Description
-一過性菌叢:一過性菌叢は、環境から腐敗した有機物を餌とする腐生菌と称される、大抵は無害な真菌、ウイルスおよび細菌からなる。この菌叢は恒久的なものではなく、行われる活動や周辺条件の変化、個体がこれらの条件に露出されることに応じて日中に変化する。最も一般的な一過性菌叢の種としては、スタフィルオコックス・アウロイス(Staphylococcus aureus)、エスケリキア・コリ(Escherichia coli)、シュードモナス・アエルギノーサ(Pseudomonas aeruginosa)およびバチルス(Bacillus)の種がある。
-常在菌叢:常在菌叢は片利共生微生物からなり、すなわちそれらの宿主に何の損傷も与えずに生きている。この菌叢の構成は固定されており、妨害された後、同じ構成に自発的に再形成される。これらの微生物(グラム陽性細菌、グラム陰性細菌など)は表皮に生息しており、主に角質層の上層ならびに汗腺や毛嚢脂腺の毛包(pilosebaceous follicle)の管、爪や毛髪の毛のような表皮付属器で見つかられる。
-細胞間の凝集結合によって表皮の強さとなる構成タンパク質であるデスモグレイン-1の形成を減少させることにより(Straseski J et al. Wound Repair Regen. 2009; 17(4): 606-616)
-また水分を表皮に輸送し、皮膚に水分補給するタンパク質であるアクアポリン3の形成を減少させることにより(Straseski J et al. Wound Repair Regen. 2009; 17(4): 606-616)
-皮膚の構成成分や細胞を変化させる活性酸素種(ROS)の産生を刺激することにより(Nys K et al. Free Radic Biol Med. 2012;52(6):1111-20)
-ケラチン10などの分化のためのケラチンの形成を抑制することにより(Straseski J et al. Wound Repair Regen. 2009; 17(4): 606-616)、および/または
-マトリックスメタロプロテアーゼ(MMP)の存在を増加させることにより(Xia YP et al. J. Invest. Dermatol. 2001; 116: 50-56)、ラーゲンなどの皮膚の構成成分を悪化させ、皮膚密集度や弾力に影響を与え、その結果として、毛穴の開きおよび細かい線やしわの形成をもたらし得る。
-皮膚バリアの強化や皮脂産生の制御などが可能な皮膚状態を提供することにより、微生物叢およびその多様性を維持すること、
-表皮の凝集を増強させ、皮膚の抵抗力を強化すること、
-皮膚バリアの形成を刺激し、皮膚に十分な水分を維持すること、
-皮膚の活性酸素種を無毒化すること、
-MMPの形成を抑制すること、および/または
-皮膚細胞を低酸素症から保護する低酸素症-誘導できる因子(HIF)の形成を刺激すること。
a)モル質量が5~630kDa、好ましくは5~120kDa、より好ましくは8~80kDaのガラクトマンナンおよびモル質量が7~3000kDa、好ましくは7~1100kDa、より好ましくは8~200kDaの架橋硫酸化ガラクタンからなる化粧剤、ならびに
b)プレバイオティクスおよびプロバイオティクス。
本発明によれば、「ケラチン物質」とは皮膚、粘膜(特に唇)および外皮を意味する。特定の実施態様によれば、組成物は皮膚、唇または外皮、好ましくは皮膚への局所適用を目的とする。
したがって、本発明は、生理学的に許容できる媒体中に少なくとも以下の物質を含む化粧品組成物に関する:
a)モル質量が5~630kDa、好ましくは5~120kDa、とり好ましくは8~80kDaのガラクトマンナンおよびモル質量が7~3000kDa、好ましくは7~1100kDa、より好ましくは8~200kDaの架橋硫酸化ガラクタンからなる化粧剤、ならびに
b)プレバイオティクスおよびプロバイオティクス。
本発明の意味での化粧剤は、バイオポリマー、すなわち化学合成によって得られる植物原料物質由来のポリマーからなる。
-モル質量が5~630kDaのガラクトマンナン、および
-モル質量が7~3000kDaの架橋硫酸化ガラクタン。
-平均モル質量が5~120kDaのガラクトマンナン、および平均モル質量が7~1100kDaの架橋硫酸化ガラクタン。
-平均モル質量が8~80kDaのガラクトマンナン、および平均モル質量が8~200kDaの架橋硫酸化ガラクタン。
-天然ガラクトマンナン粉末または天然硫酸化ガラクタンそれぞれの水への溶解化
-化学的加水分解または酵素加水分解;
-不溶性相を除去するための、可溶性相または不溶性相の分離
-規定のモル質量のガラクトマンナンの膜濾過による選択
「プレバイオティクス」とは、ヒトの健康に有益な腸のプロバイオティクス菌株(ラクトバシリ(lactobacilli)、ビフィズス菌など)およびほかの細菌の菌株の増殖や活性を調節する性質を有する非生物性食品成分を意味する。プレバイオティクスは、一般に果物や植物、蜂蜜などに自然に存在する抽出可能な単糖類、二糖類またはオリゴ糖類である。他は、多糖(例えばフルクト-オリゴ糖(FOS))の加水分解により工業的に生産するか、またはラクトースなどの二糖類に転移活性を有するラクターゼなどの酵素の作用を受けさせ、トランス-ガラクト-オリゴ糖(TOS)を生成するか、もしくは化学異性化反応によりラクツロースを生成することによって合成する。現在、プレバイオティクス効果が認められているものとしては、トランス-ガラクト-オリゴ糖(GOS)やイヌリン型フルクタン類がある。
本発明によるテオブロマカカオL.の加水分解物は、主にペプチドおよび糖を含むテオブロマカカオL豆のペプチドおよび糖加水分解物である。「主にペプチドおよび糖」とは、50%を超える量、好ましくは60%を超える量、好ましくは70%を超える量、最大90%(重量/重量)のペプチドおよび糖の乾燥物質、好ましくは90%重量の乾燥物質を意味する。
a)挽いたテオブロマカカオL豆を水相中に分散させ;
b)工程a)で得られる水分散液を酵素処理し;
c)固液分離により酵素加水分解物を回収し;
d)超濾過およびナノ濾過により加水分解物精製し、その後、場合により
e)工程d)で得られる加水分解物を凍結乾燥させる。
本発明による組成物は、より詳しくは、ケラチン物質、特に皮膚への局所適用を目的とする。
-皮膚微生物叢の生物多様性および恒常性(活性成分であるActibiome(C)(水および海水およびグリセリンおよびラミナリア・ディギタータ(Laminaria digitata)抽出物およびクロレラ・ブルガリス(Chlorella vulgaris)抽出物および異性化糖(saccharide isomerate)およびフェノキシエタノールおよびエチルヘキシルグリセリン)のように、ストレスの期間によってあらかじめ低下された皮膚のpHを再調整することにより);
-細胞再生(酸性pHで活性成分単独または組み合わせでピーリングする方法により):
・化学的に(αヒドロキシ酸(AHA)(グリコール酸、乳酸、クエン酸...)、βヒドロキシ酸(BHA)(サリチル酸)、ポリヒドロキシ酸(PHA)(グルコノラクトン)など);
・生物学的に(Exfolactive C EL PX(C)(オプンティア・コチネリフェラ(Opuntia coccinellifera))、DERMOCH DP(C)(Chlorella vulgaris)、ALGOWHITE(C)(アスコフィラム・ノドサム(Ascophyllum nodosum)など)
・酵素的に(MELACLEAR(C)(グルコン酸、スティラインズ)など);
-顔の肌の再生または再活性化(輝き)(以下の活性成分を用いて):
・平滑化(レチノール(ビタミンA様)など)
・メラニン生成に作用することによる美白化(lightening)(ビタミンC誘導体)
・エネルギー付与(energizing)(栄養素および/または抗酸化剤であるビタミンCおよびビタミンE(SEPIVITAL(C)(リン酸アスコルビルトコフェロールカリウム)など)など);
-皮脂産生の減少(皮脂キャプター(sebum captor)(クレイ)または、グルコン酸亜鉛(Mineralis GU/Zn)および親油性アボカド抽出物(5αavocuta)などの皮脂調節活性成分を用いて);
-外部からの侵入からの保護(汚染、ブルーライト、UV、化学的または鉱物フィルターを使用および/または反射粒子を拡散させることで)、
およびこれらの混合。
これらの活性成分を単独または組み合わせて用いた転写効果に関する研究は、TaqMan Low Density Array(TLDA)を用いて行った。この技術によって、正常なヒトケラチン生成細胞の処置に応じた、特定のタンパク質をコードする遺伝子の発現の変調が研究され、これは、活性成分の単独または組み合わせでの遺伝子発現に対する効果のマッピングを可能にする。
用いられた細胞:正常ヒトケラチン生成細胞
培地:HKGS(ヒトケラチン生成細胞増殖サプリメント)が補充されたEpiLife(R)
細胞の皮膚由来:腹部サンプル、47歳の女性
研究モデル:TaqMan Low Density Array (TLDA)
化合物での細胞の処理:24時間
試験化合物:
・0.25%のECOSKIN(R)(α-グルカンオリゴ糖およびヤーコン(POLYMNIA SONCHIFOLIA)根の搾汁およびマルトデキストリンおよびラクトバシラス)
・0.25%のFILMEXEL(R)(カエサルピニア・スピノサ(CAESALPINIA SPINOSA)抽出物およびカッパフィカス・アルベレッチー抽出物および水)
・0.25%のECOSKIN(R)+0.25%のFILMEXEL(R)
以下の表1は、研究した遺伝子の活性の変動を示している。
使用される実験条件は以下の通りである:
-各試験は、3回実施する(未処理の3回、同一条件で処理される3回);
-Fischer F-検定は、2つのデータ行列を比較することによって最初に適用する。値がα=0.05より大きい場合、スチューデントのt-検定の分散は2で、FischerのF-検定がα= 0.05より小さい場合、分散は3になる。
-用いられる転写変動(有意)は、スチューデントのt検定においてα(またはp)=0.05未満またはそれに等しく対応するものになる。刺激は濃い灰色(下記参照)で示し、白色の背景は、有意でない変動、すなわちスチューデントのt検定においてα(またはp)=0.05より大きく対応するものを示す。
-IVLおよびPI3遺伝子の転写活性を刺激することにより、皮膚バリアの形成、特に角質細胞(細胞性エンベロープ)の形成を増強させ、
-細胞間水分の通過を制限し(不感水分の損失)、表皮脂質を皮膚表面に維持させる細胞間密着結合の形成に関与するCLDN1およびOCLN遺伝子の転写活性を刺激し、表皮脂質輸送に関与するSLC27A4遺伝子の転写活性を刺激することにより、皮膚バリアンお性能を改善し、
-特にSOCS3、TGFBR1およびKRT5遺伝子の転写活性を刺激することにより、表皮適当な発達およびその強度を促進させ、
-毛穴への収れん性効果および皮膚起伏(relief)の不規則性の充填における皮膚の維持に関与する皮膚のタンパク質構造の安全性を維持し、SERPINB3、SERPINB7およびPI3のようなプロテアーゼインヒビター遺伝子を活性化による、これらのタンパク質構造の劣化を防止し、
-皮膚の酸素含有量が低いときに誘発されるHIF1aの転写活性を刺激し、代謝がこの新しい組織環境に適応できるようにする、低酸素(酸素不足)状態に抵抗する皮膚の能力を活性化し、
-抗菌性殺菌ペプチドであるPI3遺伝子の転写活性を刺激することにより、皮膚が微生物叢に及ぼす制御を増加させ、
-皮膚細胞の全体的な代謝を確保するために使用される細胞エネルギーを貯蔵するための分子であるATPの形成に寄与する遺伝子ATP2C1の転写活性を刺激することにより、皮膚のエネルギー恒常性を確保し、
-PI3遺伝子の転写活性を刺激することにより炎症を制限しながら、グルタチオン分子とのカップリングによりそれらを排除するGSTZ1の転写活性の刺激により毒素から皮膚を保護することにより、ストレスに対する皮膚の抵抗力を改善および/または強化する。
Claims (13)
- 生理学的に許容できる媒体中に少なくとも以下の物質を含む化粧品組成物:
a)モル質量が5~630kDaの加水分解されたガラクトマンナンを含むカエサルピニア・スピノサ抽出物およびモル質量が7~3000kDaの加水分解された架橋硫酸化ガラクタンを含むカッパフィカス・アルベレッチー抽出物からなる化粧剤、ならびに
b)α-グルコ-オリゴ糖、ヤーコン根抽出物、ラクトバシラス溶解物およびマルトデキストリンを含むプレ/プロバイオティクス複合体の形態のプレバイオティクスおよびプロバイオティクス。 - ガラクトマンナンのモル質量が5~120kDaであり、架橋硫酸化ガラクタンのモル質量が7~1100kDaであることを特徴とする、請求項1に記載の化粧品組成物。
- ガラクトマンナンのモル質量が8~80kDaであり、架橋硫酸化ガラクタンのモル質量が8~200kDaであることを特徴とする、請求項2に記載の化粧品組成物。
- 上記化粧剤a)が、組成物総重量に対して0.01重量%~2重量%の範囲の量で組成物中に存在することを特徴とする、請求項1~3のいずれか一項に記載の化粧品組成物。
- 上記化粧剤a)が、組成物総重量に対して0.4重量%~0.8重量%の範囲の量で組成物中に存在することを特徴とする、請求項4に記載の化粧品組成物。
- 上記プレバイオティクスおよびプロバイオティクスが、組成物総重量に対して0.05重量%~3重量%の範囲の活性成分の含有量で組成物中に存在することを特徴とする、請求項1~5のいずれか一項に記載の化粧品組成物。
- 上記プレバイオティクスおよびプロバイオティクスが、組成物総重量に対して0.1重量%~1重量%の範囲の活性成分の含有量で組成物中に存在することを特徴とする、請求項6に記載の化粧品組成物。
- 皮膚、唇または眼のためのケアおよび/またはメイクアップ製品の形態で存在することを特徴とする、請求項1~7のいずれか一項に記載の化粧品組成物。
- 顔の肌のためのケアおよび/またはメイクアップ製品の形態で存在することを特徴とする、請求項8に記載の化粧品組成物。
- ケラチン物質をケアおよび/またはメイクアップするための化粧方法であって、請求項1~9のいずれか一項で定義される化粧品組成物を、該ケラチン物質に少なくとも適用することを特徴とする化粧方法。
- 化粧品組成物が、顔の肌のための化粧品組成物であることを特徴とする、請求項10に記載のケラチン物質をケアおよび/またはメイクアップするための化粧方法。
- 皮膚生態系を維持すること、および/または皮膚微生物叢のバランス、微生物多様性を維持すること、皮膚バリアおよびそのハイドロリピッドフィルムを強化すること、ストレスに対する皮膚の完全性および抵抗力を改善すること、低酸素状態に対するその適応能を促進させること、および/または低酸素環境の皮膚への影響を制限すること、落屑および/または表皮再生を促進させること、弾力の損失を減少させること、毛穴弾性の損失を減少させること、表面不規則性を減少させること、顔の肌の均一性を改善すること、皮膚のエネルギー恒常性を促進させること、および/またはメイクアップ持ちおよびその付着の均質性を促進および/または改善することを目的とする、請求項10または11に記載のケアおよび/またはメイクアップするための化粧方法。
- 請求項1~3のいずれか一項に記載の化粧品組成物の非治療的使用であって、皮膚生態系を維持すること、および/または皮膚微生物叢のバランス、微生物多様性を維持すること、皮膚バリアおよびそのハイドロリピッドフィルム(hydrolipidic film)を強化すること、ストレスに対する皮膚の完全性および抵抗力を改善すること、低酸素状態に対するその適応能を促進させること、および/または低酸素環境の皮膚への影響を制限すること、落屑および/または表皮再生を促進させること、弾力の損失を減少させること、毛穴弾性の損失を減少させること、表面不規則性を減少させること、顔の肌の均一性を改善すること、皮膚のエネルギー恒常性を促進させること、および/またはメイクアップ持ちおよびその付着の均質性を促進および/または改善することを組み合わせて目的とする使用。
