新規タキサン誘導体、それらの製造法及びそれらを含む製薬学的組成物
本発明は一般式:
[式中、
−記号R1又はR2の一方は水素原子を示し、他方はヒドロキシル基を示し、
−R3は水素原子又は一般式:
の基を示し、ここで
Arはアリール基を示し、
R4はベンゾイル基又は基R5−O−CO−を示し、ここでR5はアルキル、ア
ルケニル、シクロアルキル、シクロアルケニル、ビシクロアルキル、フェニル又
はヘテロ環式基を示し、
−R6はアルキル、アルケニル、シクロアルキル、シクロアルケニル、ビシクロ
アルキル、場合により置換されていることができるアリール又はヘテロ環式基を
示す]
の新規タキサン誘導体に関する。
さらに特定的には、本発明はR、R1及びR2が上記と同義であり、R3が水素
原子又は一般式(II)の基を示し、ここで:
Arはアリール基を示し、
R4はベンゾイル基又は基R5−O−CO−を示し、ここでR5は:
−炭素数が1〜8の直鎖状もしくは分枝鎖状アルキル基、炭素数が2〜8のアル
ケニル基、炭素数が3〜6のシクロアルキル基、炭素数が4〜6のシクロアルケ
ニル基又は炭素数が7〜10のビシクロアルキル基を示し、これらの基は場合に
よりフッ素もしくは塩素原子、及びヒドロキシル、炭素数が1〜4のアルコキシ
、各アルキル部分の炭素数が1〜4のジアルキルアミノ、ピペリジノ、モルホリ
ノ、1−ピペラジニル(場合により4−位において炭素数が1〜4のアルキル基
もしくはアルキル部分の炭素数が1〜4のフェニルアルキル基により置換されて
いることができる)、炭素数が3〜6のシクロアルキル、炭素数が4〜6のシク
ロアルケニル、フェニル、シアノ、カルボキシル又はアルキル部分の炭素数が1
〜4のアルコキシカルボニル基から選ばれる1つ又は複数の置換基により置換さ
れていることができるか、
−場合により炭素数が1〜4のアルキル基及び炭素数が1〜4のアルコキシ基か
ら選ばれる1つ又は複数の基により置換されていることができるフェニル基を示
すか、
−あるいは場合により炭素数が1〜4の1つ又は複数のアルキル基により置換さ
れていることができる5もしくは6員の飽和窒素性ヘテロ環式基を示し、シクロ
アルキル、シクロアルケニル又はビシクロアルキル基は場合により炭素数が1〜
4の1つ又は複数のアルキル基により置換さ
れていることができると理解され、
R6は:
−炭素数が1〜8の直鎖状もしくは分枝鎖状アルキル基、炭素数が2〜8のアル
ケニル基、炭素数が2〜8のアルキニル基、炭素数が3〜6のシクロアルキル基
、炭素数が4〜6のシクロアルケニル基又は炭素数が7〜11のビシクロアルキ
ル基を示し、これらの基は場合によりハロゲン原子、及びヒドロキシル、炭素数
が1〜4のアルキルオキシ、各アルキル部分の炭素数が1〜4のジアルキルアミ
ノ、ピペリジノ、モルホリノ、1−ピペラジニル(場合により4−位において炭
素数が1〜4のアルキル基もしくはアルキル部分の炭素数が1〜4のフェニルア
ルキル基により置換されていることができる)、炭素数が3〜6のシクロアルキ
ル、場合により置換されていることができるフェニル、シアノ、カルボキシル又
はアルキル部分の炭素数が1〜4のアルキルオキシカルボニル基から選ばれる1
つ又は複数の置換基により置換されていることができるか、
−あるいは場合によりハロゲン原子及び炭素数が1〜4のアルキル基及び炭素数
が1〜4のアルキルオキシ基から選ばれる1つ又は複数の原子又は基により置換
されていることができるアリール基を示すか、
−あるいは場合により炭素数が1〜4の1つ又は複数のアルキル基により置換さ
れていることができる4〜6員の飽和又は不飽和窒素性ヘテロ環式基を示し、シ
クロアルキル、シクロアルケニル又はビシクロアルキル基は場合により炭素数が
1〜4の1つ又は複数のアルキル基により置換されていることができると理解さ
れる
一般式(I)の生成物に関する。
Ar及びR6により示されるアリール基は、場合によりハロゲン原子(フッ素
、塩素)又はアルキル、、アルケニル、アルキニル、アリール、アラルキル、ア
ルコキシ、アルキルチオ、アリールオキシ、アリールチオ、ヒドロキシル、ヒド
ロキシアルキル、メルカプト、ホルミル、アシル、アシルアミノ、アロイルアミ
ノ、アルコキシカルボニルアミノ、アミノ、アルキルアミノ、ジアルキルアミノ
、カルボキシル、アルコキシカルボニル、カルバモイル、ジアルキルカルバモイ
ル、シアノ及びトリフルオロメチル基から選ばれる1つ又は複数の原子もしくは
基により置換されていることができるフェニル又はα−もしくはβ−ナフチル基
であり、アルキル基及び他の基のアルキル部分の炭素数は1〜4であり、アルケ
ニル及びアルキニル基の炭素数は2〜8であり、アリール基はフェニル又はα−
もしくはβ−ナフチル基であると理解されるのが好ましい。
Arにより示されるヘテロ環式基は5員であり、窒素、酸素又は硫黄原子から
選ばれる同一又は異なる1つ又は複数の原子を含み、場合によりハロゲン原子(
フッ素、塩素)又は炭素数が1〜4のアルキル、炭素数が6〜10のアリール、
アミノ、炭素数が1〜4のアルキルアミノ、各アルキル部分の炭素数が1〜4の
ジアルキルアミノ、炭素数が1〜4のアルコキシカルボニルアミノ、炭素数が1
〜4のアシル、アリール部分の炭素数が6〜10のアリールカルボニル、シアノ
、カルボキシル、カルバモイル、アルキル部分の炭素数が1〜4のアルキルカル
バモイル、各アルキル部分の炭素数が1〜4のジアルキルカルバモイル又はアル
キル部分の炭素数が1〜4のアルキルオキシカルボニルから選ばれる1つ又は複
数の基により置換されていることができる芳香族ヘテロ環式基であるのが好まし
い。
さらに最も好ましくは、本発明はR1及びR2が上記と同義であり、R3が一般
式(II)の基を示し、ここでArは場合によりフッ素もしくは塩素原子により
、あるいはアルキル(メチル)、アルキル(メトキシ)、ジアルキルアミノ(ジ
メチルアミノ)、アシルアミノ(アセチルアミノ)、アルコキシカルボニルアミ
ノ(tert−ブトキシカルボニルアミノ)又はトリフルオロメチル基、あるい
は2−もしくは3−チエニル又は2−もしくは3−フリル基により置換されてい
ることができるフェニル基を示し、R4はベンゾイル基又は基R5−O−CO−
を示し、ここでR5はt−ブチル基を示し、R6が場合によりハロゲン原子により
置換されていることができるフェニル基を示す一般式(I)の生成物に関する。
R1、R2及びR6が上記と同義であり、R3が一般式(II)の基を示す一般式
(I)の生成物は顕著な抗腫瘍性及び抗白血病性を有する。
本発明に従えば、一般式(I)の新規生成物は一般式:
[式中、R3及びR6は上記と同義である]
の生成物の電解還元により得ることができる。
本発明に従えば、カルボニル基−CO−の還元は以下の反応に従って電気化学
的に行われる:
一般式(III)の生成物からの電解還元は、アンモニウムイオン(NH4 +)
を含む支持電解質及び一般式(III)の生成物が0.1g/lから溶液の飽和
までの濃度で溶解する溶媒又は溶媒の混合物から成る陰極液を含む電解槽(el
ectrolyser)において行われる。
一般式(III)の生成物において7−位のヒドロキシル基が7α形態にある
場合、本質的にR1が水素原子を示し、R2がヒドロキシル基を示し、7−位にお
けるヒドロキシル基が7α形態にある一般式(I)の生成物が生成される。
カルシウムイオンの存在中における酸媒体中で行う場合、一般式(III)の
生成物の電解還元は本質的に7−位におけるヒドロキシル官能基の立体配置に関
係なく、R1が水素原子を示し、R2がヒドロキシル基を示す一般式(I)の生成
物に導く。
一般式(III)の生成物から一般式(I)の生成物への還元を行うために理
論的に必要な電気の量は1モル当たり2ファラデー(又は193,000クーロ
ン)である。
還元はダイアフラム電解槽において行うのが好ましい。
本発明の方法の態様に従えば、電解還元は陰極、陰極室、分離ダイアフラム、
陽極室及び陽極を含む電解槽において行われ、それらの特徴は以下の通りである
:
a)陰極は導電性材料から成り、その上で溶媒又は支持電解質の成分の1つの還
元電位より大の電位において、あるいは溶媒又は支持電解質の成分の1つの還元
が生成物の還元と抵触するのに十分な程有意でない電位において還元が起こる。
b)陰極室は有機又は水性/有機媒体中の一般式(III)の生成物の溶液及び
アンモニウムイオン(NH4 +)を含む電解質から成る陰極液を含む。pHはアン
モニア水溶液の添加によりわずかなアルカリ性、すなわち好ましくは7〜9に維
持される。陰極液は、R1が水素原子を示し、R2がヒドロキシル基を示す一般式
(I)の生成物を選択的に得る目的で酸性であることもでき、例えば塩化カルシ
ウムにより与えられるカルシウムイオンを含むこともできる。
c)分離ダイアフラムは焼結ガラス又は磁器から作られた平板、管又はキャンド
ルなどの多孔質材料、又はイオン交換膜、好ましくはカチオン交換膜から成り、
d)陽極室は好ましくは陰極室で用いられたものと同一の溶媒又は溶媒の混合物
及び同一の支持電解質から成る陽極液を含む。陽極液はカチオン交換膜の介在に
より電解の間の陰極液のpHを維持できるようにする酸溶液から成ることもでき
る。メタノール中で希釈された塩酸の使用が好ましい。
e)陽極は導電性材料から成り、その性質は方法の実行にとって重要ではない。
一般に陽極は電解の条件下で攻撃され得ない導電性材料、例えば正味の、又は
導電性支持体上のブライト白金(bright platinum)、グラファ
イト又はガラス質炭素(vitreous carb
on)から成る。
陰極は水銀のシートから成るのが好ましい。
支持電解質は溶媒又は溶媒の混合物に溶解性の塩化アンモニウムなどのアンモ
ニウム塩から成る。一般に一般式(I)及び(III)の生成物を容易に溶解し
、一般式(I;R1=H及びR2=OH)ならびに(I;R1=OH及びR2=H)
の生成物を容易に単離できるようにするプロトン性溶媒が用いられる。溶媒はメ
タノール、エタノール、イソプロパノール又はt−ブタノールなどの炭素数が1
〜4の脂肪族アルコールから選ばれるのが好ましい。
pHは基質の安定性に適合しなければならず、電解をアンモニウムイオンの存
在中で行う場合はアンモニア水溶液の添加により、又はアンモニアガスを散布す
ることにより、それを電解の間わずかにアルカリ性に、すなわち好ましくは7〜
9に維持することができ、あるいは他の場合、電解を例えば塩化カルシウムによ
り与えられるカルシウムイオンの存在中で行う場合、塩酸の添加により酸性に維
持することができる。
陽極を陰極から分離しているダイアフラムの性質は本発明の必須の特性ではな
い。かくして反応物の拡散を制限する導電性ゲルを含む、又は含まない焼結ガラ
ス又は磁器などの多孔質材料、あるいはイオン交換膜、好ましくはカチオン交換
膜から成るいずれの既知の種類のダイアフラムを用いることもできる。膜は均質
な又は不均質な種類のものであることができ、場合により枠により強化されてい
ることができる。陽極液及び陰極液の種々の成分の存在中で膨潤せず、収縮せず
、安定な膜を用いるのが好ましい。
本発明の好ましい態様に従えば、陽極、陰極及び分離ダイアフラムは
平行な平面に沿って、好ましくは水銀のシートから成る陰極の場合水平に配置す
る。
電解浴(electrolysis bath)の温度は一般に0〜30℃で
ある。
電解は制御された電位で行い、それはカロメル参照電極に対して約−1.75
〜−1.9ボルト、好ましくは−1.85ボルトにおいて固定することができる
。
用いられる電気の理論的量は一般式(III)の生成物の1モル当たり2ファ
ラデー(又は193,000クーロン)である。実際には、用いられる電気の量
は理論的量の2〜5倍であることができる。
陰極液は、例えばポンプの作用下で循環させることができる。回路は熱交換器
又は伸縮容器(expansion vessels)などの付随装置をさらに
含むことができ、そのような伸縮容器は特に陰極液を一般式(III)の生成物
と共に供給できるようにし、一般式(I)の生成物を抽出のために取り出せるよ
うにもする。
陽極液も循環させることができる。本発明の好ましい態様に従えば、陰極の回
路は陽極の回路と類似であり、分離ダイアフラムの両側の圧力を均衡させること
を可能にする。
本発明の他の特定の態様に従えば、陽極室と陰極室に挿入物が配置される。こ
れらの挿入物は一方でイオン交換膜の変形を防ぎ、他方でこの膜が電極と接触す
るのを防ぐのに用いられる。それらは又、陰極液濃度の均一性を増すためにも用
いられる。
挿入物がない場合、陰極室における陰極液の循環速度は通常10cm/秒より
大、好ましくは50cm/秒より大である。挿入物を用いる場
合、陰極液の見掛けの速度(挿入物なしで見積もられる陰極室における速度)は
通常1cm/秒より大、、好ましくは10cm/秒より大である。
本発明の他の態様に従えば、槽は単に電解質の成分に関して不活性な材料から
作られた平行六面体又は円筒状受け器(receptacle)から成ることが
できる。この受け器は作用電極を含み、その性質は第1の型の槽の場合に定義さ
れた性質と同じである。この作用電極の形は受け器の形に適応させる。
一般に、イオン伝導性を保証する1つ又は複数のダイアフラムにより分離され
た陽極及び陰極を含むいずれの電解槽も用いることができ、部品の配置は方法の
実行に重要ではない。
本発明の方法の実行により得られる一般式(I)の生成物は、通常の方法の適
用により分離される。
7−位のヒドロキシル基の立体配置が7β−形態にある一般式(III)の特
定の生成物から、一方でR1が水素原子を示し、R2がヒドロキシル基を示す一般
式(I)、他方でR1がヒドロキシル基を示し、R2が水素原子を示す一般式(I
)の対応する生成物の混合物に導く本発明の方法の実行の場合、かくして得られ
る生成物の分離はクロマトグラフィーなどの既知の方法に従って行うことができ
る。
R6がフェニル基を示し、R3が水素原子を示す一般式(III)の生成物はそ
れぞれ10−デアセチルバッカチンIIIに対応し、それは既知の方法に従って
種々の種類のいちい(タキスス種(Taxus sp.)の抽出物から単離する
ことができる。
[脱文]及びR3が一般式(II)の基を示し、そこでArがアリール基を示
し、R4がベンゾイル基を示す一般式(III)の生成物は1
0−デアセチルタキソール及びその類似体に対応し、それはヨーロッパ特許EP
0,253,739、EP 0,336,840、EP 0,400,971
及びEP 0,428,376又は国際出願PCT WO 9209589に記
載の方法に従って得ることができる。
R6がフェニル基を示し、R3が一般式(II)の基を示し、そこでArがアリ
ール基を示し、R4がt−ブトキシカルボニル基を示す一般式(III)の生成
物は、ヨーロッパ特許EP 0,253,738及びEP 0,336,841
又は国際出願PCT WO 9209589に記載の方法に従って得ることがで
きる。
R6がフェニル基を示し、R3が一般式(II)の基を示し、そこでR4がR5−
O−CO−を示す一般式(III)の生成物は、一般式:
R5−O−CO−X (IV)
[式中、Xはハロゲン原子(フッ素、塩素)又は残基−O−R5もしくは−O−
CO−R5を示す]
の反応性誘導体を一般式:
[式中、Arは上記と同義であり、G1は水素原子あるいはヒドロキシル官能基
の保護基、例えば2,2,2−トリクロロエトキシカルボニル基又は各アルキル
部分の炭素数が1〜4であり、各アリール部分が好ましくはフェニル基を示すト
リアルキルシリル、ジアルキルアリールシリ
ル、アルキルジアリールシリルもしくはトリアリールシリル基を示し、G2は水
素原子あるいはヒドロキシル官能基の保護基、好ましくは2,2,2−トリクロ
ロエトキシカルボニル基を示す]
の10−デアセチルバッカチンIII誘導体と反応させ、一般式:
[式中、Ar、R5、G1及びG2は上記と同義である]
の生成物を得、その後保護基G1及びG2を水素原子で置換することにより得るこ
とができる。
一般に一般式(IV)の反応物と一般式(V)のバッカチンIII又は10−
デアセチルバッカチンIII誘導体の反応はエステル、例えば酢酸エチルなどの
有機溶媒中で、重炭酸ナトリウムなどの無機又は有機塩基の存在中で、0〜50
℃、好ましくは20℃近辺の温度において行う。
一般に、一般式(VI)の生成物の保護基G1及びG2の水素原子による置換は
、G1及び/又はG2が2,2,2−トリクロロカトキシカルボニル基を示す場合
、30〜60℃の温度で酢酸の存在中における亜鉛を用いた処理により、又は亜
鉛の存在中において炭素数が1〜3の脂肪族アルコール中で塩酸又は酢酸などの
無機もしくは有機酸を用いて、あるいはG1及び/又はG2がシリル化又はアルコ
キシアセチル基を示す場合、炭素数が1〜3の脂肪族アルコール(メタノール、
エタノール、イソプ
ロパノール)又はフッ化水素酸中において0〜40℃の温度で酸媒体、例えば塩
酸中で処理することにより行われる。
R3が一般式(II)の基を示し、R6が上記と同義である一般式(III)の
生成物は、一般式:
[式中、Ar、R4、G1及びG2は上記と同義であり、R7は水素原子を示し、R8
はヒドロキシル官能基の保護基を示すか、あるいは他の場合R7及びR8は一緒
になって5−もしくは6−員飽和ヘテロ環を形成する]
の生成物を一般式

の酸、又は他の場合この酸の誘導体を用いてエステル化し、一般式:
[式中、Ar、R4、R6、R7、R8、G1及びG2は上記と同義である]
の生成物を得、そこにおいて場合によりR4、R7及びR8の意味に依存して一般
式:

の生成物を通過し、それを場合により基G1及びG2を水素原子により置換する前
にベンゾイルクロリド又は一般式(IV)の生成物を用いてアシル化し、R7が
水素原子を示す場合は保護基R8、又はR7及びR8が一緒になって5−もしくは
6−員飽和ヘテロ環を形成する場合はR7及びR8、G1及び場合によりG2を水素
により置換して一般式(III)の生成物を製造することにより得られる。
一般式(VII)の生成物は一般式:
[式中、Ar、R4、R7、R8、G1は水素原子又はシリル化基であることが好ま
しいヒドロキシル官能基の保護基を示し、G2は水素原子又はアルコキシアセチ
ル基であることが好ましいヒドロキシル官能基の保護基を示す]
の生成物を電解還元し、場合によりその後保護基R7、R8、G1及びG2を水素原
子で置換することにより得ることができる。
R3が水素原子を示す一般式(I)の生成物は、ヨーロッパ特許EP 0,2
53,738、EP 0,253,739、EP 0,336,840、EP
0,336,841、EP 0,400,971又はEP 0,428,376
あるいは国際出願PCT WO 9209589に記載の方法と類似の方法に従
ってR3が一般式(II)の基を示す一般式(I)の生成物を製造するのに有用
である。
R1及びR2が上記と同義であり、R3が一般式(II)の基を示す一般式(I
)の新規生成物は異常な細胞増殖に関して有意な阻害活性を現し、異常な細胞増
殖に伴う病理学的状態を有する患者を処置できる治療性を有する。病理学的状態
には、制限を意図するものではないが筋肉、骨もしくは結合組織、皮膚、脳、肺
、性器官、リンパもしくは腎系、乳もしくは血液細胞、肝臓、消化系、すい臓及
び甲状腺もしくは副腎を含む種々の組織及び/又は臓器の悪性又は良性細胞の異
常な細胞増殖が含まれる。これらの病理学的状態には乾せん、充実性腫瘍、卵巣
、乳、脳、前立腺、大腸、胃、腎臓又は精巣の癌、カポジー肉腫、胆管癌、繊毛
癌、神経芽腫、ウィルムス腫瘍、ホジキン病、黒色腫、骨髄腫、慢性リンパ性白
血病ならびに急性及び慢性顆粒球性リンパ腫も含まれる。本発明の新規生成物は
卵巣の癌の処置に特に有用である。本発明の生成物は病理学的状態の発現又は再
発の予防又は遅延に、あるいはこれらの病理学的状態の処置に用いることができ
る。
本発明の生成物は選ばれた投与経路に適した種々の形態に従って患者に投与す
ることができ、投与経路は非経口的経路が好ましい。非経口的投与には静脈内、
腹腔内、筋肉内又は皮下投与が含まれる。腹腔内又は静脈内投与がさらに特に好
ましい。
本発明は人又は動物の治療に用いるために適した十分な量で少なくとも1種の
一般式(I)の生成物を含む製薬学的組成物も含む。組成物は通常に方法に従い
、1種又はそれ以上の製薬学的に許容し得る添加物、ビヒクル又は賦形剤を用い
て製造することができる。適したビヒクルには希釈剤、無菌水性媒体及び種々の
無毒性溶剤が含まれる。組成物は水性の溶液又は懸濁液、注射可能な溶液の形態
をとるのが好ましく、それらは乳化剤、着色剤、防腐剤又は安定剤を含むことが
できる。
添加剤又は賦形剤の選択は、生成物の溶解度及び化学的性質、投与の特定の様
式及び優れた製薬学的習慣により決定することができる。
非経口的投与の場合、無菌の水性又は非水性溶液又は懸濁液を用いる。非水性
溶液又は懸濁液の製造の場合、オリーブ油、ごま油又は液体パラフィンなどの天
然の植物油、あるいはオレイン酸エチルなどの注射可能な有機エステルを用いる
ことができる。無菌の水溶液は水中に溶解した製薬学的に許容し得る塩の溶液か
ら成ることができる。水溶液は、pHが適当に調節され、溶液が例えば十分な量
の塩化ナトリウム又はグルコースを用いて等張としてあれば、静脈内投与に適し
ている。滅菌は加熱により、又は組成物に悪影響を与えない他の手段により行う
ことができる。
本発明の組成物に関与する生成物はすべて純粋で、使用量において無毒性でな
ければならないのは明らかである。
組成物は少なくとも0.01%の治療的活性生成物を含むことができる。組成
物中の活性生成物の量は、適した投薬量が処方されるのを可能にする量である。
組成物は、非経口的投与の場合1回の投薬量が約0.01〜1,000mgの活
性生成物を含むように製造ずるのが好ましい。
治療的処置は抗悪性腫瘍医薬品、モノクローナル抗体、免疫療法又は放射線療
法あるいは生体応答調節物質を含む他の治療的処置と共同で行うことができる。
応答調節物質にはインターロイキン、インターフェロン(α,β又はδ)及びT
NFなどのリンホカイン及びサイトカイン(cytokines)が含まれるが
これらに制限するものではない。異常な細胞増殖による疾患の処置に有用な他の
化学療法薬にはナイトロジェンマスタードなどのアルキル化剤、例えばメクロル
エタミン、シクロホスファミド、メルフアラン及びクロラムブシル、ブスルファ
ンなどのアルキルスルホネート、カルムスチン、ロムシン、セムスチン及びスト
レプトゾシンなどのニトロソウレア、ダカルバジンなどのトリアゼン、葉酸類似
体などの代謝拮抗物質、例えばメトトレキセート、フルオロウラシル及びシタラ
ビンなどのピリミジン類似体、メルカプトプリン及びチオグアニンなどのプリン
類似体、ビンカアルカロイドなどの天然生成物、例えばビンブラスチン、ビンク
リスチン及びベンデシン、エトポシド及びテニポシドなどのエピポドフィロトキ
シン、ダクチノマイシン、ダウノルビシン、ドクソルビシン、ブレオマイシン、
プリカマイシン及びミトマイシンなどの抗生物質、L−アスパラギナーゼなどの
酵素、白金の配位錯体などの種々の薬剤、例えばシスプラチン、ヒドロキシウレ
アなどの置換ウレア、プロカルバジンなどのメチルヒドラジン誘導体、ミトタン
及びアミノグリテチミドなどのアドレノコルチコイド抑制剤、ホルモン及び拮抗

に制限するものではない。
本発明の方法を実行するために用いられる投薬量は予防処置又は最大治療応答
を可能にする投薬量である。投薬量は投与の形態、選択された特定の生成物、処
置される患者に固有の特徴に従って変化する。一般に投薬量は、異常な細胞増殖
による疾患の処置に治療的に有効である量である。本発明の生成物は所望の治療
効果を得るために必要なだけ頻繁に投与することができる。いくらかの患者は比
較的高い又は低い投薬量に対して急速に応答することができ、かくして必要な維
持量が低いか、又はゼロである。一般に処置の始めに低投薬量を用い、必要なら
最適効果が得られるまで徐々に高い投薬量を投与する。他の患者の場合、1日に
1〜8回、好ましくは1〜4回の維持量を、問題の患者の生理学的要求に従って
投与することが必要であり得る。いくらかの患者の場合、1日の必要投与回数が
1回又は2回だけであることも可能である。
人の場合、投薬量は一般に0.01〜200mg/kgである。腹腔内の場合

35℃より低い温度において減圧下で溶媒を除去した後、抽出物を25cm3の
脱イオン水に取り上げ、25cm3の酢酸エチルで3回抽出し、次いで有機相を
飽和塩化ナトリウム水溶液で洗浄する。沈降により分離された有機相を硫酸マグ
ネシウム上で乾燥した後、溶媒を35℃より低い温度において減圧下で蒸発させ
る。かくして510mgの粗生成物が得られ、その成分を直径が2cmのカラム
に含まれる50gのシリカ(0.04〜0.063mm)上のフラッシュクロマ
トグラフィーにより、4バールのアルゴン圧下でジクロロメタン/メタノール(
