Claims (1)
- 【特許請求の範囲】 1.Saccharomyces cerevisiaeソルビトールデヒドロ ゲナーゼ遺伝子由来のDNAセグメントであって、ベクターが宿主酵母細胞によ って複製され保有されるようにベクター中で異種ポリペプチドをコードする遺伝 子に作動可能に連結されたとき、前記異種ポリペプチドの発現を増加するDNA セグメント。 2.前記セグメントの機能的活性部分が、 (a)Saccharomyces cerevisiaeのソルビトールデ ヒドロゲナーゼ遺伝子の転写調節配列、 (b)Saccharomyces cerevisiaeのソルビトールデ ヒドロゲナーゼ遺伝子の転写開始調節配列、及び (c)Saccharomyces cerevisiaeのソルビトールデ ヒドロゲナーゼ遺伝子の終結調節配列 を含む請求項1に記載のDNAセグメント。 3.前記転写調節配列がSaccharomyces cerevisiaeの ソルビトールデヒドロゲナーゼ遺伝 子のプロモーターであり、前記終結調節配列がSaccharomyces c erevisiaeのソルビトールデヒドロゲナーゼ遺伝子のターミネーターで ある請求項2に記載のDNAセグメント。 4.ソルビトールデヒドロゲナーゼポリペプチドのN末端の11個のアミノ酸残 基をコードするDNA配列を含む請求項1に記載のDNA配列。 5.前記機能的活性部分が、図1の核酸番号1〜771及び核酸番号1848〜 2760を含む請求項2に記載のDNAセグメント。 6.図1の核酸番号772〜805を含む請求項4に記載のDNAセグメント。 7.前記宿主酵母がSaccharomyces cerevisiaeである 請求項1に記載のDNAセグメント。 8.前記異種ポリペプチドがスーパーオキシドジスムターゼである請求項1に記 載のDNAセグメント。 9.前記異種ポリペプチドが肝炎デルタ抗原である請求項1に記載のDNAセグ メント。 10.酵母中で外来ポリペプチドを発現し得る発現ベクターであって、(a)S accharomyces cer evisiaeのソルビトールデヒドロゲナーゼ遺伝子の転写調節配列、転写開 始調節配列及び終結調節配列の制御下にあるDNA配列、及び(b)酵母複製系 を含む発現ベクター。 11.前記転写及び終結調節配列が、Saccharomyces cerev isiaeのソルビトールデヒドロゲナーゼ遺伝子のプロモーター及びターミネ ーターを含む請求項10に記載の発現ベクター。 12.ソルビトールデヒドロゲナーゼポリペプチドのN末端の11個のアミノ酸 残基をコードするDNA配列を含む請求項10に記載の発現ベクター。 13.前記DNA調節配列が、図1の核酸番号1〜771及び核酸番号1848 〜2760を含む請求項10に記載の発現ベクター。 14.前記DNA調節配列が更に、図1の核酸番号772〜805を含む請求項 13に記載の発現ベクター。 15.請求項10に記載の発現ベクターを用いて形質転換された酵母宿主細胞。 16.Saccharomyces cerevisiaeである請求項15に 記載の酵母宿主細胞。 17.酵母中で異種ポリペプチドを生産する方法であって、ベクター中で外来ポ リペプチドをコードする遺伝子に作動可能に連結されており、ソルビトールによ って誘導されるSaccharomyces cerevisiaeソルビトー ルデヒドロゲナーゼ遺伝子由来の調節ヌクレオチドセグメントを導入するステッ プを含み、それによって前記ベクターが前記細胞によって複製され保有され、前 記遺伝子が誘導条件下で発現される方法。 18.前記調節ヌクレオチドセグメントが、Saccharomyces ce revisiaeのソルビトールデヒドロゲナーゼ遺伝子の転写調節配列、転写 開始調節配列及び終結調節配列の制御下にある請求項17に記載の方法。 19.前記転写開始調節配列が前記ソルビトールデヒドロゲナーゼ遺伝子のプロ モーターであり、前記終結調節配列が前記ソルビトールデヒドロゲナーゼ遺伝子 のターミネーターである請求項18に記載の方法。 20.前記調節ヌクレオチドセグメントが更に、ソルビトールデヒドロゲナーゼ ポリペプチドのN末端の11個のアミノ酸残基をコードするDNA配列を含む請 求項17に記載の方法。 21.前記調節ヌクレオチドセグメントが、図1の核酸番号1〜771及び核酸 番号1848〜2760を含む請求項20に記載の方法。 22.前記DNA調節配列が更に、図1の核酸番号772〜805を含む請求項 17に記載の方法。 23.図1の配列またはその縮重均等物からなるDNA分子であって、該DNA 分子によってコードされるポリペプチドが酵母ソルビトールデヒドロゲナーゼの 生物学的活性を有するDNA分子。 24.Saccharomyces cerevisiaeから単離及び精製さ れた請求項23に記載のDNA配列。 25.図1に示したポリペプチド配列をコードする請求項24に記載のDNA配 列。 26.Saccharomyces cerevisiae由来のソルビトール デヒドロゲナーゼ活性を有する実質的に純粋な形態のポリペプチド。 27.図1に示したアミノ酸配列を有する、ソルビトールデヒドロゲナーゼ活性 を有する実質的に純粋な形態のポリペプチド。 28.SDH発現異種ポリペプチドまたはその特異的結合 メンバーを含むアッセイ試薬。 29.前記SDH発現異種ポリペプチドが酵素である請求項28に記載のアッセ イ試薬。 30.前記SDH発現異種ポリペプチドがスーパーオキシドジスムターゼである 請求項29に記載のアッセイ試薬。 31.前記SDH発現異種ポリペプチドが特異的結合対のメンバーである請求項 28に記載のアッセイ試薬。 32.前記SDH発現異種ポリペプチドが肝炎デルタ抗原である請求項28に記 載のアッセイ試薬。 33.試験試料中の特異的結合対のメンバーを検出する方法であって、試験試料 を、少なくとも1種のSDH発現異種ポリペプドまたはその特異的結合メンバー を含むアッセイ試薬に接触させることを含む方法。 34.前記SDH発現異種ポリペプチドが酵素である請求項33に記載の方法。 35.前記SDH発現異種ポリペプチドがスーパーオキシドジスムターゼである 請求項34に記載の方法。 36.前記SDH発現異種ポリペプチドが特異的結合対のメンバーである請求項 33に記載の方法。 37.前記SDH発現異種ポリペプチドが肝炎デルタ抗原 である請求項36に記載の方法。 38.試験試料中の特異的結合対のメンバーの存在を検出することに使用する試 験キットであって、少なくとも1種のSDH発現ポリペプドまたはその特異的結 合メンバーを含むアッセイ試薬を有する容器を具備する試験キット。 39.前記SDH発現異種ポリペプチドが酵素である請求項38に記載の試験キ ット。 40.前記SDH発現異種ポリペプチドがスーパーオキシドジスムターゼである 請求項39に記載の試験キット。 41.前記SDH発現異種ポリペプチドが特異的結合対のメンバーである請求項 38に記載の試験キット。 42.前記SDH発現異種ポリペプチドが肝炎デルタ抗原である請求項41に記 載の試験キット。
