CD44エキソン6に対応するペプチド、そのペプチドに特異的な抗体、および
、腫瘍診断にそれらの抗体を使用する方法背景
本発明は、CD44遺伝子またはCD44遺伝子の一部の発現を用いて、腫瘍
形成を検査することに関する。このような検査には、体組織、体液、その他の試
料を患者から採取し、診断を行い、予後を定め、既に実行されている療法を評価
することを含む。特に、本発明は、患者に苦痛を与えずに得ることができる体液
試料またはその他の試料を用いて、腫瘍形成をルーチンとしてスクリーニングす
る単純な方法を提供する。
現在、腫瘍を診断する通常の方法は、細胞または組織薄片を顕微鏡で観察する
ことによって行われ、非常に効果的である場合が多いが、いくつかの顕著な制約
がある。試料が小さい場合、診断が非常に困難になって、多数の細胞が利用でき
なくなるが、患者から大型の試料を得ることは望ましくなかったり、不可能であ
ったりする。信頼できる診断を下すことができずじまいの例が50%にも及び、
明かに癌であるという証拠や、患者が実際に癌ではないという確信が持てない場
合がある。確たる診断方法を下すには、患者に苦痛を与えるような検査が必要で
ある。
予後の判定は、顕微鏡下で検視された場合の細胞の外観に依存する。一般に、
一次腫瘍の細胞が異様であるほど、転移の可能性が多くなるが、相関関係は決し
て完全ではない。何が最も効果的な処置であるかを判断するには、転移が起こる
可能性があるか否かを正確に予知できることが有効であることは明かであろう。
ヒトCD44遺伝子は、サイズの異なる種々のグリコシル化細胞表面タンパク
質をコードし、それらの機能の多くは未だ十分に確証されていないが、CD44
モノクローナル抗体(mAb)の存在によってそれと認識されるエピトープを共
有する。ヒトCD44遺伝子は、造血細胞や他の多くの細胞タイプに発現される
標準的な部分から成り、その中へさらにエキソンから成る生成物が種々に組み合
わされてスプライシングされ、様々なタンパク質を生成することが知られている
。これは、核生物においてはっきりと認められるメカニズムであり、機能上は関
係のない場合が多い複数のタンパク質を同じ遺伝子から生成するためのもので、
オルタナティヴスプライシングとして知られている。
これまでに、広く知られる2つのCD44のイソ型、すなわち、i)重クリコ
シル化された37kDの中心コアから成る90kD型と、ii)基部膜外ドメインに挿入さ
れた135個のアミノ酸を余分に有し、さらに重グリコシル化された180kD型が精製
され特徴付けられている(Stamenkovic et al. 1989)。免疫細胞化学的試験と
免疫沈降試験によって、多種多様な細胞および組織に90kD型と180kD型が広範囲
に分布することがわかっている。90kD型は、循環白血球、骨髄細胞およびその他
多数の細胞タイプに存在する造血型または標準型として知られている。180kD型
は、上皮変異型として知られており、複数の上皮細胞タイプで検出することがで
きる。両イソ型とも、同型接着相互作用および異型接着相互作用を仲介する受容
体として機能し、細胞を相互にまたは隣接細胞外スカホールドに接着すると考え
られる。
以前、ある種のHermes-3モノクローナル抗体の存在によって認識されたCD4
4エピトープは、循環白血球が周辺リンパ節を認識し通り抜けすることを可能に
する周辺リンパ節受容体を構成するものとして特定された。さらに、後にこの抗
原に対するモノクローナル抗体が利用できるようになり、Stamenkovic et al.(
1989)は、その一つを使用してCOS細胞の発現ライブラリーから取り出した標
準型分子をコードするcDNAをクローニングした。彼らは、さらにノーザンブ
ロット法によって、この遺伝子がリンパ系細胞だけではなく、種々の癌細胞系や
代表的な固形癌腫によっても発現され、その中で2つのクローン癌腫が正常なク
ローン上皮に比べ多く発現するように思われた。
Birch et al.は、ヌードマウスの場合、Hermes-3抗体によって認識される80-9
0kD型のCD44抗原を強く発現するメラノーマ細胞クローンは、CD44抗原
を弱く発現するクローンよりも相当転移性が高いことを報告した(1991)。Sy e tal.
は、ヌードマウスにおけるヒトリンパ腫細胞の転移能力は、上皮変異型をコ
ードする構成体によってトランスフェクトされた後ではなく、ヒトリンパ腫細胞
が標準型CD44遺伝子によってトランスフェクトされた後に穏やかに高まるこ
と
を述べた(1991)。Gunthert et al.は、ラットCD44抗原に刺激された新た
な抗体によって認識されるリンパ腫帰巣受容体の変異型は、ラット膵臓腺癌細胞
が転移行動するために必要であることを示唆する結果を得た(1991)。この抗体
を使用して、彼らは、CD44の変異型に対応するcDNA配列をクローニング
し、以前に確認されていないエキソンを含むことを発見した。転移細胞系に特有
のcDNA配列を過度に発現するよう設計された構成体によって同一細胞系由来
の非転移性クローンをトランスフェクトすると、転移行動が誘発されると思われ
る(Gunthert et al.、1991)。
上述の所見のため、種々の組織発生源から由来するその他の培養された転移性
ヒト腫瘍細胞系および非転移性ヒト腫瘍細胞系は、CD44生成物を特異的に発
現させた。細胞または組織における遺伝子の発現は、遺伝子生成物に対応するc
DNA部分に優先してアニーリングさせるために選択された特異的オリゴヌクレ
オチドプライマーを用いて、cDNAを細胞メッセンジャーRNAから生成し当
該の領域をポリメラーゼ連鎖反応によって拡大することによって最高の効率と感
受性で研究することができる。しかし、この方法を用いたHofmann et al.および
本出願人による後の研究では、CD44の発現は、ヌードマウスの転移能力、あ
るいは、上述の細胞培養系の腫瘍形成能力でさえ、終始一貫として確実に相関す
るわけではないという結果を得た。このころ、3つのグループ(Hofmann et al.
、1991;Stamenkovic et al.、1991;Jackson et al.、1992)は、彼らが種々の
ヒト細胞系におけるこの遺伝子によって発現されることを発見した別のスプライ
ス変異型に関する配列のデータを公表した。本発明
本発明は、乳癌患者および結腸癌患者とその転移部分から採取した新鮮組織試
料および新鮮体液試料におけるCD44遺伝子の種々の部分の発現を調査する試
験結果から生ずる驚くべき発見に基づく。試験結果は、CD44の発現について
i)転移(悪性)腫瘍、ii)非転移性の局部侵入性腫瘍および良性腫瘍、iii)正
常組織の間には、一目瞭然の差異があることを示した。グループi)とグループi
i)の差異は、療法の適否判断に重要であり、グループii)とグループiii)の差
異は、初期
の診断とスクリーニングに重要である。
本発明の一部は、1993年7月20日に提出された発明者らの国際特許出願PCT/GB
93/01520の対象であり、ひとつの態様として、腫瘍形成診断方法を提供し、その
方法は試料中のCD44遺伝子発現の分析から成る。
さらにひとつの態様として、上述の出願は、CD44遺伝子またはその一部か
ら成る生成物について試料を検定する方法を提供し、この検定方法は、cDNA
を試料中のメッセンジャーRNA(mRNA)から生成し、CD44遺伝子また
はその一部に対応する相補的DNA(cDNA)部分を増幅させ、増幅されたc
DNAの検出から成り、増幅されたcDNAを腫瘍形成診断に使用することを特
徴とする。
腫瘍形成の診断は、腫瘍組織の初期検出にほかならないとも言えれば、また転
移腫瘍と非転移腫瘍を識別するステップであるとも言える。従って、本出願で使
用される用語「診断」については、理解できよう。
この方法は、特に固形腫瘍、特に、具体的には癌などの悪性腫瘍の診断に利用
できる。検定が実行される試料は、体組織または体液であり、in vitro培養細胞
ではないことが好ましい。試料は、固形腫瘍から採った組織の小片または細胞の
微細針状吸引物(FNA)で良い。あるいは、血液、尿またはその他の体液から
成る試料や、頚管掻爬や、痰、尿または便など患者に苦痛を与えずに得られた試
料でも良い。
cDNAは、標識付けされた特異的オリゴヌクレオチドプローブを一つ以上使
用することによって検出され、その際、このプローブは増幅されたcDNA配列
の一部にアニーリングさせるように選択される。あるいは、標識付けされたオリ
ゴヌクレオチドプライマーおよびモノヌクレオチドまたはその一方を用いること
もできよう。放射性標識など、使用できる適切な検出可能な標識は多数ある。
次に添付の図面について説明する。
図1から図5までは、以下に報告される種々の実験結果を示すオートラジオグ
ラフである。
図6は、エキソン、プローブおよびプライマーを示すCD44遺伝子のマップ
である。エキソンの番号はG・R・Screaton et al.のつけたものと一致する。
図7は、図6に示されるエキソン6の核酸配列と、それに対応するアミノ酸配
列を示す図である。

図6は、エキソン6−14を示すCD44遺伝子のマップである。理論的には
、塩基性タンパク質すなわち標準タンパク質は、これらの9つの特別なエキソン
のいずれか1つ、複数またはすべてに由来する転写物の挿入によって修飾するこ
とができる。エキソン6は、本特許出願の優先日には未知であったが、本発明に
係る別の態様を構成するものである。エキソン6は、腫瘍において過度に発現さ
れるが、正常組織においては発現されず、エキソン7−9付近に位置する。エキ
ソン6の配列は、図7に示されている。この配列は、129の塩基対を含み、標準
CD44配列はこの配列の5'側にあり、エキソン7は常に3'側にある。
エキソン9−11とは対照的に、エキソン6から成る生成物(新たに配列され
たエキソン)は、正常組織の試料ではほとんど検出できない。これは、エキソン
6が腫瘍形成の診断において無類の価値があることを示唆する。
別の態様では、本発明は、新規の化合物として、図7に示すようなエキソン6
の核酸配列、その標徴フラグメント、縮重された配列および対立遺伝子変異を示
す配列またはその一方、相同核酸配列、プローブ、プライマーおよびそれらの配
列および相同体を用いてハイブリッド形成することができるその他の試薬を提供
する。これらの化合物および試薬は、すべて、上述の方法においても有用であろ
う。
本発明によれば、図7に示されるようなCD44エキソン6に対応するペプチ
ド配列、対立遺伝子変異、その二次修飾体、その標徴フラグメントを使用し、in
vitro診断とin vivo診断に有用な抗体を生成することができる。上記の抗体は
、図7に示されるCD44エキソン6、その対立遺伝子変異、二次修飾体および
その標徴フラグメントに対応するペプチドに特異的であり、すなわち、このよう
な
抗体はこのペプチドに結合し、その他関連するCD44タンパク質やその他のタ
ンパク質に対する交又感受が低い。上記の抗体は、モノクローナル抗体かポリク
ローナル抗体である。この抗体は、単数の抗原として図7に示されるCD44エ
キソン6に対応する完全なペプチド配列を使用することによって、また、複数の
抗原として、好ましくはCD44エキソン6に対応するペプチド配列の部分をコ
ード化する最低の長さである6個のアミノ酸から成る短いペプチドを使用するこ
とにによって生成することができる。抗原として使用されるペプチドは、当業者
に知られている方法によって、組換えてまたは化学的に生成することができる。
例えば、本発明によるペプチド抗原は、Merryfield,JACS 85(1964),2146に
従って合成することができる。免疫原合成の場合、これらのペプチドをキーホー
ルリンペットヘモシアニン(KLH)またはウシ血清アルブミン(BSA)など
の担体分子に結合することができる。ビオチン化が必要な場合、これは、PNAS U
SA 80(1983),4045などに従って実行することができる。
CD44エキソン6またはその対立変異型に対応するペプチドを原核宿主に発
現させるには、CD44エキソン6の融合遺伝子または原核宿主における発現レ
ベルが高い遺伝子を持つ対立遺伝子変異を用意することが好ましい。例えば、HI
V2のタンパク質gp32をコード化する遺伝子の一部は、大腸菌(E.Coli)に適切で
ある。それによって、エキソン6またはその対立遺伝子変異に準じてペプチド配
列を一部として有する融合タンパク質が得られる。融合遺伝子のCD44エキソ
ン6遺伝子を複製するなどして、融合タンパク質などのCD44エキソン6に対
応するペプチドエピトープ数を増加させることも好ましい。
CD44エキソン6、その対立遺伝子変異、その二次修飾体、標徴フラグメン
トに対応するペプチド配列に対抗するポリクローナル抗体は、適切な実験動物に
効果的な量のペプチドまたは抗原成分を注射し、その動物から血清を採取し、既
知の特異抗体吸着体技術によって特異抗原を単離することによって、調製される
。この方法によって生成されるポリクローナル抗体は、免疫検定のいずれにも使
用することができるが、一般には不均質になることがあるので、あまり好ましく
はない。
in vitro診断テストにモノクローナル抗体を使用することは、大量の抗体すべ
てが同様の特異性を持つように生成することがてきるので、特に好ましい。モノ
クローナル抗体生成のためのハイブリドーマ細胞系の調製は、不死化された細胞
系と抗体産生リンパ球を融合することによって行われる。これは、当業者によっ
て知られる既知の技術によって行われる(Harlow,E.and Lane,D.,Antibodie
s:A Laboratory Manual,Cold Spring Harbour Press 1988,Bessler et al.I
mmnobiol.170(1985),239-244,Jung et al.,Angew.Chimie 97(1985),8

と)。
図7に示されるCD44エキソン6の発現産物または標徴エピトープなどのそ
のフラグメントに対抗する産生モノクローナル抗体である以下のハイブリドーマ
細胞系は、DSM(Deutsche Sammlung von Mikroorganismen und Zellkulturen
GmbH,Mascheroder Weg lb,D-3300 Braunschweig,Federal Republic of Germ
anyにおけるブダペスト条約に基づき1993年11月16日に寄託された。

MAK〈CD44〉M-1.1.12については、アミノ酸9-23に対応する合成ペプチドを、M
AK〈CD44〉M-2.42.3についてはアミノ酸29-43に対応する合成ペプチドを、MAK〈
CD44〉M-4.3.16については図7に示すアミノ酸配列を有するCD44エキソン6
のアミノ酸1-13に対応する合成ペプチドを使用した。細胞系MAK〈CD44〉M-1.1.1
2によって生成される抗体は、肺腫瘍、結腸腫瘍、膀胱腫瘍と、免疫組織化学に
よって検出された細胞系ZR75-1(ヒト乳癌種−−ATCC CRL 1500)の細胞に特異
性を示す。特異的反応と表現したのは、強い反応が腫瘍組織を用いて観察される
が、正常組織では弱い反応しか示さないということである。同じ系統では、MAK
〈CD44〉M-4.3.16によって生成される抗体が、結腸腫瘍の組織およびZR75-1細胞
に特異性を示す。
CD44タンパク質または試料におけるCD44エキソン6に準ずるペプチド
配列の存在は、免疫検定のタイプを問わず実質的にはモノクローナルまたはポリ
クローナルのいずれかの上述のように調整された抗体を使用して検出することが
できる。さまざまな免疫検定技術が、Harlow et al.の参考文献(Antibodies: A
Laboratory Mamual,Cold Spring Harbour Press 1988)に見られるように利用
できる。これには、当然、非競合的なタイプの単一点検定、二点検定、「サンド
イッチ」法や、競合的な結合測定法などを含む。サンドイッチ法は、その中で最
も有用で一般に使用されている検定方法である。多数のサンドイッチ法の変種が
存在し、そのすべてが本発明によって含まれるように意図されている。上述の検
定の例には、ラジオイムノアッセイ、エンザイムイムノアッセイ、FPIAやエ
レクトロケミルミネッセンスなどの免疫蛍光検定、金ゾル粒子などの色素粒子な
どの直接標識を用いる免疫検定、CEDIAあるいはEMITなどの均質検定、
ラテックス粒子凝集検定のような濁度測定法、比濁測定法がある。サンドイッチ
法では、本発明に係る2つの抗体を使用することができる。この場合、この2つ
の抗体は、CD44エキソン6に応じたペプチド配列の異なるエピトープまたは
部位に結合しなければならない。これら2つの抗体は、例えば、異なる2つの合
成ペプチドをCD44エキソン6に対応するペプチドの異なる標徴フラグメント
に対応する免疫原として使用することによって、調製することができよう。サン
ドイッチ法では、本発明に係る抗体一つだけを用いることもできる。もうひとつ
の抗体は、別のCD44エキソンに対応する別のペプチドまたはCD44の標準
型に対応する別のペプチドに対する抗体であっても良い。このような抗体は在来
技術の中にあることが知られている。
例えば、尿、全血、子宮頚部スミア、便や、試料としての生検用体組織、痰細
胞などを使用することができる。多くの場合、CD44タンパク質は、天然の形
態で検出することができる。好ましくは、CD44タンパク質は、本発明に係る
抗体には線状エピトープまたはその天然の形態でCD44分子内に隠ぺいされて
いるエピトープに好んで結合するものがあるので、検出の前または間に変性させ
ることが好ましい。変性方法としては、デタージェントまたはカオトロピック剤
を用いる処置など当業者に知られている方法が適切である。ある場合には、CD
44分子が固相に吸着することによって、抗体の結合に十分な部分的な変性が起
こる。
CD44タンパク質は、ほとんどの細胞タイプで発現されるが、本発明に係る
抗体を使用した場合、腫瘍組織と正常組織の識別が多くの場合に可能であること
が認められた。従って、この抗体を癌診断に使用することができる。抗体は、結
腸組織、膀胱組織または肺組織の癌診断に使用することが好ましい。例えば、ハ
イブリドーマ細胞系MAK〈CD44〉M-1.1.12から得られた抗体を用いた場合、強い
反応が、結腸癌組織、膀胱癌組織、肺癌組織を用いた免疫組織化学的方法に観察
されるが、これに対し、前記の部位から採られた正常細胞組織は、かすかに反応
するだけである。
本発明に係る抗体を、Wong et al,Arch.Surg.125(1990),187-191に従っ
た方法などの免疫複合体分析に使用することもできる。このため、CD44タン
パク質またはその標徴部分および自己抗体の腫瘍関連免疫複合体の検出が可能で
ある。
本発明に係るペプチド抗原は、CD44定量法のための免疫テストに標準化合
物として使用することもできる。従って、本発明は、さらに、本発明に係るペプ
チド抗原をCD44を定量するための免疫テストにおける基準として使用するこ
とに関する。ある場合、凝集テストなどでは、配列の異同を問わず本発明に係る
複数のペプチドを担体分子に結合させるためのは好都合である。本発明に係るペ
プチドは、競合的な免疫検定において本発明に係る抗体に対する結合相手として
使用することもできる。この場合、ペプチドは標識付けされ、当業者に知られる
方法によって直接もしくは間接に(ストレプト)アビジン/バイオチンなどの特
異的結合相手を介し固相に結合される。
本発明のもう一つの態様は、緩衝剤、デタージェント、安定剤、固相などの免
疫検定に必要なその他の化合物がある中で、図7に示されるアミノ酸配列を有す
るCD44エキソン6、その対立遺伝子変異、その二次修飾体、その標徴フラグ
メントに対応するペプチドに対抗すべく意図された抗体を少なくとも1つ含むテ
ストキットである。必要に応じて、本発明に係るペプチド抗原を標準として含む
こともできる。
さらに別の態様では、本発明は、腫瘍の療法およびin vivo撮像のための手段
を提供する。この手段に有用な薬剤は、現在の技術水準で製造することができる
。悪性疾患患者から得られた試料のCD44遺伝子の種々の部分の発現を調査す
る試験から得られたデータは、驚いたことに、CD44の可変部のエキソン6を
相当過度に発現することを示している。正常組織と比較して悪性腫瘍にエキソン
6
を含むCD44スプライス変異型が相対的に多量であり、正常組織と比較して腫
瘍細胞の表面上でエキソン6によってコード化されたペプチド配列を含むCD4
4タンパク質が増加することによって、治療、in vivo診断、in vitro診断、in
vivo撮像のための腫瘍特異抗原としてエキソン6によるコード化されたペプチド
配列を使用する道が開ける。
好ましくは、モノクローナル抗体(Kohler and Milstein(1975),Nature 15
6,495-497)またはその誘導体を診断または治療の目的で使用する。本発明では
、CD44のエキソン6によってコード化されたエピトープに対抗するモノクロ
ーナル抗体が提供される。さらに、これらの抗体が腫瘍細胞に選択的に結合する
ことを示すデータが提示される。
本発明に係る抗体は、図7に示すアミノ酸配列を有するCD44エキソン6、
その対立遺伝子変異、燐酸化生成物、グリコシル化生成物およびその標徴フラグ
メントに対応するペプチドを認識する。このような抗体は、その他のCD44エ
キソンに対応するその他のペプチドの存在下においても、CD44エキソン6に
対応するペプチドに特異的である。治療の目的上、この特異性は本発明に係る抗
体が、エキソン6によってコード化されるタンパク質以外のタンパク質にはわず
かしか結合しないだけに決定的である。この非特異的な結合は、本発明に係る抗
体が腫瘍治療またはin vivo診断に使用される場合に、大きな損傷が健康な細胞
に生じない程度にとどめなければならない。
抗体は、適切な方法でエキソン6タンパク質に結合している限りは、全体とし
て抗体そのもの、そのフラグメント(Fv、(Fv)2、Fab、Fab’、F
(ab)2など)、キメラ抗体、ヒューマナイズド抗体またはキメラヒト型抗体
を使用することができる。エキソン6ペプチドを特異的に結合するCDR領域ま
たはその部分しか含まない短連鎖フラグメントも、特に、抗体が標識付けされた
ものである場合に、適切である。
本文では、抗体をそのまま悪性疾患の療法に使用する(Hale et al.,Lancet2
(1988)1394-1399; Cobbold et al.,Prog.Clin.Biol.Res.(1990)333,139
-151)。別の方法では、抗体またはその一部を、毒素分子(immunotoxin)に接
合または翻訳融合するため、腫瘍細胞の特異的な死滅がもたらされる(Brinkman
n
et al.1991,Proc Natl.Acad.Sci.USA 88,8616-8620; Pastan et al.(199
1),Cancer Res.51,3781-3787; FitzGerald and Pastan(1989),J.Natl.C
ancer Inst.81,1455-1461)。本発明に係る好ましい実施例では、ポリペプチ
ド鎖のin vitro再結合、ハイブリッドハイブリドーマ生成またはdiabodyの構成
によって(Holliger et al.(1993),Proc.Natl.Acad.Sci.USA 90,6444-6
448; Holliger and Winter(1993),Current Opin.Biotechnol.4,446-449)
、構成することができる二重特異性抗体を腫瘍治療に使用する(Bonino et al(
1992),BFE 9,719-723)。
さらに、放射性物質または蛍光性物質に結合された抗体は、呼吸器系、胃腸系
、尿性器の癌腫や、眼球癌、皮膚癌などの腫瘍の検出および治療に好ましい(Pr
ofio(1988),Proc.Soc.Photoopt.Instr.Eng.907,150-156; Jiang et al
.(1991),J.Natl.Cancer Inst.83,1218-1225)。
免疫応答を阻止するために、ヒト由来の抗体に可及的に近似する抗体を使用す
ることが好ましい(Glaasy and Dillman(1988),Mol.Biother.1,7-13)。
例えば、このような抗体は、キメラ抗体、ヒューマナイズド(CDR移植)抗体
である。このような抗体は通常、げっ歯類のモノクローナル抗体から作られてい
る(Morrison(1992),Annu.Rev.Immunol.10,239-265; Winter and Milste
in(1991),Nature 349,293-299などを参照)。本発明に係る具体的に好まし
い実施例では、腫瘍特異性ヒト型抗体(Borrebaeck et al.(1988),Proc.Nat
l.Acad.Sci.USA 85,3995-3999; Borrebaeck(1988),Immunol.Today 9,3
55-359)が治療目的で使用される。さらに、ヒトMabsを、Griffithらによる
EMBO J.12(1993)725-734などに記載されているように、ファージ表示ライブ
ラリによって調製することが特に好ましい。
治療の目的上、エフエクター機能(ADCC、CDC)を提供する抗体を使用
することが特に好ましい(Bruggemann et al.,J.Exp.Med.166(1987)1357-
1361)。特に好ましくは、ヒト同位体IgG 1抗体を使用する。
免疫毒素に関しては、本発明に係る抗体をシュードモナス外毒素、ジフテリア
毒素や、その他の毒素に結合することが好ましい(FitzGerald and Pastan(198
9))。抗体をドキソルビシンなどの化学療法剤や、細胞障害効果を有する放射
性標
識物質に結合することも好ましい。
放射性物質または蛍光性物質を用いるなどしてin vivo撮像する場合、特に、
ヒトの抗体においては、本発明に係る抗体の接合体も好ましい。
本発明の治療用化合物は、既知の薬学的技術を用いて、脈管内、腹腔内、皮下
、筋内などに非経口投与することができる。本発明に係る活性薬剤成分は、液状
、粉末状、凍結乾燥状の形態で使用され、水、生理食塩水、水溶性デキストロー
ス、水溶性緩衝剤などの希釈剤や担体と組み合わせることができる。保存薬も添
加することができる。
選択された投与経路のいかんに関わらず、本発明に係る化合物は、当業者らに
知られる従来の薬学的に許容できる剤形に構成される。この化合物は、薬学的に
許容できる酸添加塩または塩基添加塩を用いて形成することもできる。さらに、
化合物またはその塩は、適切に水酸化させた形態にして使用することもできる。
選択された投与経路のいかんに関わらず、本発明に係る毒性がないが治療的に
効果的な量の一つ以上の化合物をいかなる治療にも使用することができる。治療
用の投薬方法は、体型、年齢、体重、性別、患者の医学的状態、腫瘍タイプ、投
与経路、治療に使用された特定の化合物などの種々の因子に従って選択される。
一般的な水準の医師ならば、既知の抗体治療方法の際に必要な効果的な薬剤量を
を即座に決定し処方することができる(Hale(1988),Cobbold(1990))。そ
のような処置では、医師は初め相対的に低量の投薬をし、後に、最大の反応が得
られるまで増量する。
腫瘍におけるCD44遺伝子の多数のスプライシングされた変異型が無秩序に
過発現するということは、特定のエキソンが特定の細胞試料によって過発現され
たり、過発現されなかったり、全く発現されなかったりするということである。
従って、どのようなエキソンによってでも発現されるペプチドに対する抗体を使
用する免疫検定は、間違った結果を導くこともある。従って、本発明は、腫瘍形
成の免疫学的診断のため、2つ以上のCD44エキソン好ましくは、9つすべて
のエキソンに対する抗体を混合使用する方法を含む。
以下の例では、ヒトCD44遺伝子の発現が、正常な体組織と比較すると、種
々の固形腫瘍において、一貫して弁別的に増加することが認められた。悪性(す
なわちすでに転移している)腫瘍は、局所侵入性腫瘍や良性の腫瘍とは、観察さ
れる変化のパターンおよび大きさにおいて異なる。試験は、46のの腫瘍から得ら
れた試料について行われ、そのうち、44の腫瘍が局所侵入性、あるいは転移性で
あり、2つの腫瘍が良性であった。CD44の発現に関する分析は、PCRを用
いて新鮮な外科的生検試料から抽出されたRNAの逆転写によって作成されたc
DNAを増幅することによって行われた。特異的にCD44の遺伝子のいくつか
の部分をアニーリングさせるオリゴヌクレオチドプライマーを選択すると、上述
の結果から、診断および予後の上で重要な意味を持つCD44遺伝子部分を増幅
することができる。
今述べたような修飾CD44の発現と腫瘍形成の間の強い関連を見たからとい
って、遺伝子の個々のエキソンのいずれもが腫瘍形成時または転移性腫瘍に進行
する際にのみ発現されることを示していると考える必要はない。近年、多くの実
験室で得られた証拠(Knudson 1985,Tarin 1992,Hayle et al 1992などを参照
)は、これらの異常プロセスは細胞増殖および移動などの正常な細胞活動をコー
ドする遺伝子の調節が妨げられた結果であろうと示唆する。従って、どのような
遺伝子または遺伝子部分でも、腫瘍形成または転移をプログラムするだけしか機
能を持ってないとは思われない。
正常組織のエキソン10/11から得られた転写物の本試験の所見は、転移能力が
ある腫瘍からのPCR生成物において放射性標識付けされたプローブE4をハイ
ブリッド形成するバンドの数およびシグナル強度が著しく増加した場合でも、こ
のエキソンが転移行動にのみ関与しているわけではないことを示唆する。従って
、別の裏付けとなる現象が、CD44エキソン10/11が発現して転移活動が行わ
れると考えるためには必要であると考えられる。それにも関わらず、このエキソ
ンから得られた転写物が転移性腫瘍から得られた試料において過度に発現したと
の観察結果は、予後判断の非常に有用な指標として有望である。
さらに調査しても、個々のエキソンのいずれかからの天然の(非突然変異の)
生成物が、腫瘍細胞にだけユニークに存在し、正常な細胞には存在しないことが
発見されるとは思われない。それどころか、CD44遺伝子座に関して今に報告
されたように、過発現と遺伝子の産物の不適切な組み合わせから成る遺伝子活動
の異常パターンが、悪性化に関与すると思われる。これらの変化自体が悪性変換
のために必要なのかもしれないし、またこのような変換を生じるその他の遺伝的
障害の結果なのかもしれない。たとえそうであるとしても、この問題を解決せず
ともこれらの技術を使用する観察者は、腫瘍形成範ちゅうのものなのか、それと
も非腫瘍形成範ちゅうのものなのか、試料を振り分ける方法に関して情報を得る
ことができる。
例 方法
直径0.5-1cmの新鮮な組織試料を乳癌および結腸癌の患者34名から治療時に除
去された外科的切除標本から得た。試料は、病理学的標本受入場所に到着後10分
以内に液体窒素で急速冷凍して、使用するまで液体窒素に保存した。診断用に切
除された組織にリンパ節転移巣および血行性転移巣があれば、その一部も収集し
た。正常な胸部組織、正常な結腸粘膜、胸の腫瘍に隣接する正常なリンパ節、正
常な肝臓も外科的に切除された標本やその他の非腫瘍形成状態との比較のために
切除された標本から収集された。正常の末梢血白血球を、10名の志願者から得て
、骨髄を3名の志願者から得た。腫瘍およびその臨床段階の組織学的特徴は、表
1に記載した。
組織試料からの細胞性RNAの抽出は全て、ChomizynskiおよびSacci(1987)
によって記載された方法に従って行われた。液体試料からの抽出は、Invitrogen
によって市販されているMicrofasttrackキットを使用した。cDNA合成および
その後のポリメラーゼ連鎖反応(PCR)による増幅は、このキットに供給され
る緩衝剤および試薬を用いてSuperscriptTM前増幅装置(BRL Life Technologies
Inc.,Middlesex,UK)を使用して行われた。すなわち、これは、テンプレート
および供給されたヌクレオチドトリホスフェートとしてRNA試料を使い、逆転
転写酵素を用いて行う一次鎖cDNA合成の最初のステップに関係する。後のP
CRの場合、各試料は、油を薄く塗って、5分間94℃で加熱し、核酸を変性させ
、PCRの30サイクルを、以下のサイクルパラメータで行った。すなわち、94℃
で1分間、55℃で1分間、72℃で2分間で行った。反応混合物にテンプレートc
D
NAが存在しない負の遺伝子制御を各バッチに対し型通りに実施した。ヒトCD
44cDNAに関して公表された配列情報(Hofmann et al,1991,Stamenkovic
et al,1991,Jackson et al,1992)(図6)を用いて発明者らが考案したプ
ライマーおよびプローブ配列は以下の通りであった。

P1は(1989年にStamenkovic et al.によって公表された配列のヌクレオチド
782および783の間の)標準CD44分子の挿入部位から324bp上流に起点を置き
、P4はこの部位から158bp下流に起点を置く。試料が標準CD44(いわゆる
造血CD44)を発現する場合は、これらのプライマーは、482bpのPCRフラ
グメントを生成し、試料が選択的にスプライシングされた転写物を含む場合には
、CD44およびその他のバンドの上皮形態に関しては878bpのPCRフラグメ
ントを生成する。10μlの各PCR生成物は、1.2%の寒天ゲル中で電気泳動され
、Hybond N+(Amersham UK,Little Chalfont,UK)ナイロン膜に移され、オリ
ゴヌクレオチド・プローブE4(=5'TGAGATTGGGTTGAAGAAATC-3')をハイブリッ
ド形成する(図6参照)。ブロッティングおよびオートラジオグラフィーを、検
出感度および解像力向上のために行われた。プローブは、ポリヌクレオチドキナ
ーゼの存在下でy32P-ATPで放射性標識を付けた。プレハイブリッド形成後、10%
のデキストラン、6×NET、5×Denhardt溶液、0.5%のNP40および100μg/ml
のサケ***DNA内で42℃で一晩のハイブリッド形成を行った。次に、フィルタ
ーを2×SSC、1×SSCおよび0.1%SDSを含めた0.5%SSC内で2度連
続各15分間42℃で洗浄した。フィルターをKodakX線フィルムに2-16時間暴露し
た。この後、フィルターは、プローブをストリッピングするために0.5%SDS
で煮沸し、別の放射性標識プローブすなわち発明者らがCD44の標準部分をア
ニーリングさせるように設計したP2(=5'CCTGAAGAAGATTGTACATCAGTCACAGAC)
(図6)で再ハイブリッド形成した。ハイブリッド形成、洗浄およびオートラジ
オグラフィに使用される条件は、上述と同様である。
小数の細胞を検出するための方法の感度校正は次のように行った。塩化アンモ
ニウム緩衝液(沈層赤血球1ml当たり50mlの溶菌緩衝液)を添加することによっ
て沈層赤血球を溶解した15分後に遠心分離することによって末梢血リンパ球(P
BL)は全て20mlの全血から精製した。白血球ペレットを4本の試験管に分け、
HT29結腸癌腫細胞の懸濁液(1ml当たり5000細胞)をそれぞれの試験管に0μl、
1μl、10μl、100μずつ入れた。次に、RNAすべてを抽出したが、各試験管の
収量は約20μgだった。cDNA合成は、各試験管から得られたRNAの4μgの
分別アリコートに対して行い、分別した試料一点当たりそれぞれの腫瘍細胞は、
0個、1個、10個、100個であった。PCRは、上述の試料、正の遺伝子制御(腫
瘍細胞のみ)、負の遺伝子制御(DNA無)に対して、発明者らが図6のエキソ
ン6および14を特異的にアニーリングさせる設計をしたプライマーD1および
D5を用いて行われた。先の試験から、HT29細胞は、両方のエキソンやその他の
エキソンをPBLから容易に区別できるパターンで発現し、発明者らは増幅すべ
きセグメントを短縮することによって感度を高めたかったので、オリゴヌクレオ
チドプライマーD1およびD5を選択した。これらのプライマーを使用すること
で、この特異的な目的のためにプライマーP1およびP4を用いる問題が回避で
きると結論されたが、その理由は、これらの大半が遺伝子の標準部分をアニーリ
ングさせることによって浸漬されるからである。PCRのサイクルパラメータ、
ブロッティング条件、プローブ条件、洗浄条件は上述の通りである。プローブに
使用されるオリゴヌクレオチド配列は、32P標識付けしたE4であった。結果の概観
プライマー(P1、P4)は、スプライス生成物挿入部位を越えて増幅するの
で、標準分子の介在部分に加え、付加的なエキソンド・メインから得られた転写
物を含む選択的にスプライシングされた変異型のいずれかが増幅される。このた
め、この座から同定される配列の可能な組み合わせすべてを考慮すれば標準型に
対するプローブ(例えば、P2)などを用いて検出できると思われる生成物の総
数は大きい。プローブE4を用いて、上述の16の組み合わせ、すなわち、エキソ
ン11から得られたE4転写物を含む組み合わせは、電気泳動によって解像され
る異なる分子サイズのバンドとして視覚化することができると思われる。実際に
は、可能な組み合わせのすべての範囲を上述の結果では検出できなかったが、複
数(9つまで)の選択的スプライシング変異型が各プローブでハイブリッド形成
した腫瘍形成組織において観察された。胸部、結腸部、およびリンパ節から得ら
れた体組織は、ある種のE4含有転写物(図1および図3)の他、標準分子(図
2および図4)も発現したが、末梢血リンパ球(図5)および肝蔵(図4)は、
プローブおよびプライマーのこの組み合わせを用いた場合には、後者のみ検出可
能状態で発現した。
例1 胸部組織資料
胸部組織試料の試験から得られた結果を図1および図2に示す。転移腫瘍沈着
物とすべての症例から得られたそれら沈着物に対応する原発腫瘍は、エキソン1
1から得られた転写物含有の複数の選択的スプライシング生成物を過発現した(
図1a)。少なくも、8つのバンドが、すべての腫瘍において1500bpと1650bpで
存在する二重縞と一緒に頻繁に観察された。正常胸部組織および正常リンパ節は
、このプローブによって2つのバンド(1150bpおよび860bp)を呈した。上述の
二重縞は、正常試料には一切認められなかった。
バンドの数、サイズ、結合プローブからの信号強度の差異は、正常範ちゅうと
悪性範ちゅうの間では、試験されたすべての試料において明かであった。偶々採
取した予定外の試料については、長期間X線フィルムにフィルターを暴露し、弁
別的な差異を観察しなければならなかったが、この所見は試験されたすべてにつ
いて確認された。
転移関連の臨床的証拠がない局所侵入性腫瘍から得られた試料と2つの繊維腺
腫から得られた試料も、正常組織に比べてエキソン10/11から得られた転写物を
含むスプライス生成物を過度に発現したが、この程度は、悪性腫瘍とその転移巣
を用いて得られた結果とは容易に区別された。正常組織に認められるパターンか
らの区別も容易である(図1b)。ただし、一つの試料だけは、悪性腫瘍と類似
す
る結果を示した(レーン14)(以下参照)。2つの繊維腺腫は非転移性癌腫か
ら得られたパターンと類似したバンド・パターン示し、胸部の嚢胞疾患例から得
られた試料は、正常の非腫瘍形成胸部組織のパターンと類似していた。これは、
本方法による最も確実な診断の第1の例である。組織片は、当直の死亡診断医に
よって良性腫瘍すなわち肉眼によるむ初期の視診では肉眼的外観上は繊維腺腫か
ら得られたものとして提供された。次に、この組織片は、そのcDNAのPCR
増幅後、明確に非腫瘍性であるとして特徴付けられ、その後、組織片顕微鏡検査
によってこれが確認された。
プローブE4によって認められる差異が有効であり、技術的な人工産物でない
ことを確認するために、同じフィルターがプローブP2でハイブリッド形成され
た場合に得られた結果を図2に示す。これは、i)すべての試験組織が遺伝子の
標準型を発現し、ii)エキソン10/11由来の転写物を含まないその他のエキソン
スプライス生成物が腫瘍および転移巣に存在し、iii)上述の差異がこの複合フ
ィルター上の種々のパネルおよびレーンのトラック(track)を不均等にローデ
ィングしたことによるものではなく、選択的なスプライシングから生じたもであ
ることを示す。この実験におけるすべての条件は、フィルターをX腺フィルムに
暴露する露光時間(図1の場合、10時間露光、図3では1.5時間)を除いて、
E4を用いてハイブリッド形成した場合の条件と同じである。
例2 結腸試料
結腸癌腫に関する所見は、乳癌に関する所見と一致した。このため、すべての
場合において、結腸癌腫組織は、正常の結腸粘膜やその他の正常組織よりもプロ
ーブE4よって強く標識付けられた分子量の大きいバンド数の増加を示した。乳
癌腫の場合のように、プローブP2によるハイブリッド形成は、分子の標準型の
発現度において差異を示さなかった(図4)。
例3 方法の感度の校正
既知数のHT29結腸癌腫細胞を接種した末梢血白血球のPCR産物のオートラジ
オグラムを精査した結果、107個の白血球中の腫瘍細胞が10個のレベルまで腫瘍
細胞特有のバンドの存在を示せることがわかった。検定条件を微細に調整するこ
とによって、10mlの血液中のただ1つの腫瘍細胞をも検出することが可能である
と考えられる。
上述のシリーズでは、腫瘍形成細胞の試料すべてが、CD44遺伝子の選択的
スプライシングされた生成物を過発現し、非腫瘍性組織から得られた試料では、
全く発現を示さなかった。従って、正常組織由来または腫瘍組織由来の試料とC
D44の発現パターンは完全に対応した。ある例では、転移巣の臨床的証拠が現
在ない患者(患者B16、図1のレーン14)から除去された腫瘍は、転移能力
を示す発現パターンを有することが認められた。現在、これが偽陽性であるのか
、転移が差し迫っている徴候であるのかを知ることは不可能である。この患者は
、現在フォローアップクリニックで観察下にある。

E1およびE2は、エキソン6上に存在する。
直径0.5-1cmの新鮮な組織片試料を外科的切除標本からまたは剖検時に得た。
この研究に使用される試料はすべて診断試料が採取された後に残存する組織残部
から得られたが、本来ならば廃棄されたはずのものである。試料は、病理学的標
本受付場所に到着後10分以内に液体窒素で急速冷凍され、使用されるまで液体窒
素で凍結される。cDNAは、テンプレートとして細胞RNAの全量5μgを用い
てウイルス逆転転写酵素によって合成し、プライマーP1およびプライマーP4
を用いるPCRを行った。PCR増幅、電気泳動およびハイブリッド形成は、標
準条件下で行われた。
PCR産物を放射性標識付けしたE2またはE4によってハイブリッド形成し
た場合、癌腫から得られた試料すべてが複数のスプライス変異型を過発現したが
、各プローブによって観察されたバンドのパターンは異なった。このため、エキ
ソン6生成物に対するオリゴネクレオチドプローブは、腫瘍形成試料を非腫瘍性
試料から区別する場合に有効であるが、少なくとも固形組織から得られた試料に
ついてはE4に比べて著しく感受性が大きいというわけでなく、播種性転移細胞
または確定した転移巣の由来器官を検出するために有用であろう。その後、同じ
フィルターをストリッピングし、プローブP2によってハイブリッド形成したと
ころ、正常組織を含むすべての試料がCD44の標準部分を産生することがわか
った。これによって、E2およびE4を用いて探査された正常試料および腫瘍試
料を用いて得られた結果間に観察された差異は、PCR生成物の不均等なローデ
ィングによるものではないことが確認された。累積結果を表3にまとめたが、こ
れは、これらのの変化がさまざまな普通型癌に観察されることを示すものである
。
発明者らは、ある種の骨格筋の悪性腫瘍を調査し、骨肉腫にいて多数のスプラ
イシングされた変異型が著しく過度に発現することを観察した。
例5 臨床的に収集された尿試料のPCR検定による 癌診断
約50mlの自然排尿を各人から得て、実験室に可能な限り急速に運んだ。患者90
名から得た標本を試験したが、その内訳は、生検済膀胱癌患者から44例と膀胱が
非腫瘍性炎症(膀胱炎)患者および正常志願者46から得られた46例である。各尿
試料を完全に混和した後1mlずつ採取し、細胞学的検査を行った。別の1mlの尿を
フルオレセイン・ジアセテート・エチジウム・ブロミド染色によってチェックし
、試料中の細胞の生存度を評価した。残りの尿を2000rpmで、10分間、遠心分離
を行い、細胞ペレットを使用するまで-70℃で保持した。mRNAの抽出をオリ
ゴdTセルロース錠(Invitrogen)を用いて行った。cDNAは、AMV逆転写
酵素(Invitrogen)を用いて合成された。完成したcDNA溶液を均等に2本の
試験管に分配したが、一方はE1およびE5を用いたPCR用であり、特定のc
DNA転写物を増幅し、発明者らは、診断価値の高いことを認め、他方は、P1
およびP4を用いるPCR用であり、内的制御物としてスプライス変異型すべて
を有するか有しないCD44の標準型を増幅した。
PCRの35サイクルを実行した。サイクル条件は、95℃1分間、55℃1分間、
72℃2分間であった。「ホットスタート」法をすべての試料に採用した。結果は
図8に示されている。
等しい容積のPCR生成物を1.2%の寒天ゲルの各レーンにローディングしエ
チヂウムブロミドで染色した。尿中の細胞がエキソン6からエキソン14までの
すべてを発現するものであれば、現在のPCRプロトコルの場合、プライマーE
1およびE4を使用することによって735bpのバンドを生じることが予想された
。正常尿から得られたcDNAまたは非腫瘍性膀胱炎患者のcDNAを含むトラ
ック(track)にはバンドがないが(レーン1−8)、PCRをプライマーE1
およびE5を用いて行った場合(上部パネル)、鮮明な735バンドが膀胱癌患者
から得ら
れた尿試料すべてにおいて観察されている(レーン9−16)。
CD44型の標準型を表す482bpのバンドは、PCRをP1およびP4を用い
て実行した場合(下部のパネル)、すべての場合にほぼ均等に得られた。これは
、膀胱癌患者から得られた尿と対照から得られた尿との間の診断的に有意な差異
は、トラック(track)の不均等なローディングによって引き起こされるのでは
なく、CD44遺伝子の選択的スプライシングによって生じることを示唆する。
レーン1−4:正常尿。レーン5−8:膀胱炎の尿。レーン9−16:膀胱癌患
者1−8から得られた尿。
全体の結果では、735bpバンドは、正常な7尿標本中7標本と膀胱炎の9尿標
本中9標本には存在しなかった。すなわち、0%の偽陽性である。また、膀胱癌
患者から得られた19尿試料中14試料(74%)は、陽性の結果(すなわち26%の偽
陰性)を示した。偽陰性の試料では、フルオレセイン・ジアセテート・エチジウ
ム・ブロミド染色によって示されるように生存可能な癌細胞が不足していた。
例6
患者12名から得られた便を本文に記載された技術によって検定した。結腸直腸
がん患者9名から得られた試料中5試料は、陽性の結果を示した。正常な患者3
名から得られた試料すべてが陰性の結果を示した。前処理を課さない細菌に満ち
た試料から得られた数値は、細菌生存可能診断検定法を容易に開発することがで
きると確信させるものである。
この方法を用いて腫瘍細胞を検出する発明者の更なる実験では、以下の観察結
果がその診断的ポテンシャルを調査するその他の実験に有用となろう。結果の信
頼性および再現性は試料から得られたmRNAの質および技術を実施する際のケ
アに決定的に依存することを重大な考察として認識すべきである。主な必要条件
は、質の高いmRNAが日常的に得られることを確実にし、内部標準をすべての
反応に用いてPCR増幅ステップをモニタリングして偽陰性の結果が出ることを
排除することである。偽陽性の結果はあまり多く観察されないのであれば、同じ
試料または更に採取した試料に対する複製検定とその他の臨床データを参照すれ
ば、偽陽性と認識することができるので、重要な問題ではない。
発明者らは、腫瘍細胞を含有する小型臨床試料の中から異常なCD44遺伝子
の活性を機械的にPT−PCR検出することを確実にするために必要な手順を検
討した。組織片試料をアリコートに分割し、その半数を液体窒素中で直後に凍結
し、残りは周囲温度で放置した場合、CD44スプライス変異型を検出する能力
が、標本処理の時間とモード次第ではどのように低下するかを示すことができる
。切除30分以内にmRNA抽出した新鮮試料は、最も信頼できる結果を示し、新
鮮組織片を周囲温度に放置された場合には、次の数時間で質が次第に低下する。
試料が最初に急速冷凍された場合には、解凍直後にRNAを抽出しさえすれば満
足できるものである。だが、最初の15分間で急速に低下し始め、大型の変異転写
物が最初に損失し最終的には標準型までも損失する結果が得られた。急速に冷凍
された細胞および組織片の試料が-70℃で保管された場合、mRNAが解凍直後
に抽出された場合でさえも、結果は4週間後に低下した。従って、冷凍中に破壊
された細胞から放出されたリボヌクレアーゼによるRNAの質低下は、この温度
でさえも、速度は遅いとは言え、継続する。さらに、RNA抽出用に採取された
試料は、壊死部分または繊維症部分から得られる場合には、生存可能な腫瘍組織
に観察される典型的な結果を得られないとも考えられる。このため、試料選択お
よび標本処理における注意は、信頼できるデータを得るために必要である。
このようなことから、発明者らは新鮮な試料が、mRNAを抽出するために凍
結または処理される前に24時間以上放置されないこと、解凍試料がmRNA抽出
のために処理される前に2時間以上放置されないことが好ましいと考える。
本文に記載される診断方法は、一日で行うことができ、数時間でおこなうこと
ができる場合もあり、自動化することもできる。この診断方法の使用は、さまざ
まな試料に関して多種多用な癌を検出するために、また、血液試料および尿試料
に関しても証明してきた。従って、発明者らは、この方法を癌のスクリーニング
および診断にとって便利で実践的な方法として提供する。原則として、この診断
方法は、癌の検出プログラムと予防プログラムに広く一般的に適用できるので、
疫学的価値および公衆衛生的価値がある。この方法の感度、特異度、簡易性を適
切に利用することによって、初期の癌診断の他、体内の疾患の程度の評価、処置
の効率の判定、腫瘍再発の早期検出ができるようになろう。図の記号説明

胸部組織片試料から得られたPCR生成物のオートラジオグラムをE4を用い
て探査そた(試料フィルターをX線フィルムに10時間照射)。パネルA:転移

矢印)は、トラック15、16、18では露光の長さにおいて容易に認識するこ
とができ、長い露光についてはその他すべての腫瘍含有トラックにおいて検出で
きるようになった。しかし、図では、強いシグナルを有するトラックがかぶり映
像となるのを防ぐためには短い露光がよいことを示す。パネルC:繊維腺腫瘍(
FA)および嚢胞性乳腺症(Cyst)。トラック21、22は、良性腫瘍試料
(試料B22、23)を含み、トラック23(試料B24)における非腫瘍性(
嚢胞性疾患)試料よりも多く発現した。図2
プローブP2を用いて探査された胸部組織片試料から得られたPCR生成物の
オートラジオグム(試料フィルターをX線フィルムに1.5時間露光)。この結果
は、図1に使用されたものと同じフィルターを、以前のプローブをストリツピン
グした後再探査することによって得られた。ここでは、i)図1に観察される差
異は、トラックの不均等なローディングによるものではなく、ii)分子の標準型
の発現は、どの変異型よりも量的に多く、iii)標準型は、調査されたすべての
組織片に発現され、iv)エキソン3転写物を含まない変異型も更に存在し、腫瘍
細胞に過発現される。1500/1650bpダブレットは、パネルAの腫瘍に認められる
が、パネルBおよびパネルCで検出できるようにするには長時間の露光が必要で
ある。

図7に示されるCD44エキソン6に対応するペプチド配列のアミノ酸1−1
3、9−23、19−33、29−43、1−43に対応する5つのペプチドを
、Applied Biosystems,Inc.社のモデル431Aペプチドシンセサイザーにかけて専
売の標準スケール(0.25mmol)Fmoc化学オプションを用いて9-フルオロエニルメ
チロキシカルボニル(Fmoc)化学固相ペプチド合成(Atherton and Sheppard,1
989)によって合成した。このために、403mgの4-(2',4'-ジメトキシフェニル
−Fmoc-アミノメチル)-フェノキシ樹脂(Rink,1987)を0.62mmol/gの樹脂と置
換して用いる。アミド樹脂は、最初のカップリングサイクル以前にN,N-ジメチル
ホルムアミド(DMF)中の20%のピペリジンを用いて処理することによって脱
保護化(Fmoc開裂)する。ペプチドを合成するには、4mol過剰な以下のFmoc-ア

カップリングを行う前に、アミノ酸誘導体をDMFに溶解し、1当量のN-ヒド
ロキシベンゾトリアゾル(HOBt)をN-メチルピロロリジノン(NMP)に添加し
、1当量のN,N'-ジシクロカルボジイミド(DCC)をNMPに添加することに
よって活性化させる。HOBt-エステルアミノ酸の20分間のカップリングをDMF
中で実行する。カップリングに続き、N-末端の脱保護化(Fmoc開裂)をDMFに
20%のピペリジンで3分、続いて10分処置することによって達成する。ペプチド
鎖は、活性化/カップリング/脱保護化サイクルを繰り返すことによって延伸す
る。後に免疫原合成に使用されるペプチドにN-末端アミノカプロン酸スペーサお
よびシステインを供給し、それによってペプチドは担体タンパク質につながれる
。スクリーニング試薬として使用されるペプチドに関し、異なるN-末端が合成さ
れ、3つのγ-アミノブチル酸成分、リシン、(リシンのε−アミノ基に取り付
けられている)ビオチンを含む。合成に続き、ペプチドは、トリフルオロ酢酸(
TFA)切除によって樹脂支持体から除去される。ペプチド含有樹脂を1時間室
温(RT)で20mlのトリフルオル酢酸、1mlの水、1mlのチオアニソール、0.5ml
のエタンジチオール、1.5gのフェノールを含む開裂カクテルと反応させる。アル
ゴン下で行われた酸-不安定側鎖保護基の除去は、上述のカクテル溶液内でさら
に2.4時間の反応時間後室温で完了される。短時間冷却した後、脱保護化された
ペプチドは、ジイソピルエーテルの添加によって沈降させる。沈澱物をろ過し、
ジイソプロピルエーテルで洗浄し、50%酢酸で溶解し、凍結乾燥させた。ペプチ
ドの純度は、逆相HPLC(カラム−Vydac 218tp54、C18、300Å、5μm、4.6
×250mm、移動相−A:0.1%TFA含有の水、B:0.1%TFA含有の水/アセ
トニトリル(35/65、容積/容積);勾配−90分間で0-100%B、流量−1ml/分、
検出−226nm)。純度が60%未満の上述のペプチドを逆相HPLC(カラム−Wat
ers DeltaPak C18、100Å、15μm、50×300mm、移動相−A:0.1%TFA含有
の水、B:0.1%TFA
含有の水/アセトニトリル(35/65、容積/容積);勾配−130分間で0.50%B、
流量−15ml/分、検出−226nm)。ペプチドの素性は、プラズマ脱着質量分析によ
って検証される。免疫原合成に使用されるペプチドに関する特徴的なHPLC維
持時間特性および質量分析データを表2に記載した。
II.担体タンパク質の活性化
免疫原分析に関して、キーホールリンペットヘモシアニン(KLH)またはウ
シ血清アルブミン(BSA)のいずれかの担体タンパク質をヘテロ二官能架橋試
薬であるN-スクシニミジル3-マレイミドプロピオネート(MPS)を用いてリシ
ンのε-アミンを介して修飾する。このことがスルフヒドリルペプチドが後に共
役される「ハンドル」付きの担体タンパク質を付与する。KLHの場合、10μM
のKLH溶液をpH8.35の0.1M炭酸水素ナトリウム溶液によって調製する。懸濁
液のpHは、8.3に調整され、簡単に遠心分離される。bicinchoninic酸(BCA)
タンパク質検定(Smith,et al.,1985)によってタンパク質濃度を測定した後
、ジメチルスルホキシドに含めた3000当量の0.3モルMPS溶液をかくはんされ
たKLH溶液に滴下し、室温で1時間反応させた。溶液のpHは、0.1モルのHCLに
よって7.0に調整され、活性化された担体タンパク質は、サイズ排除クロマトグ
ラフィーによって過剰のMPSから分離された(カラム−AcA202、IBF Biotechn
ics、5×12cm)室温;緩衝液−0.1MのKH2PO4/K2HPO4、pH7.0、0.1MのNaCl;流量
-6ml/分、検出−226nm)。タンパク質濃度は、BCAタンパク質検定によって再
度測定され、活性化KLHのマレイミドプロピオアミド(MP)置換の程度(KLH-M
P)をエルマン試薬DTNBを用いて測定した(Ellman、1959)。BSAに関し
ては、190μMのBSA溶液を0.1モルのKH2PO4/K2HPO4、pH7.0に含めて調製し、1
00当量のMPS(1,4-ジオキサン中40mM)に滴下した。反応混合物を2時間室温
でかくはんした後、サイズ排除カラムにローディングした。活性化されたBSA
(BSA−MP)を精製し、KLH−MPに似たような方法で分析した。20-35:
1および200-600:1の置換値は、活性化された担体タンパク質であるBSA−MP
とKLH−MPに関して各々機械的に達成された。
III.ペプチドと活性担体タンパク質の
共役
チオエーテル結合の形成によって、チオ含有ペプチドは、MP活性化担体に共
役される。BSA−MPの場合、pH7.0の0.1MのKH2PO4/K2PO4に含めた74μMのBS
A-MP溶液を(MPに関して)1当量の4mMペプチド溶液を同じ燐酸緩衝液に含め
て反応させた。溶液をゆっくりとかくはんし、室温で一晩反応させた。遠心分離
後、可溶性BSA-MPペプチド共役体をサイズ排除クロマトグラフィー(II.
節のと同じクロマトグラフィー条件)によって未結合ペプチドを分離した。タン
パク質共役体の分析には、BCAによるタンパク質濃度決定の他、エルマン試薬
を用いての未反応MP-基の残存数を確認することを含む。KLH−MPペプチ
ド共役体は、活性担体タンパク質およびペプチドの濃度それぞれ3μMと18μMを

組換HIV2(gp32)-CD44エキソン6抗原/免疫原の
製造
CD44遺伝子のエキソン6は、図7に示すように43個のアミノ酸から成るペ
プチドをコードする。
ペプチドおよびアミノ酸100個未満の小型タンパク質は、概して、む微生物の
細胞質発現によって組換えることができない。このため、大腸菌において容易に
発現可能な遺伝子(HIV2レトロウイルスのエンベロープタンパク質gp32の一部)
とCD44エキソン6-DNAを含む融合遺伝子を作成した。
融合タンパク質のCD44エキソン6エピトープを増加させるために、(CD
44のエキソン5の3つのC末端アミノ酸をコードする)適切なリンカーを用い
てDNAレベルでCD44エキソン6を複製した。
金属キレートアフィニィティクロマトグラフィによってより簡単に抗原/免疫
原を単離するために、6個のヒスチジン残基(コドン)をDNAレベルでHIV2(
gp32)-CD44エキソン6融合タンパク質のN-末端領域に挿入した。
組換DNA技術
標準的な方法を用いてMolecular Cloning: A laboratory manualCold Spring
Harbor Press,Cold Spring Harbor,New York(1989)においてSambrook,J.e t al.
によって記載されたようにDNAを操作した。分子生物学的試薬を、製造
業者のマニュアルに従って使用した。
HIV2(gp32)-部分的遺伝子(プラスミドpUC18 HIV2-gp32)の作成
HIV2-gp32遺伝子のアミノ酸48-162のコード部をオーバーラップ化学遺伝子合
成(overlap chemical gene synthesis)によって合成した後、プラスミドpUC18
を用いてサブクローニングした。プラスミドpUC18HIV2-gp32の生成と分類記載に
ついては、欧州特許出願第0 440 207号に開示されている。大腸菌
発現ベクターpDS56-6HIS-HIV2-gp32の
作成
以下のプラスミド作成では、アミノ酸配列MRGSHHHHHHTDPEFのN-末端(ポリ-Hi
sテイル)をコードする融合遺伝子と、選択されたHIV2-gp32抗原を作成した。
この目的のため、ベクターpQE-10を制限エンドヌクレアーゼBamHIおよびHindI
IIによって消化し、約3.4kbpの長さのBamHI/HindIII-pQE-10ベクターフラグメン
トを寒天ゲル電気泳動によって単離した。pQE-10ベクター[同義:pDS56/RBSIII
、6×His(-1)]は、ドイツ国のDiagenから供給され、Stuber,D.et al.(1
990)Immunol.Methods IV: 121-152に記載されているものである。別の調製では
、プラスミドpUC18 HIV2-gp32を制限エンドヌクレアーゼBamHIおよびHindIIIに
よって消化し、約400-bpの長さのBamHI/HindIII-HIV2-gp32フラグメントを単離
し、約3.4bpの長さのBamHI/HindIII-PQE-10ベクターフラグメントに結合した。
所望のプラスミドは、制限マッピングを用いた同定により、pDS56-6HIS-HIV2-gp
32であると判明した。大腸菌
発現ベクターpDS56-HIV2-CD44エキソン6の
作成
アミノ酸配列MRGSHHHHHHTDPEFに対するN-末端(ポリ-Hisティル)をコードする
融合遺伝子と、選択されたHIV2-gp32抗原と、CD44エキソン6抗原の2つの
コピーを作成した。
CD44エキソン6遺伝子の2つのコピーは、ポリメラーゼ連鎖反応(PCR
)によって生成した[Mullis,K.B.and Faloona,F.A.(1987)Methods Enzym
ol. 155: 355-350]。
最初のPCR反応では、塩基対の位置397-538から得られたCD44エキソン
5−6のDNA配列を増幅し(図9:HIV2(gp32)-CD44エキソン6融合遺伝子
のDNA配列)、適切な個別の部位に制限エンドフクレアーゼ開裂(BamHIおよ
びHaeIII)を形成した。増幅はサブクローニングされたCD44cDNA(エキ
ソン5−
11)および以下のプライマー対を用いて行った。

NA配列)を増幅し、テンプレートDNAとしてサブクローニングされたCD4
4cDNAエキソン5−11を用いるとともに下記のプライマー対を使用して、
増幅されたDNA配列に対して適切な単一の制限ヌクレアーゼ開裂部位を与えた
。

を3つのフラグメントで結合することによって約3.8bpの長さのBgIII/HundIII-p
DS56-6HIS-HIV2-gp32ベクターフラグメントとした。所望のプラスミドは、制限
マッピングによって同定され、PCR合成DNA領域は、DNA配列(構造:pD
S56-HIV2-CD44エキソン6)によってチェックされた。大腸菌
におけるHIV2(gp32)-CD44エキソン6抗原
の発現
大腸菌においてHIV2(gp32)-CD44エキソン6抗原を発現するために、大 腸菌
K12株RM82(メチオニン復帰突然変異体であるEX8654、Murry、N.E.et a l
.(1977)Mol.Gen.Genet.150: 53-61)を、HIV(gp32)-CD44エキソン
6発現プラスミドpDS56-HIV2-CD44エキソン6(耐性アンピシリン)およびI
acIリプレッサープラスミドpUHA1(耐性カナマイシン)を用いて形質転換した。
プラスミドpUHA1の生成および分類記載はStuber,D.et al.(1990)
45 Immunol.Methods IV: 121-152に記載されている。
RM82/pUHA1/pDS56-HIV2-CD44エキソン6細胞をDYT媒体(1%(w/v)酵母エ
キス、1%(w/v)Bacto Tryptone、Difco、0.5%塩化ナトリウム)内で、50mg/
lのアンピリシンおよび50mg/lのカナマイシンを用いて、550nmにおいて光密度が
0.6-0.9になるまで培養し、IPTG(最終濃度1-5mmol/l)を用いて誘導した。4-8
時間の誘導期の後、細胞を遠心分離によって収集し、pH6.8、10mmol/lの燐酸緩
衝液で洗浄し、さらに処理を行うまで−20℃で保管した。
1mlの培地から得られた細胞ペレット(RM82/pUHA2/pDS56-HIV-CD44エキソ
ン6細胞)を0.25ml,10mmol/l、pH6.8の燐酸緩衝液と1mmol/l EDTAに含めて再度
懸濁し、細胞をフレンチプレスによって機械的に溶菌した。遠心分離後、1/5容
量の5×SDS試料緩衝液:pH6.8、50mmol/lのトリス-塩酸、1%SDS、1%メルカプ
トエタノール、10%グリセロールおよび0.001%ブロモフェノールブルー)を上
澄みに添加した。不溶性細胞破片分画を0.3mlの1×SDS試料緩衝液中で6-8M尿素
を用いて
再懸濁した。次に、試料を5分間95℃でインキュベートし、遠心分離を行った。
その後、タンパク質をSDS-ポリアクリルアミドゲル電気泳動(PAGE)によって分
離して(Laemmli,U.K.(1070)Nature227: 680-685)、クーマシーブリリアン
トブルーR色素を用いて染色した。
大腸菌で合成されたHIV(gp32)-CD44エキソン6抗原(図10)は均質であり
、不溶性細胞分画にのみ観察される。HIV2(gp32)-CD44エキソン6抗原に
関する発現レベルは、大腸菌の総タンパク質に対して30-50%であった。大腸菌
由来のHIV2(gp32)-CD44エキソン6抗原の
調製
封入体(IB)の細胞溶菌および調製。
20g(生重量)のRM82/pUHA1/pDS56-HIV2-CD44エキソン6細胞をpH7.0、10
0m10.1mol/lのトリス-塩酸で0℃で再懸濁した。30mgのリゾチームを添加し、混
合物を20分間0℃でインキュベートした。次に、細胞を機械的高圧分散によって
完全に溶菌し、DNAを2mmol/lの塩化マグネシウムおよび1mgのデオキシリボヌ
クレアーゼを添加することによって30分25℃で消化した(Boehringer Mannheim
,Germany,Cat.No.154709)。次に、50m,160mmolのEDTA、6%のトリトンX100と
、1.5mmol/lの塩化ナトリウムをpH7.0で消化溶液に添加し、この混合物をさらに
30分間0℃でインキュベートした。不溶性成分(細胞破片およびIB)をSorvall
遠心分離器にかけて遠心分離した。ペレットを100ml0.1molの燐酸緩衝液内でpH8
.5で再懸し、25℃30分間インキュベートし、IB生成物を遠心分離によって単離
した。
金属キレートクロマトグラフィーを用いるHIV(gp32)-CD44エキソン6抗原
の精製
2.5gのIBペレット(生重量)を、25℃2時間かくはんすることによって、25m

室温で添加した。さらに洗浄ステップ後、基剤(DAB)を添加し試料を用いて
インキュベートした(15-20分)。水道水で洗浄後、核をヘマトキシリンで30分
間染色した。試料を乾燥させ、Cristal Mountに埋封した(Kaiserゼリー)。
細胞系MAB〈CD44〉M-1.1.12および4.3.16由来のモノクローナル抗体を用いて
得られた結果を表4に示した。方法Bでは、MAB〈CD44〉1.1.12は、肺、結腸お
よび膀胱由来の腫瘍組織に対して高い特異性を示し、MAB〈4.3.16〉は結腸由来
の腫瘍組織に対して特異性を示した。方法Aは、MAB1.1.12およびMAB4.3.16は、
細胞系MDA4A4(エキソン6低生成物)に比べ、細胞系統ZR-75-1(エキソン6高
