Description
この発明は薬用ニンジンより単離された新規な
サポニン成分及びその単離法に関する。
薬用ニンジン中特にウコギ科に属するオタネニ
ンジン(パナツクス ギンゼング シーエー メ
イヤー,Panax ginseng C.A.Meyer)は一名チ
ヨウセンニンジンと呼ばれ、古来より強壮、強
精、消炎、利尿、抗糖尿用の薬剤として用いられ
てきたことは広く知られているところである。
近時薬用ニンジンの含有成分の研究が進めら
れ、トリテルペンのダンマラン系配糖体に属する
サポニンとしてギンゼノサイドRb1、Rb2、Rb3、
Rc、Rd、Re、Rf、Rg1、Rg2、Rh1及びRh2並び
にギンゼノサイド20−グルコ−Rfなどが知られ
ている(Chem Pharm.Bull.22(2),421〜428
(1974)および薬学雑誌94(2),252〜260(1794)
参照)。
従来上記のような公知のギンゼノサイド類は、
原料を水、低級アルコール類又は含水低級アルコ
ール類で抽出した後、その抽出物を水−n−ブタ
ノールで分配抽出しそのn−ブタノール層から単
離精製して得られている。一方水層の方は何らか
えりみられることなく排棄されていた。この発明
の発明者らはこの従来排棄されていた水層に着目
し、これに文献未記載の特に水に易溶性の新規な
サポニン物質が含有されていることを見出し、こ
れを単離しその構造を確認してこの発明をなすに
至つた。
この発明による新規サポニン類は、下記式
()で表すことができる。

上式においてR1がβ−D−グルコピラノシル
(1→6)−β−D−グルコピラノシル基の化合物
の名称は、3−O−〔6−O−マロニル−β−D
−グルコピラノシル(1→2)−β−D−グルコ
ピラノシル〕−20−O−〔β−D−グルコピラノシ
ル(1→6)−β−D−グルコピラノシル〕20
(S)−プロトパナキサジオール,(以下マロニル
−ギンゼノサイドRb1と称す);R1がα−L−ア
ラビノピラノシル(1→6)−β−D−グルコピ
ラノシル基の化合物の名称は、3−O−〔6−O
−マロニル−β−D−グルコピラノシル(1→
2)−β−D−グルコピラノシル〕−20−O−〔α
−L−アラビノピラノシル(1→6)−β−D−
グルコピラノシル〕20(S)−プロトパナキサジオ
ール,(以下マロニル−ギンゼノサイドRb2と称
す);R1がα−L−アラビノフラノシル(1→
6)−β−D−グルコピラノシル基の化合物の名
称は、3−O−〔6−O−マロニル−β−D−グ
ルコピラノシル(1→2)−β−D−グルコピラ
ノシル〕−20−O−〔α−L−アラビノフラノシル
(1→6)−β−D−グルコピラノシル〕20(S)−
プロトパナキサジオール(以下マロニル−ギンゼ
ノサイドRcと称す);及びR1がβ−D−グルコピ
ラノシル基の化合物の名称は3−O−〔6−O−
マロニル−β−D−グルコピラノシル(1→2)
−β−D−グルコピラノシル〕−20−O−β−D
−グルコピラノシル20(S)−プロトパナキサジオ
ール(以下マロニル−ギンゼノサイドRdと称す
る)である。
この発明の新規サポニン類を含有するニンジン
としては、オタネニンジンが最も好ましいもので
ある。原料として用いられる他の植物としては、
オタネニンジンと類縁植物であるトチバニンジン
(パナツクス・ヤポニカス,シー・エー・メイヤ
ー、Panax japonicus C.A.MEYER)、アメリカ
ニンジン(パナツクス・キンキユホリウム,リン
ネ、Panax quinquefolium LINNE)、三七ニン
ジン(パナツクス・プソイド・ギンゼング,ワー
リツヒPanax pseudo−ginseng WALICHまた
はパナツクス・ノトギンゼング・バーキル、
Panax notoginseng BURKILL)が挙げられる。
この発明の新規サポニン物質は、上記のごとき
ニンジンの根部、地上部もしくはその乾燥物から
抽出分離、精製するか又は上記のごときニンジン
の根部切片を組織培養し、次いで抽出分離、精製
することによつて得ることができる。また具体的
な原料としてはオタネニンジンの生薬の白参、紅
参などが挙げられるがこのうち特に白参が好まし
い。
この発明の新規サポニン物質は上記のような原
料を用いて次のようにして得ることができる。す
なわち原料となるニンジンを脱脂せずにあるいは
通常の脂溶性有機溶媒を用いて脱脂後、水、メタ
ノールやエタノールやプロピルアルコールのよう
な低級アルコール、又はこれら低級アルコールを
含有する含水アルコールで抽出される。この抽出
は加温または加熱下に行うのが好ましい。得られ
た抽出液を蒸発濃縮して抽出エキスとする。
次いでこの抽出エキスを水と脂肪族エーテル及
び/又は脂肪族中級アルコールによつて分配抽出
される。脂肪族エーテルとしてはエチルエーテル
など用いられ脂肪族中級アルコールとしては、n
−ブタノール、n−ペンチルアルコールなどが用
いられる。
上記分配抽出で得られた水層を下記のようにシ
リカゲルカラムクロマトグラフイに付し分離精製
してこの発明のサポニンが得られる。すなわち上
記水層を、例えばボンダパツクC18のようなシリ
カゲルを用いメタノール/水混合溶媒で溶離する
逆相シリカゲルカラムクロマトグラフイに付され
る。この処理は複数回行つてもよい。そして薄層
クロマトグラフイ(TLC)を指標として1〜3
に分画する。そして分画2を、例えばシリカゲル
60(メルク社製,70〜230メツシユ)を用いクロロ
ホルム−メタノール−水(65:35:10)の下層で
溶離する順相のシリカゲルカラムクロマトグラフ
イに付してマロニル−ギンゼノサイドRb1及びマ
ロニル−ギンゼノサイドRb2、並びにマロニル−
ギンゼノサイドRb2、Rcの混合物を得る。次いで
マロニル−ギンゼノサイドRb2、Rcの混合物はさ
らに例えばシリカゲル60(メイク社製70〜230メツ
シユ)を用い、n−ブタノール−酢酸エチル−メ
タノール−水(4:2:1:1)で溶離する順相
カラムクロマトグラフイに付してマロニルギンゼ
ノサイドRb2と同Rcが分離される。
なお分画1からは同様の順相シリカゲルカラム
クロマトグラフイ(クロロホルム−メタノール−
水及びn−ブタノール−酢酸エチル−メタノール
−水で溶離)に付して公知のギンゼノサイドRo、
Re、Rf及びRg1が得られる。
また分画3からは同様にして公知のギンゼノサ
イドRb1、Rb2、Rc、Rdが得られる。
このようにして得られる新規のサポニン物質
()は細胞賦活作用を有する。そしてこれらの
物質は医薬そして用いる場合には、これらの混合
物を用いてもよく、又は個々のサポニン物質を有
効成分そして使用することができる。
次にこの発明を実施例によつて説明する。
人参からマロニル−ギンセノサイドRb1,Rb2,
Rc,Rdの抽出単離
実施例 1
人参(長野県産白参,125Kg,粉末)を80%含
水メタノール(3)と25℃で5時間、撹拌す
る。過してメタノール抽出液を得、残渣に新た
に80%含水メタノール(3)を加え、同様の抽
出操作を計5回行う。得られたメタノール抽出液
を合し、減圧下溶媒留去後、エーテルと水に分配
抽出する。エーテル移行部および水移行部をそれ
ぞれ減圧下溶媒留去して、エーテル移行部エキス
(6.7g),水移行部エキス(282g)を得る。
水移行部エキス(282g)を逆相シリカゲルカ
ラムクロマトグラフイー〔担体:ボンダパツク
C18(ウオーターズ社製)100g;溶出溶媒:メタ
ノール−水(1:3〜3:1)〕で分離し、人参
オリゴ配糖体混合物(77.6g)を得た。
人参オリゴ配糖体混合物(77.6g)を逆相シリ
カゲルカラムクロマトグラフイー〔ボンダパツク
C18,250g;メタノール−水(1:2〜2:1)〕
で分画し、分画1(31.3g),分画2(23.5g)お
よび分画3(18.1g)を得た。
分画2(23.5g)をシリカゲルカラムクロマト
グラフイー〔担体:シリカゲル60(メルク社製,
70〜230メツシユ)1.2Kg;溶出溶媒:クロロホル
ム−メタノール−水(65:35:10,下層)〕で分
離し、マロニル−ギンセノサイドRb1(10.23g),
マロニル−ギンセノサイドRd(1.45g)およびマ
ロニル−ギンセノサイドRb2,Rc混合物(9.5g)
を得た。マロニル−ギンセノサイドRb2、Rc混合
物(9.5g)をシリカゲルカラムクロマトグラフ
イー〔シリカゲル60(70〜230メツシユ)500g;
n−ブタノール−酢酸エチル−メタノール−水
(4:2:1:1)〕で分離し、マロニル−ギンセ
ノサイドRb2(5.10g)およびマロニル−ギンセノ
サイドRc(3.81g)を得た。
なお、分画1(31.1g)からは、同様のシリカ
ゲルカラムクロマトグラフイー(クロロホルム−
メタノール−水およびn−ブタノール−酢酸エチ
ル−メタノール−水)を行い、ギンセノサイド
Ro(3.29g),Re(11.56g),Rf(2.74g)および
Rg1(10.85g)を得た。
分画3(18.1g)からは、同様にしてギンセノ
サイドRb1(7.65g)、Rb2(3.80g),Rc(2.67g),
Rd(1.16g)を得た。
実施例 2
長野県産白参(10Kg,粉末)をメタノール(70
)で3時間加熱還流する。過してメタノール
抽出液を得、残渣に新たにメタノール(70)を
加え、同様の抽出操作を計3回行う。メタノール
抽出液を合し、減圧下溶媒留去し、メタノール抽
出エキス(1.45Kg)を得る。メタノール抽出エキ
ス(1.45Kg)を水(4)に懸濁し、エーテル抽
出ついでn−ブタノール抽出する。エーテル抽出
液,n−ブタノール抽出液および水移行部につい
て、それぞれ減圧下溶媒留去しエーテル抽出エキ
ス(210g),n−ブタノール抽出エキス(430
g),水移行部エキス(810g)を得た。
水移行部エキス(103g)を逆相シリカゲルカ
ラムクロマトグラフイー〔ボンダパツクC18200
g,メタノール−水(1:3〜3:1)〕ついで
シリカゲルカラムクロマトグラフイー〔シリカゲ
ル60(70〜230メツシユ)500g;クロロホルム−
メタノール−水(65:35:10,下層)〕で分離し、
ギンセノサイドRo(2.68g),マロニル−ギンセ
ノサイドRb1(7.82g),マロニル−ギンセノサイ
ドRd(0.88g)およびマロニル−ギンセノサイド
Rb2、Rc混合物(7.1g)を得た。
マロニル−ギンセノサイドRb2,Rc混合物
(7.1g)をシリカゲルカラムクロマトグラフイー
〔シリカゲル60(70〜230メツシユ)500g,n−ブ

および中国産生干し人参、および島根県産オタネ
ニンジン(Panax ginseng C.A.MEYER)の新
鮮根中にも同様に存在することが認められた。
なお、マロニル−ギンセノサイドRb1、Rb2、
Rc及びRdの特性は次のとおりである。

また、1aを40%酢酸水溶液と75℃、3時間加
熱すると、マロニル−ギンセノサイドRg3メチル
エステル〔5a,20(R,S)混合物〕が得られ
た。5aをアルカリ加水分解すると、ギンセノサ
イドRg3〔20(R,S)混合物〕およびマロン酸が
得られる。
以上の検討の結果および1a,4a,5aの炭素13
核磁気共鳴スペクトル等の物理データの考察から
1はギンセノサイドRb1の3位に結合したオリゴ
糖〔β−D−グルコピラノシル(1→2)−β−
D−グルコピラノシル〕における一級水酸基のい
ずれかに、マロン酸の結合した構造を有すること
が判明した。
5aにおける1個の一級水酸基をp−アニシル
クロロジフエニルメタン−ピリジンで処理し5b
に導く。5bをアルカリ加水分解して脱マロニル
化後、ヨウ化メチル−ジメチルスルホキサイド−
水素化ナトリウムでメチル化して5cを得る。5cを
9%塩化水素−乾燥メタノールでメタノリシスす
ると、メチル化糖として、メチル2,3,4,6
−テトラ−O−メチルグルコピラノサイドおよび
メチル−3,4−ジ−O−メチルグルコピラノサ
イドが検出同定されたことから、5a,5b,5cに
おけるマロニル基は6″位であることが判明した。
以上、得られた知見を総合することによつて、
マロニル−ギンセノサイドRb1の化学構造は1式
で現わされる。

の化学構造の検討
マロニル−ギンセノサイドRb2(2)をアルカ
リ加水分解すると、ギンセノサイドRb2およびマ
ロン酸が得られる。2をジアゾメタンでメチル化
すると、マロニル−ギンセノサイドRb2メチルエ
ステル(2a)〔mp 179〜182℃,(エタノールから
結晶化,無色微細結晶),〔α〕23 D+11.2゜(c=
1.59,メタノール),C57H94O25・2H2O,赤外線

1739 1074〕が得られた。また2aを酢酸部分加水

ル等の物理データの考察から、マロニル−ギンセ
ノサイドRb2は2式で現わされることが判明し

マロニル−ギンセノサイドRc(3)をアルカリ
加水分解すると、ギンセノサイドRcおよびマロ
ン酸が得られる。3をジアゾメタンでメチル化す
ると、マロニル−ギンセノサイドRcメチルエス
テル(3a)〔mp 15〜18℃,(エタノールから結晶
化,無水微細結晶),〔α〕23 D+1.6゜(c=1.90,メ
タノール),C57H94O25・2H2O,赤外線吸収スペ

が得られた。3aを酢酸部分加水分解すると、マ
