Abstract
Description
本発明は生物学的試料、例えば体液や***物中の過酸化
活性物質を検出するための試薬組成物及びそれを用いた
試験片に関するものである。
更に詳しくは、本発明は試料中に存在するアスコルビン
酸等の還元物質による妨害を回避するためのヨードキシ
ベンゼン誘導体を含有するレドックス反応に基づく過酸
化活性物質測定用組成物及びそれを用いた試験片に関す

尿、血液、糞便等の体液や***物中の過酸化活性物質、
特に血液(潜血)の存在を検出することは種々の疾病を
早期に発見するという観点から臨床医学上非常に重要で
ある。
今日、臨床化学の分野では、これら過酸化活性物質のレ
ドックス反応を利用した測定法が重要な位置を占めてお
り、多(の迅速測定法が知られている。
近年では試薬組成物を吸収性担体に担持させる試験片等
の、いわゆるドライケミストリーの分野が簡易・迅速で
あることより汎用されている。
代表的な例として、試料中の潜在性血液、いわゆる増血
の存在を知るために、試料中に含まれるヘモグロビンの
ペルオキシダーゼ様触媒作用を利用してのレドックス反
応に基づき、色原体を酸化呈色せしめる測定法、あるい
は試料中のグルコースのグルコースオキシダーゼ/ペル
オキシダーゼ/レドックス系反応に基づく測定法等が挙
げられる。
これら臨床化学の分野において、しばしば試料となる尿
、血液等はアスコルビン酸あるいは他の還元性物質を含
有しており、特にアスコルビン酸の存在による偽陰性の

【発明が解決しようとする課題]
アスコルビン酸等の還元性物質による悪影響を排除しよ
うとする多くの試みが報告されている。
例えば、***国特許第2,625,834号には、反応
系にアスコルビン酸オキシダーゼを添加することによっ
てアスコルビン酸を酸化して、影響を回避する手段が記
載されている。
しかしながら、アスコルビン酸オキシダーゼによるアス
コルビン酸の消費には時間がかかり、時間を短縮するに
は多量のアスコルビン酸オキシダーゼが必要となり、不
経済的であるという欠点を有していた。
又、公開特許公報昭56−63262号には、水銀イオ
ンと配位子との錯体を添加した組成物の使用が開示され
ているが、多量のアスコルビン酸を消費できないことや
、水銀イオンが有害物質である等の欠点を有する。
更に、公開特許公報昭56−151358号には、沃素
酸塩を添加した診断剤の使用が開示されている。
しかしながらヨウ素酸塩は、カチオンの種類に拘らず一
定の酸化ポテンシャルを有しており、従って過酸化活性
物質を検出する際に用いられる色原体が、試験前に酸化
されてしまわないよう特別な配慮が必要となる。その最
良の解決策としてはヨウ素酸塩により酸化されにくい色
原体を厳密に選ぶことが挙げられるが、このことは試薬
組成物が著しく制限されることになり、結果的に検出系
の感度が大きく左右されることにつながる。
又、公開特許公報昭57−13357号には、Co (
m )錯体を含有する組成物が、公開特許公報昭59−
193354号には、Fe3“−ポリカルボキシアルキ
ルアミン誘導体の金属キレートを含む組成物が開示され
ている。しかしながら、両者とも多量のアスコルビン酸
を消費できないという欠点を有していた。
これに対し本発明の試薬組成物においては、ヨ−トキシ
ベンゼン環上の置換基を変化させることにより、望まし
い酸化力を有する化合物を得ることが可能であり、これ
に伴って利用される色原体も自由に選択できる利点を有
する。
従って本発明の過酸化活性物質測定用組成物は多量のア
スコルビン酸の存在下においても、短時間で迅速なアス
コルビン酸の消去を可能にし、しかも経済的に優れてお
り、更に本発明の組成物を使用した試験片は、簡便で安
定した品質並びに長期保存性に優れた作用を奏するもの
である。
これらのヨードキシベンゼン誘導体の内、1部の化合物
は文献に記載されている。
例えば、2−ヨードキシベンゾエイトを用いた薬剤中の
イソニアシトの測定方法[K、に、ベルマ(Verma
lら、アナリティカル ケミストリーfAnalyti
calc:hemistry) 、 54巻、 250
0〜2552頁(1982年)参照〕が挙げられる。こ
れには硫酸酸性中、ヨウ化カリウムとデンプンの共存下
でアスコルビン酸を2−ヨードキシベンゾエイトにより
酸化させ、ヨウ素・デンプン反応により青呈色させるこ
とによってアスコルビン酸の定量を行うことが記載され
ている。
しかしながら、この反応は特定の条件下に限定されたも
のであり、本発明の測定対象であるレドックス反応に基
づく過酸化活性物質の測定を目的としたものではなく、
まして種々の置換基を有する広範囲の化合物が、本発明
の測定対象に応用可能であることは開示されていない。
【課題を解決するための手段】
本発明の組成物及びそれを用いた試験片は、その組成中
に少なくとも緩衝物質および色原体を含んで成るレドッ
クス反応に基づく過酸化活性物質測定用組成物において
、更に、一般式:(式中、R,、R2、R3、R4およ
びR5は同一、または異なり、各々水素原子、ハロゲン
原子、カルボキシ基、スルホン基、ニトロ基、シアノ基
、ヒドロキシ基、ヨードキシ基、ポロン基、アルソン基
、ホスホン基および置換されているか、または未置換の
アルキル基、アミン基、アルコキシ基、アリール基、ア
リールスルホニル基、アリールオキシ基、アリールアゾ
基よりなる群から選択された置換基を表し、且つ隣接し
ている2個の置換基は縮合環を形成していてもよく、そ

更に、本発明の試薬組成物を酵素、緩衝物質および色原
体と共に吸収性担体に含有させ、該担体をプラスチック
等の支持体に貼付せしめ、試験片とした場合、特に簡便
で貯蔵安定性に優れた試験具を得ることができる。もっ
とも多用される吸収性担体は濾紙であるが、不織布、綿
、木片等も使用可能である。又、試薬組成物を所望によ
り適当な接着剤、例^ばゼラチン、合成樹脂等とともに

クロマトグラフィー用の濾紙(ワットマン社製:3MM
)に上記第1含浸溶液を含浸させてがら50℃で60分
間乾燥し、次いて同じく上記第2含浸溶液で含浸し、5
0℃で20分間乾燥した後プラスチック板に両面テープ
で貼着し、これを短冊上に裁断して、幅5mm、長さ8
0mmのプラスチック片からなる基体の片隅に、5X5
mmの濾紙が貼着されている本発明の1実施例品たるヘ
モグロビン測定用試験片(以下、A試験片とする)を得
た。
実施例2゜
実施例1.で調整した第1含浸溶液中の3−ヨードキシ
ベンゼンカルボン酸の代わりに3.5−ヨードキシベン
ゼンジカルボン酸を用い、他は同一処方にて調整した。
この第1含浸溶液を用る他は実施例1.と同様にして本
発明の1実施例品たるヘモグロビン測定用試験片(以下
、B試験片とする)を得た。
実施例3゜
実施例1.で調整した第1含浸溶液中の3−ヨードキシ
ベンゼンカルボン酸の代わりに5−ブロモ−3−ヨード
キシベンゼンカルボン酸を用い、他は同一処方にて調整
した。この第1含浸溶液を用る他は実施例1.と同様に
して本発明の1実施例品たるヘモグロビン測定用試験片
(以下、C試験片とする)を得た。
実施例4゜
実施例1.で調整した第1含浸溶液中の3−ヨードキシ
ベンゼンカルボン酸の代わりに3−ヨードキシベンゼン
スルホン酸を用い、他は同一処方にて調整した。この第
1含浸溶液を用る他は実施例1.と同様にして本発明の
1実施例品たるヘモグロビン測定用試験片(以下、D試
験片とする)を得た。
実施例5゜
実施例1.で調整した第1含浸溶液中の3−ヨードキシ
ベンゼンカルボン酸の代わりに2−二トロー4−ヨード
キシベンゼンカルボン酸を用い、他は同一処方にて調整
した。この第1含浸溶液を用いる他は実施例1.と同様
にして本発明の1実施例品たるヘモグロビン測定用試験
片(以下、E試験片とする)を得た。
実施例6゜
実施例1.で調整した第1含浸溶液中の3−ヨードキシ
ベンゼンカルボン酸の代わりに2−ヨードキシベンゼン
アルソン酸ナトリウムを用い、他は同一処方にて調整し
た。この第1含浸溶液を用いる他は実施例1.と同様に
して本発明の1実施例品たるヘモグロビン測定用試験片
(以下、F試験片とする)を得た。
実施例7゜
実施例1.で調整した第1含浸溶液中の3−ヨードキシ
ベンゼンカルボン酸の代わりに4−ヨードキシベンゼン
ボロン酸ナトリウムを用い、他は同一処方にて調整した
。この第1含浸溶液を用いる他は実施例1.と同様にし
て本発明の1実施例品たるヘモグロビン測定用試験片(
以下、C試験片とする)を得た。
実施例8゜
実施例1.で調整した第1含浸溶液中の3−ヨードキシ
ベンゼンカルボン酸の代わりに2−ヨードキシ−5−[
2−(2−メトキシエトキシ)エトキシ]ベンゼンカル
ボン酸を用い、他は同一処方にて調整した。この第1含
浸溶液を用いる他は実施例1.と同様にして本発明の1
実施例品たるヘモグロビン測定用試験片(以下、H試験
片とする)を得た。
〈性能比較試験〉
実施例1.から実施例8.で作成した試験片A〜Hの性
能をヨードキシベンゼン誘導体無添加の他は実施例1.
と同様にして作成した従来の試験片(対照)と比較した
