Description
本出願は、その全体が参照により本明細書に組み込まれている、2021年4月29日に出願した米国特許仮出願第63/181,963号の利益を主張する。
本出願は、ASCIIフォーマットで電子提出されており、その全体が参照により本明細書に組み込まれている配列表を含有する。前記ASCIIコピーは、2022年4月28日に作成され、632WO_SL_ST25.txtと名付けられ、22,918バイトのサイズである。
本出願は、とりわけ新規トポイソメラーゼ阻害薬、薬物リンカー、抗c-Met抗体薬物複合体(ADC)及びそれらを作製する方法に関する。
c-Metは、上皮細胞及び内皮細胞の表面上に発現されるシグナル伝達チロシンキナーゼ受容体である。その唯一の公知のリガンドである肝細胞増殖因子(HGF)によるc-Metの活性化は、細胞増殖、血管新生、生存、及び細胞運動性を制御することが示されている。
本開示は、ヒトc-Metに特異的に結合する抗体薬物複合体を提供する。例示的なCDRのアミノ酸配列、並びに例示的な抗c-Met ADCの抗体の重鎖及び軽鎖のVH及びVL領域のアミノ酸配列は、以下の詳細な説明に提供される。
本発明の様々な態様は、トポイソメラーゼI(TOP1)阻害剤、及びそのようなTOP1阻害剤を含む抗c-Met抗体薬物複合体に関する。ある特定の実施形態において、本発明は、TOP1阻害剤を含む抗c-Met ADCを含む抗c-Met ADC、ADCを合成するために有用な合成等価体、ADCを作製する方法及びADCを使用する様々な方法を提供する。
トポイソメラーゼ1(TOP1)は、DNA複製の際に形成されたスーパーコイルを除去する。TOP1阻害剤(TOP1i)は、TOP1-DNA複合体に結合して安定化し、DNA鎖切断及びアポトーシスを誘導することができる。本明細書において、抗体とのコンジュゲーションによる細胞への標的化送達が目的とされ得る、構造式(I)で表されるトポイソメラーゼI阻害薬(「TOP1i薬」)が提示される。
本明細書に記載されたトポイソメラーゼ阻害剤は、抗c-Met抗体にコンジュゲートされて、抗c-Met TOP1i抗体薬物複合体(ADC)を形成し得る。抗体薬物複合体は、ADCが、1つ以上の薬物部分(複数可)を標的組織、例えば腫瘍関連抗原、例えばc-Met発現腫瘍に選択的に送達する能力のために、疾患の処置における抗体の治療有効性を増加させ得る。このように、複数の実施形態において、本開示は、例えば非小細胞肺がんの処置における治療的使用のための抗c-Met TOP1i ADCを提供する。
7.2.1.リンカー薬物LD1(構造式(V))
本明細書に開示されるADCは、抗体の立体配置、及び少なくとも部分的に、コンジュゲーションを行うために使用された方法に応じて、様々な化学量論的モル比で抗体部分に連結された薬物分子を含む。
AbAは、マウスモノクローナル抗体224G11のヒト化型であり、これは米国特許第8,329,173号明細書において初めて開示され、具現化された。マウス抗体m224G11は、配列番号13として示される可変重ドメイン(variable heavy domain)及び配列番号14として示される可変軽ドメイン(variable light domain)を有する:

複数の実施形態において、本明細書に開示される方法は、非扁平上皮NSCLCを有する患者を本発明の抗c-Met ADCによって処置することを伴う。

細胞増殖
TOP1i薬が細胞生存率に及ぼす効果は、代謝的に活性な細胞に関するATPの存在のモニタリングによって評価された。Calu-6細胞(肺腺癌)及びA375細胞(悪性黒色腫)の増殖は、CellTiter-Glo(登録商標)発光を使用して定量された。図2A及び表1に示されるように、式I(実施例1G)の化合物は、増殖中のCalu-6細胞(0.57nM、図2A)及びA375(0.50nM、図2B)の生存率の低減に関してナノモル以下の効力を示した。このナノモル以下の効力は、同じアッセイにおいて6.58nM(Calu-6)及び2.30nM(A375)のEC50を実証したTOP1i薬SN-38の成績と比較して改善した。
AbAの調製及び精製
配列番号9及び配列番号10の重鎖及び軽鎖配列をそれぞれ有するAbAを、チャイニーズハムスター卵巣(CHO)細胞において発現させた。AbAは、コンジュゲーションの前に単離及び精製された。
コンジュゲーションのための手順(ADC-1)
還元の前に、抗c-Met抗体AbAの溶液を、4℃で少なくとも12時間インキュベートした。次に、0.5Mエチレンジアミン四酢酸二ナトリウム塩溶液(EDTA、Sigma Aldrich)を、最終濃度5mMで加えた。トリス(2-カルボキシエチル)ホスフィン(TCEP、100mM、2.67モル当量、Bond Breaker(商標)、Thermo Scientific(商標))を、溶液(1×DPBS(ダルベッコリン酸緩衝食塩水)中でおよそ10mg/mL)に加え、プレートを5分間軽く攪拌して(225rpm)混合した。4℃の冷蔵庫において17時間インキュベートした後、プレートを225rpmで攪拌して混合し、溶液を22℃にした。次に、10容量/容量%のN,N-ジメチルアセトアミド(DMA、Sigma Aldrich)共溶媒を加え、続いて20容量/容量%の調製された1Mホウ酸、pH8.0(Sigma Aldrich)を加え、次に6モル当量の50mMリンカー薬物LD1(構造式V、実施例1J)のN,N-ジメチルアセトアミド中溶液を滴下して加えた。混合を20分後に中止し、溶液を22℃で3.3時間インキュベートした。インキュベーション後、1×DPBS中で調製された2モル当量の50mM N-アセチル-L-システイン(NAC、Sigma Aldrich)を溶液に加え、容器を手で攪拌することにより軽く混合した。22℃で1時間インキュベーション後、溶液を、691mL G25 Fine Desalting(Cytiva(商標))カラムを使用して、1×DPBS中で脱塩した。
還元前、抗c-Met抗体AbAの溶液を、4℃で少なくとも12時間インキュベートした。次に、0.5Mエチレンジアミン四酢酸二ナトリウム塩溶液(EDTA、Sigma Aldrich)を、最終濃度5mMで加えた。トリス(2-カルボキシエチル)ホスフィン(TCEP、100mM、1.00モル当量、Bond Breaker(商標)、Thermo Scientific(商標))を、抗体溶液(1×DPBS中でおよそ4mg/mL)に加え、プレートを軽く攪拌しながら(225rpm)5分間混合した。4℃の冷蔵庫で17時間インキュベートした後、プレートを225rpmで攪拌して混合し、溶液を22℃にした。次に、10容量/容量%のN,N-ジメチルアセトアミド(DMA、Sigma Aldrich)共溶媒を加え、続いて20容量/容量%の1Mホウ酸、pH8.0(Sigma Aldrich)を加え、次にN,N-ジメチルアセトアミド中の4モル当量の50mMリンカー薬物LD1(構造式V、実施例1J)を滴下して加えた。混合を20分後に中止し、溶液を22℃で2.1時間インキュベートした。インキュベーション後、1×DPBS中で調製された2モル当量の10mM N-アセチル-L-システイン(NAC、Sigma Aldrich)を溶液に加え、容器を手で攪拌しながら軽く混合した。22℃で5時間インキュベーション後、溶液を濃縮し、緩衝液を、限外ろ過ダイアフィルトレーション(UF/DF)を介して交換した。
Pellicon(登録商標)3 0.22m2カセット(Millipore)に、滅菌水2Lを一度に流した後、製剤緩衝液0.5Lを流した。限外ろ過(UF)は、250mL/分の流速で実施され、膜間差圧(TMP)は12~13psiであり、開始容量は、1.3Lであった。最終的な透過容量は1.1Lであった。ダイアフィルトレーション(DF)は、最終製剤緩衝液の25ダイア容量(diavolume)(DV)で、UFステップと同じ流速及びTMPで実施された。DF後、第2のUFステップを実施して、ADC溶液の容量を100mLへとさらに低減させた。ADC溶液を除去し、カセットを、製剤緩衝液125mLによって2回すすぎ、次に各20mLへと限外ろ過した。次に、これらのすすぎ液をバルクADC溶液に加えた。最終的な溶液を、特性評価の前に0.22μmフィルターによってろ過滅菌した。
コンジュゲーションのための手順(ADC-2)
還元の前に、抗c-Met抗体AbAの溶液を氷中で35分間インキュベートした。次に、0.5Mエチレンジアミン四酢酸二ナトリウム塩溶液(EDTA、Sigma Aldrich)を、最終濃度5mMで加えた。トリス(2-カルボキシエチル)ホスフィン(TCEP、25mM、3.50モル当量、Bond Breaker(商標)、Thermo Scientific(商標))を溶液に加え(1×DPBS中でおよそ10mg/mL)、チューブを数回ゆっくり上下させて軽く混合した。次に、溶液を氷中に入れ、4℃の冷蔵庫に24時間入れた。インキュベーション後、10容量/容量%のN,N-ジメチルアセトアミド共溶媒を加え、続いて10モル当量の10mMリンカー薬物LD2(構造式VI、実施例2)のN,N-ジメチルアセトアミド(DMA、Sigma Aldrich)中溶液を加え、チューブを数回ゆっくり上下させて軽く混合した。次に、溶液を氷中に30分間入れ、22℃で90分間インキュベートした。インキュベーション後、PBS中で調製した8モル当量の10mM N-アセチル-L-システイン(NAC、Sigma Aldrich)を溶液に加え、チューブを数回ゆっくり上下させて軽く混合した。22℃で60分間インキュベーション後、溶液を、HiPrep(商標)26/10脱塩カラムによって1×DPBS(Cytiva(商標))中で脱塩した。次に、脱塩されたADCを、10容量/容量%の1Mホウ酸、pH8.0(Sigma Aldrich)の調製された溶液を加えて22℃で96時間インキュベートすることによって加水分解した。加水分解されたADCを、アフィニティクロマトグラフィーと1×DPBSの最終製剤中での脱塩クロマトグラフィーとのカップリング(2×1mL HiTrap MabSelect SuRe、HiPrep(商標)26/10脱塩、Cytiva(商標))を通して精製した。収集した分画をプールし、特性評価の前に0.22μmフィルターを通してろ過滅菌した。
コンジュゲーションのための手順(ADC-3)
エチレンジアミン四酢酸二ナトリウム塩水溶液(EDTA、0.5M、21.3μL、Sigma Aldrich)を、抗c-Met抗体(AbA、1×DPBS、pH7.4中で13.34mg/mL)溶液5.337mLに加えた。トリス(2-カルボキシエチル)ホスフィン(TCEP、10mM、3.0モル当量、142.4μL、Bond Breaker(商標)、Thermo Scientific(商標))を溶液に加え、軽く攪拌し、37℃で75分間維持した。溶液を周囲温度に冷却し、リンカー-薬物LD3(構造式VII、実施例3C)(10当量、LD3(構造式VII、N,N-ジメチルアセトアミド中の実施例3C、90%純度))の10mM溶液527μL)を加えた。コンジュゲーション物を軽く攪拌し、周囲温度で1.2時間放置した。ADC溶液を脱塩によって精製した。脱塩後、ADC溶液を、0.22μmフィルターを通してろ過し、得られた試料を4℃で保存した。得られたADCを、10容量/容量%の1.0Mホウ酸緩衝液、pH8.0を加え、周囲温度の暗所で72時間インキュベートすることによって加水分解した。加水分解後、得られたADC溶液を、プロテインA樹脂カラム(MabSelect(商標)SuRe(商標)LX、GE Healthcare)に吸着させること;1×DPBS、pH7.4の4カラム容量によって洗浄すること;IgG溶出緩衝液(Thermo Scientific(商標))の5カラム容積によって樹脂から溶出させること;及び1×DPBS、pH7.4中で脱塩することによって精製した。脱塩後、ADC溶液を、0.22μmフィルターを通してろ過し、得られた試料を4℃で保存した。
コンジュゲーションのための手順(ADC-4)
エチレンジアミン四酢酸二ナトリウム塩水溶液(EDTA、0.5M、974μL、Sigma Aldrich)を、MSL109-C6v1抗体の精製溶液(20mMトリス緩衝液、pH7.4中で12.3mg/mL)243.3mLに加えた。MSL109-C6v1は、CMV糖タンパク質Hに結合するモノクローナル抗体である。MSL109-C6v1は、配列番号11として示される重鎖及び配列番号12として示される軽鎖を含み、非標的化対照抗体として使用されている。トリス(2-カルボキシエチル)ホスフィン(TCEP、500mM、6.0モル当量、242.4μL、Bond Breaker(商標)、Thermo Scientific(商標))を、2mM EDTAを含むMSL109-C6v1の溶液に加え、軽く攪拌し、4℃で20時間維持した。4℃のトリス緩衝液(1.0M、pH8.0)を溶液(24.5mL、10容量/容量%)に加えた。リンカー-薬物LD1(実施例1J、(2S)-2-(2-ブロモアセトアミド)-N-[(2S)-1-({3-[(7S)-7-エチル-7-ヒドロキシ-8,11-ジオキソ-7,8,11,13-テトラヒドロ-2H,10H-[1,3]ジオキソロ[4,5-g]ピラノ[3’,4’:6,7]インドリジノ[1,2-b]キノリン-14-イル]ビシクロ[1.1.1]ペンタン-1-イル}アミノ)-1-オキソプロパン-2-イル]-3-メチルブタンアミド)を、還元された抗体(10当量、10mM LD1、構造式V、実施例1JのN,N-ジメチルアセトアミド中溶液22.5mL、90%純度)にpH8.0で加えた。コンジュゲーション物を軽く攪拌し、周囲温度の水浴中で1.5時間放置した。コンジュゲーション物を、4当量のN-アセチル-L-システイン水溶液(NAC、Sigma Aldrich、1×DPBS中の100mM溶液、0.8mL)を加えることによってクエンチした。混合物を軽く攪拌し、周囲温度で1時間放置した。トリス緩衝液(15%、Sigma、1.0M、pH7.4、44mL)を加えて、ADC溶液をpH7.5に調整した。ADC溶液を4℃で一晩保存した。ADC溶液を濃縮し、緩衝液を、限外ろ過ダイアフィルトレーション(Pellicon(登録商標)3 0.22m2カセット(Millipore))を介して交換した。最終的なADC溶液を、0.22μmフィルターを通してろ過し、得られたADC試料を4℃で保存した。ADCの最終容量は138mLであった。
DAR(薬物抗体比)の決定
DARは、LC-MSによって決定された。LC-MS解析は、Agilent LC/MSD TOF 6220 ESI質量分析計にインターフェースで接続されたAgilent 1100 HPLCシステムを使用して実施された。ADCは、5mM(最終濃度) Bond-Breaker(商標)TCEP溶液(Thermo Scientific(商標)、Rockford、IL)によって還元され、Protein Microtrap(Michrom Bioresources、Auburn、CA)脱塩カートリッジにロードされ、10%B~75%Bの0.2分間の勾配によって周囲温度で溶出した。移動相Aは、0.1%ギ酸(FA)を含むH2Oであり、移動相Bは、0.1%FAを含むアセトニトリルであり、流速は0.2mL/分であった。同時に溶出する軽鎖及び重鎖のエレクトロスプレーイオン化飛行時間型マススペクトルを、Agilent MassHunter(商標)獲得ソフトウェアを使用して獲得した。抽出された強度をm/zスペクトルに対して、MassHunter(商標)ソフトウェアの最大エントロピー特色を使用してデコンボリューションし、各還元された抗体断片の質量を決定した。DARは、軽鎖及び重鎖に関する裸のピーク及び改変されたピークの強度を合計することによってデコンボリューションされたスペクトルから計算され、結合した薬物の数を強度に乗算することによって正規化された。合計した正規化された強度を、強度の合計で除算し、2つの軽鎖及び2つの重鎖の合計した結果は、完全なADCに関する最終の平均DAR値を生じた。バイオコンジュゲートのチオスクシンイミド加水分解は、水をコンジュゲートに加えると、コンジュゲートの観察可能な分子量に対し18ダルトンの増加をもたらすことから、エレクトロスプレー質量分析によってモニターされ得る。

腫瘍細胞を、96ウェルプレートにおいて、10%FBS(仔ウシ胎児血清)を含有する増殖培地180μl中、2000~5000個/ウェルで播種し、5%CO2の湿潤インキュベーターにおいて37℃で培養した。18~24時間後、20μL中の抗体又はADCの滴定量を加え、細胞を、HCC827、NCI-H820、及びHEK-293(5日間)及びMIA PaCa2(4日間)を除き、6日間インキュベートした。細胞生存率は、CellTiter-Glo(登録商標)発光細胞生存率アッセイ(Promega)を使用して、製造元の使用説明書に従って決定された。非結合の無関係な陰性対照ADC(ADC-4、構造式VIII、AbはMSL109-C6v1である)もまた、細胞の殺滅が抗原依存的であることを確認するために全てのアッセイに含めた。ADCは全て、およそ等価な平均DARを有した。
受容体密度の決定
c-Met細胞表面密度(細胞あたりの抗原結合能)は、QIFIKIT(登録商標)(Dako/Agilent)を使用して培養細胞上の細胞表面抗原の間接的免疫蛍光染色によって決定された。簡単に説明すると、細胞を、FACS(蛍光活性化細胞選別)解析のために、上記のように培養フラスコから採取し、96ウェル丸底プレートに100μL/ウェルで加え、3μg/mL c-Met抗体m224G11と共に4℃でインキュベートした。同じアイソタイプ(mIgG1)の無関係なマウスモノクローナル抗体3μg/mLによって処置したウェルを対照として含めた。一次抗体と共に1時間インキュベーション後、細胞を、300×gで3分間遠心分離し、FACS緩衝液によって2回洗浄した。QIFIKIT(登録商標)ビーズの間接的免疫蛍光染色のために、バイアル1(セットアップビーズ)及びバイアル2(較正ビーズ)からの再懸濁したビーズ100μLを、個別のウェルに加え、300×gで3分間遠心分離し、FACS緩衝液によって1回洗浄した。全てのウェルを、FACS緩衝液中で1:250に希釈した抗マウスAlexa Fluor(登録商標)488コンジュゲート抗体(Invitrogen、カタログ#A-11029)100μLと共に4℃で1時間インキュベートした。細胞を、300×gで3分間遠心分離し、FACS緩衝液によって2回洗浄し、100μL/ウェルの1%ホルムアルデヒドのPBS(リン酸緩衝食塩水)中溶液によって固定した。データは、BD(商標)LSRIIフローサイトメーター上で獲得し、5つのビーズ集団に関して幾何平均値を記録し、これを使用してビーズあたりのロット特異的抗体分子に基づいて標準曲線を作成した。標準曲線を使用して、ABC(抗体結合能又は受容体の数)を、染色された細胞試料に割付した。
c-Met発現レベルとADC-1に対する感受性との可能性がある相関を決定するために、9個の細胞株のパネルを、インビトロでの増殖アッセイにおいて試験した。FACS解析は、これらの細胞株が、細胞表面のc-Met分子数を表すc-Met抗体結合能を介して定量された場合に、一定範囲のc-Met発現レベルを保有することを実証した(表3)。細胞増殖アッセイにおけるADC-1に対する感受性は、最大殺滅及びIC50として定量された(表3)。ADC-1は、MET増幅細胞株を含む、c-Metを過剰発現するがん細胞の増殖を阻害した(表3)。比較として、非コンジュゲートAbAは、MET増幅を有する細胞、すなわちHs 746T、SNU-5、及びEBC-1(図3A~C)の増殖を阻害したが、MET増幅を有しない細胞株、すなわちNCI-H441、NCI-H1573、HCC827、NCI-H820、及びCalu-3の増殖は阻害せず、ADC-1は同様に活性であった(図3D~H)。非コンジュゲートAbAもADC-1も、低c-Met発現細胞であるHEK-293及びMIA PaCa2において活性ではなく(表3、図3I及び図3J)、ADC-1による有意な殺滅のために、c-Met発現レベルの閾値が存在することを示唆した。
c-Met標的化TOP1i ADCは、インビボでがん異種移植片の成長を阻害する
細胞株、Hs 746T及びNCI-H441は、ATCC(American Type Culture Collection)から得られた。細胞は、供給元の推奨に従って3回以下の継代で単層培養として維持された。Matrigel(登録商標)と混合した培養培地(1:1、容量:容量)中の細胞2×106個の懸濁液を、雌性C.B-17 SCIDマウスの右脇腹に皮下注射し、胃癌細胞株Hs 746Tからの異種移植片を生成した。NSCLC細胞株であるNCI-H441からの異種移植片を生成するために、Matrigelと混合した培養培地(1:1、容量:容量)中の細胞5×106個の懸濁液を、雌性SCID/bgマウスに皮下注射した。脇腹の腫瘍のサイズがおよそ200mm3に達すると処置を開始した。
c-MET処置TOP1i ADCは、インビボで患者由来がん細胞の成長を阻害する
NSCLC患者由来異種移植片(PDX)モデル、LU450、LU120、LU572、LU123、及びLU413は、雌性免疫欠損NOD/SCIDマウス(Charles River Laboratories)に患者の生検組織断片を皮下に植え込むことによって内部で確立された。腫瘍が確立された後、PDX腫瘍細胞を、腫瘍を細胞懸濁液へと解離することによって増大させ、培養培地中の細胞5×104個を、Matrigel(登録商標)と混合し(1:1、容量:容量)、雌性NOD/SCIDマウスの乳腺脂肪体領域に皮下注射した。有効性試験に関して、腫瘍を有するマウスを処置群に無作為化し、各群は、等しい平均腫瘍体積(130~200mm3)を有した。c-Met-ADCは、6つの用量レベル(最も低い用量から最も高い用量まで、用量A~F)で単一用量として腹腔内注射によって投与された。c-Met標的化有効性を、ビヒクルの類似の投与と比較した。腫瘍体積を週に1~2回測定し、有効性は、腫瘍体積(mm3)を時間に対してプロットし、腫瘍進行までの時間(TTP)を計算することによって評価された。日数で表記されるTTPは、腫瘍が再成長して処置前(無作為化時)のサイズを倍加させるまでの1回用量処置後の時間である。持続性の応答(治癒)に関するTTPは、数の前に「>」の印を有する試験期間として示される。

- 前記抗体Abが、配列番号9として示されるアミノ酸配列を含む重鎖及び配列番号10として示されるアミノ酸配列を含む軽鎖を含む、請求項1に記載の抗c-Met抗体薬物複合体。
