し、2回目の免疫の後少なくとも10週間は高いレベルにとどまった。CBA
マウスを用いても同様の結果が得られた。
MT4細胞及びHIV−I−MNを用いた中和検定において分析した場合(表
2)、免疫から2週間後に得られた4つのBalb/c血清のうちの2つは、1
00中1の希釈度で、ウイルスに誘発される融合細胞形成を完全に阻害した。
HIV−I中和抗体力価を、HIV−IMNを用いて融合細胞形成阻害検定で
決定した(9)。中和力価は、vMLN450で免疫したBALB/cマウスか
らの70日目の血清では1:100〜1:400の範囲内にあったが、一方、対
照血清についての値は1:100未満であった。動物におけるV3特異的応答は
、現在測定中である。
カニクイザルを用いて、匹敵する実験を行った。106pfuの生きたvM16又
はvMLN450を1回筋内注射して、動物を免疫した。HIV−I特異的抗体
を、1ヵ月後に、精製された組換えgp160MN−LAI(表4)又は市販の
ELISAキット(ELAVIA)のいずれかを用いて(データ示さず)、EL
ISAにより測定した。両方の試験共、vMLN450で免疫したサルには上昇
したgp160特異的抗体が存在したが、一方、vM16で免疫した動物は全く
反応性を示さなかったことを明らかにした。
例7
gp120特異的細胞傷害性免疫応答
B細胞及びT−ヘルパー細胞エピトープに加えて、HIV−I MNのgp1
20のV3ループ配列は、H−2dハプロタイプ(10)でマウスで認識される
MHCクラスIに制限された細胞傷害性Tリンパ球(CTL)エピトープを含む
ことが示されてきた。したがって、我々は、vMLN450で免疫したBALB
/cマウスにおける、このエピトープに対する細胞傷害性細胞性免疫応答の存在
について調べた。免疫したマウスからの脾細胞を、インビトロでP18−MNペ
プチドで刺激し、V3 CTLエピトープ配列(10)を含むペプチドPI8−
MNでパルスされた同系の標的細
胞(P815)に対する細胞傷害活性について検定した。表5に示されていると
おり、vMLN450で免疫した動物においては、25:1というエフェクター
対標的比で、明らかなHIV−IMN特異的細胞傷害活性を立証することができ
たが、親であるvM16で免疫した動物では立証できなかった。この活性はMH
CクラスIの制限を受けていた。参考文献11を参照のこと。
vM16又はvMLN450で免疫したBALB/cマウスからの脾細胞をプ
ールし、その後、「材料及び方法」の中で記述したとおりにインビトロで再刺激
した。細胞溶解活性は、25:1というエフェクター:標的比率で、P18−M
Nペプチドでパルスされた又はされていないP815細胞に対して測定した。
細胞傷害性検定は以下のとおりに行った。BALB/cマウスを、0日目及び
21日目に、106pfuのvM16又はvMLN450を用いて腹腔内で免疫した
。1ロットにつき3匹のマウスからの脾細胞を10日後に回収し、プールし、7
日間P18−MNペプチド(10)で、次に5日間、成長因子供給源としての5
%コンカナバリンA上清含有培地の存在下の、P18−MNペプチドでインビト
ロで再刺激した。刺激を受けた脾細胞の細胞溶解活性を、5h51Cr−放出検定
によって決定した。標的細胞はペプチドによ
るパルスを受けた、51Crで標識されたP815腫瘍細胞であった。特異的51C
rの放出の百分率は、〔(実験上の放出−自然発生的放出)/(最大放出−自然
発生的放出)]×100として計算した。自然発生的放出は、1%トリトンX−
100とのインキュベーションによって得られた最大放出の20%未満であった
。

vMQG−1の構築
この構築において使用したHIV−I−MN gp120配列は、vMLN4
50構築の説明の中で前述したとおりに生成した。ただし、アミノ酸配列は、H
IVインサートのC末端及びL−ペプチドのN末端で、図14に記述されている
とおりに変更した。
L−ペプチドのN末端に非相同短縮型gp120又はそのフラグメントを含み
、かつ例えば3Cのようなプロテアーゼ切断部位(単数又は複数)によってLペ
プチドから分離された組換えメンゴウイルスvMQG−1の生成は、vMLN4
50に比べて、gp120配列の発現及び免疫原性を増大させる潜在的可能性を
秘めている。gp120のフラグメントは、動物における免疫応答を惹起するよ
う選択する。特定の態様においては、これらのフラグメントは20〜100個の
アミノ酸のペプチドで構成されている。
vMQG−1のcDNAであるpMRA−5の構築の流れ図を、図15に表す
。
図15中の合成二本鎖オリゴヌクレオチドで標識されたLQGd/sの配列は
、以下のとおりである:
簡単に言うと、PCRで増幅された配列Δgp120−VCNをHincII及
びNcoIで制限消化し、451bpのフラグメントを分離した。このフラグメン
トを、3C切断部位をコードする合成二本鎖オリゴヌクレオチドLQGd/sに
連結した。結果として得られた連結されたフラグメントをNcoIで制限し、p
MRA−1のNcoI制限部位に挿入した。結果として得られたプラスミドをp
MRA−4と名付けた。上述のpM16の5.8kbのSphIフラグメントをS
phI(1926)でpMRA−4に挿入して全長メンゴウイルス配列を回復さ
せ、pMRA−5を作成する。pMRA−5をインビトロで転写させ、結果とし
て得られるRNAを用いてHeLa細胞に感染させ、ウイルスを生成させる。
vMQG−1の場合において、メンゴウイルスゲノム中への486の付加的な
塩基の挿入は、vMLN450の場合に459塩基の挿入が完全に生存可能な組
換えメンゴウイルスを生み出したことから、ウイルスの生存可能性と干渉する確
率は低い。非相同アミノ酸配列とLペプチドとの間への3C切断部位の導入と共
に、図14に示されているメンゴウイルスLの突然変異体L*のN末端にミリス
チル化シグナルを保持させた。さらに、L*内のN末端アミノ酸配列の変化は、
この変更がアミノ酸配列GNSによる最初の2つのアミノ酸(MA)の置換しか
含んでいないことから、L*ペプチドの機能に干渉しないものと予想される。N
末端アミノ酸の同様の欠失及び非相同アミノ酸との融合は、vMLN450の場
合、ウイルスの生存可能性に対する劇的な効果を全くもたないことが示された。
vMQG−1タンパク質の発現は、プロテアーゼ3Cによるタンパク質分解的
切断の後にタンパク質Δgp120−Q及びL*を生じるはずである。gp12
0−Qとgp120−VCNとの差は、タンパク質のC末端にある。
Δgp120−Qの全配列は、以下のとおりである:
Δgp120−Qの細胞での局在は、細胞質であり、L*ペプチドの局在とは
無関係であると予想されている。
さらに、この戦略は、Δgp120配列のN末端におけるシグナル配列又はそ
の他のあらゆる非相同インサートのクローニングに適用可能であるという情報を
提供し、外来性インサートの分泌を導く免疫原性の増大を結果としてもたらす可
能性が高い。

メンゴウイルスDNAの供給源としてのpM16−1
メンゴウイルス核酸供給源としてプラスミドpM16−1を用いても、前述の
例を実施することができる。pM16−1は、プラスミド領域内にヌクレオチド
10,931〜10,950の欠失を含んでおり、その転写物の感染力の増大と
いう結果がもたらされている。pM16−1の配列を、図17に示し、pM16
とpM16−1の配列比較を図18に示す。vMLN450のような組換えウイ
ルスの構築のために、pM16−1又はメンゴウイルスcDNAの
その他のあらゆるバージョンを使用することが可能である。
例10
組換えメンゴウイルスを用いた
リンパ球脈絡髄膜炎感染からのマウスの防御
1つのモデルとして、リンパ球脈絡髄膜炎ウイルス(LCMV)核タンパク質
(NP)からの充分に特徴づけされた免疫優性細胞傷害性Tリンパ球(CTL)
エピトープを使用した。ワクチニアウイルスへのこのエピトープの導入がBAL
B/c(H−2α)マウスにおける致死的LCMV感染に対する防御免疫の誘発
を可能にする、ということは以前に示されている。J.C.Whitton et al.,J.Vir
ology 67:348-356(1993)。9アミノ酸(aa)のCTLエピトープ;Pro Gln
Ala Ser Gly Val Tyr Met Glyを含むLCMV NPの14個のアミノ酸を発現
するメンゴウイルスキメラvLCMG4を構築した。
材料と方法
vLCMG4の構築と産生
DNA操作、転写、トランスフェクション、HeLa細胞の組織培養、及びメ
ンゴウイルスの産生は、上述のとおりに実施した。
細胞傷害性検定
基本的に、P.L.Gossens,H.JOUIN及びG.Milon,「マウスのリステリア症の
間に肝臓に動員されるリンパ球及び炎症性細胞の動力学」,J.Immunol.,147,
3514-3520により記述されているとおりに、免疫した動物からの肝臓中で、LC
MVに対する一次的細胞傷害性活性を調べた。
各々の肝臓を、10mlのHBSS+抗生物質を含む陶工用ガラス粉砕機中でホ
モジナイズし、ナイロンふるいにかけ、4℃において150gで10分間遠心分
離した。各ペレットを、0.03%トリプシン及び33μg/ml DNAse−1
に調整した45mlのHBSS中に再懸濁させた。混合物を10%ウシ胎児血清(
FCS)に調整する前に、細胞を、穏やかに振とうしながら37℃で45分間イ
ンキュベートした。細胞をHBSSで2回洗浄し、最終ペレットを、1mlの完全
RPMI 1640培地(10%FCS、1%L−グルタミン、4×10-5Mの
β−メルカプトエタノール)中に再懸濁させた。細胞を計数し、CTL検定のた
め適当な濃度に調整した。感染を受けていない又はLCMVで感染させたJ77
4標的細胞を、37℃で1時間、200μCiの51Crで放射線標識し、洗浄し、
希釈されたエフェクター細胞を含む、1ウェル当り105細胞/100μlの割
合で、96ウェルプレートに分配した。プレートを、37℃での4時間のインキ
ュベーションの前に、2分間100gの遠心分離に付した。もう1回遠心分離し
た後、上清を集め、ガンマカウンターで計数した。特異的細胞傷害性は、実験上
の放出−自然発生的放出/最大放出−自然発生的放出、という公式にしたがって
計算した。なお、ここで、自然発生的放出は、エフェクター細胞の不在下で決定
し、100%の放出は、1%トリトンの存在下で決定した。
免疫
Iffa-Credoから得た8週齢の雌のBALB/cマウスを、0.2mlのPBS又
は異なる用量のvM16、vLCMG4、もしくは
LCMVのアームストロング株(LCMV−ARM;Scripps Research Institu
te,ラ・ホーヤ、米国のOldstone博士から入手した)のいずれかを用いて、腹腔
内で免疫した。LCMV−ARMは、American Type Culture Collection(米国
基準株収集機関)12301 Parklawn Drive,Rockville,MD 20852に受託番号AT
CC VR−134として寄託されている。免疫後(p.i.)10日目に、一
次防御応答について試験するため、そしてp.i.43日又は45日目に、記憶
応答について試験するため、30μl中の102.8pfuのLCMV−ARMを用い
て、マウスに頭蓋内抗原投与した。
結果
vLCMG4の構築及び生育特性
117〜130としてLCMV NPをコードする二本鎖合成オリゴヌクレオ
チドを、メンゴウイルスcDNA pMCSの制限部位SnaBI(747)と
NheI(754)との間で、フレーム内に挿入し、プラスミドpLCMG4を
生成した(図1)。プラスミドDNAを、突然変異を受けた領域を通して配列決
定した。pLCMG4のRNA転写物をHeLa細胞にトランスフェクションし
、組換えウイルスvLCMG4を生成した。トランスフェクション及び感染の上
清からのウイルスは、親であるvM16又はvMCSと同じプラーク表現型を示
した(図2)。HeLa細胞における継代後、vLCMG4は、vM16又はv
MCSに匹敵する高い力価まで生育する。
LCMV特異的細胞傷害性
PBS及びvM16で免疫した動物が同系のLCMV感染を受けたJ775標
的細胞に対していかなる溶解活性も示さないのに対して、LCMV及びvLCM
G4で免疫した動物は、低いエフェクター/標的比においてさえ特異的な細胞傷
害性を示す(表6)。
LCMV抗原投与に対するBALB/cマウスのワクチン接種単一用量による
免疫
106pfuのvM16又はvLCMG4、2×105pfuのLCMV−ARM、又
はウイルス無しの対照としての組織培養培地を用いて、腹腔内でマウスを免疫し
た。「材料と方法」の中で記述されているとおりに10日目又は43日目にLC
MVの頭蓋内注射により、動物に抗原投与した。死亡は、全て10日目以前に起
こった。vM16又は培地を与えた動物については、防御は全く得られず、一方
、LCMV及びvLCMG4で免疫した動物については完全な防御が観察された
(表7)。
用量依存性防御
vLCMG4での有効なワクチン接種のための最小必要用量を評価するために
、異なる用量のvLCMG4を用いてBALB/cマウスを免疫した。100%
の防御は、免疫から10日後に100pfuという低い用量で観察でき、10pfuで
動物を免疫した場合、防御は、なお60%(5匹のうち2匹死亡)であった。免
疫から45日後で、防御レベルは非常に高い状態にとどまり、10、103及び
104pfuで単一の死亡例が見られた(表8)。
考察及び結論
Mengo−LCMV組換え体vLCMG4について得られた結果は、HIV
組換え体vMLN450について得られた結果を確認し、拡張している。すなわ
ち、
1)14アミノ酸の外来性配列がメンゴウイルスゲノムに挿入されたが、生存可
能性の損失はなかった。vLCMG4はvM16及びvMCSに匹敵する高い力
価まで生育し、プラーク表現型は親ウイルスについてと同じものである。
2)LCMVに対してvLCMG4で免疫された動物において、特異的免疫応答
を誘発することができる。インビトロでのエフェクター細胞の二次抗原刺激なし
に、免疫された動物の肝臓中に、LCMV感染標的細胞に対して強い一次CTL
活性を検出することができる。外来性抗原に対する体液性応答を誘発するメンゴ
ウイルスの能力に加えて、この結果は、非相同配列に対する細胞性免疫応答を誘
発するためのベクターとしてメンゴウイルスを使用する可能性を確認するもので
ある。
3)vLCMG4での免疫の後、動物は、致死用量のLCMV抗原投与に対して
防御された。
考察
ポリオウイルス、ライノウイルス又はメンゴウイルスのようなピコルナウイル
スは、そのRNAゲノムが感染を受けた細胞の細胞質内で排他的に発現されるこ
とから、組換えワクチンの開発にとって魅力あるモデルである。我々は、新しい
ウイルスベクターとしてのメンゴウイルスを開発した。メンゴウイルスは、カル
ディオウイル
スであり、脳心筋炎ウイルス(EMCV)、コロンビアSK及びMaus-Elberfeld
ウイルスと同じ血清型を共有している(1、2)。メンゴウイルスは、霊長類を
含む広範囲の動物種の体内で複製することができる(3、4)。遺伝子工学は、
メンゴウイルスの病原性能力がゲノムの5′非コーディング領域内のホモポリマ
ー状ポリ(C)トラクト(C50UC10)により支配されるということを示してき
た(5、6)。ポリ(C)トラクトの短縮又は欠失は、細胞培養で容易に増殖で
き、復帰突然変異に対して高い耐性をもつ、安定した形で弱毒化されたメンゴウ
イルス株を導く。
全てのピコルナウイルスと同様に、メンゴウイルスゲノムは、一連の成熟した
構造及び非構造タンパク質へとタンパク質分解によって切断される大きいポリタ
ンパク質をコードする(2)。
エンテロウイルス及びライノウイルスとは異なり、カルディオウイルス及びア
フトウイルスのP1キャプシド領域には、リーダー(L)ポリペプチドが先行し
ている。メンゴウイルスLポリペプチドの長さは、67アミノ酸である。ウイル
スに対するその機能的関連性は、それがアフトウイルスにおけるLタンパク質の
ようなプロテアーゼではないことから、わかっていない。ポリタンパク質からの
Lの遊離には、ウイルスゲノム内で下流にコードされる酵素であるウイルスプロ
テアーゼ3Cによるタンパク質分解が必要とされる。弱毒化されたメンゴウイル
スが、ウイルスベクターとして使用されうるか否か、そして組換え生ワクチンと
して役立つ潜在能を有するか否か、を見極めるために、我々は、Lポリペプチド
をコードするvM16ゲノムの領域へとHIV−IMN gp120の
V3−C4ドメインをコードするセグメントを工学処理した。結果として得られ
た組換えウイルスは、正常なメンゴウイルスタンパク質と共にgp120−L融
合タンパク質を発現し、免疫された動物においてHIV−Iに対する強い体液性
ならびに細胞性免疫応答を惹起した。
我々はここで、メンゴウイルスを免疫原性の外来性タンパク質配列の発現のた
めのベクターとして使用できるということを実証してきた。この場合、HIV−
IMN gp120からの147のアミノ酸を、メンゴウイルスのvM16株の
LポリペプチドのN末端へとフレーム内で融合させた。組換え体は、vM16に
比べて幾分か小さいプラークサイズ及び低いウイルス収量を示すものの、生存可
能であった。vMLN450ウイルスは、HIV−I配列を完全に保持しながら
細胞培養で少なくとも4サイクルについて安定した形で継代され得た。
ポリオウイルスゲノムのサイズを組換えバイシストロン構造の中で最高17%
まで増大させ、生存可能性は減少するものの、それでもなお包膜させることがで
きる、ということが報告されていた。野生型に比べて31%長いポリオウイルス
ゲノムは、包膜されない(13)。vMLN450のビリオンはHIV−I/メ
ンゴ組換え体RNAゲノムを担持するが、ウイルスキャプシドは親メンゴウイル
スのものと同一である。HIV−I配列は、その他のメンゴ非構造タンパク質に
ついて標準的であるように、感染中にのみ複製及び発現される。融合タンパク質
とP1領域との間のウイルス3C切断部位は、通常の要領で認識され、プロセシ
ングされる。5′非コー
