発明の目的及び構成
本発明のスルフアイド化合物は下記一般式
()で表わされる。
(式中R1は低級アルキル基を、R2を水素原子、
水酸基、低級アルコキシ基、アラルキルオキシ
基、アラルキルオキシメトキシ基、低級アシルオ
キシ基、低級アシルアセトキシ基、低級アルコキ
シアセトキシ基、フエノキシアセトキシ基、低級
アルキルアミノアセトキシ基、低級アシルアミノ
アセトキシ基、カルボキシエチルカルボニルオキ
シ基、ラクトイルオキシ基、ベンゾイルオキシ
基、低級アルコキシカルボニルオキシ基、フエノ
キシカルボニルオキシ基、アラルキルオキシカル
ボニルオキシ基、テトラハイドロフラニルオキシ
基又はテトラハイドロピラニルオキシ基を、R3
は水素原子、水酸基、低級アルコキシ基、シクロ
アルキルオキシ基、低級アルコキシ置換エトキシ
基、テトラテイドロフラニルオキシ基、テトラハ

チル、イソブチル、t−ブチル、ペンチル、ヘキ
シル基等を例示できる。
一般式()中、R2及びR3で表わされる低級
アルコキシ基としては炭素数1〜6のアルコキシ
基、例えばメトキシ、エトキシ、プロポキシ、イ
ソプロポキシ、ブトキシ、ヘキシロキシ基等を、
低級アシルオキシ基としては炭素数2〜6のアシ
ルオキシ基、例えばアセチルオキシ、プロピオニ
ルオキシ、ブチリルオキシ、ピバロイルオキシ、
カプロイルオキシ基等の低級アルキルカルボニル
オキシ基を、低級アルコキシカルボニルオキシ基
としては炭素数2〜5のアルコキシカルボニルオ
キシ基、例えばメトキシカルボニルオキシ、エト
キシカルボニルオキシ、プロポキシカルボニルオ
キシ、ブトキシカルボニルオキシ基、イソブトキ
シカルボニルオキシ、t−ブトキシカルボニルオ
キシ基等を、アラルキルオキシカルボニルオキシ
基としては炭素数8〜10のアラルキルオキシカル
ボニルオキシ基、例えばベンジルオキシカルボニ
ルオキシ、フエネチルオキシカルボニルオキシ、
フエニルプロポキシカルボニルオキシ基等のフエ

R2で表わされるアラルキルオキシ基としては
炭素数7〜9のアラルキルオキシ基、例えばベン
ジルオキシ、フエネチルオキシ、フエニルプロポ
キシ基等のフエニル低級アルキルオキシ基を、ア
ラルキルオキシメトキシ基としては炭素数8〜10
のアラルキルオキシメトキシ基、例えばベンジル
オキシメトキシ、フエネチルオキシメトキシ、フ
エニルプロポキシメトキシ基等のフエニル低級ア
ルキルオキシメトキシ基を、低級アシルアセトキ
シ基としては炭素数4〜6のアシルアセトキシ
基、例えばアセチルアセトキシ基、プロピオニル
アセトキシ、ブチリルアセトキシ基等の低級アル
キルカルボニルアセトキシ基を、低級アルコキシ
アセトキシ基としては炭素数3〜6のアルコキシ
アセトキシ基、例えばメトキシアセトキシ、エト
キシアセトキシ、プロポキシアセトキシ、ブトキ
シアセトキシ基等を、低級アルキルアミノアセト
キシ基としては炭素数3〜6のアルキルアミノア
セトキシ基、例えばメチルアミノアセトキシ、エ
チルアミノアセトキシ、プロピルアミノアセトキ
シ、ブチルアミノアセトキシ基等を、低級アシル
アミノアセトキシ基としては炭素数4〜6のアシ
ルアミノアセトキシ基、例えばアセチルアミノア
セトキシ、プロピオニルアミノアセトキシ、ブチ
リルアミノアセトキシ基等の低級アルキルカルボ
ニルアミノアセトキシ基を夫々例示できる。
R3で表わされるシクロアルキルオキシ基とし
ては例えばシクロペンチルオキシ、シクロヘキシ
ルオキシ、シクロヘプチルオキシ基等を、低級ア
ルコキシ置換エトキシ基としては炭素数3〜8の
アルコキシ置換エトキシ基、例えばメトキシエト
キシ、エトキシエトキシ、プロポキシエトキシ、
イソプロポキシエトキシ、ブトキシエトキシ、ヘ
キシロキシエトキシ基等を、低級アルキルカルバ
モイルオキシ基としては炭素数2〜5のアルキル
カルバモイルオキシ基、例えばメチルカルバモイ
ルオキシ、エチルカルバモイルオキシ、プロピル
カルバモイルオキシ、ブチルカルバモイルオキシ
基等を夫々例示できる。
本発明のスルフアイド化合物()は、例えば
抗アレルギー作用を有する一般式
(式中R4はR1と同一又は相異なつて低級アルキ
ル基を、Yは酸残基を、R1、R2、R3、n及びA
は前記と同一の意味を示す。)
で表わされるスルホニウム誘導体の製造原料とし
て有用である。
上記スルホニウム誘導体()の製造方法につ
いては、後記参考例において具体的に述べる。
本発明のスルフアイド化合物()はその有す
るR1、R2、R3及びAにより下記各種の方法で製
造される。
<Aがエーテル結合の場合の本発明化合物(
a)、(a′)、(a″)の製造>
一般式()中Aがエーテル結合である本発明
化合物は次に示す製造法A〜Cのいずれかにより
製造される。
〔製造法A〕
(式中R1、R2、R3及びnは前記と同一の意味を
示し、Halはハロゲン原子を示す。)
上記スルフアイド化合物(a)(一般式()
中Aがエーテル結合を示すもの)は、上記反応式
に示すように、ハライド化合物()に、フエノ
ール誘導体()を、好ましくは塩基性化合物、
例えばナトリウム、カリウム等のアルカリ金属、
これらの水素化物、これらの水酸化物又はこれら
の炭酸塩、或はピリジン、モルホリン、ピペリジ
ン、ピペラジン、トリエチルアミン等の存在下、
無溶媒又は溶媒中0〜200℃程度の温度下に反応
させることにより製造される。ハライド化合物
()はフエノール誘導体()に対し過剰量を
使用してもよいが、好ましくは1〜4倍モル使用
される。塩基性化合物はハライド化合物()に
対し過剰量使用してもよいが、好ましくは1〜4
倍モル使用される。溶媒としては、本反応に関与
しない溶媒、例えばメタノール、エタノール等の
アルコール類;テトラハイドロフラン、ジオキサ
ン等のエーテル類;ベンゼン、トルエン等の芳香
族炭化水素類;アセトン、メチルエチルケトン等
のケトン類;ジメチルホルムアミド;水及びこれ
らの混合溶媒等を使用できる。
〔製造法B〕
(式中R1、n及びHalは前記と同一の意味を示
し、OR′3は水素原子及び水酸基以外のR3を示
す。)
上記スルフアイド化合物(a′)(一般式()
中Aがエーテル結合を、R3が水素原子及び水酸
基以外の基を示すもの)は、上記反応式に示すよ
うに、スルフアイド化合物()に、ハライド化
合物()を、好ましくは塩基性化合物の存在
下、無溶媒又は溶媒中、−20〜200℃程度の温度下
に反応させることにより製造される。原料とする
スルフアイド化合物()は、上記製造法Aに従
い製造される。ハライド化合物()はスルフア
イド化合物()に対し、過剰量使用してもよい
が、好ましくは1〜4倍モル使用される。塩基性
化合物としては製造法Aに例示したものと同じも
のを使用でき、これはハライド化合物()に対
して通常1〜4倍モル使用される。溶媒は、反応
に関与しない溶媒、例えばクロロホルム、ジクロ
ルメタン等のハロゲン化炭化水素類;テトラハイ
ドロフラン、エチルエーテル、ジオキサン等のエ
ーテル類、;ベンゼン、トルエン等の芳香族炭化
水素類;アセトン、メチルエチルケトン等のケト
ン類;酢酸エチルエステル、酢酸メチルエステル
等のエステル類;ジメチルホルムアミド、ジメチ
ルスルホキサイド等の極性溶媒類;水及びこれら
の混合溶媒等を使用できる。
〔製造法C〕
(式中R1、R3、n及びHalは前記と同一の意味
を示し、OR′2は水素原子及び水酸基以外のR2を
示す。)
上記スルフアイド化合物(a″)(一般式()
中Aがエーテル結合を、R2が水素原子及び水酸
基以外の基を示すもの)は、上記反応式に示すよ
うに、スルフアイド化合物()に、ハライド化
合物()を、好ましくは塩基性化合物の存在
下、無溶媒又は溶媒中、−20〜200℃程度の温度下
に反応させることにより製造される。原料とする
スルフアイド化合物()は、上記製造法A又は
Bに従い製造される。ハライド化合物()はス
ルフアイド化合物()に対し過剰量使用しても
よいが、好ましくは1〜4倍モル使用される。塩
基性化合物としては製造法Aに例示したものと同
じものを使用でき、これはハライド化合物()
に対して、通常1〜4倍モル使用される。溶媒
は、反応に関与しないものであればよく、上記製
造法Bに例示したものと同一の各種溶媒を使用で
きる。
<Aがアミド結合の場合の本発明化合物(b)、
(b′)、(b″)の製造>
一般式()中Aがアミド結合である本発明化
合物は次に示す製造法D〜Fのいずれかにより製
造される。
〔製造法D〕
(式中R1、R2、R3、n及びHalは前記と同一の
意味を示す。)
上記スルフアイド化合物(b)(一般式()
中Aがアミド結合を示すもの)は、上記反応式に
示すように、ハライド化合物()を、アミノ誘
導体()に、好ましくは塩基性化合物、例えば
ナトリウム、カリウム等のアルカリ金属、これら
の水素化物、これらの水酸化物又はこれらの炭酸
塩、或はピリジン、モルホリン、ピペリジン、ピ
ペラジン、トリエチルアミン等の存在下、無溶媒
又は溶媒中0〜150℃程度の温度下に反応させる
ことにより製造される。ハライド化合物()は
アミノ誘導体()に対し過剰量使用してもよい
が、好ましくは1〜4倍モル使用される。塩基性
化合物はハライド化合物()に対し過剰量使用
してもよいが、好ましくは1〜4倍モル使用され
る。溶媒としては、本反応に関与しない溶媒、例
えばクロロホルム、ジクロルメタン等のハロゲン
化炭化水素類;テトラハイドロフラン、エチルエ
ーテル、ジオキサン等のエーテル類;ベンゼン、
トルエン等の芳香族炭化水素類;アセトン、メチ
ルエチルケトン等のケトン類;酢酸エチルエステ
ル、酢酸メチルエステル等のエステル類;ジメチ
ルホルムアミド、ジメチルスルホキサイド等の極
性溶媒類;水及びこれらの混合溶媒等を使用でき
る。
〔製造法E〕
(式中R1、n、Hal及びOR′3は前記と同一の意
味を示す。)
上記スルフアイド化合物(b′)(一般式()
中Aがアミド結合を、R3が水素原子及び水酸基
以外の基を示すもの)は、上記反応式に示すよう
にスルフアイド化合物(XI)に、ハライド化合物
()を、好ましくは塩基性化合物の存在下、無
溶媒又は溶媒中、−20〜200℃程度の温度下に反応
させることにより製造される。ハライド化合物
()はスルフアイド化合物(XI)に対し、過剰
量使用してもよいが、好ましくは1〜4倍モル使
用される。塩基性化合物としては製造法Dに例示
したものを使用でき、これはハライド化合物
()に対して通常1〜4倍モル使用される。原
料とするスルフアイド化合物(XI)は上記製造法
Dに従い製造される。溶媒としては、反応に関与
しない溶媒であればよく、製造法Dに例示したと
同一の各種溶媒を使用できる。
〔製造法F〕
(式中R1、R3、n、OR′2及びHalは前記と同一
の意味を示す。)
上記スルフアイド化合物(b″)(一般式()
中Aがアミド結合を、R2が水素原子及び水酸基
以外の基を示すもの)は、上記反応式に示すよう
に、スルフアイド化合物(XII)に、ハライド化合
物()を、好ましくは塩基性化合物の存在下、
無溶媒又は溶媒中、−20〜200℃程度の温度下に反
応させることにより製造される。ハライド化合物
()はスルフアイド化合物(XII)に対し過剰量
使用してもよいが、好ましくは1〜4倍モル使用
される。塩基性化合物としては製造法Dに例示し
たものと同じものを使用でき、これはハライド化
合物()に対して、通常1〜4倍モル使用され
る。原料のスルフアイド化合物(XII)は上記製造
法D又はEに従い製造される。溶媒は、反応に関
与しないものであればよく、上記製造法Dに例示
したものと同一の各種の溶媒を使用できる。
上記製造法A〜Fに従い得られる本発明のスル
フアイド化合物は、反応終了後、通常の分離方法
に従つて単離できる。該分離方法としては、例え
ば再結晶法、抽出法、濃縮法、蒸留法、カラムク
ロマトグラフイー等を採用できる。
実施例
次に本発明を更に説明するため、参考例並びに
実施例を示す。
参考例 1
4−(3−エトキシ−2−ハイドロキシプロポ
キシ)フエニルカルバモイルメチルジメチルス
ルホニウムP−トルエンスルホネートの合成
4−(3−エトキシ−2−ハイドロキシプロポ
キシ)フエニルカルバモイルメチルメチルスルフ
アイド2.99gをメチレンクロライド20mlに溶解
後、これにP−トルエンスルホン酸メチル5.50g
を加えて、室温で24時間撹拌する。反応液にエー
テルを加えて不溶部を取し、メチレンクロライ
ド−エーテルより再結晶して、4−(3−エトキ
シ−2−ハイドロキシプロポキシ)フエニルカル
バモイルメチルジメチルスルホニウムP−トルエ
ンスルホネート4.75g(収率97.7%)を得る。

フアイド(化合物1)の合成
ジメチルホルムアミド4mlに、トリエチルアミ
ン2.00g及び4−(3−エトキシ−2−ハイドロ
キシプロポキシ)アニリン2.11gを溶解する。こ
れに氷冷下メチルメルカプトアセチルクロライド
1.25gを加える。室温で12時間撹拌後、反応液を
濃縮し、残渣をクロロホルム抽出する。クロロホ
ルム層を水洗した後、濃縮する。残渣をカラムク
ロマトグラフイー(シリカゲル、エタノール:ク
ロロホルム=1:5)にて精製し、4−(3−エ
トキシ−2−ハイドロキシプロポキシ)フエニル
カルバモイルメチルメチルスルフアイド(化合物
1)2.85g(収率95.3%)を得る。

アイド(化合物2)の合成
実施例1で合成した4−(3−エトキシ−2−
ハイドロキシプロポキシ)フエニルカルバモイル
メチルメチルスルフアイド4.00g及びピリジン
2.10gをベンゼン50mlに溶解する。氷冷下アセチ
ルクロライド1.60gを滴下する。室温で12時間撹
拌後、濃縮し、残渣をクロロホルム抽出し、クロ
ロホルム層を1N−HCl、1N−NaOHついで水で
順次洗浄する。クロロホルム層を濃縮し、残渣を
エーテル−石油エーテルより精製して、4−(2
−アセトキシ−3−エトキシプロポキシ)フエニ
ルカルバモイルメチルメチルスルフアイド(化合
物2)4.12g(収率90.4%)を得る。

水酸化ナトリウム0.40g及び90%メタノール10
mlに4−(2,3−ジハイドロキシプロポキシ)
フエノール1.84gを溶解後、これに2−メチルメ
ルカプトエチルクロライド1.11gを加え、6時間
還流する。反応液を濃縮し、残渣をクロロホルム
抽出する。クロロホルム層を水洗し、芒硝で脱水
後、濃縮する。残渣をベンゼン−石油エーテルよ
り再結晶して、2−{4−(2,3−ジハイドロキ
シプロポキシ)フエノキシ}エチルメチルスルフ
アイド(化合物3)2.25g(収率87.2%)を得
る。

77)の構造を表1に示すと共に、各例における収
率(%)並びに各化合物のmp(℃)、元素分析値
及び核磁気共鳴スペクトル(NMR)分析結果
(δ値、ppm)を下記表2に示す。尚表2中元素
