本発明はヒト免疫不全症ウィルス(HIV)に特異的な抗体に関する。
HtVは後天性免疫不全症候群(AIDS)の原因因子であると言われている(
Poρ−povic eL al、、 1984+ 5ctence 224:
497) ++これは、そのゲノムが少なくとも6ケの遺伝子産物をコードし得
るヒトの病原性レトロウィルスである。そのenv遺伝子1良蛋自分解によって
、120 KD%層蛋白(gpl加)と41に口軽膜蛋白(gp41)とに処理
される160 KDaのグリコジル蛋白(gp160)をコードする。 gp1
20は、非共有結合によってgp41と共に、ピリオンに磁路される。 gpL
20 とgp4Lとは、ピリオン粒子ならびにウィルス感染細胞双方の表面に存
在している。
「■vの異なる株は、ウィルスのゲノムによってコードされる蛋白のアミノ酸配
列部、それも特に、外層糖蛋白1p120のアミノ酸配列順序(Stareic

全長に亙って、ひとつのIIIV変異体から他の変異体へ約20−25凌化する
。全エンベロープ蛋白に及ぼす変化の度合いは一定していない。
保存領域と可変領域にはひとつのパターンがあり、これは蛋白が、明確な機能の
源泉になっている領域に分割されていることを示唆している。多くの異なった領
域については、例えば、CD4の結合領域、主要な中和決定因子それに細胞障害
性のT細胞認識決定因子が、これまでに確認されている。
癌や他の病気の治療には、これまで標的細胞に対する抗体が用いられてきた。
Zarling等(EPo 308936)は、[lIVに感染した細胞を選択
的に死滅させるGP120の主要な中和領域に特異的な抗体の異種結合体(ヘテ
ロコンジュゲート)を開示している* Pincus等(J、ras+unol
−(1989) 142;3070)は、gp120の免疫支配領域をも認識す
る抗体毒素結合体について記述しており、fill等(Proc、 Mat、^
Q、Sci、、1989.86;1981ンは、抗gp41毒素結合体を開示し
ている。

本発明は、HIVエンベロー1蛋白の非免疫支配エピトープを認識することが可
能な抗体を特色としており、この場合、エンベロープ蛋白への抗体の結合は、E
lVに感染した患者の血清によって阻止されない0本川tiMs中で使用する[
抗体1とは、全抗体分子あるいはフラグメントもしくは抗体の修飾体を指してお
り、例えば、抗体のフラグメントは〜分子のFabgフラグメント、Fab+フ
ラグメントまたは長鎖もしくは短鎖のみであってよく、例えば修飾体は、flu
s−等が匍羽/的詞4で、また1、adner等がwo88101649で記述
しているように、長鎖と短鎖の双方を含む線状ポリペプチド分子であってもよい
0本明細書で使用する「非免疫支配エビドーグ」とは、有意に免疫原性で、ない
、つまり、少なくともヒト患者の75zで、抗体反応を誘引しない天然蛋白の構
造内におけるアミノ酸の配列を意味している0本発明によれば、HIVエンベロ
ー1蛋白の非免疫支配領域に向けられた抗体は、潜在的に競合するWI環抗体が
不在であったり、低戯であるためにその鏝体に結合することが可能である。対照
的に、エンベロープ蛋白の免疫支配領域に向けられた抗体は、愚者の、[IIV
IJ染に対する自然な免疫反応により、患者に抗体が存在するため、標的エン
ベロープ蛋白への結合から、部分的に、あるいは完全、gp12oもしくはgp
41部分内にあると思われる。非免疫支配性c3 [IIV陽性ヒト血清の存在
下で、標的抗原に試験管内で抗体を結合させることによって、測定することが可
能である。非免疫支配エピトープに特異的な抗体は、IIIV陽性血清の存在下
もしくは不在下で、比較可能な結合効率を実証しよう、 nrv陽性直清の存在
下における標的抗原に対する抗体の結合効率は、少なくとも、[l[V陽性血清
の不在下におけるその結合効率の80χである。
好ましい態様において、抗体によって認識される非免疫支配エピトープは、グル
ープに共通する0本明細書において使用する「グループに共通する決定因子」と
は、その株にのみ特異的でなく、少なくとも、もうひとつのHri株上にも存在
する■■v株によってコードされる蛋白の、抗原部分を意味する。好ましくは、
抗体は、HIVのエンベロープ糖蛋白を発現する細胞の表面に結合し得るもので
あり、RaLmr等の1985. Nature 313:277における番号
順の申し合わせによる、アミノ酸残基473から759まで(両端を含む)にお
けるエンベロープ蛋白の領域を認識することができ、かつ、473から759ま
でのアミノ酸領域内に含まれるap120の部分を認識することができるもので
ある。このような抗体の一例は^0丁、C,C,N。
、 He 10321の細5.′胞系によって産生される抗体である。
他の好ましい態様としては、抗体は毒素に共有結合して結合体を形成するもので
あり、その結合体が、ヒ)HIV+血清の存在下で、[ITVに感染したII胞
を死滅させ得るものである。II胞の殺傷作用は、[IIV vMIA胞による
抗体毒素結合体の、II胞内への取込みによって生じるであろう、この結合体は
、抗体と毒素分子を化学的に結合する蛋白レベルで、またはDNAレベルで上記
Hustanの抗体をコードするDMAの配列をクローニングし、毒素をコード
するDNAの配列に連結することによって作られる。
本明細書中で使用する用語’trill Jとは、有毒レクチン、リシン、アブ
リン、モデクシンCwwietcin)、ジフテリア毒素、シェードモナス外毒
素または好ましくはそのトキシンA11部分といった=般に呼称される毒素、並
びに放射性同位元素、IIl!lil害剤ならびに制癌剤のような他の毒性剤を
も含む意味に使用されている、「毒素」は、またひとつの抗体分子に結合し、そ
れによって種々の細胞毒性を提供し得るさまざまな毒素の組合せを指すこともあ
る。
本発明の、他の、好ましい態様では、[lIV特異抗体は、異種凝集体(ヘテロ
アグリゲート)もしくは異種抗体(ヘテロ抗体)としても知られる、抗体の異種
結合体(ヘテロコンジュゲート)を形成させるために、エフェクター細胞に特異
的な第二の抗体に連結される。異種結合体の抗■IV抗体は、[IIV感染棒胞
つまり、死滅させるべき標的細胞に結合する一方で、異種結合体の抗エフェクタ
ー抗体は、例えば、σ細胞としても知られる)it胞傷害性T@胞、単核球(特
にマクロファージ)、顆粒球もしくは、ナチュラルキラー作用または抗体依存性
m胞傷害作用を伴った細胞を含む大型顆粒リンパ球といった末梢血リンパ球(P
BL)の集団内に見られるエフェクター!11j!に結合し、結果的に、異種結
合体の抗体成分がエフェクターと標的maとを架橋し、それによってm胞傷害エ
フェクター細胞による標的細胞の死滅を促進する。
[11Vに感染した患者は、[vFJl!を死滅せしめるに十分な抗体毒素結合
体もしくは本発明の抗体異種結合体の一定量を投与することにより治療すること
が可能である。ウィルスの活発な産生中、ウィルスのエンベロー1蛋白は、感染
細胞の表面に発現される。ウィルスが増殖している細胞を、ウィルス特異表面抗
原と反応する抗体結合体を用いて選択的に死滅せしめると、ウィルスの感染サイ
クルは中断されるであろう。
本発明の抗体または抗体毒素結合体もしくは抗体異種結合体のもつひとつの利点
は、その抗体が特異的であるHIVエンベロープ糠蛋白エピトープの、非免疫支
配性である。非免疫支配性の結果、もしくは、ヒトの免疫系がエピトープに対す
る検出可能な応答を増大し得ないということの結果、そうしたエピトープ、つま
りurv 感染患者におけるエンベロープ蛋白の、gp160非免疫支配エピト
ープに対する#R抗体は、もしあるとしても、掻く僅かにすぎない、従って、本
発明の抗体毒素結合体もしくは抗体異種結合体を用いて、HIV FKIJ、者
を治療した場合、標的非免疫支配エピトープに対する循環抗体と競合することな
く、HtV fi染l1Ili!を選択的に標的とすることが可能になる。
本発明による特定の抗体、抗体毒素結合体ならびに抗体異種結合体が存するもう
ひとつの利点は、tItVエンベG−711蛋白の、グループに共通した決定因
子を認識し得る能力である。グループに共通した決定因子と云うのは、grvの
異なった株間で、本質的に不変なエンベロープポリペプチド部分である。従って
、グループに共通した決定因子を認識し得る抗体は、[ltVの、いずれの株の
gp160を認識することも可能である。そうした理由から、本発明に従って行
なう[111/感染患者の治療は、HIVのいずれかひとつの株に限定されない
ばかりか、その標的決定因子が共通するすべての株を包含することになる。
本発明の、他の特色や利点は、その好ましい態様に関する下記の説明や請求の範
囲から明らかになろう。

次に、本発明による抗体の調製と使用方法とを説明する。
免疫源
本発明による抗体の産生には、g9L60 (Repltgea Corp、+
Cambridge、 M幻と残基473から残基7Jに及ぶエンベロープ蛋
白フラグメントで、pEliV (rva薗ff eLal、、米国特許No−
4,861,707> と命名された2種類の免疫源を使用した。
ap16Gは、標準技法に従って完全フロインドアジエバンド(CFA) (口
1fco Labs、 Grand l5land、NY)と乳化して免疫処理
のために調製した。
モノクロ−ル の
!k 1 h/cJ系雌マウス(Jackson Labs、、 [Lar b
rbor、 ME)に、−6当たりgpL60/CF^70μgの腹腔内投与に
よる免疫化を行なった。3週間後、マウスにフロイントの不完全アジュバントに
よる乳化物で、ブースター免疫処理を行なったマウスを採血し、その血清につい
て、免疫原と反応する抗体の有無を検査し九強力な直情学的反応を示したマウス
には、最初のブースター免疫の5週間後、溶解状(A、T、C,C,lio、
CRIJ21?7.^、T、C,C,Jio、 C孔額圓)の骨髄腫細胞CKt
sx+rney at al、、 J、Iwunol、、1979.123:1
548)と、5:1の比率で、i:ohlerならびにMi[5tein(Na
ture (1975) 256:495)の方法に準拠した標準的な方法によ
り融合した。
融合の10−21日後に出現したハイプリドーマから得た上清を、pEにv9の
蛋白フラグメントと反応する抗体の産生についてスクリーニングにかけた。
%−ウェルのCos ta r平底マイクロタイター板のウェルはいずれも、各
ウェルでの最終濃度が2−10μg/曽lのpENV9蛋白フラグメントを含む
PaS溶液父マイクロリフドルを加えて被覆した。 pENvi液は吸引し、ウ
ェルは洗浄して、PBS + 0.5X紹^と入れ替えたの52時間インキュベ
ートした。インキュベート後、ウェルは吸引、洗浄したのち、ハイブリドーマ上
滑液50μ【を加えて2時間インキュベートした。インキュベート後、ウェルを
PBSで3回洗浄したのち、西洋ワサビペルオ牛シダーゼ(HRP、 Boeh
ringer Mannheig、 ’4est Genta−を結合したヤギ
抗マウス免疫グロブリンの適切な希釈tf、50u+を加えて、1時間インキュ
ベートした。ウェルは再び、PBSで洗浄し、そして結合抗体検出のためと30
2 HtOtの1: tooo希釈液を加えたO、1Mりzン酸ナトリウムpf
14.2中(F)L a+M ABTS(2,27ジノ、ビス(3−エチル ベ
ンズアゾリン6−スルホ>to tooμIt西洋ワサビペルオキシダーゼの基
質として加えた。 3olI+後、Dynatech分光光度計の自動読みとり
器(Virginia)によりODagmで吸光度を測定した。
試験の結果最初の免疫抗原gp16G−\の結合が陽性だった4uのバイブリド
−?(ICI、 11(9,207および2D) ニツイて、第2の免疫抗原、
pENV9.と他の異なった11株から得た。160への、結合能力の有無を試
験した。 ELrS^法により、pENV9フラグメントとの反応性を示したI
CIと呼ばれるひとつのハイブリドーマクローンは、1株以上のI(rv内に存
在する、つまり、下記に説明するようなグループに共通した決定因子であるエン
ベロー1自決定因子をも認識し得る抗体を産生じた。
上記4種類の抗体は、■IV−rl[B分離株からのgp160あるいはIII
V−RF分離株からのgρ160の、いずれかに結合するかどうかをELISA
法によって試験した。下記の表1に提示した結果は、pENII9に特異なIC
I抗体が、IITBならびに[lF分離株双方のgp160とpENV9とに結
合するが、残る3種類の抗体が、3種類の蛋白のいずれをも認識しないことを示
している。

ICIクローンのイソタイプは、主要な免疫グロブリンの各イソタイプに対応す
るヤギー抗マウスHRP (Zyged Labs、 San Francrs
co、 CA)製品を用い、ELISh法によってrgGtaであることを確認
した。 l(:Iクローンは、サブクローニングし、上記抗原への結合能力を再
度スクリーンニングした。ブリスタンによる処理を受けたBa1b/cマウスに
、ICIサプクa−ンを腹腔内注射して増殖させた。腹水をマウスから採取し、
抗体は下記のようにしてプロティン^親和性クロマトグラフィーによって精製し
た。
モノクローナル の ]1襄
精製ICI抗体は、ブリスタン処理した同系マウス(syngeneic濡1c
e)に、繰り返しELISA法による試験結果が陽性だったハイブリドーマのサ
ブクローンを腹腔内注射して調製しf、生じた腹水は、注射の2−3週間後に回
収し、抗体を下記のごとく精製したのちPBSに対する透析を行なった。
IgG2aのIcI抗体を含む腹水は、0.1 ?l Tris/3 M Na
CL p[I8.9で5倍に希釈し、同じ緩衝液と平衡化したプロティン−^−
セファロース親和性カラムに結合させたのち、0.15M NaC[、0,1M
l!N!、pH3,0を用いてカラムから溶出させた。溶出後、抗体は直ちにI
M haJω、を加えて中和した。
ICI抗生の桔金特興性
ICI抗体は、それが結合するエピトープを位置づけるため、aIsA法により
pEにV9gp120ならびにp121との結合の有無を試験した。 p121
(Chag et al、、米国特許No、 4,774,175)は、gp
160のgp41部分内で、アミノ酸約566−648にまたがるおアミノ酸蛋
白フラグメントであり、完全に、ρENV9の配列内に含まれている(gρ16
0のこれら領域1図1に模式的に説明されている) 、 p121は、客p4L
蛋の主要な免疫支配エピトープを包含している(Chang ec al、+米
国特許No、 4,724,175ならびに−ang et al、、 198
6. Proc、 Nat、^ca、 Sci、 a3:6159) 、図2は
、ELISA法による測定結果を示している。これらの結果は、ICIがpH,
’lV9とgp120とに特異的に結合するが、ρ121には結合しないことを
示している。従って、ICI抗体は、gpt2o内にも存在するpFjff9の
N域に結合するが、それはp121部分内に含まa’nlない、対照実験では、
g941に特異的な抗体(Epitope、 Inc、、Beaverton、
01?)が、pENV9 、p121また番よgp120に結合するがどうか
を試験したところ、予想通り、&p41抗体はpENν9およびp12]に結合
したがgp120には結合しなかった。
竺上血消のm−におけるICI −〇°今旧シエンベロープ蛋白に特異的で、治
療的に有用な外因性の抗体は、競合する循環抗体の存在下で、エンベロープ蛋白
を発現する)[IVもしくは[l[V感染細胞に結合し得る筈である。
ICI抗体を、HTV感染患者から得た血清の存在下で、標的抗原に結合し得る
その能力を試験し、gp41の免疫支配領域と結合することが知られている対照
抗体と比較した0図3(a) −3(d)は、マイクロタイターのウェルを、捕
捉抗原で被覆して行なったEIJSA法による結果を示している。その後、Ic
1または対照抗体を(1)非希釈ttrv陰性血清、(2)非希釈fHV陽性血
清または(3) 0.51 BSAの各50μm中に加えた。2時間後、ウェル
を洗浄し、ヒトIgと交叉反応しなかった二次抗体(ヒツジ抗マウスIl′RP
)を加えた。lvF間後、二次抗体を除去し、ウェルを洗浄したのちABTSを
加え、x背後に00.、、を測定した。
図3(a)では、ICI抗体が、lfl捉抗原pENV9に結合するかどうかを
検査している。その結果は、ntv陽性血清もしくは)IIV陰性血清の存在下
で、ICIがほぼ等しい効率でpI!1iV9に結合したことを示しているΦ0
3a)、 ICL抗体は、抗体濃度が0.01Mg/mlのHIV陰性血清の存
在下において、最高の効率でpENV9に結合したが、■■V陽性血清の存在下
での、この抗体のpENV9への結合は、抗体濃度1ttgi纏1および10M
g/mlの■【ν陰性血清の存在下でのpENV9への結合効率と比べると、そ
れぞれほぼ8濱および95%以上であっL他の4人の愚者から得たITIV陽性
血清の存在下で、IcIの結合を試験した時にも同様な結果が得られている。こ
れらの結果は、HIV陽性血清中にpENV9特異抗体が存在しているとしても
、その力価は十分に低いか結合親和性が弱く、あるいは異なったpENV9のエ
ピトープと反応するため、pEW9へのICI抗体の結合を有意に干渉しないこ
とを示している。従って、ICLは、一種のを用な治療剤としての可能性がある
。
図3(b)では、IcI抗体のp121蛋白への結合能力を試験しているが、I
CI抗体は全< p121に結合していない(0,5zBAS中でO,lui■
1以上のICIの濃度で観察されたp121との最lJlの反応性は、非特異的
な結合によるものであろう)。
対照抗体、抗gp41 (Epitope、 Inc、)ならびに2種類の標的
抗原、p121とpENV9とを用いて2種類の追加対照実験を行なった0図3
(c)および3(d)に示した結果は、抗gp4tが標的抗原のいずれにも結合
するが、抗体濃度が0.1lg/mlの場合、pENV9への結合より、p12
1への結合がより効率的であることを示している。この結果はまた、これらの同
じ抗体濃度では、抗g941抗体の結合力qIn陽性血清によって部分的に阻止
されている。これらの結果は、抗gp41抗体の結合を部分的に阻止することが
可能なρ121およびp[!IIV9に特異的な抗体が、旧V陽性血清中に存在
することを示している。

より下記のごとく確認した。
ICI抗体は、その表面にgρ12(とgp41の両方を発現する:[V eo
v遺伝子を含むワクチニアウィルス組換え体により感染したcvl II胞(A
、T、C,C,No、 CCL70) (CVI−EnV) 、もしくは陰性対
照として、HIVenv遺伝子を含まないワクチニアウィルス組換え体によって
感染したCv1細1m(CVI−Lac)のいずれにも結合した。 IIIVエ
ンベロープ遺伝子全長を発現することが可能な組換え体の構造に7いては、参照
文献としてここに含まれるEPO243029に記載されている。 Vl 4(
a)−4(d)に結果を示す。
図4(a)では、LCI抗体はCVI 、Efiv細胞に結合したが、[N4(
a)をICI抗体とインキュベートされたCVI−Lac細胞に対するFACS
のプロフィールを示す図4(b)に重ね合わせると、CVI−1ae al飽と
比較して、CVI−Env細胞においては蛍光強度に右寄りの移動(つまり、増
大)があり、このことはICI抗体が、エンベロープ蛋白を発現しない細胞より
も、HIVエンベロープ蛋白を発現する細胞に有意に良好に結合することを意味
している。このような結果は、IcIが自然状態の標的抗原に結合することを示
し、かつ、HIVエンベロー1蛋白を発現する細胞が、毒素に連結したIcIか
らなる免疫毒素結合体に対する特異標的である可能性を示唆するところから重要
である0図4(c)と図(d)とは、それぞれICI抗体を感染していないCv
l細胞に結合させ、緩衝液のみをIcI−Env細胞に結合させた対照である、
(CVI EnvおよびCVL4adM胞のFAC5分析の明らかな背景の蛍
光は、恐らく異種ウィルス蛋白の発現によって生じた細胞膜の変化によるもので
あろう)。
路種結金体■抗体桔金体調製
抗体は、例えば、Vi Leica at al、、 1987.5cienc
e 238:1098に記載されているように紺胞毒性剤に結合して免疫毒素と
して用い、あるいは抗−111V薬剤もしくは毒素を含むリボゾームの表面に付
加して、こうした薬剤や毒素を[l[VIfi染細胞に対して特異的に集中させ
ることができる0本明細書に使用した免疫毒素と云う用語は、抗体と−MWもし
くはそれ以上の毒素との結合体を意味している。様々な毒素を抗体分子に結合さ
せた場合、異なった化学的機序によって、例えば、共有結合、親和性結合、挿入
(インタルカレーション)、配位結合および錯体形成といった結合が利用できる
。しかしながら、抗体と毒素との好ましい結合は、化学的なあるいは遺伝子融合
による共有結合である。
好ましい態様では、免疫毒素は、縁■菌(Pseud国唖oas aerugi
nosa)の外毒素に連結したHrvエンベロープ蛋白の、非免疫支配性グルー
プ共通エピトープに反応する抗体である。圃面の外毒素(PE)は、容易に大量
jjl製でき、ヒトがそれに対する中和抗体を通常持っていないこと、および結
合に先立ってサブユニトに分層する必要がないことなどの理由で他の青票に比べ
て特に好ましい、PEは緑厘菌によって分泌され、真核細胞内での蛋白合成を抑
制する極めて活性な単量体蛋白(分子量66にD)である、 PEの好ましい形
態は、細胞への結合領域が除去されたPE40と呼ばれる切断分子(Pasuo
et al、、 EP出願公開No、 0261671)である。
PE40は、例えば、ヘテロ三官能性架橋リンカ−5pop <s−スクシニミ
ジル−3,(2−ピリジルジチオール)プロピオネート)、 51gm、 SL
、 Louis、 Mo)(Pastao et al、。
1986、 Ce1l 47:641)を用いたり、遺伝子融合(Cha+xl
hary eL al、、) 1989. NaLure 339:394)に
よって、本発明の抗体に、化学的な結合により連結することが可能である。抗体
の異種結合が好ましい場合は、抗体結合に適切な任意の方法が使用でき、例えば
好ましい方法には、にarpovsky QCal、(1984,J、 Exp
、 Med、 160:1686)の方法による架橋リンカ−5popを用いて
、抗体を架橋連結する方法がある。
架橋連結に引き続いて、異種結合体はサイズ排除クロマトグラソイ−によって遊
離抗体から分離される。
本発明の抗体毒素結合体または異種結合体は、該結合体をHIVエンベローフ1
蛋白を発現するHIVに慢性感染した細胞またはJIJfi細胞と共にインキュ
ベートし、該細胞を1トチミジンまたは+4(−ロイシンで短時間標識すること
によって、標的特異性や死滅効率を確認し得る。毒性は、非感染対照細胞と対比
した感染細胞における細胞***もしくは蛋白合成の低下によって測定できる。I
I胞の死滅効率は、感染細胞を本発明の結合体と共にインキュベートし、限界希
釈して平板にまき、生存細胞数を同様に処理した非患染編胞と比較するクローン

用途
免疫毒素として、本発明の抗体を用いる典型的な治療では、H[vvMll胞の
みで発現される蛋白に結合する抗体を、HIV感染膳胞に(および非感染細胞に
も同様に)毒性である毒素(例えば、シュードモナス外毒素)に結合させる。抗
体に細胞傷害側を結合することにより、非特異的毒性レベルを顕著に低めて標的
細胞に対し特異的に作用する高度な毒性効果を得ることが可能である。毒性剤は
、その結合する抗体が、標的(この場合は、HIV m胞)に対して特異的に酸
剤を運び、それによってIPI!vJIPI−t−毒素から遠避けるから、その
使用が可能である、II胞傷害剤に抗体を結合させるために使用できる技法は、
上記Vitetu et al。
および1988年8月24日に頌されたヨーロバ特許出jl No、 279,
668に、詳細に記述されている。
本発明の抗体は、HIVに感染した個人の治療用途への使用のため、常用の製薬
処方に取り入れることが可能である。更に、こうした処方は、製薬的に受け入れ
得る担体−希釈側、塩およびこの科学の分野でよく知られた他の様々な物質を含
むことができる0等張食塩水、減菌水、IOzマルトース、ヒト血清アルブミン
、グリシンまたは他の製薬的に受け入れ得る物質が、本発明の抗体から成る製薬
処方の調製上、希釈液、担体もしくは溶媒として使用できる。
薬躬組成物は、固形、半固形、液体、粉末、ビル、錠側、溶液もしくば懸濁液、
坐剤、ポリマーマイクロカプセル、リポソームまたは注射もしくは注入可能な形
態といった様々な剤型からなることができる。製薬処方は、静注、経口、皮下、
腹腔内もしくはり゛ンパ系内を含む、常用の方法で投与されるが、それらのみに
限定されない6更に、本発明の抗体、免疫毒素または異種結合体は、治療の効果
を高めるために他の治療と関連して投与することもでさる。

他の態様は、丁記の請求範囲内にある0例えば、はとんどの場合、モノクローナ
ル抗体は、ヒト以外の種から産生されているため、ヒトに対してしばしば免疫原
性である。これらのモノクローナル抗体を、ヒトの治療に有効に使用するために

種に由来し、構造的な安定性や他の生物学的機能の提供に関与する部分(不変領

可変領域がひとつの宿主に由来し、不変領域が第二の宿主に由来するキメラ抗体
産生の方法を開示している。
別法として、可変領域の相補性決定領域(CDRs)のみを、所望する抗原特異
性の免疫グロブリンからのC0R5と置換することによって、抗体を産生ずる方
法は、Winter (GB 出願公開No、 2188638)に記載されて
いる0例えば、グループ共通決定因子と非免疫支配領域とを認識するρE!iV
9に特異なマウスのモノクローナル抗体のCDR5は、組み換え口N^技法によ
ってヒト抗体のフレームワークに移植することが可能である。こうした調製は、
モノクローナル抗体を治療に応用する場合、特に有利である。
寄託
細胞系IC1,IH5は、1990年1月IO日にアメリカン・タイプ・カルチ
ャー・コレクションに寄託ざ顛寄託番号HB 10321が付与された。出願者
の譲受人であるRepl igen Corporationは、A、T、C,
C,が寄託の永続性を提供する保管者であり、特許が認可された場合、公衆によ
る人手が可能である旨を保証する。かくのごとく寄託された物質の、公衆による
人手上の制約はいずれも、特許が認可された時点で除去さ娠その撤回は不可能と
なるものとする。当該物質の人手は、37 C,F。
R,1,15および35t1.S、C,122のもとに米国特許庁長官がその資
格を有すると認定した個人に対して、本特許出願の係属中可能となる。寄託され
た物質は、その寄託微生物のサンプル提供に関する鰻も最近の要請後、少な(と
も5年間は、その生存を可能にし、かつ汚染させないため入念に必要なあらゆる
処置を講じて保管し、またいかなる場合においても、寄託日後少なくとも30年
間、あるいは特許権の有効期間のいずれか長い期間は保管されるものとする。出
願人の譲受人は、万一、寄託物質の状態により、要請があった時点で寄託機関が
サンプルを提供し得ない場合、寄託物質を!き換える義務を認めるものである。

出されるものとする。

Claims (10)
- 1.HIVエンベロープ蛋白の非免疫支配エピトープを認識することができ、且 つ前記エンベロープ蛋白への結合がHIV感染患者からの血清によって阻止され ない抗体。
- 2.前記エピトープがグループに共通なものである請求項1の抗体。
- 3.HIVエンベロープ糖蛋白発現細胞の表面に結合することが可能な請求項2 の抗体。
- 4.HIVエンベロープ蛋白の473から759のアミノ酸残基間(両端を含む )の領域を認識する請求項3の抗体。
- 5.前記領域内のgp120の部分を認識する請求項4の抗体。
- 6.ATCC No.HB10321の細胞系によって産生される請求項5の抗 体。
- 7.更に毒素を結合して含むことにより、ヒトHIV陽性血清の存在下でもHI V感染細胞を死滅させ得る結合体を形成した請求項1−6のいずれか1項の抗体 。
- 8.前記結合体が、HIV感染細胞の内部に移行することができる請求項7の抗 体毒素結合体。
- 9.更に、異種結合体を形成するため前記抗体に共有結合した第二の抗体を含む 請求項1−6のいずれか1項の抗体。
- 10.HIV感染細胞を死滅せしめるに十分な重で、請求項6の抗体毒素結合体 もしくは請求項8の抗体異種結合体を投与することからなる、HIV感染患者の 治療方法。
