Description
本発明は凝固防止剤として有効な化合物に関す
る。本発明はまたその製造も包含している。この
ような化合物は殺鼠剤として特に有効な化合物を
包含しており、そして本発明はまた本発明の凝固
防止剤化合物が殺鼠剤成分または主要な殺鼠剤成
分を提供する殺鼠剤組成物(えさ)も包含してい
る。
凝固防止剤は血液の凝固作用を妨げ、そのよう
に働く能力はその化学的な性質に依存している。
下記の一般式で表わされる或種の4−ヒドロキシ
クマリン化合物は凝固防止剤の性質を有するもの
として知られ、殺鼠剤の主成分として成功裡に使
用されてきた。
その殺鼠剤としての活性はそれの凝固防止剤の
性質に依存している。上記の式においてRが−
CHPh−CH2−CO−CH3である化合物は周知の
殺鼠剤、ワルフアリンである。ワルフアリンに代
わる「第二の世代」として追加の4−ヒドロキシ
クマリン化合物を提供するという目的によつて、
米国特許第3957824号明細書は上記の式における
Rが下記の式を意味する、或種の殺鼠剤4−ヒド
ロキシクマリン化合物を開示している。
この式においてR1およびR2は同じかまたは異
つていて、水素またはハロゲン原子、好ましくは
塩素または臭素または、好ましくは6個以下の炭
素原子を有する、アルキル基またはアルコキシ基
であり、R3は同じかまたは異つていて、ハロゲ
ン原子、好ましくは少なくとも2個、より好まし
くは5ないし12個の炭素原子を含む、直鎖または
分枝鎖のアルキル基またはアルコキシ基、シクロ
アルキル基、アラルキル基、好ましくはα−アラ
ルキル基、フエニル基またはフエノキシ基または
ハロゲノ、好ましくはパラハロゲノ、その置換さ
れた誘導基であり、そしてmは1または2であ
る。
米国特許第4035505号明細書は活性成分として
上に丁度定義した化合物と組合せた担体を含む殺
鼠剤組成物(えさ)を開示している。
このような4−ヒドロキシクマリン、特にそれ
ぞれジフエナカウム(Difenacoum)およびブロ
ジフアカウム(Brodifacoum)として知られて
いる、ジフエニル構造およびp−ブロム−ジフエ
ニル構造を含む4−ヒドロキシクマリンは、ワル
フアリン−抵抗性のけつ歯動物に対して有効な殺
鼠剤組成物の活性成分、すなわち殺鼠剤としてか
なりの間商業的に重要となつてきた。
従来技術の中で最も有効な凝固防止剤である殺
鼠剤はブロジフアカウムであつて、これは上記の
一般式のR3がパラ−ブロム−フエニル基を含む
分子構造を有する。その構造はテトラリン環の遊
離端に本来ハロゲン原子が存在するところに注目
を集めており、そのハロゲン原子はそれ自身フエ
ニレン基を経てテトラリン環に必然的に結合して
いるベンゼン環と連結している。換言すれば末端
にハロゲンを有するビフエニル構造がみられる。
ブロジフアカウムはワルフアリン−抵抗性のけ
つ歯動物に対するその有効性からみると、けつ歯
動物の制御には最も重要な商業製品である
(MR.HadlerおよびR.S.Shadbolt著、Nature、
第253巻(5489号)、第275頁、1975年を参照)。
凝固防止剤の4−ヒドロキシクマリン化合物の
開発は、上記の一般式においてRが最初のワルフ
アリンにおけるような限定されたフエニル構造ま
たは後のジフエナカウム型およびブロジフアカウ
ム型の化合物におけるような限定されたビフエニ
ル構造を含む基を表わす、或種の化合物に制限さ
れてきており、今までに凝固防止剤の性質を有す
ることが判明した4−ヒドロキシクマリン化合物
のR基が許容される性質の中では、上記の制限が
凝固防止剤の4−ヒドロキシクマリン化合物を見
出すことができる2つの領域−そして僅か2つの
領域−を限定するように、当該技術において受け
入れていることが、従来技術から明らかである。
したがつて、当該技術にたずさわる者はそれらの
領域の外側の調査がやりがいのあるものと考える
理由を持たなかつたことは明らかであり、そして

本発明の基礎となつている、構造と凝固防止剤
活性との間に関する出願人の研究者の基礎的な研

様な分子特性が、2個の連結したフエニレン基を

この追加の4−ヒドロキシクマリン化合物の予

本発明によれば、ねずみに関して凝固防止剤の
性質を有し、かつ下記の一般的な分子式を有する
化合物が提供される。

上記の式の(a)におけるDが−(CH2)m−であ

る発見に加えて、本出願人の発明者による2つの

本明細書において先に認めた従来の特許明細書
が、テトラリン環をその分子構造内で具体的に表
現する性質を有する、4−ヒドロキシクマリン化
合物凝固防止剤に関する先行の技術を開示してい
るにすぎないことは出願人による文献調査から明
らかであり、追加の、および或場合には、(殺鼠
剤として)より魅力的な本発明の凝固防止剤を発
見するのが遅れたことは、ブロジフアカウムによ
つて例示される先行技術の化合物が知られるよう
になり、そして当該技術において利用されてきた

米国特許第4035505号明細書の中の凝固防止剤

生物学的試験
本発明の化合物、およびブロジフアカウム、ジ
フエナカウムおよびワルフアリンを包含する、本
発明の範囲内に入らない他の置換された4−ヒド
ロキシクマリン化合物を次の実施例に示したよう
に試験した。プロトロンビン時間ED50値は凝固

実施例
下記の第表は次の方法によつて得られた本発
明の化合物における試験データと、同様に得られ
た比較データを含んでいる。

液に少しづつ加え、この混合物を室温において
2時間撹拌した。水を加えて分離したジクロル
メタン溶液を水洗し、無水硫酸マグネシウム上
で乾燥させた。蒸発によつて油が生成し、これ
を氷酢酸(30ml)中で4−ヒドロキシクマリン
(8.1g)と共に3時間還流させた。冷却した混
合物に水を加え、次いでこれをジクロルメタン
で抽出した。ジクロルメタン抽出液を蒸発させ
ると固体が生成し、これをシリカゲルカラム上
でクロマトグラフイーにより分離した。ジクロ
ルメタンによる溶離によつて3〔3−(4′−トリ
フルオルメチル−4−ビフエニル)−1,2,
3,4−テトラヒドロ−1−ナフチル〕−4ヒ
ドロキシクマリンの異性体を含む留分が得られ
た。最初に溶離した異性体(化合物24トラン
ス)はほぼテトラヒドロナフチルの1,3位
(0.89)、融点207.5℃であつた。

C.S.Perkin 1190頁、1976年に詳しく記載され
ている。
化合物8
1 パラブロムアニリン(500g)の希塩酸(水
3150mlで濃塩酸727mlを希釈したもの)溶液に
亜硝酸ナトリウム溶液(水290mlに亜硝酸ナト
リウム200gを溶解)を5℃において加えた。
反応温度が10℃よりも低く保たれるように添加
の速さを制御した。添加が完了した後混合物を
3℃において1時間撹拌した。次に塩化亜鉛の
溶液(水1000mlに塩化亜鉛396gを溶解)を室
温において撹拌しながら加えた。更に10分間撹
拌した後固体の沈澱を別し、真空炉の中で乾
燥した。更に特性を表示しないでジアゾニウム
塩化亜鉛塩(688g)を使用した。
2 上記1の塩の撹拌されているフラン(600ml)
懸濁液に、粉末状の水酸化ナトリウム(49g)
と酢酸ナトリウム(40g)を、25℃において
0.3時間にわたつて加えた。その混合物を24時
間撹拌してから、それを水(200ml)で希釈し、
有機層を分離した。水溶液もジエチルエーテル
で抽出し、フラン溶液と混合した。抽出液を水
洗し、無水硫酸マグネシウム上で乾燥させた。
抽出液の蒸発によつて暗色の油が生成し、これ
をシリカゲル上のクロマトグラフイーによりジ
クロルメタンで分離した。画分(32g)は核磁
気共鳴によつて2(4−ブロムフエニル)フラ
ンであることが同定された。
3 (ビルスマイヤー法)クロルベンゼン(100
ml)と混合した2(4−ブロムフエニル)フラ
ン(20.4g)にオキシ塩化燐POCl3(12.5g)と
ジメチルホルムアミド(7ml)を加え、この混
合物を10℃において1時間撹拌した。次いで更
にPOCl3(1.5g)とDMF(0.7g)を加え、混合
物を室温において更に1時間撹拌した。混合物
を氷の上に注ぎ、炭酸ナトリウムで中和し、沈
澱した固体をクロロホルム(119ml)から結晶
させた。この固体をクロマトグラフイーにより
シリカゲル上でクロロホルムで分離すると2
(4−ブロムフエニル)フルフラール(11g)、
融点153.5℃が得られた。

ジルクロライド(4g)を添加した。この混合物
を室温において一晩中撹拌してから水で希釈し、

て固体が生成し、これをエタノールから結晶させ
て3(p−フルオルベンジルオキシフエニル)テ

1 撹拌されたベンゼン中の(38gの4−クロル
ブロムベンゼンから誘導された)4−クロルフ
エニルマグネシウムブロマイドと塩化カドミウ
ム(37g)との混合物に塩化フエニルプロピオ
ニル(33.7g)を0℃において少しづつ加え
た。その混合物を室温において6時間撹拌した
後、塩化アンモニウム水溶液を加えた。エーテ
ルによる抽出によつてp−クロルフエニルフエ
ニルエチルケトン(25g)、融点78〜79℃が得
