Description
本発明は、ベイロネーラ属に属する菌を培養
し、この培養液から新規な制癌性物質TV−110
を製造する方法に関する。
近年、各種の癌患者の治療法において、宿主の
免疫機能を亢進させ、免疫機能の助けを借りなが
ら制癌効果を発現させる治療法が盛んとなつて来
た。かかる療法に使用される薬剤としては、各種
細菌の菌体、菌体培養物から得られる成分、ある
いは担子菌の子実体又はその培養菌体から得られ
る多糖体が知られている。しかし嫌気性菌を培養
し、その培養液から優れた制癌作用を有する成分
の研究は少なく、特にベイロネーラ属に属する菌
を培養し、その培養液から得られる成分並びにそ
の制癌作用については未だ知られていない。
本発明者はヒト口腔内より分離したベイロネー
ラ属に属する菌を培養し、その培養液から菌体を
除去した上清液について、その薬理作用を調べて
いたところ、この上清液から採取される特定の成
分が強い制癌作用を有すること、しかもこれは、
コロニー形成抑制法においては癌細胞の集落形成
阻止作用は極めて小さく、殺細胞による制癌作用
ではなく、宿主介在性あるいは宿主の免疫力を亢
進させ、免疫力の助けを借りながら間接的に制癌
作用を発現させる作用を有するものであること、
更にこの成分は毒性が極めて弱いことを見出し、
本発明を完成した。
本発明で利用される菌としては、ベイロネーラ
属に属するTV−110生産菌であればよく、好適
なものとしては、ベイロネーラ・アルカレツセン
ス、ベイロネーラ・パルブラが挙げられる。具体
的には、例えばベイロネーラ・アルカレツセンス
TF−V、ベイロネーラ・パルブラTF−V−1お
よび微生物学の一般常識としてそれらの性質を有
する菌株、すなわち自然変異株あるいは人工的に
改良された菌株等が利用される。
次にベイロネーラ・アルカレツセンスTF−V
及びベイロネーラ・パルブラTF−V−1の菌学
的性状を記載すれば以下のとおりである。

以上の諸性状をバージーズ・マニユアル・オ
ブ・デイタミネイテイブ・バクテリオロジー第8
版に照して検討すると、これらの菌株はベイロネ
ーラ・アルカレツセンス(Veillonella
alcalescens)及びベイロネーラ・パルブラ
(Veillonella parvula)
に酷似するので、それぞれ、これに属する菌であ
ると同定し、これらの菌株をベイロネーラ・アル
カレツセンスTF−V(Veillonella alcalescens
TF−V)及びベイロネーラ・パルブラTF−V−
1(Veillonella parvula TF−V−1)と命名し
て、工業技術院微生物工業技術研究所にそれぞれ
受託番号微工研菌寄第5876号(FERM−PNo.
5876)及び第5877号(FERM−PNo.5877)とし
て寄託した。
本発明の制癌性物質TV−110は、例えば次の
ようにして製造される。
(a) 培養
ベイロネーラ・アルカレツセンス及びベイロ
ネーラ・パルブラの培養は、通常の嫌気性菌の
培養法によつて行われる。すなわち、各種ペプ
トン、牛の脳、心臓抽出物等の窒素源;グルコ
ース、ラクトース等の炭素源;イースト・エク
ストラクト等のビタミン源;L−シスチン、亜
硫酸ソーダ、チオグリコレート・ナトリウム等
の還元剤;塩化ナトリウム等の無機塩を含む培
地を水酸化ナトリウムでPH7付近、特に好まし
くはPH7に調整したものを用いて嫌気的条件
下、好ましくは35〜40℃の温度で1〜7日間静
置あるいは撹拌培養を行う。特に次の成分を含
む培地(以下TF培地と称する)を使用するの
が好ましい。しかし窒素源として牛の脳、心臓
抽出物のブレイン・ハート・インヒユージヨン
は必ずしも必要でなく、牛の心臓抽出物である
ハート・インヒユージヨン、牛肉エキス、魚肉
エキス、トウモロコシより抽出されたコーンス
テイプリカ等を代用してもよく、また、各種ペ
プトンにおいてプロテオース・ペプトン、フア
イトン・ペプトンは必ずしも必要ではなく、ま
たトリプトケース・ペプトンをポリペプトンで
代用することもできる。なお、寒天を使用しな

セトン等のケトン類が挙げられるが、アルコー
ル類特にエタノールが最もよい結果を与える。
この親水性有機溶媒はその濃度が30〜80%(容
量比)、好ましくは60%(容量比)前後になる
ように添加するのが好適である。親水性有機溶
媒を加えた後、低温、好ましくは約4〜5℃の
温度で数時間〜数日間放置し、沈殿物の生成を
完結させる。
このようにして生じた沈殿物をデカンテーシヨ
ン、遠心分離、過等の通常の操作で分離する。
次いで、上で得られた沈殿物に10〜40倍量の水
を加え、生ずる水不溶物を遠心分離、過等の通
常の方法で除去する。培養液を使用したときはこ
の処理により菌体が除去される。
上で得られた分離液を透析又は限外過し、そ
の内液を採取し、これを乾燥すれば制癌性物質
TV−110が得られる。
斯くして得られるTV−110は次のような性状
を有する。
(イ) 白灰色〜淡褐色の粉末。
(ロ) マウスのエールリツヒ復水型癌、エールリツ
ヒ結節型癌、ザルコーマ180癌細胞の増殖を阻
止し、免疫賦活作用を有する。
(ハ) メタノール、エタノール、アセトン、ベンゼ
ン、クロロホルム、酢酸エチル、ジエチルエー
テルに不溶。
(ニ) 明確な融点を示さず、約110℃より分解を始
め、200℃以上で著しく分解する。
(ホ) KBr錠剤法による赤外線吸収スペクトルは、
3500〜3300、2920〜2880、1660〜1630、1550〜
1520、1460〜1370、1240〜1220、1120〜1020お
よび980cm-1の近傍に吸収帯を有する。
(ヘ) PH7の水溶液の紫外線吸収スペクトルは、吸

(1) 免疫賦活作用
一群3匹のICR系マウスを用い、制癌性物質
TV−110を生理食塩水に溶解させ、その溶液
0.2mlを腹腔内投与した。投与24時間後に

本発明方法で得られた制癌性物質TV−110の
マウスにおける急性毒性(LD50)は腹腔で100
mg/Kg以上である。
以上の薬理実験の結果から明らかなように本発
明方法で得られた制癌性物質TV−110は制癌剤
として有用なものであり、各種の癌疾患に使用さ
れ効果が期待されるものである。
本発明方法で得られたTV−110は常法により
経口、注射、坐薬等の剤形にして使用することが
できる。経口剤としては種々の賦形剤を含んでも
よく、カプセル剤、錠剤、散剤、顆粒剤とするこ
とができる。また、注射剤としては皮下、筋肉
内、静脈注射剤のいずれでもよく、懸濁液、溶液
もしくは使用時溶解させる粉末等の剤形が用いら
れる。また注射剤には局所麻酔剤を含んでいても
よい。
本発明方法で得られたTV−110の投与量は患
者の症状に応じて適宜選択されるが、一般に成人
では0.01〜50mg/Kgを1日1〜数回に分け投与す
るのが好ましく、投与方法としては経口又は皮
下、筋肉内、静脈内もしくは患部への注射による
のが好ましい。
次に本発明の実施例及び製剤例を挙げて説明す
る。
実施例 1
(1) 2容のスクリユーキヤツプ付培養瓶1本に
トリプトケース・ペプトン34g、フアイトン・
ペプトン6g、プロテオース・ペプトン20g、
ブレイン・ハート・インヒユージヨン70g、イ
ースト・エクストラクト6g、食塩15g、グル
コース12g、ラクトース10g、L−シスチン
0.5g、亜硫酸ソーダ0.2g、チオグリコレー
ト・ナトリウム1.0g、寒天1.4gを加え、PH7
に調整した培地2を入れ、120℃で15分間加
圧滅菌したのち、ただちに水冷冷却し、予め同
組成の培地で前培養しておいたベイロネーラ・
アルカレツセンスTF−V〔受託番号微工研菌寄
第5876号(FERM−PNo.5876)〕の前培養液を
培養瓶1本につき100mlの割合で滅菌条件下に
接種し、37℃のふ卵器中で48時間静置培養を行
う。培養終了後、4℃で4000rpm、20分間この
培養液を遠心分離し、菌体を除去して上清液約
1.82を得た。
(2) (1)で得た上清液に5℃で撹拌下にエタノール
2.73を加え、無定形の沈殿物が完全に沈殿す
るまで低温室で放置する。ついで5℃で、
6000rpm、15分間遠心分離し沈殿物を採取し、
エタノールで洗浄し、減圧乾燥して約2.0gの
粗粉末を得た。
(3) (2)で得た粗粉末2.0gを水60mlに溶解させ、
この際生じる水不溶物を7500rpm、10分間遠心
分離で除去して得た上清液と、水不溶物を水10
mlで2回洗浄した洗浄液とを合した溶液75mlを
蒸留水に対して透析し、内液を凍結乾燥し、白
灰色〜淡褐色のTV−110の粉末1.50gを得た。
このものは次の理化学的性質を有する。
(イ) メタノール、エタノール、アセトン、ベン
ゼン、クロロホルム、酢酸エチル、ジエチル
エーテルに不溶。
(ロ) 明確な融点を示さず、約110℃より分解を
始め、200℃以上で著しく分解する。
(ハ) KBr錠剤法による赤外線吸収スペクトル
は、3500〜3300、2920〜2880、1660〜1630、
1550〜1520、1460〜1370、1240〜1220、1120
〜1020および980cm-1の近傍に吸収帯を有す
る(第3図)。
(ニ) PH7の水溶液の紫外線吸収スペクトルは、
吸収末端に強い吸収があり、また247〜268n
mの近傍に吸収を示す(第4図)。
(ホ) モーリツシユ反応、フエノール硫酸反応、
アンスロン硫酸反応、インドール塩酸反応、
ロウリー・フオリン反応は陽性。
(ヘ) 元素分析値
C:37%〜40%、H:4%〜6%、N:5
%〜7%
(ト) フエノール硫酸法による糖の含有率は約45
%〜65%(グルコース換算)、およびロウリ
ー・フオリン法による蛋白質の含有率は約15
%〜25%(牛血清アルブミン換算)である。
実施例 2
(1) 2容のスクリユーキヤツプ付培養瓶1本に
トリプトケース・ペプトン34g、ブレイン・ハ
ート・インヒユージヨン70g、イースト・エク
ストラクト6g、食塩15g、グルコース12g、
ラクトース10g、L−シスチン0.5g、亜硫酸
ソーダ0.2g、チオグリコレート・ナトリウム
1.0g、寒天1.4gを加え、PH7に調整した培地
2を入れ、120℃で15分間加圧滅菌したのち、
ただちに水冷冷却し、予め同組成の培地で前培
養しておいたベイロネーラ・パルブラTF−V
−1〔受託番号微工研菌寄第5877号(FERM−
PNo.5877)〕の前培養液を培養瓶1本につき100
mlの割合で滅菌条件下に接種し、37℃のふ卵器
中で120時間静置培養を行う。培養終了後、4
℃で4000rpm、20分間この培養液を遠心分離
し、菌体を除去して上清液約1.83を得た。
(2) (1)で得た上清液に5℃で撹拌下にエタノール
2.75を加え、無定形の沈殿物が完全に沈殿す
るまで低温で放置する。ついで4℃で、
6000rpm、15分間遠心分離し沈殿物を採取し、
エタノールで洗浄し、減圧乾燥して約1.6gの
粗粉末を得た。
(3) (2)で得た粗粉末1.6gを水50mlに溶解させ、
この際生じる水不溶物を7500rpm、10分間遠心
分離で除去して得た上清液と、水不溶物を水10
mlで2回洗浄した洗浄液とを合した溶液65mlを
蒸留水に対して透析し、内液を凍結乾燥し白灰
色〜淡褐色のTV−110の粉末1.0gを得た。
このものは次の理化学的性質を有する。

イン0.5%含有溶液等に溶解させ注射液として用
いる。
