[Prior Art] A "plate" is a synthetic resin substrate that is regularly arranged and used for culturing cells, in which holes are regularly arranged.
It has been commonly referred to as "," and has the configuration shown in the perspective view of FIG.
In FIG. 3, 1 is a synthetic resin substrate and 2 is a hollow cylindrical hole. The holes 2 are regularly arranged in the vertical and horizontal directions on the substrate 1, and a plate having 12 × 8 = 96 holes is commonly used. The one hole has a diameter of 6.5 mm and a depth of about 10 mm as shown in the longitudinal sectional view of FIG. 4, and the distance between adjacent holes is about 1 mm. 0.2 ml of culture medium containing cell groups such as lymphocytes and blood
At the same time, insert it into the hole at the specified position from above, and
Incubate for a predetermined time under humidity conditions. When inspecting the culture result, the plate can be brought under the optical microscope while the plate is in the horizontal position, and the objective lens can be moved above the hole to perform the inspection.

Note that a plate commonly called a Terasaki plate, which has an extremely smaller shape than the holes shown in FIGS. 3 and 4 and whose longitudinal sectional shape is shown in FIG. 5, is also known. Bottom diameter is approx.
It is 1 mm in diameter and the diameter of the upper end of the hole is about 4 mm, and it has the shape of a funnel as a whole. The slope has a shape that spreads rapidly upward. The distance between the holes is about 2 mm.

[Problems to be Solved by the Invention] The plates shown in FIGS. 3 and 4 required a large amount of the culture solution / cell group, and there were cases where the plates after the culture were too large. When using a Terasaki plate, the required size of the cell group is small because of its small shape, but when the different shape of the cell is cultivated in the adjacent holes of the same plate, the shape of the holes is too wide. For example, since the cell group is dropped on the synthetic resin substrate intermediate between the adjacent holes and scattered, it is easy to contaminate the cell group in the adjacent hole. In addition, because of the shape of the Terasaki plate, the culture solution was easily evaporated in the air.

An object of the present invention is to improve the above-mentioned drawbacks and to provide a culture plate provided with a hole having a shape capable of culturing only by using a small amount of cells.

[Means for Solving the Problems] Means adopted by the present invention for solving the above problems are as follows. That is, in a cell culture plate in which a substantially transparent substrate made of a synthetic resin and holes of a substantially cylindrical shape are regularly arranged in the vertical and horizontal directions, the bottom of the cylindrical hole is circular and narrower than the upper end. The shape is such that the hole expands upward in a funnel shape, and the angle between the inner wall and the bottom is about 60 degrees.

[Operation] Since the shape of the hole of the cell culture plate according to the present invention is funnel-shaped from the middle of the hole, the amount of cells used for culture can be much smaller than before.

[Embodiment] FIG. 1 is a schematic view showing a vertical cross-sectional shape of a part of a plate as one embodiment of the present invention, and FIG. 2 is a view showing a cross-sectional shape of one hole. In FIG. 1, reference numeral 1 is a transparent substrate made of synthetic resin, and 3-1, 3-2 ... Show holes according to the present invention. As shown in FIG. 2, the bottom 4 such as the hole 3-1 is
For example, an inclined surface 5 having a diameter of 2 mm and extending in a funnel shape from both ends thereof forms an angle of about 60 degrees with the bottom portion 4, for example. The depth of the funnel-shaped part is about 3.5 mm. The cylindrical hole portion 6 has a depth of about 8 mm, and the hole 6 has a slight inclination so that the hole 6 expands slightly toward the upper end. It is convenient when taking out from the mold including the case.

[Advantage of the Invention] According to the present invention, the area of the bottom of the hole of the cell culture plate is narrower than the area of the upper end of the hole, and the funnel shape expands from the bottom of the hole. Even if the volume of the culture solution put in the holes is smaller than the conventional one, the predetermined culture treatment can be performed in the same manner as the conventional one. For example, according to the above-mentioned embodiment, the amount is about 0.02 ml, which is 1/10. This is because the conventional plate shown in FIG. 3 does not spread over the entire bottom of the hole when the amount of culture solution is small, the area in contact with air is large, and the amount of evaporation is large.

In addition, since a small amount of the culture solution is processed, it is easy to partially remove the culture solution or add it as necessary while observing it using an optical microscope during the process, and thus cell cloning is possible. It's easy to do. It was very difficult to add the culture solution to the Terasaki plate shown in FIG.

Next, since the hole has a cylindrical portion, when a cell group is put into the hole, it is less likely to mistakenly insert a cell other than a predetermined one into the gap or the adjacent hole, and contamination with bacteria or the like can be prevented.
