PCPレセプター・リガンドおよびその用途発明の分野
本発明は、動物における神経分解および他の神経病理学的状態の予防および/ま
たは処置に有用な医薬組成物分野に関するものである。
発明の背景
アミノaL−グルタメートは、中枢神経系内の興奮性シナプスにおける化学的伝
達物質として作用すると広く考えられている。グルタメートに対する神経細胞応
答は複雑であり、少なくとも3つの相異なるレセプター・タイプ、すなわちKA
SQAおよびNMDAサブタイプにより伝達されると思われる。前記サブタイプ
は、各々それらの比較的特異的なりガント、すなわち各々カイニン酸、キスクア
ル酸およびN−メチル−D−アスパラギン酸にちなんで命名されている。
NMDAレセプターは、脳虚血または低酸素症後に生じる神経細胞死に深く関与
する。虚血/低酸素性脳発作、例えば背骨または頭部外傷、卒中または心臓発作
中に起きる発作が発生すると、神経伝達物質の保持に必要とされるエネルギー供
給が奪われた神経終末部から、内在性グルタメートの過剰放出が生じる。過剰量
のグルタメートは、近くの神経細胞に対するNMDAレセプターの過剰刺激を誘
発する。NMDAレセプターに伴うのはイオン・チャンネルである。認識部位、
すなわちNMDAレセプターは、イオン・チャンネルに対して外部のものである
。グルタメートがNMDAレセプターと相互作用するとき、それがイオン・チャ
ンネルを開口させることにより、細胞膜を横切るカチオンの流れ、例えば細胞へ

1987年7月29日公開のヨーロッパ特許出願公開第0230370号は、式
(1)を有する化合物を開示している。
式中、R1、R2、R1およびR′がHである場合、化合物はMK−801であ
る。この化合物およびその誘導体は、アンダーソン等によるアメリカ合衆国特許
第4399141号(1983)の特許対象である。
アンダーソン等によるアメリカ合衆国特許第4374838号(1983)は、
式(II)で示されるMK−801に関連した化合物を開これらは、筋肉弛緩剤
、抗うつ薬、抗けいれん薬として、並びに混合不安−うつ病、微小脳機能障害お

アンダーソン等によるアメリカ合衆国特許第4064139号(1977)は、
式(II+)で示されるMK−801に関連した化合物を開これらは、弱トラン

キンソン病の処置において有用である。

IV)テ示される10.5−(イミノメタ/)−10,11−ジヒドロ−5H−

ベンゾ[a、dl−シクロへブテン−1O−オール誘 ゛導体を開示している。

X)を有す65.6.1−1.12−テトラヒドロジベンゾ[a、e]ンクOオ
クテン−5,11−イミンを開示している。
これらの化合物は、鎮咳およびマスクロトロピック(musculotropi
c)鎮けい活性を有することが報告されている。
不デレック等によるアメリカ合衆国特許第3892758号(1975)は、式
(XI)を有する5、IO−イミノ−ジベンゾ−シクロヘプテン類を開示してい
る。

アンダーンン等によるアメリカ合衆国特許第4064139号(1977)は、
式(Xn)を有する置換9.10−ジヒドロアントラセン−9,10−イミン類
を開示している。

上述の誘導体が開発されたにも拘わらず、依然として、卒中、虚血、CNS外傷
および低血糖症と関連した神経細胞喪失の処置または予防、並びにアルツハイマ
ー病、筋萎縮性側索硬化症、ハンチントン舞踏病およびダウン症候群を含む神経
変性疾患の処置または予防に関する新しい方法が要望され続けている。
発明の要旨
本発明は、卒中、虚血、CNS外傷および低血糖症と関連した神胚細胞喪失の処
置または予防、並びにアルツノ1イマー病、筋萎縮性側索硬化症、ハンチントン
舞踏病およびダウン症候群を含む神経変性疾患の処置方法であって、処置を必要

得る。勿論、正確な処置レベルは、処置される動物、例えばヒトの病歴により異
なるものと理解される。正確な処置レベルは、過度の寅験をせずとも当業界の一
熟練者により決定され得る。

当な有機溶媒まI;は溶媒混合物を含み得る。胃液に耐性を示すコーティングを
製造するためには、適当なセルロース製剤、例えばアセチルセルロースフタレー
トまたはヒドロキシプロピルメチルセルロースフタレートの溶液が使用される。
例えば活性化合物用量の組み合わせを同定または特性確認するため、染料または
色素が錠剤または糖衣錠コーティングに加えられ得る。
経口使用され得る他の医薬製剤には、ゼラチンでてきたブツシュ−フィツト・カ
プセル並びにゼラチンおよび可塑剤、例えばグリセリンまたはソルビトールでで
きた密封軟カプセルがある。ブツシュ−フィツト・カプセルは、増量剤、例えば
乳糖、結合剤、例えば澱粉、および/または滑沢剤、例えばタルクまたはステア
リン酸マグネシウムおよび所望により安定剤と混合され得るか粒形態で活性化合
物を含み得る。軟カプセルでは、活性化合物は、好ましくは適当な液体、例えば
脂肪油または液体パラフィンに溶解または懸濁される。さらに、安定剤が加えら
れ得る。
直腸投与に使用され得る可能な医薬製剤には、例えば1種またはそれ以上の活性
化合物と量刑基剤の組み合わせから成る止剤がある。
適当な止剤基剤は、例えば天然または合成トリグリセリドまたはパラフィン族炭
化水素である。さらに、活性化合物と基剤の組み合わせから成るゼラチン直腸カ
プセルの使用も可能である。可能な基剤材料には、例えば液体トリグリセリド、
ポリエチレングリコールまたはパラフィン族炭化水素がある。
非経口投与に適した製剤には、水溶性形態の活性化合物、例えば水溶性塩類の水
溶液がある。さらに、適当な油状注射懸濁液として活性化合物の懸濁液も投与さ
れ得る。適当な親油性溶媒または賦形剤には、脂肪油、例えばゴマ油または合成
脂肪酸エステル類、例えばオレイン酸エチルまたはトリグリセリドがある。水性
注射懸濁液は、懸濁液の粘ちゅう性を高める物質を含み得、例えばカルボキシメ
チルセルロース、ソルビトールおよび/まt;はデキストランが含まれる。所望
により、懸濁液はまた安定剤を含み得る。
以下、実施例により本発明の方法および組成物について説明するが、限定的なも
のではない。当業界の熟練者には明らかであり、臨床的治療で通常直面する多様
な状態およびパラメーターの他の適当な修飾および適応も本発明の精神および範
囲内に含まれる。
実施例
実施例1: IDDCの合成
A、N−(2,2−ジェトキシエチル)−ジフェニルエチルアミンの合成
N−(2,2−ジェトキシエチル)−ジフェニルエチルアミンは、タカヤマ、H
3、ケミストリー・レターズ865頁(1978年)に準じて製造した。1.2
−ジフェニルエチルアミン(3,94g、20゜0ミリモル、アルドリッヒ・コ
ーポレーション、そのまま使用)のN、N−ジメチルホルムアミド(DMFXI
Oml)溶液を撹拌し、80−90℃で1時間かけて用時蒸留したブロモアセト
アルデヒド−ジエチルアセタール(4,50g、22.5ミリモル、アルドリッ
ヒ・コーポレーション)を流加した、1時間後、炭酸カリウム(2,76g。
20.0ミリモル)を添加した。13時間後、褐色の混合物を25℃まで冷却し
、次に混合物をI N Na0H(200m1)で希釈し、CH2Cl2(全量
100m1)で2回抽出し、そして乾燥した(MgSO+)。
溶媒を蒸発させ、残留物を蒸留し、N−(2,2−ジェトキシエチル)−ジフェ
ニルエチルアミン(5,29g、84%)を得た:沸点150−160°c/
0.50mm; ’HNMR(CD CIs)、51.10 (t、 3)、
1.12 (t、 3)、 ]、7゜(bs・ ]ン・ 2.50 (dd、I

ヒ・コーポレーション)を流力aした。NMR分光学より、反応は54時間後J
こ完結したことが示された。黒色の溶液を水(50ml)で希釈し、IN Na
OH(200ml)で塩基を作り、CH、CI、で抽出した。抽出物は濃縮乾固
し、残留物をシリカゲルのフラッシュクロマトグラフィーで精製した。lO・1
エーテル: THFで溶出した液より最初の少量の1.2−ジフェニルエチルア
ミンが得られ、統いて無色の分画が得られたが、この分画を170’C!/ 0
.5mmで蒸留すると油状物(1,15g、75%)が得られ、放置すると固化

5!施例2: 5−メチルIDDCの合成A、N−プロパルギル−1,2−ジフ
ェニルエチルアミン1.2−ジフェニルエチルアミン(930mg、4.20ミ
リモル)のエタノール(30ml)溶液を撹拌し、3−ブロモ−1−プロピン<
700 mgs 5.30ミリモル)および炭酸カリウム(1,38g、10.
0ミリモル)を添加した。混合物を16時間還流し、次いで3−プロモー■−プ
ロピン(100mg)を添加した。混合物をさらに6時間還流し、次イテ冷却し
、l N Na0H(200m1)テ希釈し、CH,C12で抽出した。抽出物

実施例3−(+)−1DDCの製造
A、(±)−1,2−ジフェニルエチルアミンの分解V、M、ポタポフらの一般
法を採用し、以下の分解を行った(ポタポフ、V、M、ら、ジャーナル・才ブ・
オルグ・ケム、USSR% 16巻、683頁(1980年))。L−(+)−
酒石酸(マリンクロツッ、そのまま使用)9.5013g(63,2ミリモル)
の水400m1溶液を撹拌し、45℃で(±)−1,2−ジフェニルエチルアミ
ン(アルドリソヒ、そのまま使用)24.7673g(125,5ミリモル)を
滴加した。
即座に白色沈澱を生じた。25℃で2.5時間撹拌した後、沈澱を収集し、不完
全な空気乾燥し、白色固形物94gを生成した。この94gを沸騰水300m1
に溶解し、次に熱い間に濾過し、透明な無色の溶液を得た。20分以内にこの溶
液から結晶化が始まった。収集した結晶を不完全な空気乾燥すると、31.6g
になった。続いてさらに5回、この物質を比例した量の沸騰水を用いて再結晶す
ると、(+)−L−酒石酸および(+)−1(R)、2−ジフェニルエチルアミ
ンの不完全に分解したジアステレオ異性体塩の白色微小結晶905゜2mgを得
た:融点222.5−223.5℃分解、[σ]D”−51。
3’ 、co、92、H200ナカザキら、ブチレン・オブ・ザーケミカル・ソ
サエティー・オブ・ジャパン、36巻、161N(1963年)によると以下の
物理定数が報告されている=(−)−1(R)。
2−ジフェニルエチルアミンの(+)−酒石酸塩、融点229−230°C1[
g]D”−−55,3°、c−0,92、H,o; ()−1(R)。
2−ジフェニルエチルアミン[αlD”−−51,2’ 、c−3,7、エタノ
ール。最後の4回の再結晶の母液は一緒にした。水溶液を濃縮し、固体の水酸化
ナトリウムを添加して塩基を作り、CH,CI□3分画で抽出した。有機層を一
緒にし、乾燥しくK xc o i)、減圧下溶媒を除去し、不完全に分解した
(−)−1(R)、2−ジフェニルエチルアミン2.5397g生成した; [
σ]D’″−−44−08、c−3゜7、エタノール(ナガサキ1M、ら、上述
)o L−(+)−酒石酸(マリンクロッツ、標準品)1.8116g(12,
1ミリモル)の水40m1溶液を撹拌し、45°Cでアミン(2,5055g、
l 2−7ミリモル)を滴加した。その結果、生じた白色沈澱を、混合物を環境
温度で12時間撹拌した後収集した。収集した固形物を沸騰水で3回再結晶する
と、(+)−L−酒石酸および(−)−1(R)、2−ジフェニルエチルアミン
の白色微小結晶のジアステレオ異性体塩815.2mgを生成した:融点224
−225°C分解、〔α]D1″−−54.8°、c−0,92、H,O,ナガ
サキら、上述。塩(806,6mg)をIN NaOH50m1およびCHz
Clx 25 mlに溶解した。層分離し、水層はCHCH2C1x25で2回
抽出した。有機層を一緒にし、IN Na○H15mlで洗浄し、乾燥しくK、
C03)、減圧下溶媒を除去し、(−)−1(R)、2−ジフェニルエチルアミ
ンの無色液体449.1mgを得た;[a]D’ラー−50,1’ 、c−3,
7、エタノール。
B、N−(1(R)、2−ジフェニルエチルアミン)アミノアセトアルデヒド−
ジエチルアセタールの製造
スズキ、T、ら、ケミカル・アンド・ファーマシューテイカル・ブチレン、34
巻、1988頁(1986年)の−膜性を採用して以下のアルキル化を行った。
(−)−1(R)、2−ジフェニルエチルアミン249.5mg(1,3ミリモ
ル)および無水炭酸カリウム(ペーカー、そのまま使用)193.6mg(1,
4ミリモル)の混合物をN、N−ジメチルホルムアミド(ベーカー、そのまま使
用)2.5ml中撹拌し、90℃でブロモアセトアルデヒドジエチルアセタール
(アルドリツヒ、蒸留物、沸点83℃/ 40mm)284.1mg(14ミリ
モル)の溶液を2時間で滴加した。結果的に得られた混合物を撹拌しながら加熱
した(95−105℃)・16時間後、反応混合物を10℃まで冷却し、IN
NaOH50m1を加え、続いてCI(2C1z 25mlを加えた。層分離し
、水層をCH2Cl215m+で2回抽出した。有機層を一緒にしてIN Na
OHlomlで洗浄し、乾燥しくK、C○、)、減圧下溶媒を留去し、褐色油状
物346.1mgを得た。油状物はグーゲルロー装置を用いて蒸留しく180−
190℃、0.05mm/Hg)、淡黄色の油状物315.5mgを得た。黄色
の油状物(304,2mg)をクロマトグラフィー(log、シリカゲル)にか
け、ジエチルエーテル25m1、続いて3:lジエチルエーテル/ T HF
40 ml、次いで2.lジエチルエーテル/THF40mlを溶出液として用
いた。
極性がほとんどない物質(RrO,56)を含有する分画を一緒にし、減圧下溶
媒を除去し、N−(1(R)、2−ジフェニルエチルアミン)アミノアセトアル
デヒド・ジエチルアセタールの透明な淡黄色の液体283.3mgを生成した(

ンゾ[a、b]シクロへブタン(IDDC)の製造T、スズキらの上述の一般法
を採用し、以下の環形成を行った。
過塩素酸(70%水溶液)6mlに、24°Cで撹拌しなからN−(1(R)、
2−ジフェニルエチル)アミノアセトアルドヒト−ジエチルアセクール280m
g(8,9ミリモル)を5分間で滴加した。添加中1:褐色油状物を分離した。
混合物を16時間、環境温度で撹拌し、2NNaOH50m1上に注加し、CH
,C1,15m1で3回抽出しlコ。有機層を一緒にしてIN NaOH15m
1で洗浄し、乾燥しくK2CO、)、減圧下溶媒を除去し、褐色油状物225.
4mgを得Iこ。TLC(THF)では2個のスポフトが認められ、Rf値は0
.67およびθ、23であった。油状物(225−g)はフラッジ1クロマトグ
ラフイー(8g1 シリカゲル)に供し、ジエチルエーテル25m1,3:1ジ
エチルエーテル/THF 40ml、 2 : lジエチルエーテル/THF2
5ml、l:lジエチルエーテル/THF25mlおよびTHF25mlで溶出
した。より極性の高い物質を含有する分画(Rfo、22)を−緒にし、溶媒を
減圧下留去し、褐色油状物172.1mgを得た。この油状物をクーゲルロー装
置を用いて二重蒸留(175−185℃、0.05mm/ Hg) L、(+)
−1DDC153,4mgの透明な淡黄色油状物を得、これを放置すると固化し

D、(+)−1DDCのマレイン酸塩の製造(+)−1DDC50,6mg(0
,23ミリモル)のエタノール1.0ml溶液を43℃で撹拌し、これにマーレ
イン酸(アルドリッヒ、そのまま使用)26.4mg(0,23ミリモル)のエ
タノール0.3ml溶液を加えた。結果的に得られた溶液は約10分以内に結晶
化を始めた。この混合物を環境温度で撹拌した。14時間後、白色結晶を収集し
、熱エタノール0.4mlから再結晶し、(+)−TDDCのマレイン酸塩(融
点168−168.5°C分解)31.1mgを得た。再結晶後、母液からマレ
イン酸塩に続く2分画、lo、2mg(融点167−167゜5分離)および1
3.8mg(融点1.64−165.6℃分解)を単離しIこ。
実施例4−光学活性N−メチルIDDCの製造(+)−1DDC(9,3mg、
0.04モル; [α]D−+ l l 6.3°、エタノール、c−1)のア
セトニトリル(0,4m1)および37%ホルムアルデヒド水溶液(0,61ミ
リモル)溶液を25°Cで撹拌し、ソディウム・シアノポロハイドライド(5,
1mg、0.08ミリモル)を加えた。得られた混合物を15分間撹拌し、氷酢
酸1滴加えて−pHを7に下げた(湿潤pH紙で検査した)。混合物を20時間
撹拌し、溶媒を減圧下除去した。残留物を2N水酸化ナトリウム(4ml)およ
びエーテル(4ml)で処理した。層分離し、水層をエーテルで抽出した。有機
層を一緒にして乾燥(K、C○、)、濾過し、蒸発させて、透明の油状物(l1
mg)を得、これを予備のシリカゲルのTLCで精製した。エタノールで溶出す
ると、2本の帯が現れ、Rf−0,41および0.83であった。0.41の帯
をとり、アセトンで抽出した。
溶液を乾燥しくK ICOs)、蒸発させて光学活性N−メチルI DDC(1

(+)−1DDCが、細胞培養中維持されるラット海鳥神経におけるNMDA誘
起(+グリシン)反応を使用回数に依存して妨害する能力を試験した。この化合
物は、MK−801が呈する使用回数依存的妨害作用と極めてよく似た結果を呈
した。
海鳥神経は、l−3日齢新生ラット(ロング−エバンス)の海馬のCA1部位か
ら得た。組織の小塊(1mm’未満)をパパイン(20単位/ml、ウォーンン
グトンークーバ:)中30分間恒温培養した。組織は2 、5 mg/mlウシ
血清アルブミンおよび2.5mg/mlトリプシン阻害剤(シグマ)を含有する
完全成長培地(アールのMEM、20mMグルコース、50単位/mlペニシリ
ン/ストレプトマイシン、5%加熱して不活化したウシ胎児血清、コラポラテイ
ブ・リサーチのセリューム・エクステングー)中、火で滑らかにした(fire
−polished)パスツールピペットで細かく砕いて単一細胞の懸濁液にし
た。細胞をガラスのカバースリップ(coverslips)上に置き、コラー
ゲン/ポリ−D−リジンで被覆した。3日毎に培地の半量を置き換えて、培養物
に栄養補給した。細胞を置いた後、最初の1週間の1または2日間、培養物にア
ラビノシルサイトシン(5X I O−’M)を加え、非神経性細胞の増殖を抑
制した。
培養物中i3週間成長した神経から採った、パッチ・クランプ記録の全形細胞法
[ハミル、O,D、、マーティー、A1、ネアー、E。
、サックマン、B、およびシグウォース、F、J、、フユーガーズ・アーチ7.
391巻、85−100頁(1981年)〕を用いて全ての電気生理学的実験を
行った。作用薬および作用薬/拮抗薬を組み合わせたものをU管器具(フェンウ
ィックら、ジャーナル・オブ・フィジオロジー、331巻、577−597頁(
1982年))により全形細胞実験の神経に供給した。外部溶液はNaC114
0、KCl3.5、CaCl、 l 、グルコース5、ピクロトキシニン0.0
2、テトロドトキシン(TTX)5X I O−’およびN−2−ヒドロキシエ
チルベビラジンーN’−xタンスルホン酸(HEPES)l O(mM)を含有
した。この溶液のpHをNaOHて7.4に調整した。内部(パンチ電極)溶液

有した。膜電流は400ヘルツ(−3デンペル; 8極ベツセル)で濾過しくf
iltered)、後者の分析用に磁気テープに記録した。実験は室温で行った
(20−25°C)。[化学物質の入手先:N−メチル−D−アスパルテート、
ケンブリッジ・リサーチ・バイオケミカルズ:ビロトキ/ニン、シグマ・ケミカ
ル・カンパニー;記録用溶液(recording 5olutions)の塩
、アルドリッヒ(ゴールド・ラベル)またはアルファ(プラトロニツタ)。[3
H]カイネートおよび[3H]CPPおよび[’H]AMPAはデュポン/NE
N(ボストン、マサチューセッツ州)より購入。]lμMグリシン存在下、50
μM NMDAを3秒間適用すると、その結果、保持電位−60mVで100−
1000pAの内側方向の全形細胞電流が流れた。繰り返し同じ細胞に適用(3
0秒毎)すると、少なくとも30分間5%未満しか変化しない電流を生じた。M
K−801(10μM)をNMDAと一緒に適用した場合、内側方向の電流は、
