Description
【発明の詳細な説明】
本発明は新規なプロスタグランジン類似化合物
に関する。
プロスタグランジンは次の構造をもつプロスタ
ン酸の誘導体である。
種々のタイプのプロスタグランジンが知られてお
り、そのタイプは脂環式環の構造と置換基に依存
する。例えば、プロスタグランジンF(PGF)、
E(PGE)D(PGD)及びA(PGA)の脂環式環
はそれぞれ次の構造をもつ。

前記の構造式中もしくは本明細書中の他の構造式
中の点線は、一般的に認められている命名法の規
則に従い、それについている基が環平面のうしろ
側すなわちα−配置であり、太線はそれについ
ている基が環平面の前側すなわちβ−配置であ
り、波線はそれについている基がα−あるいは
β−配置、またはそれらの混合物であることを示
す。
それらの化合物は脂環式環の8位と12位につい
ている側鎖上の二重結合の位置によつて副分類さ
れる。PG−1化合物はC13−C14間にトランス二
重結合(トランス−Δ13)をもちPG−2化合物は
C5−C6間にシス二重結合とC13−C14間にトランス
二重結合(シス−Δ5、トランス−Δ13)をもつて
いる。例えば、プロスタグランジンD1(PGD1)
及びプロスタグランジンD2(PGD2)は、それぞ
れ次の構造式で示される。

PG−1化合物及びPG−2化合物の13位と14位
の炭素原子間がエチレン基でおきかえられた化合
物はジヒドロプロスタグランジン、例えばジヒド
ロプロスタグランジンD1(ジヒドロ−PGD1)及
びジヒドロプロスタグランジンD2(ジヒドロ−
PGD2)として知られている。
更にプロスタグランジンの脂環式環の12位につ
いている脂肪族基から1つまたはそれ以上のメチ
レン基が加えられたり、あるいは除去されたりし
たとき、その化合物は、有機命名法の一般則に従
つてホモ−プロスタグランジン(メチレン基が加
えられた場合)、または、ノル−プロスタグラン
ジン(メチレン基が除去された場合)として知ら
れており、そしてそれ以上のメチレン基が加えら
れるか、もしくは除去されたとき、その数は、
「ホモ」あるいは「ノル」の接頭語の前にジ−あ
るいはトリ−等を付けて示される。
プロスタグランジンは一般に薬理的性質を有す
る。例えばそれらは、平滑筋の収縮を刺激する作
用、血圧降下作用、利尿作用、気管支拡張作用、
脂肪分解阻害作用、血小板凝集抑制作用、胃酸分
泌抑制作用を有する。したがつて高血圧症、血栓
症、喘息及び胃腸の潰瘍の治療、妊娠哺乳動物の
分娩誘発及び中絶、動脈硬化の予防、または利尿
剤として有効である。それらは、動物体内の生体
内へプロスタグランジンを分泌する各組織中にご
く微量存在する脂溶性物質である。
例えば、PGE及びPGAは胃酸の分泌を抑制す
るので胃潰瘍の治療に用いられる。それらはま
た、エピネフイリンによつて引き起こされる遊離
脂肪酸の放出を阻害し、その結果、血中の遊離脂
肪酸濃度を下げるため動脈硬化症と高脂血症の予
防に有効である。PGE1は、血小板の凝集を抑制
し血栓を除去し血栓症を予防する。PGEとPGF
は平滑筋の刺激作用を有し腸管の蠕動運動を増大
させる。これらの作用は手術後の腸管閉塞症の治
療や下剤として有効であることを示している。更
にPGEとPGFは分娩促進剤、妊娠初期及び中期
における中絶剤及び出産後の胎盤の排出に用いら
れまた、雌の哺乳動物の性周期を調節するため経
口避妊剤としても用いられる。PGE及びPGAは
血管拡張及び利尿作用を有する。PGEは脳血流
量を増加させるため脳脈管系の病気の改善に有効
である。そしてまた、その気管支拡張作用のため
喘息の治療に有効である。
本発明者らは、「天然」のプロスタグランジン
の薬理作用を有するか、またはそれらの性質のひ
とつまたはそれ以上の性質で、より強い活性を有
するか、あるいは「天然」のプロスタグランジン
には全く見られないような性質を有する新規な化
合物を見い出すために幅広い研究を行つた結果、
脂環式環の構造がPGD、PGA又は【式】
(9−デオキシ−Δ9−PGD)であり、15位に
【式】(基中、R3、R4及びR5は同じ
であつてもあるいは異なつていてもよく、それぞ
れ水素原子または炭素数1〜4の直鎖あるいは分
枝鎖アルキルを表わす。)を有するプロスタグラ
ンジン類似化合物が、「天然」のプロスタグラン
ジンが有する薬理的性質は有さないか、または有
していても非常に弱いのに反し、抗腫瘍作用に驚
くほど強いことを見い出し本発明を完成した。
従来より、PGD、PGA及び9−デオキシ−Δ9
−PGD類似化合物に関する出願はいくつかなさ
れているが、その範囲には本発明化合物は全く含
まれない。また、本発明の特徴である15位に
【式】基(基中、全ての記号は前記と
同じ意味を表わす。)を導入することは、これら
公知のプロスタグランジン類似化合物から容易に
考えられるものではない。
さらに重要なことは、本発明化合物は、公知の
プロスタグランジン類似化合物には全く見い出さ
れていなかつた新規な作用、すなわち抗腫瘍作用
を有している点にある。公知のプロスタグランジ
ン類似化合物に関する公開明細書では、それら化
合物が一般のプロスタグランジンに知られていた
薬理的性質を有することを述べているにとどま
り、プロスタグランジン類似化合物が抗腫瘍作用
を有することに着目したものは皆無である。
本発明は、一般式

ス−ビニレン基を表わし、Yはエチレン基または
トランス−ビニレン基を表わし、Rは水素原子ま
たは炭素数1〜12の直鎖あるいは分枝鎖アルキル
基を表わし、R1は単結合または炭素数1〜5の
直鎖あるいは分枝鎖アルキレン基を表わし、R2
は炭素数1〜8の直鎖あるいは分枝鎖アルキル
基、置換されていないかあるいは少なくとも1つ
の炭素数1〜8のアルキル基で置換されている炭
素数4〜7のシクロアルキル基または置換されて
いないかあるいは少なくとも1つのハロゲン原
子、トリフルオロメチル基あるいは炭素数1〜4
の直鎖あるいは分枝鎖アルキル基で置換されてい
るフエニル基あるいはフエノキシ基を表わし、
R3、R4及びR5は前記と同じ意味を表わし、波線
はそれについている基がα−あるいはβ−配置
またはそれらの混合物であることを表わす。ただ
しR1が単結合を表わす場合、R2は置換または無
置換のフエノキシ基を表わさないものとする。〕
で示されるプロスタグランジン類似化合物、及び
それらのシクロデキストリン包接化合物、及びR
が水素原子を表わす場合には、その酸の非毒性塩
に関する。
本発明化合物としては、一般式
(式中、全ての記号は前記と同じ意味を表わ
す。)
で示されるPGD類似化合物、及び一般式
(式中、全ての記号は前記と同じ意味を表わ
す。)
で示される9−デオキシ−Δ9−PGD類似化合物、
及び一般式
(式中、全ての記号は前記と同じ意味を表わ
す。)
で示されるPGA類似化合物が含まれる。
一般式()で示される化合物には、いくつか
の不斉炭素が存在する。例えば一般式(a)で
示される化合物には、4個の不斉炭素(すなわ
ち、8位、9位、12位及び15位の炭素原子)が存
在し、一般式(b)及び(c)で示される化
合物では、8位、12位及び15位の炭素原子が不斉
炭素であり、さらにR、R1、R2、R3、R4、及び
R5が表わす種々の置換基のアルキル部分が分枝
鎖である場合には、その他の不斉炭素が生じる可
能性がある。しかしながら、本発明の一般式
()で示される化合物は、不斉炭素によつて生
じる各々の異性体及びそれらの混合物をも含有す
るものである。
一般式()において、Rが表わす炭素数1〜
12のアルキル基としては、メチル、エチル、プロ
ピル、ブチル、ペンチル、ヘキシル、ヘプチル、
オクチル、ノニル、デシル、ウンデシル、ドデシ
ル基及びそれらの異性体が挙げられ、好ましいR
は水素原子または炭素数1〜4のアルキル基であ
り、より好ましいRは水素原子またはメチルまた
はエチル基である。
一般式()において、R1が表わす炭素数1
〜5のアルキレン基としては、メチレン、エチレ
ン、トリメチレン、テトラメチレン、ペンタメチ
レン基及びそれらの異性体が挙げられ、好ましい
R1は単結合またはメチレンまたはエチレン基で
ある。
一般式()において、R2が表わす炭素数1
〜8のアルキル基としては、メチル、エチル、プ
ロピル、ブチル、ペンチル、ヘキシル、ヘプチ
ル、オクチル基及びそれらの異性体が挙げられ、
好ましくは、無置換であるかあるいは1個または
2個のメチルまたはエチル基で置換されたブチ
ル、ペンチル、ヘキシル基である。
一般式()において、R2が表わす置換また
は無置換のシクロアルキル基としては、シクロブ
チル、シクロペンチル、シクロヘキシル、シクロ
ヘプチル基及びひとつまたはそれ以上のメチル、
エチル、プロピル、ブチル、ペンチル、ヘキシ
ル、ヘプチルまたはオクチル基で置換されたシク
ロブチル、シクロペンチル、シクロヘキシル、シ
クロヘプチル基が挙げられ、好ましくは、無置換
であるかあるいは1個のメチル、エチル、プロピ
ルまたはブチル基で置換されたシクロブチル、シ
クロペンチルまたはシクロヘキシル基である。
一般式()において、R2が表わす置換また
は無置換のフエニルまたはフエノキシ基として
は、フエニル、フエノキシ基及びひとつまたはそ
れ以上のフツ素原子、塩素原子、トリフルオロメ
チル基、メチル、エチル、プロピルまたはブチル
基で置換されたフエニルまたはフエノキシ基が挙
げられ、好ましくは、無置換であるかあるいは1
個の塩素原子、トリフルオロメチル基、メチル基
またはエチル基で置換されたフエニルまたはフエ
ノキシ基である。
一般式()において、好ましいR1−R2とし
ては、ブチル、ペンチル、1−メチルペンチル、
2−メチルペンチル、3−メチルペンチル、1,
1−ジメチルペンチル、1−エチルペンチル、2
−エチルペンチル、ヘキシル、1−メチルヘキシ
ル、2−メチルヘキシル、1−エチルヘキシル、
2−エチルヘキシル、ヘプチル、2−メチルヘプ
チル、2−エチルヘプチル、シクロブチル、1−
メチルシクロブチル、1−プロピルシクロブチ
ル、1−ブチルシクロブチル、3−エチルシクロ
ブチル、3−プロピルシクロブチル、シクロペン
チル、シクロペンチルメチル、2−シクロペンチ
ルエチル、3−エチルシクロペンチル、3−プロ
ピルシクロペンチル、3−ブチルシクロペンチ
ル、シクロヘキシル、シクロヘキシルメチル、2
−シクロヘキシルエチル、3−エチルシクロヘキ
シル、3−プロピルシクロヘキシル、3−ブチル

4−エチルフエノキシメチルが挙げられる。
一般式()において、R3、R4およびR5が表
わす炭素数1〜4のアルキル基としては、メチ
ル、エチル、プロピル、ブチル基及びそれらの異
性体が挙げられ、好ましいR3は水素原子または
メチルあるいはエチル基であり、より好ましい
R3は水素原子である。好ましいR4及びR5は水素
原子またはメチルあるいはエチル基であり、より
好ましいR4及びR5はメチル基である。
さらに一般式()において、好ましいXはシ
ス−ビニレン基であり、好ましいYはトランス−
ビニレン基である。また、15位の炭素原子につい
ている基の好ましい配置はα−配置である。
本発明に従えば、全ての記号が前記と同じ意味
を表わす一般式(b)の9−デオキシ−Δ9−
PGD類似化合物は、全ての記号が前記と同じ意
味を表わす一般式(a)のPGD類似化合物を
緩和な脱水反応、すなわち15位のエステル結合が
できるだけ影響を受けないような脱水反応、例え
ば適当な有機溶媒中でトリエチルアミンのような
塩基の存在下に、アルキルまたはアリルスルホニ
ルクロライド、例えばメタンスルホニルクロライ
ドを用いて低温で、好ましくは−30℃〜0℃で反
応させることにより製造される。
これと全く同様の方法を用いて、全ての記号が
前記と同じ意味を表わす一般式(c)のPGA
類似化合物が、一般式

で示されるPGE類似化合物から製造される。
一般式(a)で示されるPGD類似化合物及
び一般式(V)で示されるPGE類似化合物は、

で示されるPGF類似化合物の9位についている
水酸基または11位についている水酸基をオキソ基
に酸化することによつて製造される。このような
酸化反応はよく知られており、例えば

化またはスワン酸化が用いられる。コリンズ試薬
を用いる酸化反応は、クロロホルム、塩化メチレ
ン、四塩化炭素の如きハロゲン化炭化水素中室温
から0℃の温度、好ましくは0℃で行なわれ、ジ
ヨーンズ試薬を用いる酸化反応は、通常室温以下
の温度で行なわれ、スワン酸化はクロロホルム、
塩化メチレンのようなハロゲン化炭化水素中−
50゜〜−60゜で反応させ、次いでトリエチルアミン
で処理することにより行なわれる。
この酸化反応によつて得られる一般式(a)
で示されるPGD類似化合物及び一般式()で
示されるPGF類似化合物は、通常の単離精製手
段、例えばシリカゲルの薄層またはカラムまたは
高速液体クロマトグラフイーを用いて、それぞれ
に単離される。
一般式()で示されるPGF類似化合物は、
次の図式に示される一連の反応工程によつて製造
することができる。図式中、R6は炭素数1〜4
の直鎖または分枝鎖アルキル基あるいはフエニル
基、好ましくはn−ブチル基またはフエニル基を
表わし、その他の記号は前記と同じ意味を表わ
す。
図式中の各工程はすべて公知の反応により行な
われる。例えば工程〔a〕は塩化メチレン、クロ
ロホルム、四塩化炭素またはテトラヒドロフラン
のような不活性有機溶媒中、モレキユラーシーブ
ス3Aまたは4Aの存在下、R6に相当するボロン
酸、例えばフエニルボロン酸〔PhB(OH)2〕ま
たはブチルボロン酸〔n−BuB(OH)2〕と還流
温度で反応させることにより行なわれる〔J.C.S.
Chem.Comm.,658ページ(1975年)及び
Prostaglandins,9巻,109ページ(1975年)参
照のこと。〕
工程〔b〕は一般によく知られているアルコー
ルとハロゲン化アシルとの反応であり、不活性有
機溶媒、例えば塩化メチレン、クロロホルムのよ
うなハロゲン化炭化水素中トリエチルアミンのよ
うな三級アミンまたはピリジン、好ましくはピリ
ジンの存在下で、一般式
〔式中、R7はハロゲン原子を表わし、その他
の記号は前記と同じ意味を表わす。〕
で示されるハロゲン化アシルを用いて室温で行な
われる。
工程〔c〕は炭素数1〜4のアルカノール水溶
液、例えば含水メタノールまたは含水エタノール
中、60℃以下の温度、好ましくは45〜50℃で過酸
化水素水と反応させることにより行なわれる。
上記工程〔a〕、〔b〕及び〔c〕は途中で精製
することなく、連続的に反応させることができ
る。
一般式(a)で示されるPGD類似化合物は、
次の図式に示される一連の反応工程によつてもま
た製造することができる。図式中、R7はトレメ
チルシリル基、トリエチルシリル基、トリブチル
シリル基、tert−ブチルジメチルシリル基、トリ
ベンジルシリル基、トリフエニルシリル基のよう
な三置換シリル基、好ましくはtert−ブチルジメ
チルシリル基を表わし、R8は置換されていない
かまたは少なくとも1個のアルキル基で置換され
ているテトラヒドロピラン−2−イル基またはテ
トラヒドロフラン−2−イル基あるいは1−エト
キシエチル基、好ましくはテトラヒドロピラン−
2−イル基を表わし、その他の記号は前記と同じ
意味を表わす。
図式中の各工程は全て公知の反応により行われ
るが、簡単に説明すると、例えば工程〔d〕は11
位の水酸基を選択的にOR7基に変換する反応であ
り、その反応は、John Wiley&Sons,Inc.,社
(米国)発行、“Protective Groups in Organic
Synthesis”,39〜50ページ(1981)(以後、文献
Aと略記する。)に詳しく記載されているが、好
ましくはジメチルアミノピリジン、ジメチルホル
ミアミド中ピリジン、イミダゾール、トリエチル
アミンのような塩基の存在下、室温から50℃の温
度で式Cl−R7(式中、R7は前記と同じ意味を表わ
す。)で示されるクロロシラン化合物と反応させ
ることによつて行われる。
工程〔e〕は不活性有機溶媒、例えば塩化メチ
レン、クロロホルム、ジエチルエーテル中、縮合
剤、例えばp−トルエンスルホン酸、硫酸、トリ
フルオロ酢酸の存在下、2,3−ジヒドロピラ
ン、2,3−ジヒドロフラン、エチルビニルエー
テル等を用いて室温から−30℃の温度で行なわれ
る。好適には塩化メチレン中、ピリジンのp−ト
ルエンスルホン酸塩またはp−トルエンスルホン
酸の存在下、2,3−ジヒドロピランを用いて室
温で行なわれる。
工程〔f〕はOR7基を水酸基に変換する反応で
あり、文献Aに詳しく記載されているが、好まし
くはテトラヒドロフラン中、室温でトラブチルア
ンモニウムフロリド(n−Bu4N+F-)を用いて
行われる。
工程〔g〕は水酸基をオキソ基に変換する酸化
反応であり、一般式()の化合物から一般式
(a)及び一般式()の化合物を得るために
前記した酸化反応を用いて行われる。
工程(h)は酸性条件下で加水分解することに
より行なわれる。この反応は、
(1) 酢酸、プロピオン酸、シユウ酸、p−トルエ
ンスルホン酸の如き有機酸の水溶液又は塩酸、
硫酸、リン酸の如き無機酸の水溶液中、好適に
は水と混和しうる有機溶媒、例えばメタノール
又はエタノールの如き低級アルカノール(好ま
しくはメタノール)、又は1,2−ジメトキシ
エタン、ジオキサンもしくはテトラヒドロフラ
ンの如きエーテル(好ましくはテトラヒドロフ
ラン)の存在下に室温から75℃の温度(好まし
くは45℃以下の温度)で、又は
(2) メタノール又はエタノールの如き無水低級ア
ルカノール中p−トルエンスルホン酸又はトリ
フルオロ酢酸の如き有機酸の存在下10〜45℃の
温度で行なわれる。加水分解は、好ましくは希
塩酸及びテトラヒドロフランの混合液、希塩酸
及びメタノールの混合液、酢酸、水及びテトラ
ヒドロフランの混合液、リン酸、水及びテトラ
ヒドロフランの混合液、又はp−トルエンスル
ホン酸及び無水メタノールの混合液を用いて行
なわれる。
一般式(b)で示される9−デオキシ−Δ9
−PGD類似化合物は一般式()の化合物から、
次の図式で示される一連の反応工程によつてもま
た製造することができる。図式中、Tはアルキル
スルホニル基あるいは置換または無置換のアリル
スルホニル基、好ましくはメタンスルホニル基あ
るいはトリルスルホニル基を表わし、その他の記
号は前記と同じ意味を表わす。

図式中のそれぞれの反応工程において、反応工
程〔b〕、〔c〕、〔d〕、〔e〕、〔f〕及び〔h〕
は、それぞれ前記した反応工程〔b〕、〔c〕、
〔d〕、〔e〕、〔f〕及び〔h〕と同様の反応工程
であり、反応工程〔g〕は前記した反応工程
〔g〕同様の酸化工程により、水酸基のオキソ基
への変換及びOT基の脱離とC9−C10間への二重
結合の導入が同時に行なわれる。
工程〔j〕は9位の水素基をOT基に変換する
反応であり、()ピリジンまたはトリエチルア
ミンのような三級アミンの存在下、塩化メチレン
のような不活性有機溶媒中、あるいは()ピリ
ジン中、−30℃から50℃の温度で、メタンスルホ
ニルクロライドのようなアルキルスルホニルクロ
ライドまたはトリルスルホニルクロライドのよう
なアリルスルホニルクロライドと反応させること
により行なわれる。
本発明において、出発物質である一般式()
の化合物はPGFまたはPGF類似化合物として、
それ自身公知であるが、次の図式で示されるそれ
ぞれの反応工程を必要に応じて行うことによつて
もまた製造することができる。図式中、R9は炭
素数1〜4の直鎖または分枝鎖アルキル基を表わ
し、R10は炭素数5〜12の直鎖または分枝鎖アル
キル基を表わし、その他の記号は前記と同じ意味
を表わす。
図式中、工程〔h〕は前記の工程〔h〕と同じ
工程であり、工程〔k〕はエステルを加水分解す
る通常の反応であり、アルカリ条件下で行なわ
れ、例えばこの加水分解は含水メタノールまたは
含水エタノールのような含水低級アルカノール
中、ナトリウムまたはカリウムのようなアルカリ
金属の水酸化物を用いて、室温以上の温度、好ま
しくは50〜60℃で行なわれる。
工程〔m〕は通常のエステル化反応であり、例
えば
(1) ジアゾアルカンを用いる方法、あるいは
(2) アルキルハライドを用いる方法、あるいは
(3) N,N−ジメチルホルムアミド−ジアルキル
アセタールを用いる方法、あるいは
(4) 日本特許第762305号明細書記載のジシクロヘ
キシルカルボジイミドと相当するアルコールを
用いる方法、あるいは
(5) 日本特許第756972号明細書記載のピパロイル
ハライドと相当するアルコールを用いる方法、
あるいは
(6) 日本特許第759351号明細書記載のアリールス
ルホニルハライド又はアルキルスルホニルハラ
イドと相当するアルコールを用いる方法が挙げ
られる。
ジアゾアルカンを用いる方法は、不活性有機溶
媒、例えばジエチルエーテル、酢酸エチル、塩化
メチレン、アセトンあるいはそれらの2以上の混
合溶媒中−10℃から室温、好ましくは0℃で相当
するジアゾアルカンと反応させることにより行な
われる。
アルキルハライドを用いる方法は、N,N−ジ
メチルアセトアミド又はN,N−ジメチルホルム
アミド中炭酸水素ナトリウム又は炭酸水素カリウ
ムの如きアルカリ金属の炭酸水素塩の存在下
〔Advan.Org Chem.,537(1965)参照〕で相当
するアルキルハライド、例えば沃化メチルを0℃
から室温で反応させて行なわれる。
N,N−ジメチルホルムアミド−ジアルキルア
セタールを用いる方法は、無水ベンゼン中N,N
−ジメチルホルムアミド−ジアルキルアセター
ル、例えばN,N−ジメチルホルムアミド−ジメ
チルアセタールを0℃から室温で反応させて行な
われる。
ジシクロヘキシルカルボジイミドを用いる方法
は、不活性有機溶媒、例えばクロロホルム、塩化
メチレンの如きハロゲン化炭化水素中ピリジン、
ピコリンの如き塩基、好ましくは4−ジメチルア
ミノピリジン存在下相当する酸と適当なアルコー
ルを室温から0℃の温度で反応させて行なわれ
る。
ピバロイルハライド又はアリールスルホニルハ
ライド又はアルキルスルホニルハライドを用いる
方法は、不活性有機溶媒、例えばクロロホルムも
しくは塩化メチレンの如きハロゲン化炭化水素類
又はジエチルエーテル中又は溶媒不存在下トリエ
チルアミンの如き三級アミン又はピリジンと、ピ
バロイルハライド、例えばピバロイルクロリド、
もしくはアリールスルホニルハライド、例えばベ
ンゼンスルホニルクロリドあるいはp−トルエン
スルホニルクロリド、もしくはアルキルスルホニ
ルハライド、例えばメタンスルホニルクロリドあ
るいはエタンスルホニルクロリドを加えて混合酸
無水物とし、次に相当するアルコールを加え温室
から0℃の温度で行なわれる。
一般式(XI)で示される化合物は、以下に示
される文献及び特許公開明細書に記載された方法
またはそれらの自明な類似方法により製造するこ
とができる。すなわち、
(1) R1−R2がペンチル基を表わす場合には、J.
Amer.Chem.Soc.,91,5675(1969)又は同誌,
92,397(1970)に記載された方法、
(2) R1−R2がアルキル基を表わす場合には、特
開昭47−42675、48−54068、48−64073、49−
124048、50−95250、50−96543又は50−101340
号の明細書に記載された方法、
(3) R1−R2のR2がシクロアルキル基を表わす場
合には、特開昭49−109353、50−95250、50−
96543、50−123647、50−148339、51−122040、
51−125256、52−27753又は53−25544号の明細
書に記載された方法、
(4) R1−R2基のR2がフエニル基又はフエノキシ
基を表わす場合には、特開昭50−95250、50−
96543、51−59841、51−101961又は52−25745
号の明細書に記載された方法により製造され
る。
一般式(c)で示されるPGA類似化合物は
一般式(XI)の化合物から次の図式で示される
一連の反応工程によつてもまた製造することがで
きる。図式中、全ての記号は前記と同じ意味を表
わし、工程〔b〕、〔g〕、〔h〕及び〔m〕は、そ
れぞれ前記した工程〔b〕、〔g〕、〔h〕及び
〔m〕と同じ工程であることを表わし、反応工程
〔n〕はPGE化合物をPGA化合物にするために、
よく知られた反応工程であり、例えば塩酸のよう
な無機酸あるいは酢酸のような有機酸の水溶液
中、50℃〜80℃の温度で行なわれる。
反応工程〔p〕はPGA化合物のエステル加水
分解のために良く知られた方法、例えばパン酵母
を用いる生物学的方法によつて行なわれる。

一般式()で示されるプロスタグランジン類
似化合物のシクロデキストリン包接化合物は、α
−、β−あるいはγ−シクロデキストリンあるい
はそれらの混合物を用いて日本特許第790979号及
び特開昭47−39057号明細書記載の方法を用いる
ことにより得られる。シクロデキストリン包接化
合物に変換することにより一般式()で示され
るプロスタグランジン類似化合物の安定性が増大
する。
Rが水素原子を表わす一般式()で示される
化合物は、所望により公知の方法で塩に変換され
る。塩は非毒性の塩であることが好ましい。ここ
で非毒性の塩とは動物の組織に対して比較的無害
で、治療に必要な量を用いたとき一般式()で
示される化合物の有効な薬理的性質がそのカチオ
ンにより生じた副作用によつて損なわれないよう
なカチオンの塩を意味する。塩は水溶性であるこ
とが好ましい。適当な塩には、例えばナトリウム
又はカリウムの如きアルカリ金属の塩、カルシウ
ム又はマグネシウムの如きアルカリ土類金属の
塩、アンモニウム塩及び薬学的に許容される(非
毒性の)アミン塩が含まれる。カルボン酸とその
ような塩を形成する適当なアミンはよく知られて
おり、例えば理論上アンモニアの1個あるいはそ
れ以上の水素原子を他の基に置き変えて得られる
アミンが含まれる。その基は1個以上の水素原子
が置換されているときは同じでも異なつてもよい
が、例えば炭素数1〜6のアルキル基、炭素数1
〜3のヒドロキシアルキル基から選ばれる。適当
な非毒性アミン塩は、テトラメチルアンモニウム
の如きテトラアルキルアンモニウムの塩、及びメ
チルアミン塩、ジメチルアミン塩、シクロペンチ
ルアミン塩、ベンジルアミン塩、フエネチルアミ
ン塩、ピペリジン塩、モノエタノールアミン塩、
ジエタノールアミン塩、リジン塩、又はアルギニ
ン塩の如き他の有機アミン塩が挙げられる。
塩は、Rが水素原子を表わす一般式()で示
される酸を公知の方法、例えば適当な溶媒中で一
般式()で示される酸と適当な塩基、例えばア
ルカリ金属又はアルカリ土類金属の水酸化物ある
いは炭酸塩、水酸化アンモニウム、アンモニア又
は有機アミンを理論量ずつ反応させて得られる。
塩は溶液を凍結乾燥するか、あるいは反応溶媒に
充分不溶であるならば過するか、あるいは必要
ならば溶媒を一部除去した後に過することによ
り単離される。
一般式()で示されるプロスタグランジン類
似化合物、及び一般式()で示される化合物の
シクロデキストリン包接化合物、及び一般式
()において、Rが水素原子を表わす場合には
その酸の非毒性塩は、プロスタグランジンに特有
の薬理作用、例えば血圧降下作用、血小板凝縮抑
制作用、子宮筋の収縮を刺激する作用、下痢作用
を全く示さないか、またはそれらの作用が非常に
弱いものであるにもかかわらず、抗腫瘍作用が驚
くほど強力で、しかも毒性は極めて少ないため、
例えば白血病及び固型ガンに対する予防効果、治
療効果及び延命効果を目的とする非常に有効で理
想的な抗腫瘍剤として用いることができる。
例えば、実験室の実験では、ヒトの口底癌細胞
(KB cell)を用いた、in vitroでの増殖抑制試験
において、本発明化合物は優れた抑制効果を示し
た。以下にその実験方法と結果を示す。
ヒトの口底癌細胞(KB cell)を用いた増殖抑
制試験
<実験方法>
ヒト口底癌細胞を用いた増殖抑制試験はよく知
られた方法で行なつた。すなわちヒトの口底癌細
胞(KB cell)を、10%の仔牛胎児血清含有のイ
ーグルMEM培養液に加え、培養液中の細胞数を
1×105個/mlに調整し、本発明化合物のエタノ
ール溶液5μg/ml又は1μg/mlを加えた後、37
℃で4日間静置培養した。対照としてエタノール
0.1%を加えた培養液も同様に培養した。培養後
トリパンブルー染色法で染色し、生細胞数を測定
して対照に対する抑制率からIC50値(50%増殖抑
制濃度)を求めた。

一般式()で示されるプロスタグランジン類
似化合物、及びそれらのシクロデキストリン包接
化合物、及びそれらの非毒性塩を抗腫瘍剤として
用いるには、通常全身的あるいは局所的に、経口
または非経口で投与される。投与量は年令、体
重、症状、治療効果、投与方法、処理時間等によ
り異なるが、通常成人ひとり当り、1回につき5
mg〜500mgの範囲で1日1回から数回経口投与さ
れるかまたは成人ひとり当り、1回につき500μ
g〜50mgの範囲で1日1回から数回非経口投与、
好ましくは静脈内投与される。もちろん前記した
ように、投与量は種々の条件で変動するので、上
記投与量範囲より少ない量で十分な場合もある
し、また範囲を越えて必要な場合もある。
本発明による経口投与のための固体組成物とし
ては、錠剤、散剤、顆粒剤等が含まれる。このよ
うな固体組成物においては、ひとつまたはそれ以
上の活性物質が、少なくともひとつの不活性な希
釈剤、例えば乳糖、マンニトール、ブドウ糖、ヒ
ドロキシプロピルセルロース、微結晶セルロー
ス、デンプン、ポリビニルピロリドン、メタケイ
酸アルミン酸マグネシウムと混合される。組成物
は常法に従つて、不活性な希釈剤以外の添加剤、
例えばステアリン酸マグネシウムのような潤滑剤
や繊維素グルコン酸カルシウムのような崩壊剤を
含有していてもよい。錠剤または丸剤は必要によ
り白糖、ゼラチン、ヒドロキシプロピルセルロー
ス、ヒドロキシプロピルメチルセルロースフタレ
ートなどの胃溶性あるいは腸溶性物質のフイルム
で被膜してもよいし、また2以上の層で被膜して
もよい。さらにゼラチンのような吸収されうる物
質のカプセル剤としてもよい。
経口投与のための液体組成物は、薬剤的に許容
される乳濁剤、溶液剤、懸濁剤、シロツプ剤、エ
リキシル剤等を含み、一般的に用いられる不活性
な希釈剤、例えば精製水、エタノールを含む。こ
の組成物は不活性な希釈剤以外に湿潤剤、懸濁剤
のような補助剤、甘味剤、風味剤、芳香剤、防腐
剤を含有していてもよい。
経口投与のためのその他の組成物としては、ひ
とつまたはそれ以上の活性物質を含み、それ自体
公知の方法により処方されるスプレー剤が含まれ
る。
本発明による非経口投与のための注射剤として
は、無菌の水性または非水性の溶液剤、懸濁剤、
乳濁剤を包含する。水性の溶液剤、懸濁剤として
は例えば注射用蒸留水及び生理食塩水が含まれ
る。非水性の溶液剤、懸濁剤としては、例えばプ
ロピレングリコール、ポリエチレングリコール、
オリーブ油のような植物油、エタノールのような
アルコール類、ポリソルベート80等がある。この
ような組成物は、さらに防腐剤、湿潤剤、乳化
剤、分散剤のような補助剤を含んでもよい。これ
らは例えばバクテリア保留フイルターを通す
過、殺菌剤の配合または照射によつて無菌化され
る。これらはまた無菌の固体組成物を製造し、使
用前に無菌水または無菌の注射用溶媒に溶解して
使用することもできる。
非経口投与のためその他の組成物としては、ひ
とつまたはそれ以上の活性物質を含み、それ自体
公知の方法により処方される外用液剤、軟コウの
ような塗布剤、直腸内投与のための坐剤及び腟内
投与のためのペツサリー等が含まれる。
本発明に含まれる一般式()で示される化合
物のうち、好ましいものとしては、例えば、一般
式()で示されるプロスタグランジン類似化合
物において、15位の置換基が【式】
であり、R1−R2がペンチル、1−メチルペンチ
ル、1,1−ジメチルペンチル、2−メチルペン
チル、2−メチルヘキシル、1−メチルシクロブ
チル、1−ブチルシクロブチル、シクロペンチ
ル、3−エチルシクロペンチル、3−プロピルシ
クロペンチル、3−ブチルシクロペンチル、シク
ロヘキシル、3−エチルシクロヘキシル、3−プ
ロピルシクロヘキシル、3−ブチルシクロヘキシ
ル、4−メチルシクロヘキシル、4−エチルシク
ロヘキシル、4−プロピルシクロヘキシル、ベン
ジル、フエノキシメチル、3−クロロフエノキシ
メチルまたは3−トリフルオロメチルフエノキシ
メチル基である、PGD1及びPGD2類似化合物、
9−デオキシ−Δ9−PGD1及び9−デオキシ−Δ9
−PGD2類似化合物、及びPGA1及びPGA2類似化
合物、およびそれら類似化合物の13,14−ジヒド
ロ化合物及びそれら類似化合物のメチルまたはエ
チルエステルが挙げられる。
以下、参考例及び実施例により本発明を詳述す
るが、本発明はこれらの実施例に限定されるもの
ではない。なお参考例及び実施例中の「TLC」、
「NMR」、「IR」及び「Mass」は、各々「薄層ク
ロマトグラフイ」「核磁気共鳴スペクトル」、「赤
外吸収スペクトル」及び「質量分析」を表わす。
クロマトグラフイによる分離の箇所に記載されて
いるカツコ内の溶媒は使用した展開溶媒を示す。
特別の記載がない場合には、IRは液膜法で測定
し、NMRは重クロロホルム(CDCl3)溶液で測
定している。
参考例 1
15−O−(3,3−ジメチル)アクリロイル−
PGF2〓メチルエステル
乾燥塩化メチレン15mlに溶かしたPGF2〓メチル
エステル1.104gとフエニルボロン酸439mgの溶液
に4Aモレキユラーシーブス2gを加えて、6時
間還流した。室温に冷却した後ピリジン1.9mlと
3,3−ジメチルアクリロイルクロライド1.35ml
を加えて、室温で1.5時間かきまぜたのちエーテ
ル200mlを加えて上澄液を分取した。得られた溶
液を水洗し、硫酸マグネシウムで乾燥後減圧濃縮
した。得られた残留物にエタノール5mlと炭酸水
素ナトリウム0.5gを加えたのち、氷冷下に35%
過酸化水素水1.0mlを加えて室温で30分間かきま
ぜた。得られた反応混合物にジエチルエーテル
200mlを加えて、飽和炭酸水素ナトリウム水溶液
及び水で洗浄して、硫酸マグネシウムで乾燥後減
圧濃縮した。得られた残留物をシリカゲルカラム
クロマトグラフイー(溶出溶媒,酢酸エチル:シ
クロヘキサン=1:1)で精製して次の物理的性
質を有する表題化合物1.15gを得た。

実施例 1
15−O−(3,3−ジメチル)アクリロイル−
PGD2メチルエステル及び15−O−(3,3−
ジメチル)アクリロイル−PGE2メチルエステ
ル
アセトン20mlに溶かした15−O−(3,3−ジ
メチル)アクリロイル−PGF2〓メチルエステル
(参考例1で製造した)910mgを−40℃に冷却し、
ジヨーンズ試薬2.3mlを滴下し、同温度で30分間
かきまぜたのちイソプロピルアルコール1mlを加

Claims (1)
ス−ビニレン基を表わし、Yはエチレン基または
トランス−ビニレン基を表わし、Rは水素原子ま
たは炭素数1〜12の直鎖あるいは分枝鎖アルキル
基を表わし、R1は単結合または炭素数1〜5の
直鎖あるいは分枝鎖アルキレン基を表わし、R2
は炭素数1〜8の直鎖あるいは分枝鎖アルキル
基、置換されていないかあるいは少なくとも1つ
の炭素数1〜8のアルキル基で置換されている炭
素数4〜7のシクロアルキル基または置換されて
いないかあるいは少なくとも1つのハロゲン原
子、トリフルオロメチル基あるいは炭素数1〜4
の直鎖あるいは分枝鎖アルキル基で置換されてい
るフエニル基あるいはフエノキシ基を表わし、
R3、R4及びR5は同じあつてもあるいは異なつて
もよく、それぞれ水素原子または炭素数1〜4の
直鎖あるいは分枝鎖アルキル基を表わし、波線
はそれについている基がαあるいはβ−配置また
はそれらの混合物であることを表わす。ただし
R1が単結合を表わす場合、R2は置換または無置
